Human cytomegalovirus(HCMV)is a double-strand DNA virus widely infected in human.Circular RNAs(circ RNAs)are non-coding RNAs with most functions of which keep unknown,and the effects of HCMV productive infection on host circ RNA transcriptions remain unclear.In this study,we profiled 283 host circ RNAs that significantly altered by HCMV productive infection in human embryonic lung fibroblasts(HELF)by RNA deep sequencing and bioinformatics analysis.Among these,circ SP100,circ MAP3 K1,circ PLEKHM1,and circ TRIO were validated for their transcriptions and sequences.Furthermore,characteristics of circ SP100 were investigated by RT-q PCR and northern blot.It was implied that circ SP100 was produced from the sense strand of the SP100 gene containing six exons.Kinetics of circ SP100 and SP100 m RNA were significantly different after infection:circ SP100 levels increased gradually along with infection,whereas SP100 m RNA levels increased in the beginning and dropped at 24 h post-infection(hpi).Meanwhile,a total number of 257 proteins,including 10 HCMV encoding proteins,were identified potentially binding to cytoplasmic circ SP100 by RNA antisense purification(RAP)and mass spectrometry.Enrichment analysis showed these proteins were mainly involved in the spliceosome,protein processing,ribosome,and phagosome pathways,suggesting multiple functions of circ SP100 during HCMV infection. 展开更多

Viruses commonly create favorable cellular conditions for their survival through multiple mechanisms. Micro RNAs(mi RNAs), which function as post-transcriptional regulators, are utilized by human cytomegalovirus(HCMV) in its infection and pathogenesis. In the present study, the DNA replication inhibitor Geminin(GMNN) was identified to be a direct target of hcmv-mi R-US5-1. Overexpression of hcmv-mi R-US5-1 could block the accumulation of GMNN during HCMV infection, and the decrease of GMNN expression caused by hcmv-mi R-US5-1 or GMNN specific si RNA reduced HCMV DNA copies in U373 cells. Meanwhile, ectopic expression of hcmv-mi R-US5-1 and consequent lower expression of GMNN influenced host cell cycle and proliferation. These results imply that hcmv-mi R-US5-1 may affect viral replication and host cellular environment by regulating expression kinetics of GMNN during HCMV infection. 展开更多

Red sandalwood(Pterocarpus santalinus)is the most precious species of Pterocarpus,and the long maturity period of its heartwood is an important factor limiting its development.The 1-year-old plants of P.macarocarpus were chosen as the rootstocks to graft red sandalwood with single-bud scions of 1-year-old shoots in spring.The 5-month survival rate reached over 80%,and the grafted plants grew well.Moreover,the grafting significantly promoted the formation and development of the heartwood of red sandlawood.At 18^(th)month after grafting,compared with the seedling plants,the diameter of heartwood was increased by 82.8%and 29.6%,respectively,and the ratio of heartwood increased by 64.8%and 29.6%,respectively.The propagation technology had outstanding practical value and scientific significance,which has not been reported at home and abroad. 展开更多

Human cytomegalovirus(HCMV)is a ubiquitous pathogen belongs to betaherpesvirus subfamily.RNA2.7 is a highly conserved long non-coding RNA accounting for more than 20%of total viral transcripts.In our study,functions of HCMV RNA2.7 were investigated by comparison of host cellular transcriptomes between cells infected with HCMV clinical strain and RNA2.7 deleted mutant.It was demonstrated that RNA polymeraseⅡ(PolⅡ)-dependent host gene transcriptions were significantly activated when RNA2.7 was removed during infection.A145 nt-in-length motif within RNA2.7 was identified to inhibit the phosphorylation of PolⅡSerine-2(PolⅡS2)by reducing the interaction between PolⅡand phosphorylated cyclin-dependent kinase 9(pCDK9).Due to the loss of PolⅡS2 phosphorylation,cellular DNA pre-replication complex(pre-RC)factors,including Cdt1 and Cdc6,were significantly decreased,which prevented more cells from entering into S phase and facilitated viral DNA replication.Our results provide new insights of HCMV RNA2.7 functions in regulation of host cellular transcription. 展开更多

