Objective:To investigate and explore the current status and correlation between psychological status and disease perception of adolescent patients with inflammatory bowel disease.Methods:Adolescent patients hospitaliz...Objective:To investigate and explore the current status and correlation between psychological status and disease perception of adolescent patients with inflammatory bowel disease.Methods:Adolescent patients hospitalized in a tertiary hospital in Zhejiang Province and some adolescent patients attending the summer camp of Zhejiang Province Aizaiyanchang Foundation for Inflammatory Bowel Disease were selected as the study subjects,and a questionnaire survey was carried out on 148 adolescent patients with inflammatory bowel disease by using the Generalized Anxiety Disorder Scale(GAD-7),Patient Health Questionnaire(PHQ-9),and the Simplified Version of the Disease Perception Questionnaire(BIPQ).questionnaire survey.Results:The depression score of adolescent inflammatory bowel disease patients was(16.40±8.30),the anxiety score was(12.55±5.34),and the total illness perception score was(43.75±10.41).Disease perception of adolescent inflammatory bowel disease patients was at moderate to high level,in which adolescent inflammatory bowel disease patients had the strongest perception of disease control in cognitive dimension as(7.20±0.98)score,followed by strong perception of treatment effect in cognitive dimension as(6.95±2.23)score.According to Pearson correlation analysis it was concluded that their disease perception was positively correlated with anxiety and depression.Conclusion:The psychological condition of adolescent patients with inflammatory bowel disease is poor,and there are significant individual differences in the level of disease perception.Healthcare professionals should provide appropriate knowledge support and psychological interventions according to the different needs of adolescent patients.In addition,an online and offline communication platform should be established to better help adolescent patients with inflammatory bowel disease.展开更多

We have previously reported that natural killer (NK) cells enter target tumor ceils, leading to either target cell death or self-destruction within tumor cells. However, it has remained elusive as to the fate of NK cells after internalization and whether the heterotypic cell-in-cell process is different from that of the homotypic cell-in-cell event recently named entosis. Here, we show that NK cells undergo a cell-in-cell process with the ultimate fate of apoptosis within tumor cells and reveal that the internalization process requires the actin cytoskeletal regulator, ezrin. To visualize how NK cells enter into tumor cells, we carried out real-time dual color imaging analyses of NK cell internalization into tumor cells. Surprisingly, most NK cells commit to programmed cell death after their entry into tumor cells, which is distinctively different from entosis observed in the homotypic cell-in-cell process. The apoptotic cell death of the internalized NK cells was evident by activation of caspase 3 and DNA fragmentation. Furthermore, NK cell death after internalization is attenuated by the caspase inhibitor, Z-VAD-FMK, confirming apoptosis as the mode of NK cell death within tumor cells. To determine protein factors essential for the entry of NK cells into tumor cells, we car- ried out siRNA-based knockdown analysis and discovered a critical role of ezrin in NK cell internalization. Impor- tantly, PKA-mediated phosphorylation of ezrin promotes the NK cell internalization process. Our findings suggest a novel regulatory mechanism by which ezrin governs NK cell internalization into tumor cells.展开更多
Objective:To describe the Chinese experience of natural orifice transluminal endoscopic surgery(NOTES)in urology.Methods:From December 2008 to May 2017,35 animal experiments and 305 clinical surgeries of NOTES or natu...Objective:To describe the Chinese experience of natural orifice transluminal endoscopic surgery(NOTES)in urology.Methods:From December 2008 to May 2017,35 animal experiments and 305 clinical surgeries of NOTES or natural orifices specimen extractions(NOSE)were performed in China.The animal experiments included five kidney biopsies,24 nephrectomies and six partial nephrectomies.The clinical surgeries included 12 transvaginal NOSE(TV-NOSE),266 hybrid transvaginal NOTES(TV-NOTES)and 27 pure TV-NOTES.The TV-NOSE procedure was performed in five transumbilical laparoendoscopic single-site(U-LESS)nephrectomies,four suprapubic-assisted laparoendoscopic single-site surgery(SA-LESS)nephroureterectomies,and three laparoscopic radical cystectomies.The hybrid TV-NOTES procedure included 210 nephrectomies,31 adrenalectomies,eight nephroureterectomies,13 partial nephrectomies,and four heminephrectomies.The pure TV-NOTES procedure included five renal cyst decortications and 22 nephrectomies.Results:A total of 29 animal experiments were successfully performed.One partial nephrectomy