AIM: To explore the effect of Obtusifolin on retinal pigment epithelial cell growth under hypoxia.METHODS: In vitro chemical hypoxia model of ARPE-19 cells was established using cobalt chloride(CoCl2). Cell viability was tested by cell counting kit-8(CCK-8) assay. Western blot and real-time quantitative polymerase chain reaction were applied to detect proteins and mRNAs respectively. Flow cytometry was used to examine the cell cycle. Secretion of vascular endothelial growth factor(VEGF) was tested by using enzyme linked immunosorbent assay(ELISA).RESULTS: Under the chemical hypoxia model established by CoCl2, hypoxia inducible factor-1α(HIF-1α) mRNA and protein levels was up-regulated. Cell viability was increased and the proportion of S phase was higher. Obtusifolin could reduce cell viability under hypoxic conditions and arrest cells in G1 phase. Obtusifolin reduced the expression of Cyclin D1 and proliferating cell nuclear antigen(PCNA) in the hypoxic environment and increased the expression of p53 and p21. The levels of VEGF, VEGFR2 and eNOS proteins and mRNA were significantly increased under hypoxia while Obtusifolin inhibited the increasing.CONCLUSION: Obtusifolin can inhibit cell growth under hypoxic conditions and down-regulate HIF-1/VEGF/eNOS secretions in ARPE-19 cells. 展开更多

Objective To construct a morphine tolerance model in primarily cultured striatal neurons, and screen the differentially expressed genes in this model using suppression subtractive hybridization (SSH). Methods Sbtracted cDNA libraries were constructed using SSH from normal primarily cultured striatal neurons and long-term morphine treated striatal neurons (10^-5 mol/L for 72 hours). To check reliability of the cell culture model, RT-PCR was performed to detect the cAMP-responsive element-binding protein (CREB) mRNA expression. The subtracted clones were prescreened by PCR. The clones containing inserted fragments from forward libraries were sequenced and submitted to GenBank for homology analysis. And the expression levels of genes of interest were confirmed by RT-PCR. Results CREB mRNA expression showed a significant increase in morphine treated striatal neurons (62.85± 1.98) compared with normal striatal neurons (28.43 ± 1.46, P〈0.01). Thirty-six clones containing inserted fragments were randomly chosen for sequence analysis. And the 36 clones showed homology with 19 known genes and 2 novel genes. The expression of 2 novel genes, mitochondrial carrier homolog 1 (Mtchl ; 96.81±2.04 vs. 44.20±1.31, P〈0.01) and thyrnoma viral proto-oncogene 1 (Akt1 ; 122.10±2.17 vs 50.11±2.01, P〈0.01), showed a significant increase in morphine-treated striatal neurons compared with normal striatal neurons. Conclusions A reliable differential cDNA library of striatal neurons treated with long-term morphine is constructed. Mtchl and Aktl might be the candidate genes for the development of morphine tolerance. 展开更多

目的初步研究痛泻要方对急性放射性肠炎(radiation enteritis,RE)大鼠肠组织的防护作用及机制.方法随机将48只♂SD大鼠分为4组:A组为正常对照组(n=12)、B组为模型照射组(n=12)、C组为痛泻要方组(n=12)、D组为谷氨酰胺组(n=12);A组不予... 目的初步研究痛泻要方对急性放射性肠炎(radiation enteritis,RE)大鼠肠组织的防护作用及机制.方法随机将48只♂SD大鼠分为4组:A组为正常对照组(n=12)、B组为模型照射组(n=12)、C组为痛泻要方组(n=12)、D组为谷氨酰胺组(n=12);A组不予任何处理,其余三组均以高能X线直线加速器给予10 Gy全腹腔照射.造模成功后第1天,A组和B组给予蒸馏水,C组给予中药痛泻要方,D组给予谷氨酰胺,各组均连续灌胃7 d.每日观察大鼠一般状况、排便情况及体重变化.各组均于灌胃结束后6 h取空肠组织,光镜下观察肠组织形态学变化.检测空肠组织匀浆一氧化氮(nitric oxide,NO)的含量,ELISA法测定空肠黏膜炎性因子白介素(interleukin,IL)-6、IL-10、肿瘤坏死因子-α(tumor necrosis factorα,TNF-α)的蛋白含量.结果灌胃结束后,除B组于造模第3天死亡1只外,其余各组均无死亡.C组和D组大鼠的一般状况及黏液血便均有不同程度改善,体重增加较B组明显(P<0.05).与B组比较,C组和D组大鼠的NO、IL-6及TNF-α含量均出现下降(P<0.05).C组和D组大鼠的IL-10含量较B组大鼠明显升高,差异均有显著统计学意义(P<0.01).结论痛泻要方对急性RE大鼠的肠黏膜具有防护作用,其机制可能与降低空肠NO、IL-6及TNF-α活性,提高IL-10含量,减轻肠组织炎症反应有关. 展开更多

