In this paper,we report the construction of two accurate mass databases and the development of a combination detection method that simultaneously screens for 733 pesticide and chemical contaminant multi-residues via h...In this paper,we report the construction of two accurate mass databases and the development of a combination detection method that simultaneously screens for 733 pesticide and chemical contaminant multi-residues via high-throughput liquid chromatography(LC)-and gas chromatography(GC)-quadru pole-time-of-flight mass spectrometry(Q-TOFMS).This work demonstrates that electronic mass spectral standards may replace chemical-source standard materials as references through one sample preparation and the combination of GC/LC-Q-TOFMS screening.This cutting-edge technique has also replaced multiresidue determination using targeted detection with non-targeted screening.The pesticide residue types,sensitivity,recovery,and reproducibility of this combination technique are evaluated in eight fruit and vegetable matrices.This technique shows three advantages:①In comparison with the discovery capability of a single technique,the combination technique shows an improvement of 51.1%(GC-QTOFMS)and 39.6%(LC-Q-TOFMS),respectively;②the combination technique can satisfy a screening limit lower than 10μg·kg^-1 and meet the requirements of“uniform standards,”although some of the pesticide residues could be optimized to further improve screening sensitivity;③over 488 pesticides with recoveries between 60%-120%and relative standard deviation(RSD)<20%at a spiked level of 10μg·kg^-1 were detected with the combination technique in eight different matrices.From 2012 to 2017,this combination technique was applied in an investigation to screen pesticide residues from 1384 sampling locations for 38138 batched samples covering 18 categories and 134 types of fruits and vegetables obtained from across the mainland of China.After statistical analysis,533 pesticides in 115891 determinations were detected,and the regularity of pesticides in the fruits and vegetables sold on the Chinese market was shown.展开更多

The morphological identification showed that Micorcystis wesenbergii and Micorcystis smithii were the main component of the bloom. Five strains of M. smithii were successfully isolated. Their 16S rRNA gene sequences based phylogenetic tree showed that the five strains of M. smithii intermixed with strains of other morphospecies in Microcystis. A fragment of mcy gene encoding for microcystin synthetase was detected in one of the five M. smithii strains (CHAB 2183), indicating its potential of microcystin production. High performance liquid chromatography analysis confirmed M. smithii CHAB 2183 to produce microcystin-RR as 1550 μg per gram dry weight cells. The present investigation, for the first time, reported the isolated strains of M. smithii and microcystin production from M. smithii.展开更多
Lake Caohai has experienced extensive Microcystis blooms in recent years,and to improve its water quality,the local government carried out a series of water control measures.To better understand the dynamics of both p...Lake Caohai has experienced extensive Microcystis blooms in recent years,and to improve its water quality,the local government carried out a series of water control measures.To better understand the dynamics of both pelagic and benthic Microcystis and their characteristics in Lake Caohai,we conducted a 1-year investigation from December 2015to December 2016 to gain a seasonal outlook on the distribution and dynamics of cell abundance,colony size and intracellular microcystins(MCs)of Microcystis.The results indicated that the Microcystis bloom occupied primarily the northeastern region and then moved gradually from lakeshore to lake center.The perennial southwesterly winds and the water inflow from northeast to southwest in Lake Caohai determined the spatiotemporal distribution of pelagic Microcystis.Benthic Microcystis was mainly distributed in the northeastern region in summer,occupied the lake center in autumn and then occupied the southeastern region in winter,determined by the sedimentation of pelagic Microcystis and the death of benthic Microcystis.Small colonies(20-60μm)overwintered more easily in both water column and sediment.The concentrations of intracellular toxin of benthic Microcystis were observed to be significantly higher than those of pelagic Microcystis.This might be because Microcystis synthesized large amount of MCs to acclimate to an unfavorable benthic environment.This knowledge on the dynamics of Microcystis expands our understanding of mechanisms underpinning the formation of Microcystis blooms.展开更多
Twenty water bodies in China were sampled,and 186 strains of different Microcystis species were isolated,from which eight morpho species were identified and 43 stains containing the mcyB gene were detected.Phylogeneti...Twenty water bodies in China were sampled,and 186 strains of different Microcystis species were isolated,from which eight morpho species were identified and 43 stains containing the mcyB gene were detected.Phylogenetic analysis based on the mcyB gene indicated that the microcystin(MC)-producing Microcystis in China could be divided into two groups(ⅠandⅡ)and showed significant differences between the two groups.The maximum sequence similarity was 69.1%.Microcystins(MCs)were measured by high-performance liquid chromatography(HPLC)analysis,and no microcystin-RR(MC-RR)was detected in some strains belonging to GroupⅡ.Compared to other regions of the world,the proportion of Chinese MC-producing was different,and the regional differences were more obvious.A whole-cell polymerase chain reactio(PCR)assay was conducted to analyze the proportion of the mcyB gene in the laboratory cultured and field cultured Microcystis.The proportion of four morphospecies(M.vividis,M.ichthyoblabe,M.novacekii,and M.aeruginosa)that contained the mcyB gene exceeded 50%in the field cultured sample s.Compared with former studies,M.aeruginosa was the mo st likely morphotype that can produce MCs in the world.This study provided new insight of Microcystis hazard assessment and field monitoring.展开更多