三类/四类暴露复发难治性多发性骨髓瘤患者的临床特征及生存分析

目的:探讨三类/四类暴露复发难治性多发性骨髓瘤(relapsed or refractory multiple myeloma,RRMM)患者的临床特征与生存预后。方法:回顾性分析2017年5月至2024年5月山西省8所医院(山西白求恩医院、山西医科大学第二医院、运城市中心医... 目的:探讨三类/四类暴露复发难治性多发性骨髓瘤(relapsed or refractory multiple myeloma,RRMM)患者的临床特征与生存预后。方法:回顾性分析2017年5月至2024年5月山西省8所医院(山西白求恩医院、山西医科大学第二医院、运城市中心医院、国药同煤总医院、晋城市人民医院、临汾市人民医院、山西省人民医院和运城市人民医院)三类/四类暴露RRMM患者的临床数据,用Kaplan-Meier法分析总生存期(overall survival,OS)和无进展生存期(progression-free survival,PFS),Cox模型及Log-rank检验分析影响因素。结果:112例三类暴露患者中16例为四类暴露。三类暴露患者高风险细胞遗传学异常及髓外病变的检出率分别为57.1%和36.6%,四类暴露患者分别为87.5%和62.5%;三类暴露患者的中位PFS和OS分别为5.6个月和12.2个月,四类暴露患者分别为9.4个月和16.9个月。Cox模型分析显示,髓外病变、多线治疗(≥3线)是影响三类暴露RRMM患者生存的独立危险因素(P<0.05);既往自体干细胞移植、后续常规药物治疗及B细胞成熟抗原(B-cell maturation antigen,BCMA)的嵌合抗原受体T细胞(chimeric antigen receptor T,CAR-T)治疗为保护因素(P<0.05)。三类耐药后,Logrank检验证实BCMA CAR-T治疗较常规药物治疗显著延长患者中位PFS(9.4个月vs.5.2个月)(P=0.0269),但OS差异无统计学意义(16.9个月vs.7.9个月)(P=0.2634)。结论:三类/四类暴露RRMM患者预后差,BCMA CAR-T细胞治疗可改善三类耐药RRMM患者生存期。 展开更多

脂肪族聚酯是重要的可降解材料,可基于生物质转化获得环酯单体进而聚合制备成的大宗树脂;环酯单体到聚酯转化的技术关键是催化剂,新型均相催化剂不仅可以高效地催化环酯开环聚合而且能够实现所得聚酯分子量及结构控制,提升树脂材料的性... 脂肪族聚酯是重要的可降解材料,可基于生物质转化获得环酯单体进而聚合制备成的大宗树脂;环酯单体到聚酯转化的技术关键是催化剂,新型均相催化剂不仅可以高效地催化环酯开环聚合而且能够实现所得聚酯分子量及结构控制,提升树脂材料的性能。目前科研人员普遍关注席夫碱配位金属化合物,而含有磷氮键配体的多齿配位金属化合物在环酯开环聚合中展示了特异性和巨大价值。虽然氮与磷元素具有相似的电子结构,然而其电负性差异较大,所以含有磷氮键的多齿配体能够帮助调控金属的配位环境并提高环酯开环聚合的催化性能。磷氮键既可以是单键也可以是双键,作为配体与金属配位时总是氮原子优先配位,磷则帮助调控邻近配位氮原子上空间位阻与电子效应,其结果不仅提高了催化活性而且能够对所得聚酯结构实现剪裁。本文集中讨论近年来含磷氮键的金属配合物催化环酯的开环聚合,基于配体结构系统地总结了配体结构与催化性能以及所得聚酯产物微结构的关联,以求推进新型高效配合物催化剂的设计合成,引导产业界选出具有实用价值的催化剂,推动相关科学发展与实现产业化。 展开更多

