Anti-RPL30 as a novel biomarker for enhanced diagnosis of autoantibody-negative primary biliary cholangitis

BACKGROUND The diagnosis of primary biliary cholangitis(PBC)remains challenging,particularly in cases where anti-mitochondrial antibody(AMA),anti-mitochondrial E2 subunit antibody(AMA-M2),anti-glycoprotein 210(anti-gp210),and anti-speckled protein 100(anti-Sp100)are all negative.In such instances,the condition is often confirmed through a liver needle biopsy.AIM To identify additional plasma biomarkers for non-invasive diagnostic methods of PBC.METHODS We utilized the Sengenics KREX^(TM)immunome protein array to identify potential biomarkers for the diagnosis of PBC.Subsequently,we validated the predictive capability of the RPL30 antibody through an ELISA and retrospectively analyzed its association with the clinical features of 17 autoantibody-negative PBC cases and 45 autoantibody-positive PBC cases.RESULTS In our study we observed that RPL30 demonstrated the highest fold-change difference in PBC,with a penetrance frequency of 40%and a penetrance fold change of 38.30147.The analysis of anti-RPL30 optical density values between patients with AMA/AMA-M2/anti-gp210/anti-Sp100-negative PBC(autoantibody-negative PBC)and healthy controls using a receiver operating characteristic curve yielded an area under the curve of 0.853.This analysis established an optimal cutoff value of 0.0708,achieving 100%specificity and 75%sensitivity.The combination of anti-RPL30 and other autoantibodies elevated the diagnosis rate of PBC from 61.29%to 79.00%(P=0.0489).Anti-RPL30 demonstrated a high positive rate in antibody-negative PBC cases,including AMA/AMAM2/anti-gp210/anti-Sp100-negative cases.Correlation analysis of anti-RPL30 optical density values with clinical data from patients with PBC revealed a positive association with both the international normalized ratio(P=0.008)and the Model for End-Stage Liver Disease score(P=0.003).CONCLUSION Our study highlighted the potential of anti-RPL30 as a promising biomarker for diagnosing PBC,particularly in autoantibody-negative cases. 展开更多

Thermal sensitization of acupoints in patients with knee osteoarthritis:A cross-sectional case-control study

Objective:Varied acupoint selections represent a potential cause of the uncertainty surrounding the efficacy of acupuncture for knee osteoarthritis(OA).Skin temperature,a guiding factor for acupoint selection,may help to address this issue.This study explored thermal sensitization of acupoints used for the treatment of knee OA.Methods:This cross-sectional case-control study enrolled cases aged 45–75 years with symptomatic knee OA and age-and gender-matched non-knee OA controls in a 1:1 ratio.All participants underwent infrared thermographic imaging.The primary outcome was the relative skin temperature of acupoint(STA),and the secondary outcome was the absolute STA of 11 acupoints.The Z test was used to compare the relative and absolute STAs between the groups.Principal component analysis was used to extract the common factors(CFs,acupoint cluster)in the STAs.A general linear model was used to identify factors affecting the STA in the knee OA cases.For the group comparisons of relative STA,P<0.0045(adjusted for 11 acupoints through Bonferroni correction)was considered to indicate statistical significance.For other analyses,P<0.05 was used as the threshold for statistical significance.Results:The analysis included 308 participants,consisting of 151 cases(mean age:[64.58±6.67]years;male:25.83%;mean body mass index:[25.70±3.16]kg/m~2)and 157 controls(mean age:[63.37±5.96]years;male:26.11%;mean body mass index:[24.47±2.84]kg/m~2).The relative STAs of ST34(P=0.0001),EX-LE2(P<0.0001),EX-LE5(P=0.0006),SP10(P<0.0001),BL40(P=0.0012)and GB39(P=0.0037)were higher in the knee OA group.No difference was found in the STAs of ST35,ST36,SP9,GB33 and GB34.Four CFs were identified for relative STA in both groups.The acupoints within each CF were consistent between the groups.The mean values of the relative STAs across each CF were higher in the knee OA group.In the knee OA cases,no factors were observed to affect the relative STA,while age and gender were found to affect the absolute STA.Conclusion:Among patients with knee OA,thermal sensitization occurs in the acupoints of the lower extremity,exhibiting localized and regional thermal consistencies.The thermally sensitized acupoints that we identified in this study,ST34,SP10,EX-LE2,EX-LE5,GB39 and BL40,may be good choices for the acupuncture treatment of knee OA. 展开更多

