Circ-PAN3 facilitates hepatocellular carcinoma growth via sponging miR-153 and upregulating cyclin D1

Background:Circular RNAs(circRNAs)play a pivotal role in the development and advancement of various cancer types.However,the involvement of circ-PAN3 in hepatocellular carcinoma(HCC)is not well understood.To shed light on this,we conducted a comprehensive study through biochemistry,cell biology,molecular biology,and bioinformatics techniques to investigate the role of circ-PAN3 and its associated pathway in the progression of HCC.Methods:Cell Counting Kit-8(CCK-8)assay and colony formation assay were utilized to evaluate cell proliferation;Quantitative real-time PCR(RT-qPCR)and Western blot were adopted for assessing mRNA and protein expression;Annexin V/propidium iodide(PI)staining was applied to detect cellular apoptosis;CircInteractome and Targetscan databases were searched to predict potential targets of circRNA and miRNA;Luciferase reporter assay and RNA pulldown assay were performed to examine the interaction of RNA molecules.Conclusions:Our findings revealed a significant increase in circ-PAN3 expression in HCC clinical specimens,which correlated with a poor survival rate in HCC patients.Knockdown of circ-PAN3 resulted in impaired cell proliferation,reduced cell survival,and inhibited tumorigenesis of HCC in vivo.Further analysis demonstrated that circ-PAN3 could serve as a sponge for miR-153,leading to a decrease in its expression level.This in turn upregulated cyclin D1 and ultimately promoted the proliferation of HCC cells.Additionally,overexpression of cyclin D1 mitigated the inhibitory effect on HCC proliferation induced by circ-PAN3 knockdown.Our study highlights the presence of a novel circ-PAN3/miR-153/cyclin D1 regulatory axis that plays a crucial role in the progression of HCC. 展开更多

Objective Castleman disease, also known as giant lymph node hyperplasia, involves lesions in the lymph nodes usually located in the chest_ENREF_1, particularly in the mediastinum. Meanwhile, sinus bradycardia is a sinus rhythm slower than 60 beats per min, and it can occur in both healthy and sick individuals. However, the comorbidity of these two disorders has not been previously reported. In this paper, we report a case of a 46-year-old woman who presented with persistent sinus bradycardia and irondeficiency anemia. Diagnostic work-up revealed hepatosplenomegaly and a giant mass near the splenic hilum. The mass was removed surgically; after which, the patient's bradycardia resolved immediately, while her anemia was corrected after subsequent chemotherapy. Pathological examination revealed lymph nodes with benign lesions, and the patient was diagnosed with hyaline-vascular variant of Castleman disease. This is the first documented case of sinus bradycardia associated with Castleman disease. In this paper, we describe the case characteristics, discuss the possible pathogenesis, and consider the appropriate treatment of symptomatic sinus bradycardia accompanying Castleman disease. 展开更多

Laparoscopic middle pancreatectomy

Pancreaticoduodenectomy and distal pancreatectomy are the traditional surgical treatments of tumors in the neck or body of the pancreas. Although resecting the lesions successfully, these procedures can also, however, lead the substantial loss of normal pancreatic parenchyma, causing endocrine and exocrine function disorder. The combination of distal pancreatectomy with splenectomy increased the risk of thrombosis sometimes. So the surgical trauma is too great, especially for benign and lower malignant tumors located in the neck or body of the pancreas. While, middle pancreatectomy may decrease operative trauma with the excision of these lesions, and can maximize the retention of pancreatic parenchyma, and maintain pancreatic endocrine and exocrine function integrity. The procedure is interesting but rarely performed. With the application and development of minimally invasive surgical techniques, laparoscopic and robotic middle pancreatectomy are available now. Researches about laparoscopic and robotic middle pancreatectomy are presented, with decreased morbidity, reduced operation time and hospital stay. There is only a few reports on the two procedures, but the security and effectiveness of them are suggested. 展开更多

Pancreaticoduodenectomy(PD) has long been used for chronic pancreatitis(CP), but greatly affects the postoperative quality of life. A new procedure called duodenum-preserving pancreatic head resection(DPPHR) has been introduced, and has little effect on the structure and function of the digestive system. With the development of minimally invasive surgical techniques, treatment of CP can be performed with laparoscopic DPPHR(LDPPHR). We present a case of CP that was successfully treated with LDPPHR. The postoperative pathological diagnosis was pancreatitis, demonstrating the feasibility of LDPPHR. We recommend this minimally invasive surgical method as preferred treatment for CP. 展开更多