This consensus on multiple myeloma-related bone diseases(MBDs)underscores the importance of a multidisciplinary approach that encompasses hematology,radiology,orthopedics,and additional specialties to tackle its intricate challenges.MBD,a prevalent and debilitating complication of multiple myeloma,leads to bone pain,fractures,and skeletal-related events(SREs),which profoundly impact patients’quality of life.The guidelines offer a thorough framework for diagnosis,treatment,and continual assessment,emphasizing early detection and consistent monitoring using imaging techniques such as positron emission tomography-computed tomography(PET-CT)and magnetic resonance imaging(MRI).Treatment strategies prioritize the careful application of antimyeloma agents,bisphosphonates,and denosumab to minimize bone loss and decrease SRE risk,complemented by surgical and radiotherapy interventions for structural or pain-related issues.Supportive care measures,including pain management,rehabilitation,nutritional support,and dental evaluations,play a crucial role in enhancing patient outcomes and preserving quality of life.This consensus advocates a standardized,evidencebased approach to managing MBD,ensuring comprehensive and coordinated care for patients. 展开更多

治疗多发性骨髓瘤(multiple myeloma,MM)的药物主要以蛋白酶体抑制剂(proteasome inhibitors,PIs)、免疫调节剂(immunomodulatory drugs,IMiDs)为主。近年来包括CD38单抗、CAR-T细胞治疗以及其他一些新药的使用,显著提高了MM患者的治疗... 治疗多发性骨髓瘤(multiple myeloma,MM)的药物主要以蛋白酶体抑制剂(proteasome inhibitors,PIs)、免疫调节剂(immunomodulatory drugs,IMiDs)为主。近年来包括CD38单抗、CAR-T细胞治疗以及其他一些新药的使用,显著提高了MM患者的治疗疗效,改善了患者的生存期。其中PIs仍然是MM治疗的基本药物。卡非佐米(carfilzomib,CFZ)作为新一代的PIs,相较于硼替佐米(bortezomib,BTZ)以及伊沙佐米(ixazomib,IXZ),由于其治疗机制的不同,更多地使用于复发难治性MM(relapsed or refractory multiple myeloma,RRMM)以及耐药MM的患者,且其疗效已被大量数据证实。但随着卡非佐米的临床应用,部分患者出现耐药现象。本文将根据近些年对于卡非佐米耐药的研究,从自噬水平、药物代谢、蛋白酶体结构及数量、基因的修饰、细胞代谢5个方面分析卡非佐米的耐药机制,并据此给出可能的新的治疗方案。根据上述新的治疗方案有望增强卡非佐米的疗效,减少耐药现象的出现,延长患者的生存时间。 展开更多

Effect of Soil p H on the Growth,Photosynthesis and Mineral Element Content in Own-Rooted Saplings and Grafted Saplings of Blueberry 被引量:4

To explore the effect and probable mechanism of the adaptation of grafted blueberry( Vaccinium spp.) saplings to the soil environment with higher p H value. We took three types of saplings,( i) own-rooted Wufanshu( V. bracteatum),an economical shrub belonging to the same genus and family with blueberry,also known as oriental blueberry,( ii) own-rooted‘Sharpblue',a southern highbush blueberry( V. corymbosum) cultivar,and( iii) grafted-bluebedrry as experimental materials,which were grown in quartz sand medium for 90 d,irrigated with nutrient solution of three levels of p H values,respectively,i. e. p H 4. 6,p H 5. 8 and p H7. 0. Results showed that root development,shoot growth and dry biomass accumulation of grafted blueberry were significantly better than those of own-rooted blueberry grown in higher p H medium; compared with own-rooted blueberry saplings,leaf chlorophyll content,chlorophyll a/b ratio,chlorophyll fluorescence chemical efficiency and photosynthetic efficiency of grafted blueberry saplings grown in higher p H medium were all significantly improved. Results also showed that mineral element content of leaves,especially Mg and Fe elemental content of grafted blueberry was significantly higher than that of own-rooted blueberry saplings grown in higher p H media. Grafting-propagation of blueberry saplings taking Wufanshu as rootstock significantly enhanced the adaptation of blueberry to soil environment with higher p H value. 展开更多