Superhydrophilic surfaces have been applied for supercapacitor;however,during energy storage reaction,how the wettability affects the process of electrochemical reaction specifically is still unclear.Herein,we demonstrate superhydrophilic surface for promotion of electrochemical reactions by liquid affinity and further explain the mechanism,where the transition of the wettability state caused by the change in surface free energy is the main reason for the obvious increase in specific capacitance.Through citric acid assistance strategy,an intrinsically hydrophobic Ni(OH)_(2)thick nanosheets(HNHTNs,16 nm)can be transitioned into superhydrophilic Ni(OH)_(2)ultrathin nanosheets(SNHUNs,6.8 nm),where the water contact angle was 0°and the surface free energy increased from 8.6to 65.8 mN·m^(-1),implying superhydrophilicity.Compared with HNHTNs,the specific capacitance of SNHUNs is doubled:from 1230 F·g^(-1)(HNHTNs)to 2350 F·g^(-1)(2A·g^(-1))and,even at 20 A·g^(-1),from 833 F·g^(-1)(HNHTNs)to 1670 F·g^(-1).The asymmetric capacitors assembled by SNHUNs and activated carbon show 52.44 Wh·kg^(-1)at 160W·kg^(-1)and excellent stability with~90%retention after5000 cycles(~80%retention after 9500 cycles).The promotion of electrochemical performances is ascribed to the change of surface wettability caused by surface free energy,which greatly increase affinity of electrode to the surrounding liquid environment to reduce the interface resistance and optimize the electron transport path. 展开更多

Recent evidence suggests that low-intensity extracorporeal shock wave therapy(Li-ESWT)is a promising treatment for chronic prostatitis/chronic pelvic pain syndrome(CP/CPPS);however,its safety in pelvic organs,particularly prostate tissues and cells,remains unclear.The current study evaluates the risks of prostate cell damage or oncogenesis following the administration of LiESWT for prostatitis.To this end,a robust in vitro model(Cell Counting Kit-8[CCK-8]assay,clone formation assay,cell scratch assay,lactate dehydrogenase[LDH]release assay,flow cytometry,and immunoblotting assay)was designed to examine the effects of Li-ESWT on cell proliferation,clonogenicity,migration,membrane integrity,and DNA damage.Exome sequencing of Li-ESWTtreated cells was performed to determine the risk of carcinogenesis.Furthermore,an in vivo rat model(n=20)was employed to assess the effects of Li-ESWT on cancer biomarkers(carcinoembryonic antigen[CEA],Ki67,proliferating cell nuclear antigen[PCNA],and gamma-H2A histone family member X,phosphorylation of the H2AX Ser-139[γ-H2AX])in prostate tissue.Based on our findings,Li-ESWT promotes cellular growth and motility without inducing significant cell membrane or DNA damage or alterations.Genetic analyses did not demonstrate an increase in mutations,and no damage to prostate tissue or upregulation of cancer biomarkers was detected in vivo.This comprehensive in vitro and in vivo assessment confirms the safety of Li-ESWT in managing prostate disorders. 展开更多

目的探讨早发冠状动脉粥样硬化性心脏病(CHD)患者焦虑状态发生的危险因素,了解焦虑状态对其短期预后的影响。方法连续筛选诊断为早发CHD患者120例,根据汉密尔顿焦虑量表(HAMA)评分分为焦虑组65例和非焦虑组55例,记录基本临床资料,并检... 目的探讨早发冠状动脉粥样硬化性心脏病(CHD)患者焦虑状态发生的危险因素,了解焦虑状态对其短期预后的影响。方法连续筛选诊断为早发CHD患者120例,根据汉密尔顿焦虑量表(HAMA)评分分为焦虑组65例和非焦虑组55例,记录基本临床资料,并检测超敏C-反应蛋白(hs-CRP)等生化指标,行心脏彩超、冠状动脉造影或支架植入治疗(PCI)。对入选者随访180 d,记录其临床事件。结果 (1)焦虑组女性、高血压、吸烟史、心肌梗死(陈旧和急性)、双支或双支以上血管病变、PCI治疗、高中以上文化程度其所占比例及hs-CRP水平高于非焦虑组(P<0.05);随着焦虑程度的增加,hs-CRP水平、高血压、双支及以上血管病变及PCI例数递增(P<0.05);(2)高血压[OR=2.352(95%CI:1.090,5.044),P=0.004]、心肌梗死[OR=2.195(95%CI:1.862,5.577),P=0.018]、PCI治疗[OR=3.680(95%CI:1.743,7.772),P=0.000]、双支或双支以上血管病变[OR=3.96(95%CI:1.895,8.304),P=0.011]是早发冠心病患者焦虑状态发生的危险因素;(3)焦虑组180 d无事件生存率低于非焦虑组(P=0.049)。结论高血压、心肌梗死、PCI治疗、双支或双支以上血管病变为早发CHD患者发生焦虑的危险因素,焦虑状态影响早发CHD患者短期预后。 展开更多