was converted to standard laparoscopic surgery.Two kidney biopsies and two nephrectomies were unsuccessful.A total of 297 clinical surgeries were successfully performed.Six patients who underwent hybrid TV-NOTES were converted to open surgery.Two patients who underwent pure TV-NOTES were converted to SA-LESS.There were 22 major complications,16 occurred intraoperatively and six postoperatively.The mean visual analog score(VAS)of 48 h after the operation was 2.5 points in TV-NOSE,2.3 points in hybrid TV-NOTES and 1.7 points in pure TV-NOTES.The mean follow-up of 50.6(3.0-87.0)months showed that all patients were in good condition.The umbilicus scars were nearly invisible in TV-NOSE and hybrid TV-NOTES.The vaginal incision healed well.Conclusions:TV-NOSE and TV-NOTES are feasible,safe,and effective with little injury,low pain,fast recovery,and good cosmetic outcomes in properly selected patients.They are worth consideration for urological clinical practice.展开更多

Population genetic diversity which is essential for adaptation to environmental change is a useful index for long-term species survival. In this paper, genetic diversity of eight Phrynocephalus forsythii population which distributed in Tarim Basin, China, were evaluated based on three mtDNA gene and its correlation with environment factors were investigated using RDA. Our result revealed that, the level of genetic diversity of P. forsythii populations was related to its location but there was no significant correlation between genetic distances and geographic distances in P. forsythii. However, we find that mtDNA of P. forsythii was subjected to selection pressure during evolution and population genetic diversity was significantly positively related to variation coefficient of rainfall(VCR) and altitude(AL), while significantly negatively related to longitude(N) and annual average temperature(AAT). Our result supported the previous prediction that excessive ambient heat is a threat to P. forsythii.展开更多

Hierarchically porous carbon materials are promising for energy storage,separation and catalysis.It is desirable but fairly challenging to simultaneously create ultrahigh surface areas,large pore volumes and high N co...Hierarchically porous carbon materials are promising for energy storage,separation and catalysis.It is desirable but fairly challenging to simultaneously create ultrahigh surface areas,large pore volumes and high N contents in these materials.Herein,we demonstrate a facile acid-base enabled in situ molecular foaming and activation strategy for the synthesis of hierarchically macro-/meso-/microporous N-doped carbon foams(HPNCFs).The key design for the synthesis is the selection of histidine(His)and potassium bicarbonate(PBC)to allow the formation of 3D foam structures by in situ foaming,the PBC/His acid-base reaction to enable a molecular mixing and subsequent a uniform chemical activation,and the stable imidazole moiety in His to sustain high N contents after carbonization.The formation mechanism of the HPNCFs is studied in detail.The prepared HPNCFs possess 3D macroporous frameworks with thin well-graphitized carbon walls,ultrahigh surface areas(up to 3200 m^2 g^-1),large pore volumes(up to 2.0 cm^3 g^-1),high micropore volumes(up to 0.67 cm^3 g^-1),narrowly distributed micropores and mesopores and high N contents(up to 14.6 wt%)with pyrrolic N as the predominant N site.The HPNCFs are promising for supercapacitors with high specific capacitances(185-240 F g^-1),good rate capability and excellent stability.They are also excellent for CO2 capture with a high adsorption capacity(~4.13 mmol g^-1),a large isosteric heat of adsorption(26.5 kJ mol^-1)and an excellent CO2/N2 selectivity(~24).展开更多

4-Hydroxyphenylpyruvate dioxygenase(HPPD)is an important target for both drug and pesticide discovery.As a typical Fe(II)-dependent dioxygenase,HPPD catalyzes the complicated transformation of 4-hydroxyphenylpyruvic a...4-Hydroxyphenylpyruvate dioxygenase(HPPD)is an important target for both drug and pesticide discovery.As a typical Fe(II)-dependent dioxygenase,HPPD catalyzes the complicated transformation of 4-hydroxyphenylpyruvic acid(HPPA)to homogentisic acid(HGA).The binding mode of HPPA in the catalytic pocket of HPPD is a focus of research interests.Recently,we reported the crystal structure of Arabidopsis thaliana HPPD(At HPPD)complexed with HPPA and a cobalt ion,which was supposed to mimic the pre-reactive structure of At HPPD-HPPA-Fe(II).Unexpectedly,the present study shows that the restored At HPPD-HPPA-Fe(II)complex is still nonreactive toward the bound dioxygen.QM/MM and QM calculations reveal that the HPPA resists the electrophilic attacking of the bound dioxygen by the trim of its phenyl ring,and the residue Phe381 plays a key role in orienting the phenyl ring.Kinetic study on the F381 A mutant reveals that the HPPD-HPPA complex observed in the crystal structure should be an intermediate of the substrate transportation instead of the pre-reactive complex.More importantly,the binding mode of the HPPA in this complex is shared with several well-known HPPD inhibitors,suggesting that these inhibitors resist the association of dioxygen(and exert their inhibitory roles)in the same way as the HPPA.The present study provides insights into the inhibition mechanism of HPPD inhibitors.展开更多