Image-guided thermal ablation(TA),which is less invasive,has been widely applied for treating various kinds of tumors.However,TA still poses the potential risk of thermal damage to sensitive tissue nearby.Therefore,an adjunctive thermoprotective hydrodissection technique with constant injection of 5%glucose(5%Glu)has currently been adopted for clinical application,but this may be hazardous to humans.In this study,a multifunctional hyaluronic acid-based hydrogel(HA-Dc)was developed for hydrodissection.Compared with 5%Glu(the most clinically used solution)and the previously reported F127 hydrogel,the HA-Dc hydrogel was studied in vitro in a porcine liver model and in vivo in a rabbit model and showed good injectability and better tissue retention,stability,and thermoprotective properties throughout the TA procedure.Furthermore,in the preclinical evaluation in a Macaca fascicularis(M.fascicularis)model,HA-Dc showed excellent performance in terms of stricter neuroprotection compared with 5%Glu.In addition,the HA-Dc hydrogel with good biocompatibility and controllable degradation behavior in vivo could be a promising platform for thermal protection during clinical TA procedures. 展开更多

Analyses on the Overall Codon Usage Pattern in Fish Rhabdovirus

The codon usage bias and the base compositions in the available 49 complete fish rhabdovirus genome sequences were investigated. The high correlation between GC12% and GC3% suggested that mutational pressure rather than natural selection was the main factor determining the codon usage and base component in fish rhabdovirns, most of the spots lying below the expected curve ( ENC vs GC3% ) also suggested that codon usage bias in these 49 genomes was influenced by mutational pressure in codon usage pattern. Through PCA analysis based on RSCU, all ORFs in the 49 fish rhabdoviurs genomes were clustered into two parts, and this result showed that hosts played a role in codon usage pattern of fish rhabdovirns genomes. Further, comparison between SVCV, INHV, VHSV and their hosts suggested the effect of natural host on the codon usage pattern took part in the evolution process of fish rhabdovirns. This study represents the most comprehensive analysis to date for fish rhabdovirus eodon usage patterns. 展开更多

Synthesis of functional polyethylene from ethylene alone is tricky and heavily dependent on both the type and structure of the precatalyst and the choice of cocatalyst used in the polymerization.In the present study,a series of cobalt precatalysts was prepared and investigated for ethylene polymerization under various conditions.By incorporation of strong electron-withdrawing groups(F and NO_(2))and a steric component(benzhydryl)into the parent bis(imino)pyridine ligand,the catalytic performance of these precatalysts was optimized.On activation with MAO or MMAO,these precatalysts with relatively open structure achieved unprecedented ethylene polymerization rates at 60℃(up to 27.6×10^(6)g mol^(−1)h^(−1))and remained effective at temperatures up to 100℃.Chain growth reactions were moderate,resulting in polyethylene with molecular weights up to 61.0 kg/mol and broad bimodal dispersity index.High crystallinity and melt temperature indicated a strictly linear microstructure,as further confirmed by high-temperature^(1)H/^(13)C NMR measurements.Of significant note that chain termination predominantly occurred throughβ-elimination(up to 84.5%),yielding vinyl-terminated long-chain olefins.These functionalα-macro-olefins are valuable as precursors for postfunctionalization,expanding the potential applications of polyethylene across various sectors. 展开更多

Although the distance between binary codes can be computed fast in Hamming space, linear search is not practical for large scale datasets. Therefore attention has been paid to the efficiency of performing approximate nearest neighbor search, in which hierarchical clustering trees (HCT) are widely used. However, HCT select cluster centers randomly and build indexes with the entire binary code, this degrades search performance. In this paper, we first propose a new clustering algorithm, which chooses cluster centers on the basis of relative distances and uses a more homogeneous partition of the dataset than HCT has to build the hierarchical clustering trees. Then, we present an algorithm to compress binary codes by extracting distinctive bits according to the standard deviation of each bit. Consequently, a new index is proposed using compressed binary codes based on hierarchical decomposition of binary spaces. Experiments conducted on reference datasets and a dataset of one billion binary codes demonstrate the effectiveness and efficiency of our method. 展开更多