目的:研究第一眼白内障手术是否会引起对侧眼炎症反应,并尝试探究该炎症反应的发展规律。方法:本课题为配对样本实验,共纳入37例双眼年龄相关性白内障患者,实验样本均来源于患者术眼房水。其中行第一眼超声乳化白内障吸除联合人工晶状... 目的:研究第一眼白内障手术是否会引起对侧眼炎症反应,并尝试探究该炎症反应的发展规律。方法:本课题为配对样本实验,共纳入37例双眼年龄相关性白内障患者,实验样本均来源于患者术眼房水。其中行第一眼超声乳化白内障吸除联合人工晶状体植入术时收集的房水样本纳为A组(共37眼),行第二眼超声乳化白内障吸除联合人工晶状体植入术时收集的房水样本纳为B组(共37眼)。并根据患者双眼白内障手术间隔时间将样本分为五组。最终,待所有样本收集完成,通过Luminex生物检测技术同时检测其中TNF-α,IL-6,IL-8,CCL2/MCP-1,IL-6 Rα,IL-1 ra,MIP-1α,MIP-1d,CCL4,IL-2,CXCL9,TIMP-1,CCL11等13种细胞因子的浓度。结果:本研究所检测的13种细胞因子中,IL-6,MIP-1α,CCL2/MCP-1,TNF-α,MIP-1d,IL-2,TIMP-1等7种因子在双眼中的浓度有显著性差异(P<0.05),且均表现为第二眼高于第一眼,其中第二眼房水中IL-6和MIP-1α的浓度较第一眼呈显著性升高(P<0.001)。与第一眼相比,CCL4,CCL11,IL-6Rα,CXCL9,IL-1ra等炎症因子在第二眼中的浓度只有轻微升高,IL-8表现为轻微下降,差异均无统计学意义(P>0.05)。且研究结果提示,第一眼后3wk,IL-6,MCP-1/CCL2,IL-2,MIP-1d等4种炎症因子在双眼房水中的浓度差异最为显著。结论:本研究提示超声乳化白内障吸除联合人工晶状体植入术可能引起对侧眼的免疫反应,且第二眼的免疫反应高峰期大约出现在第一眼术后3wk左右。 展开更多

Thin film thermocouples(TFTCs) can provide fast and accurate surface temperature measurement with minimal impact on the physical characteristics of the supporting components. In this study, NiCr and NiSi films were prepared with radio frequency(RF) magnetron sputtering and the influences of vacuum annealing on the resistivity of the films were investigated. Afterward, NiCr-NiSi films were deposited on Ni-based superalloy substrates to form TFTCs. The overall dimension of the thermocouple is 64 mm in length, 8 mm in width and 30 lm in thickness. Compared with those of as-deposited sample, the thermoelectric property and stability of the TFTC are significantly improved by vacuum annealing of NiCr and NiSi films. The variation of the Seebeck coefficient of TFTC was discussed based on the size effect of NiCr and NiSi films. And a lower Seebeck coefficient of TFTC of 38.4 μV·℃^-1 is obtained. 展开更多