The gut microbiota has gained attention because of its importance in facilitating host survival and evolution.However,it is unclear whether gut microbial communities are determined by the host(heritable factor) or env...The gut microbiota has gained attention because of its importance in facilitating host survival and evolution.However,it is unclear whether gut microbial communities are determined by the host(heritable factor) or environment(environmental factor).In this study,we investigated the gut microbial communities and potential functional signatures of two sympatric species distributed along an elevation gradient,the toadheaded lizards Phrynocephalus axillaris and P.forsythii.Our results indicated that at high elevations,the gut microbial communities of P.axillaris and P.forsythii did not significantly differ,and the phylogenetic relationships of gut microbial communities contradicted their hosts.At low altitudes,the two lizards could be distinguished based on their significantly different gut microbial communities.Compared to low-altitude populations,Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis showed that at higher altitudes,energy metabolism,such as carbohydrate,lipid,and amino acids metabolism were higher in both lizards.While a larger number of pathogenic bacteria were found in the lowaltitude population of P.forsythii.This suggests that the convergence of gut microbiota of two lizards at highaltitude stem from environmental factors,as they were exposed to the same environmental stress,whereas the divergence at low-altitude stemmed from heritable factors,as they were exposed to different environmental stresses.These results provide a new perspective regarding whether heritable or environmental factors dominate the gut microbiota during exposure to environmental stress.展开更多
Speciation has never been directly observed in nature because it is a lengthy phenomenon.Although rare,ring species are an optimal natural example of speciation and can be identified through the assessment of the geog...Speciation has never been directly observed in nature because it is a lengthy phenomenon.Although rare,ring species are an optimal natural example of speciation and can be identified through the assessment of the geographical conditions of their potential habitat.Phrynocephalus forsythii is endemic to the Tarim Basin,which compnses the Taklamakan Desert and surrounded by mountains on three sides.This study aimed to determine whether P.forsythii had a ring-species-like divergence pattern through the characterization of the genetic features of 17 populations covering the major distribution of this species.Species distribution modelling revealed that P.forsythii had a continuous circular distribution around the Tarim Basin.Gene flow was observed in most adjacent populations except for two terminal populations of the ring,which exhibit the highest differentiation.Genetic distance and geographic distance were significantly correlated,indicating that the observed differentiation resulted from genetic variation gradually accumulating during population dispersion.Although our results do not definitively indicate that P.forsythii is a ring species,our results indicate a ring-shaped diversification This phenomenon elucidates the potential mechanism underlying speciation in the presence of gene flow,providing insight into this evolutionary process.展开更多
Objective:The aim of the study was to observe the therapeutic effect of FOLFOX4 regimen plus Fufangchangtai decoction on postoperative colorectal cancers.Methods:Thirty postoperative colorectal cancer patients were al...Objective:The aim of the study was to observe the therapeutic effect of FOLFOX4 regimen plus Fufangchangtai decoction on postoperative colorectal cancers.Methods:Thirty postoperative colorectal cancer patients were allocated into control and experiment groups respectively.Patients in experiment group were given Fufangchangtai decoction combined with FOLFOX4 regimen.Patients in control group were given FOLFOX4 regimen alone.Efficacy was evaluated after 2 periods of treatment.Results:The improvement rate of symptoms were 86.6% in experiment group compared to 53.3% in control group.KPS was stable in experiment group,and decreased in control group.QOL was increased in experiment group,and stable in control group after the treatment.For impact of immunity parameters,there were enhancements of CD3+ and CD4+ in experiment group,while they did not change in control group.In experiment group,WBC reduction was slighter than that of control group.The differences were not remarkable in PLT reduction,alimentary response,and toxicity of liver and kidney and nervous system.Conclusion:The clinical observation showed that Fufangchangtai decoction plus FOLFOX4 regimen could effectively enhance KPS,improve the symptoms,the quality of life and the immunity state,and down-regulate the side effects.In conclusion,Fufangchangtai decoction can decrease the toxicity so as to increase the treatment effect.展开更多