The Gay-Berne (GB) model has been proved to be highly successful in the simulation of liquid crystal phases via both molec- ular dynamics (MD) and nonequilibrium molecular dynamics (NEMD). However, the conventional thermostats used in the simulations of GB systems, such as Nose-Hoover and Langevin thermostats, have serious shortcomings especially in NEMD simulations. Recently, dissipative particle dynamics (DPD) has established itself as a useful thermostat for soft matter simulations, whereas the application of DPD thermostat in (NE)MD simulations is limited to the spherically isotropic potential models, such as the Lennard-Jones model. Considering the virtues of the DPD thermostat, that is, local, momentum conserved, and Galilean invariant, we extend the DPD thermostat to the non-spherical GB model. It is interesting to find that the translational DPD and rotational DPD thermostats can be used in the GB system independently and both can achieve the thermostatting effects. Also, we compared the performance of the DPD thermostat with other commonly used thermostats in NEMD simulations by investigating the streaming velocity profiles and the dynamics of phase separation in a typical but simple binary GB mixture under shear field. It is revealed that the known virtues of DPD thermostats, such as Galilean invariant, shear velocity profile-unbiased, and unscreened hydrodynamic interactions, are still intact when applying to GB systems. Finally, the appro- priate parameters for the DPD thermostat in the GB system are identified for future investigations. 展开更多

The 4,6-bis(arylimino)-1,2,3,7,8,9,10-heptahydrocyclohepta[b]quinoline-iron(II)chlorides(aryl=2,6-Me_(2)C_(6)H_(3) Fe1;2,6-Et_(2)C_(6)H_(3) Fe_(2);2,6-i-Pr_(2)C_(6)H_(3) Fe_(3);2,4,6-Me3C_(6)H_(2) Fe4;and 2,6-Et_(2)-4-Me_(2)C_(6)H_(2) Fe_(5))have been prepared in good yield by a straightforward one-pot reaction of 2,3,7,8,9,10-hexahydro-1H-cyclohepta[b]quinoline-4,6-dione,FeCl_(2)·4H_(2)O,and the appropriate aniline in acetic acid.All ferrous complexes have been characterized by elemental analysis and FT-IR spectroscopy.In addition,the structure of Fe_(3) has been determined by single crystal X-ray diffraction,which showed the iron center to adopt a distorted square pyramidal geometry with the saturated sections of the fused six-and sevenmembered carbocycles to be cis-configured.In combination with either MAO or MMAO,Fe1–Fe5 exhibited exceptionally high activities for ethylene polymerization(up to 15:86×10^(6) g(PE)mol^(−1)(Fe)h^(−1) at 40℃(MMAO)and 9:60×10^(6) g(PE)mol^(−1)(Fe)h^(−1) at 60℃(MAO))and produced highly linear polyethylene(HLPE,Tm≥128℃)with a wide range in molecular weights;in general,the MMAO-promoted polymerizations were more active.Irrespective of the cocatalyst employed,the 2,6-Me_(2)-substituted Fe1 and Fe4 proved the most active while the more sterically hindered 2,6-i-Pr_(2) Fe_(3) the least but afforded the highest molecular weight polyethylene(Mw:65.6–72.6 kg mol^(-1)).Multinuclear NMR spectroscopic analysis of the polymer formed using Fe4/MMAO at 40℃ showed a preference for fully saturated chain ends with a broad bimodal distribution a feature of the GPC trace(Mw/Mn=13:4).By contrast,using Fe4/MAO at 60℃ a vinyl-terminated polymer of lower molecular weight(Mw=14:2 kg mol^(−1))was identified that exhibited a unimodal distribution(Mw/Mn=3:8).Moreover,the amount of aluminoxane cocatalyst employed,temperature,and run time were also found to be influential on the modality of the polymer. 展开更多