Major royal jelly protein 1(MRJP1), designated apalbumin 1, has been regarded as a freshness marker of royal jelly(RJ). A MRJP1-specific peptide(IKEALPHVPIFD) identified by bioinformatics analysis of homologous members of the major royal protein family was synthesized and used to raise polyclonal anti-MRJP1 antibody(antiSP-MRJP1 antibody). Western blot analysis showed that anti-SP-MRJP1 antibody only reacted with MRJP1 in RJ. In contrast, the previously reported antibody against recombinant MRJP1(anti-R-MRJP1 antibody) reacted with other members of MRJP family in RJ. Enzyme-linked immunosorbent assay(ELISA) using anti-SP-MRJP1 antibody demonstrated that MRJP1 content in RJ stored at 40 °C significantly degraded by 37.3%, 55.9%, 58.0%, 60.6%, 65.7%, 72.7%, and 73.1% at 7, 14, 21, 28, 35, 42, and 49 d, respectively, when compared with MRJP1 content in fresh RJ(0 d). Optical density analysis of MRJP bands from sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) profiles demonstrated that the degradation of MRJP1, MRJP2, MRJP3, and MRJP5 in RJ was strongly and positively correlated with the period of storage(P〈0.0001). Our results indicated anti-SP-MRJP1 antibody was highly specific for MRJP1, and ELISA using the antibody is a sensitive and easy-to-use method to determine the freshness and authenticity of RJ. 展开更多

The complete mitochondrial genome of Acanthacorydalis orientalis (McLachlan) was determined and analyzed (GenBank accession number: KF840564). This paper represents the first mitochondrial (mt) genome of the dobsonfly genus Acanthacorydalis. The mt genome is a typical circular DNA of 15 753 bp composed of 37 genes with an A+T content of 76.7%. It has an ancestral gene arrangement of the insect mt genomes. Eleven of the 13 PCGs start with codon ATT and ATG,while several exceptions such as ATA and TTG respectively for atp 8 and nad 1 are also present. Five protein-coding genes end with a single T, while others have a termination codon of TAA or TAG. Most tRNAs are folded into the typical clover-leaf structure except for the trnS 1 whose dihydrouridine arm was a simple loop. The secondary structure of rrnl consists of five structural domains and 50 helices, while the rrns includes three domains and 34 helices. The control region has a stretches of Ts with a length of 22 bp but lacks obvious tandem repeat region. Both Bayesian inference and maximum likelihood (ML) analyses, based on all 13 protein-coding genes and two rRNA genes of the mt genomes, confirm the monophyly of Corydalinae and suggest that Acanthacorydalis, together with Corydalus, which is an endemic dobsonfly genus from the New World, belong to a monophyletic lineage. 展开更多

By using first-principles calculation,we study the properties of h-BN/BC_(3)heterostructure and the effects of external electric fields and strains on its electronic and optical properties.It is found that the semiconducting h-BN/BC_(3)has good dynamical stability and ultrahigh stiffness,enhanced electron mobility,and well-preserved electronic band structure as the BC_(3)monolayer.Meanwhile,its electronic band structure is slightly modified by an external electric field.In contrast,applying an external strain can mildly modulate the electronic band structure of h-BN/BC_(3)and the optical property exhibits an apparent redshift under a compressive strain relative to the pristine one.These findings show that the h-BN/BC_(3)hybrid can be designed as optoelectronic device with moderately strain-tunable electronic and optical properties. 展开更多

Objective:The mechanism of Yupingfeng Powder in the treatment of non-small cell lung cancer was analyzed by network pharmacology and molecular docking method.Methods:Search the related literature and TCMSP database,query the chemical composition and action target of Yupingfeng powder,query the disease database of Genecards,OMIM,DisGeNET,Drugbank,etc.,use"non-small cell lung cancer"as the keyword to search the disease-related target,select the intersection target with Yupingfeng powder as the research target,and use Cytoscape 3.7.2 software to construct the active component-target network map.The protein-protein interaction network map was constructed by using STRING database,and the treatment core targets were further screened by topological parameters,and GO analysis and KEGG signal pathway enrichment analysis were carried out.Finally,molecular docking was verified.Results:37 active components of Yupingfeng Powder were screened,including 146 common targets and 44 core targets.GO analysis and KEGG analysis show that the core target participates in many biological processes such as angiogenesis and cell proliferation,and acts on a variety of signal pathways such as AGE-RAGE,IL-17,TNF,MAPK and so on.Molecular docking shows that the core compound has good docking activity with the target.Conclusion:Network pharmacological analysis shows that Yupingfeng powder in the treatment of non-small cell lung cancer involves multi-target and multi-pathway mechanism,and the drug and target have good docking activity,which can provide theoretical basis for follow-up experimental research. 展开更多
