Dynamic model uncertainty analysis and control system multi-objective optimization of space nuclear reactor

Compared to other energy sources,nuclear reactors offer several advantages as a spacecraft power source,including compact size,high power density,and long operating life.These qualities make nuclear power an ideal energy source for future deep space exploration.A whole system model of the space nuclear reactor consisting of the reactor neutron kinetics,reactivity control,reactor heat transfer,heat exchanger,and thermoelectric converter was developed.In addition,an electrical power control system was designed based on the developed dynamic model.The GRS method was used to quantitatively calculate the uncertainty of coupling parameters of the neutronics,thermal-hydraulics,and control system for the space reactor.The Spearman correlation coefficient was applied in the sensitivity analysis of system input parameters to output parameters.The calculation results showed that the uncertainty of the output parameters caused by coupling parameters had the most considerable variation,with a relative standard deviation<2.01%.Effective delayed neutron fraction was most sensitive to electrical power.To obtain optimal control performance,the non-dominated sorting genetic algorithm method was employed to optimize the controller parameters based on the uncertainty quantification calculation.Two typical transient simulations were conducted to test the adaptive ability of the optimized controller in the uncertainty dynamic system,including 100%full power(FP)to 90%FP step load reduction transient and 5%FP/min linear variable load transient.The results showed that,considering the influence of system uncertainty,the optimized controller could improve the response speed and load following accuracy of electrical power control,in which the effectiveness and superiority have been verified. 展开更多

Pancreatic ductal adenocarcinoma(PDAC)has a poor prognosis and limited first-line treatments.This phaseⅠb/Ⅱrandomized trial(NCT05218889)investigated the efficacy and safety of surufatinib plus camrelizumab and nab-paclitaxel/S-1(NASCA)versus nab-paclitaxel and gemcitabine in patients with locally advanced or metastatic PDAC.The primary endpoints were dose-limiting toxicities and the recommended phaseⅡdose(RP2D)of surufatinib in phase lb,and the objective response rate(ORR)in phaseⅡ.PhaseⅠb used a 3+3 dose-escalation design to determine the RP2D of surufatinib in six patients,which was established at 200 mg.In phaseⅡ,patients were randomized 1∶1 to receive the NASCA(45 patients)or nab-paclitaxel and gemcitabine(45 patients).NASCA group showed an ORR of 51.1%(23/45)versus 24.4%(11/45)in the nab-paclitaxel and gemcitabine group(odds ratio 3.2,95%CI 1.3-8.2,p=0.01).The median progression-free survival(PFS)was 7.9 vs.5.3 months(HR 0.63,95%CI 0.40-0.99,p=0.045).The median overall survival was 13.0 vs.11.0 months(HR 0.77,95%CI 0.47-1.28,p=0.318).The most common Grade≥3 treatment-related adverse event was decreased neutrophil count(33.3%vs.35.6%).In the NASCA group,enrichment of CD8+and CD8+PD-1+cells,a high baseline M1/M2 macrophage ratio,and a reduction in CA19-9 levels at weeks 6 and 12 were associated with improved PFS compared to patients without these features.The NASCA regimen showed promising efficacy with tolerable safety relative to nab-paclitaxel and gemcitabine for locally advanced or metastatic PDAC. 展开更多

目的:探讨二甲双胍对核糖体40S小亚基S6K蛋白激酶(P70S6k)及胰岛素受体底物-1(IRS-1)蛋白及其ser307位点磷酸化表达的影响。方法:利用0.1 mmol/L二甲双胍体外连续作用多囊卵巢综合症(PCOS)患者黄素化颗粒细胞24 h为实验组,设未加二甲双... 目的:探讨二甲双胍对核糖体40S小亚基S6K蛋白激酶(P70S6k)及胰岛素受体底物-1(IRS-1)蛋白及其ser307位点磷酸化表达的影响。方法:利用0.1 mmol/L二甲双胍体外连续作用多囊卵巢综合症(PCOS)患者黄素化颗粒细胞24 h为实验组,设未加二甲双胍培养的颗粒细胞为对照组。通过RT-PCR和Real-time PCR检测P70S6k和IRS-1的表达,细胞免疫荧光化学和Western blotting的方法检测P70S6k、p-thr389-P70S6k、IRS-1及p-ser307-IRS-1蛋白的表达。结果:Real-time PCR结果显示实验组P70S6k和IRS-1m RNA水平显著低于对照组,差异有统计学意义(P<0.05)。细胞免疫荧光化学检测IRS-1和p-ser307-IRS-1蛋白在颗粒细胞胞质表达,P70S6k和p-thr389-P70S6k蛋白在细胞核表达。Western blotting法结果显示二甲双胍作用24 h前、后卵巢颗粒细胞P70S6k、p-thr389-P70S6k、IRS-1及p-ser307-IRS-1蛋白表达有统计学差异(P<0.05),灰度值分析显示添加二甲双胍组P70S6k、p-thr389-P70S6k蛋白表达显著升高,IRS-1及p-ser307-IRS-1蛋白表达显著下降(P<0.05)。结论:二甲双胍可抑制人颗粒细胞P70S6k的表达,并通过Akt/P70S6k/IRS途径调节IRS-1的表达,从而增加颗粒细胞胰岛素的敏感性。 展开更多

Objective:The aim is to study the effects of metformin on the expression of 70 kDa ribosomal protein S6 kinase(P70S6k),insulin receptor substrate 1(IRS-1),and IRS-1Ser307 phosphorylation in human luteinized granulosa cells.Methods:Granulosa cells in the experimental group were cultured in M199 medium containing 0.1 mmol/L metformin for 24 h and those in control group were cultured in M199 medium.The expression levels of P70S6k and IRS-1 mRNA were detected by reverse-transcriptiom polymerase chain reaction(RT-PCR)and real-time PCR.P70S6k,IRS-1,p-ser307-IRS-1,and p-thr389-P70S6k protein expression levels were detected by immunofluorescence and western blotting.Results:P70S6k mRNA level was higher and IRS-1 was significantly lower in the experimental group than those in the control group.IRS-1 and p-ser307-IRS-1 were expressed in cell plasma,and P70S6k and p-thr389-P70S6k were expressed in cell nucleus.The results of Western blot analysis indicated that the expression levels of P70S6k,p-thr389-P70S6k,IRS-1,and p-ser307-IRS-1 proteins had significant difference between the experimental group and the control group.Compared to the control group,the relative intensity illustrated that the expression levels of P70S6K and p-thr389-P70S6k significantly increased in the experimental group;however,those of IRS-1 and p-ser307-IRS-1 proteins significantly decreased.Conclusion:Metformin can inhibit the P70S6k mRNA and protein expression levels in the granulosa cells and improve insulin sensitivity by regulating IRS-1 expression through Akt/P70S6k/IRS-1-dependent pathway. 展开更多
