Mold spoilage and nutrition enrichment are major concerns of Chinese steamed bread,and using functional lactic acid bacteria as starter to play the role of in-situ preservation and nutritional fortification was a prom...Mold spoilage and nutrition enrichment are major concerns of Chinese steamed bread,and using functional lactic acid bacteria as starter to play the role of in-situ preservation and nutritional fortification was a promising alternative.Hence,Lactiplantibacillus plantarum P10 capable of inhibiting spoilage microorganisms and producing γ-aminobutyric acid(GABA)(380 mg/L)was screened from Chinese pickle.Metabolomics analysis showed that P10 produced key antimicrobial metabolites mainly including ten organic acids,aldehyde,alkaloid.Besides,P10 might exhibit health beneficial effects such as degrading cholesterol,lowering blood glucose and reducing uric acid due to the production of deoxycholic acid and Ile-Pro-Ile,and the degradation of purine.Finally,the promising application potential of P10 was evidenced in steamed bread with extended shelf life,excellent anti-aging effect,and 24.5-fold enrichment of GABA.Conclusively,this investigation could provide scientific basis for the application of P10 as a functional starter in steamed bread.展开更多
Listeria monocytogenes is a worrisome food-borne pathogen threatening global food safety.Our previous study proved that lipopeptide brevilaterin B showed efficient antibacterial activity against L.monocytogenes by int...Listeria monocytogenes is a worrisome food-borne pathogen threatening global food safety.Our previous study proved that lipopeptide brevilaterin B showed efficient antibacterial activity against L.monocytogenes by interacting with the cell membrane.This research further explored the antibacterial mechanism of brevilaterin B against L.monocytogenes at the sub-minimum inhibition concentration via transcriptomic analysis.Brevilaterin B induced growth inhibition rather than direct membrane lysis in L.monocytogenes at the minimum inhibitory concentration.Transcriptomic analysis showed 1779 difference expressed genes,including 895 up-regulated and 884 down-regulated genes.Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis indicated that brevilaterin B influenced multiple pathways of L.monocytogenes,including peptidoglycan biosynthesis,membrane transport(ATP-binding cassette transports,ion transport),cellular metabolism(amino acid and lipid metabolism),ATP synthesis,and activation of the stress response(quorum sensing and bacterial chemotaxis).In conclusion,brevilaterin B affects gene expression related to biosynthesis,transport and stress response pathways on the membrane of L.monocytogenes.The present work provides the first transcriptomic assessment of the antibacterial mechanism of lipopeptide brevilaterin B at the gene level.展开更多
Plant protein beverage adulteration occurs frequently,which may cause health problems for consumers due to the hidden allergens.Hence,a novel method was developed for authentication by ultra-performance liquid chromat...Plant protein beverage adulteration occurs frequently,which may cause health problems for consumers due to the hidden allergens.Hence,a novel method was developed for authentication by ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).Almond,peanut,walnut and soybean were hydrolyzed,followed by separation by NanoLC-Triple TOF MS.The obtained fingerprints were identified by ProteinPilotTM combined with Uniprot,and 16 signature peptides were selected.Afterwards,plant protein beverages treated by trypsin hydrolysis were analyzed with UPLC-MS/MS.This method showed a good linear relationship with R2>0.99403.The limit of quantification(LOQ)were 0.015,0.01,0.5 and 0.05 g/L for almond,peanut,walnut and soybean,respectively.Mean recoveries ranged from 84.77%to 110.44%with RSDs<15%.The developed method was successfully applied to the adulteration detection of 31 plant protein beverages to reveal adulteration and false labeling.Conclusively,this method could provide technical support for authentication of plant protein beverages to protect the rights and health of consumers.展开更多

Surfactin holds significant potential as a natural emulsifier and antimicrobial agent,offering a promising alternative to synthetic ingredients in the food industry.However,its large-scale application is hindered by l...Surfactin holds significant potential as a natural emulsifier and antimicrobial agent,offering a promising alternative to synthetic ingredients in the food industry.However,its large-scale application is hindered by low production yields.Recent studies have explored the use of magnetic fields(MF)in bioprocesses due to their potential to enhance fermentation efficiency.This study investigated the effects of a static MF on Bacillus subtilis to improve surfactin production.Results showed that a 24 mT static MF increased the maximum surfactin productivity by 118.6%(p<0.05).Mechanistic studies revealed that the MF induced pH fluctuations in the fermentation broth and enhanced sugar consumption by B.subtilis.Untargeted metabolomic analysis demonstrated that MF intervention significantly up-regulated the accumulation of key intermediates and energy metabolites during the lag and logarithmic growth phases,facilitating the synthesis of surfactin precursors such as fatty acids,amino acids,and energy-related substances.Additionally,the activities of four critical enzymespyruvate kinase(PK),acetyl-CoA carboxylase(ACC),glutamic-oxalacetic transaminase(GOT),and glutamate synthetase(GOGAT)-involved in surfactin biosynthesis were notably enhanced.These findings provide a foundation for the potential application of MF in industrial-scale surfactin production for food processing.展开更多
Antimicrobial peptides(AMPs),as new antimicrobial products,have wide application prospects in food,medicine,and other fields.A strain of Paenibacillus ehimensis HD was previously identified by our research group,and t...Antimicrobial peptides(AMPs),as new antimicrobial products,have wide application prospects in food,medicine,and other fields.A strain of Paenibacillus ehimensis HD was previously identified by our research group,and the AMPs synthesized by this strain were found to be highly stable and had a broad antimicrobial spectrum.To enhance the AMPs yield,the ultraviolet-atmospheric and room temperature plasma mutagenesis(UV-ARTP)was adopted in P.ehimensis HD in this study.A stable mutant of P.ehimensis HD 8-3,with enhanced AMPs production,was obtained.Subsequently,using the resting cell culture system and untargeted metabolomics analysis,we found that the addition of L-lysine(Lys)or L-asparagine(Asn)was beneficial to the growth of P.ehimensis HD 8-3,while Asn also enhanced the AMPs production.The metabolites associated with the carbohydrate,glycolytic pathway,TCA cycle,pentose phosphate pathway(PPP),and amino acid metabolism were upregulated under the addition of two amino acids during the rapid growth phase of P.ehimensis HD 8-3,providing precursors and energy for the strain growth or AMPs production.These results indicate that the appropriate addition of exogenous Lys and Asn is an effective strategy to increase AMPs yield in P.ehimensis HD 8-3.展开更多
