Jujube(Ziziphus jujuba Mill.)is an important perennial fruit tree with a range of interesting horticultural traits.It was domesticated from wild jujube(Ziziphus acidojujuba),but the genomic variation dynamics and genetic changes underlying its horticultural traits during domestication are poorly understood.Here,we report a comprehensive genome variation map based on the resequencing of 350 accessions,including wild,semi-wild and cultivated jujube plants,at a>15×depth.Using the combination of a genome-wide association study(GWAS)and selective sweep analysis,we identified several candidate genes potentially involved in regulating seven domestication traits in jujube.For fruit shape and kernel shape,we integrated the GWAS approach with transcriptome profiling data,expression analysis and the transgenic validation of a candidate gene to identify a causal gene,ZjFS3,which encodes an ethyleneresponsive transcription factor.Similarly,we identified a candidate gene for bearing-shoot length and the number of leaves per bearing shoot and two candidate genes for the seed-setting rate using GWAS.In the selective sweep analysis,we also discovered several putative genes for the presence of prickles on bearing shoots and the postharvest shelf life of fleshy fruits.This study outlines the genetic basis of jujube domestication and evolution and provides a rich genomic resource for mining other horticulturally important genes in jujube. 展开更多

Seeds of soybeans and mung beans were soaked into five different concentrations (0, 0.05, 0.10, 0.20, 0.30 mg/L) of natural brassinolide ( NBR ) solution. According to the results, natural brassinolide treatment could improve seed germination rate and hypocotyl-radicle ratio of soybeans and mung beans and promote the growth of sprouts. To be specific, seed germination rate of soybeans reached the highest in 0.05 mg/L natural brassinolide treatment, which was im- proved by 25.0 percentage points compared with the control group, and the sprout length was improved by 4.33 em; 0.30 mg/L natural brassinolide exhibited the most significant promotion effect on seed germination and sprout growth of mung beans, seed germination rate was improved by 18.3 percentage points and sprout length was improved by 7.29 cm; hypocotyl-radiele ratio of soybean and mung bean sprouts reached the highest (2.96, 1.43 ) in 0.05 mg/L natural brassinolide treatment. 展开更多

Tree peony cultivars with yellow flowers are rare and valuable,but the molecular mechanism of pigment accumulation is not clear.In this study,the petal transcriptome of three developmental stages were sequenced to determine the differentially expressed genes(DEGs)regulating yellow flowers color.The results showed that 10,842 and 12,022 DEGs were screened in stage 1 vs.stage 2 and in stage 2 vs.stage 3,respectively.Through analysis of flavonoid structural genes(FSGs),we found that the transcription level of DFR was very low in the three developmental stages.In a small group of cultivars,the DFR transcription level of red flowers was 862 times higher than that of yellow flowers.These data suggested that the flavonoid pathway is interrupted in the later stage due to the low transcriptional level of DFR,which limits the biosynthesis of anthocyanins in yellow flowers.The transcription levels of F3’H and FLS were upregulated from stage 1 to stage 2,while those of CHI and FLS were downregulated from stage 2 to stage 3.In addition,67 MYBs and 44 bHLHs showed similar transcription profiles with different members of FSGs.The results deepen our understanding of the molecular mechanism of yellow pigment accumulation in tree peony. 展开更多