目的:探讨25G玻璃体切割术联合不同抗血管内皮生长因子(VEGF)药物治疗增殖性糖尿病视网膜病变(PDR)患者的效果观察。方法:选择2018-07/2020-07本院收治的PDR患者作为研究对象,所有患者均行25G玻璃体切割术,术前7d给予抗VEGF药物,根据治... 目的:探讨25G玻璃体切割术联合不同抗血管内皮生长因子(VEGF)药物治疗增殖性糖尿病视网膜病变(PDR)患者的效果观察。方法:选择2018-07/2020-07本院收治的PDR患者作为研究对象,所有患者均行25G玻璃体切割术,术前7d给予抗VEGF药物,根据治疗方法分为雷珠单抗组(31例31眼)、康柏西普组(30例30眼)、阿柏西普组(29例29眼)。于玻璃体腔注射抗VEGF药物前及行玻璃体切割术时采集房水检测VEGF、色素上皮衍生因子(PEDF)水平,于术前及术后3、6mo时测定最佳矫正视力(BCVA)、黄斑中心凹视网膜厚度(CMT)。结果:各组患者玻璃体腔注药后房水VEGF水平显著降低(P<0.05),PEDF水平升高(P<0.05),三组组间比较均无差异(P>0.05)。三组手术时间、术中出血、医源性裂孔发生情况比较均无差异(P>0.05)。三组患者术后3、6mo时BCVA显著优于术前(P<0.05),CMT显著低于术前(P<0.05),三组组间比较均无差异(P>0.05)。结论:PDR患者玻璃体切割术前玻璃体腔注射抗VEGF药物可降低房水内血管相关因子的表达;雷珠单抗、康柏西普、阿柏西普联合玻璃体切割术治疗PDR的临床疗效及安全性相当。 展开更多

目的:探讨不同填充物联合玻璃体切除及内界膜剥除治疗特发性黄斑裂孔(IMH)的疗效及安全性。方法:回顾性研究。选择2018-07/2020-03本院眼科收治的IMH患者117例117眼作为研究对象,两组患者均行玻璃体切除联合内界膜剥除术,根据术中填充... 目的:探讨不同填充物联合玻璃体切除及内界膜剥除治疗特发性黄斑裂孔(IMH)的疗效及安全性。方法:回顾性研究。选择2018-07/2020-03本院眼科收治的IMH患者117例117眼作为研究对象,两组患者均行玻璃体切除联合内界膜剥除术,根据术中填充材料不同分为两组:采用空气填充者65眼;采用C_(3)F_(8)填充者52眼。于术前及术后1、2、3mo时进行复查,统计患者末次随访时裂孔闭合率、闭合形态、最佳矫正视力(BCVA)、黄斑中央视网膜厚度(CRT)、外界膜层(ELM)缺损直径、椭圆体带缺损直径、眼压、术后并发症发生情况。结果:空气填充组、C_(3)F_(8)填充组患者术后气体吸收时间分别为8.55±2.17、25.74±7.41d(P<0.05);空气填充组及C_(3)F_(8)填充组黄斑裂孔闭合率(95.4%vs 98.1%)和闭合孔形态比例比较均无差异(P>0.05)。两组患者术后3mo时BCVA和CRT较术前有差异(P<0.001),ELM缺损直径、椭圆体带缺损直径较术前降低(P<0.001),两组间BCVA、CRT、ELM缺损直径、椭圆体带缺损直径比较均无差异(P>0.05)。两组患者治疗前后眼压比较无差异(P>0.05),两组患者均未发生严重并发症。结论:玻璃体切除及内界膜剥除联合空气填充及C_(3)F_(8)填充治疗IMH疗效及安全性相当,临床可根据实际情况选择。 展开更多

