Infections are frequent complications in patients with hematological disorders,and pathogen diagnosis remains challenging.Metagenomic next-generation sequencing(mNGS)is an unbiased high-throughput technology that has been widely applied in the diagnosis of infectious diseases.However,to date,there are no established international guidelines or expert consensuses regarding the use of mNGS to diagnose infections in patients with hematologic disorders.The Anti-Infection Study Group of the Chinese Society of Hematology invited experts in the fields of hematology,microbiology,and mNGS technology to draft an expert consensus focused on clinical indications,sample collection,quality control,and interpretation of results.This consensus will likely contribute to clarifying the medical indications for mNGS testing,optimizing the interpretation of reports,and becoming an inspiration for global practice. 展开更多

Solid-state fermentation with lactic acid bacteria of citric acid degrading for hawthorn fruit improvement:Sensory,flavor,antioxidant properties

Hawthorn fruit with high citric acid content leads to sour taste,and compacted flesht is not conducive to process into juice.To solve these problems,Leuconostoc citreum,Lactobacillus casei,and Lactobacillus brevis were screened out for compounding,by which the rate of degradation of citric acid reached 81.147%after solid-state fermentation(SSF).Results show that pectin solubility is improved,the cell-wall structure is dissolved and loosened,the fruit becomes softer,and the juice yield is increased to 60.63%,while the surface color is slightly affected after fermentation.Meanwhile pH value of fermented hawthorn fruit grows to 5.05,the content of citric acid reduces to 1.54 mg/kg,and the solidity-acid ratio is increased to 11.55,so sourness and astringency are alleviated and the taste is apparently improved.Additionally,total phenolics and total flavones are released and the antioxidant activity is enhanced by a factor of 1.2-1.6.Besides,the contents of undesirable flavor substances such as acids and sulpho-compounds are reduced in fermented hawthorn fruit,while the content and diversity of flavor substances including esters,phenolics,and aldehydes are increased.This is positive for improving the flavor of hawthorn fruit.Based on the KEGG(Kyoto Encyclopedia of Genes and Genomes)results,it is proven that seven pathways are mainly involved in the fermentative metabolism,in which nine metabolites are produced.Therein,the phenylalanine metabolism pathway involving benzeneacetaldehyde shows the highest enrichment.Therefor,SSF of compound LAB is feasible in improving the processing quality and the suitability of hawthorn fruit. 展开更多

The browning of Dongbei Suancai(DS)is common during storage.To study the regulation mechanism of gamma(γ)irradiation on non-enzymatic browning in DS during storage,different doses of 0,0.5,1.5,and 2.5 kGy were used to irradiate DS,and the changes in contents of vitamin C(VC),polyphenol,reducing sugar,amino nitrogen,and 5-hydroxymethylfurfural(5-HMF)were investigated.The main factors and interaction of different irradiation doses on non-enzymatic browning were studied by path analysis.The results showed that 1.5 kGy irradiation inhibited the browning by reducing the oxidative decomposition of VC and decreasing its leading role in browning.And 2.5 kGy irradiation aggravated browning due to inducing the oxidative polymerisation of polyphenols and enhancing the decisive effect of polyphenols on browning.Therefore,1.5 kGy irradiation was an effective measure with which to inhibit non-enzymatic browning in DS during storage.The present work provides a theoretical basis for effective regulation of browning in DS during storage. 展开更多

Postharvest blueberries tend to soften and rot,resulting in serious fruit waste.Gamma(γ)irradiation can delay the softening of blueberries and improve fruit quality.Calcium is a structural component of plant cell wall and membrane,and plays a role as a second messenger in the physiological process of fruit.To explore the mechanism of inhibiting the softening of cold storage blueberry fruit byγirradiation,‘Bluecrop’blueberry fruit were treated byγirradiation at a dose of 2.5 kGy,and then combined with calcium ion(Ca^(2+))inhibitors and intracellular calmodulin antagonists.The Ca^(2+)fluorescence signals,fruit firmness,protopectin and different soluble pectin contents,cellulose and hemicellulose contents,and hydrolase activities were detected.Results showed that irradiation induced Ca^(2+)release from intracellular calcium store to generate fluorescence signals.Irradiation mediated the Ca^(2+)signals to exert a positive effect on fruit firmness of blueberries during cold storage,for the degradation of protopectin and chelator-soluble pectin to water-soluble pectin was reduced by inhibiting activities of polygalacturonase and pectin methylesterase.Therefore,γirradiation-induced Ca^(2+)signals played an important role in regulation of pectin-involved fruit softening in blueberries during cold storage.This study provides a theoretical basis for the mechanism of Ca^(2+)signaling in blueberry storage. 展开更多
