Cold stress in low-temperature environments can trigger changes in gene expression,but epigenomics regulation of temperature stability in vital tissues,including the fat and diencephalon,is still unclear.Here,we explo...Cold stress in low-temperature environments can trigger changes in gene expression,but epigenomics regulation of temperature stability in vital tissues,including the fat and diencephalon,is still unclear.Here,we explore the cold-induced changes in epigenomic features in the diencephalon and fat tissues of two cold-resistant Chinese pig breeds,Min and Enshi black(ES)pigs,utilizing H3K27ac CUT&Tag,RNA-seq,and selective signature analysis.Our results show significant alterations in H3K27ac modifications in the diencephalon of Min pigs and the fat of ES pigs after cold exposure.Dramatic changes in H3K27ac modifications in the diencephalon of Min pig are primarily associated with genes involved in energy metabolism and hormone regulation,whereas those in the fat of ES pig are primarily associated with immunity-related genes.Moreover,transcription factors PRDM1 and HSF1,which show evidence of selection,are enriched in genomic regions presenting cold-responsive alterations in H3K27ac modification in the Min pig diencephalon and ES pig fat,respectively.Our results indicate the diversity of epigenomic response mechanisms to cold exposure between Min and ES pigs,providing unique epigenetic resources for studies of low-temperature adaptation in large mammals.展开更多

Moreover, if a link between the adjacent two neurons is cut, the ring neural network turns to a linear one, and the stability results are also established. In microbial ecology,there is limited understanding of the mechanisms governing patterns in community structure across space and time.Here,we studied the changes of bacterial co-occurrence network structure over four ...In microbial ecology,there is limited understanding of the mechanisms governing patterns in community structure across space and time.Here,we studied the changes of bacterial co-occurrence network structure over four primary successional soils after glacier retreat,including a sand dune system and three glacier foreland series,varying in timescale from centuries to millennia.We found that in all series,network structure was most complex in the earliest stages of succession,and became simpler over time.Richness and abundance of keystone species and network stability also declined over time.It appears that with less productive,nutrient poor and physiologically extreme conditions of early succession,closer interactions among bacterial species are ecologically selected for.These may take the form of consortia(with positive interactions)or stronger niche exclusion(with negative interactions).Additionally,we quantified the relative roles of different structuring processes on bacterial community using a bin-based null model analysis(iCAMP).With each successional series,community composition was initially governed by stochasticity,but as succession proceeded there was a progressive increase in deterministic selection over time,correlated with decreasing pH.Overall,our results show a consistency among the four series in long-term processes of community succession,with more integrated networks and greater stochasticity in early stages.展开更多
Land application of anaerobic digestion(AD)effluent as a fertilizer is desirable for nutrient recycling,but often supplies excess phosphorus(P),which contributes to surface water eutrophication.Reducing the P content ...Land application of anaerobic digestion(AD)effluent as a fertilizer is desirable for nutrient recycling,but often supplies excess phosphorus(P),which contributes to surface water eutrophication.Reducing the P content in AD effluent filtrate using calcium(Ca)treatment prior to land application is a potential strategy for improving effluent disposal and meeting the discharge standard.This study took flue gas desulphurization(FGD)gypsum,a by-product of coal-fired power plants,as a low-cost Ca source,and combined with traditional phosphorus removal agents to achieve high phosphorus removal efficiency with less chemical cost.As the results showed,FGD gypsum dosages of 20 mmol/L Ca(3.44 g/L)and 40 mmol/L Ca(6.89 g/L)removed up to 97.1%of soluble P(initially 102.8 mg/L)within 60-90 minutes.Combining FGD gypsum treatment with traditional chemical treatments using calcium hydroxide[Ca(OH)2]or ferric chloride(FeCl3)could achieve>99%P removal with reduced chemical costs.This study demonstrated that FGD gypsum is an efficient calcium-based precipitant for phosphorus removal,offering a cost-effective and sustainable approach to enhance wastewater treatment practices and meet discharge standards in wastewater management.展开更多

Here, we found benzydamine, a locally acti...Esophageal squamous cell carcinoma (ESCC) is one of the leading causes of cancer death worldwide. It is urgent to develop new drugs to improve the prognosis of ESCC patients. Here, we found benzydamine, a locally acting non-steroidal anti-inflammatory drug, had potent cytotoxic effect on ESCC cells. Benzydamine could suppress ESCC proliferation in vivo and in vitro. In terms of mechanism, CDK2 was identified as a target of benzydamine by molecular docking, pull-down assay and in vitro kinase assay. Specifically, benzydamine inhibited the growth of ESCC cells by inhibiting CDK2 activity and affecting downstream phosphorylation of MCM2, c-Myc and Rb, resulting in cell cycle arrest. Our study illustrates that benzydamine inhibits the growth of ESCC cells by downregulating the CDK2 pathway.展开更多
As an important protein acylation modification,lysine succinylation(Ksucc)is involved in diverse biological processes,and participates in human tumorigenesis.Here,we collected 26,243 non-redundant known Ksucc sites fr...As an important protein acylation modification,lysine succinylation(Ksucc)is involved in diverse biological processes,and participates in human tumorigenesis.Here,we collected 26,243 non-redundant known Ksucc sites from 13 species as the benchmark data set,combined 10 types of informative features,and implemented a hybrid-learning architecture by integrating deep-learning and conventional machine-learning algorithms into a single framework.We constructed a new tool named HybridSucc,which achieved area under curve(AUC)values of 0.885 and 0.952 for general and human-specific prediction of Ksucc sites,respectively.In comparison,the accuracy of HybridSucc was 17.84%-50.62%better than that of other existing tools.Using HybridSucc,we conducted a proteome-wide prediction and prioritized 370 cancer mutations that change Ksucc states of 218 important proteins,including PKM2,SHMT2,and IDH2.We not only developed a high-profile tool for predicting Ksucc sites,but also generated useful candidates for further experimental consideration.The online service of HybridSucc can be freely accessed for academic research at http://hybridsucc.biocuckoo.org/.展开更多