The optical properties of tri-group(B, Al, Ga, In) doped(6,6) SiC nanotubes(SiCNTs) are studied from first principles. The results show that the main absorption and dispersion of SiCNTs caused by the intrinsic transition appear in the ultraviolet-visible region(below 500 nm), and the tri-group doping increases the minimum dielectric constant value resulting in enhanced transmittance. In addition, the tri-group doping can introduce a weak absorption and dispersion region in the near-mid-infrared region, and the response peak blue shifts as the diameter of the doping atom increases. Comparative studies of reflectance, absorptivity, and transmittance show that the key factors affecting the transmittance of SiCNTs are reflectance(or refractive index) rather than absorption coefficient. 展开更多

Based on the transport theory and the polarization relaxation model,the effects of hydrogen and hydroxyl passivation on the conductivity and dielectric properties of silicon carbide nanowires(SiCNWs)with different sizes are numerically simulated.The results show that the variation trend of conductivity and band gap of passivated SiCNWs are opposite to the scenario of the size effect of bare SiCNWs.Among the influencing factors of conductivity,the carrier concentration plays a leading role.In the dielectric properties,the bare SiCNWs have a strong dielectric response in the blue light region,while passivated SiCNWs show a more obvious dielectric response in the far ultraviolet-light region.In particular,hydroxyl passivation produces a strong dielectric relaxation in the microwave band,indicating that hydroxyl passivated SiCNWs have a wide range of applications in electromagnetic absorption and shielding. 展开更多

Comparative study on transport properties of N-, P-, and As-doped SiC nanowires: Calculated based on first principles

According to the one-dimensional quantum state distribution, carrier scattering, and fixed range hopping model, the structural stability and electron transport properties of N-, P-, and As-doped SiC nanowires(N-SiCNWs, P-SiCNWs, and As-SiCNWs) are simulated by using the first principles calculations. The results show that the lattice structure of NSiCNWs is the most stable in the lattice structures of the above three kinds of doped SiCNWs. At room temperature,for unpassivated SiCNWs, the doping effect of P and As are better than that of N. After passivation, the conductivities of all doped SiCNWs increase by approximately two orders of magnitude. The N-SiCNW has the lowest conductivity. In addition, the N-, P-, As-doped SiCNWs before and after passivation have the same conductivity–temperature characteristics,that is, above room temperature, the conductivity values of the doped SiCNWs all increase with temperature increasing.These results contribute to the electronic application of nanodevices. 展开更多

Silicon carbide nanosheets(SiCNSs)have a very broad application prospect in the field of new two-dimensional(2D)materials.In this paper,the interlayer interaction mechanism of bilayer SiCNSs(BL-SiCNSs)and its effect on optical properties are studied by first principles.Taking the charge and dipole moment of the layers as parameters,an interlayer coupling model is constructed which is more convenient to control the photoelectric properties.The results show that the stronger the interlayer coupling,the smaller the band gap of BL-SiCNSs.The interlayer coupling also changes the number of absorption peaks and causes the red or blue shift of absorption peaks.The strong interlayer coupling can produce obvious dispersion and regulate the optical transmission properties.The larger the interlayer distance,the smaller the permittivity in the vertical direction.However,the permittivity of the parallel direction is negative in the range of 150-300 nm,showing obvious metallicity.It is expected that the results will provide a meaningful theoretical basis for further study of SiCNSs optoelectronic devices. 展开更多