Tree species diversity is vital for maintaining ecosystem functions,yet our ability to map the distribution of tree diversity is limited due to difficulties in traditional field-based approaches.Recent developments in...Tree species diversity is vital for maintaining ecosystem functions,yet our ability to map the distribution of tree diversity is limited due to difficulties in traditional field-based approaches.Recent developments in spaceborne remote sensing provide unprecedented opportunities to map and monitor tree diversity more efficiently.Here we built partial least squares regression models using the multispectral surface reflectance acquired by Sentinel-2 satellites and the inventory data from 74 subtropical forest plots to predict canopy tree diversity in a national natural reserve in eastern China.In particular,we evaluated the underappreciated roles of the practical definition of forest canopy and phenological variation in predicting tree diversity by testing three different definitions of canopy trees and comparing models built using satellite imagery of different seasons.Our best models explained 42%–63%variations in observed diversities in cross-validation tests,with higher explanation power for diversity indices that are more sensitive to abundant species.The models built using imageries from early spring and late autumn showed consistently better fits than those built using data from other seasons,highlighting the significant role of transitional phenology in remotely sensing plant diversity.Our results suggested that the cumulative diameter(60%–80%)of the biggest trees is a better way to define the canopy layer than using the subjective fixeddiameter-threshold(5–12 cm)or the cumulative basal area(90%–95%)of the biggest trees.Remarkably,these approaches resulted in contrasting diversity maps that call attention to canopy structure in remote sensing of tree diversity.This study demonstrates the potential of mapping and monitoring tree diversity using the Sentinal-2 data in species-rich forests.展开更多

High sensitivity and good selectivity were observed. The detection linear range was from 0.01 μIU/mL to 10 μIU/mL, in which the peak currents increased along with the concentration. The detection limit was 0.005 μIU/mL at S/N = 3. The immunosensor was also applied for TSH detection in human serum with recoveries from 98.0% to 101.8% and relative standard deviations from 1.3% to 3.1%, demonstrating potential value in clinical diagnosis.展开更多

Doxorubicin(DOx)is the most effective chemotherapeutic for breast cancer,but it is usually associated with severe cardiotoxicity.Further investigation to alleviate its side effects is essential.The present study inves...Doxorubicin(DOx)is the most effective chemotherapeutic for breast cancer,but it is usually associated with severe cardiotoxicity.Further investigation to alleviate its side effects is essential.The present study investigated the mechanism of the cross-organ communication between tumors and the heart and potential intervention targets.Morphological bubble-like protrusions were observed in both adult murine ventricular cardiomyocytes(AMvCs)and human induced pluripotent stem cell-derived cardiomyocytes(hiPSC-CMs)cocultured with breast cancer cells(BCCs),along with elevated expression of pyroptosis-related proteins.Exosomes(EXOs)from DOX-treated BCCs aggravated DOX-induced cardiotoxicity(DOXIC)in an orthotopic mouse model of breast cancer.Blocking miRNAs by knocking down Rab27a or inhibiting the release of EXOs in cancer tissue by Dicer enzyme knockout attenuated this additional injury effect.Exosomal miRNA sequencing revealed that miR-216a-5p is especially upregulated in EXOs from DOX-induced BCCs.Mechanistically,miR-216a-5p was upregulated by enhanced transcription mediated by DOXinduced AMP-dependent transcription factor 3(ATF3)and packaged into EXOs by splicing factor 3b subunit 4(SF3B4)in BCCs.Itchy E3 ubiquitin-protein ligase(ITCH)was identified as a novel downstream target mRNA of miR-216a-5p.ITCH negatively mediated thioredoxin-interacting protein(TXNIP)ubiquitination to activate the NOD-,LRR-and pyrin domain-containing protein 3(NLRP3)inflammasome pathway,ultimately leading to cardiomyocyte pyroptosis.Our findings revealed novel cross-organ pathogenic communication between breast cancer and the heart through the exosomal miR-216a-5p-mediated ITCH/TXNIP/NLRP3 pathway,which drives cardiomyocyte pyroptosis.These findings suggest that targeting myocardial miR-216a-5p or blocking harmful EXOs from breast cancer is a potential therapeutic strategy for alleviating DOXIC.展开更多
Introduction:To investigate the prevalence of elevated blood pressure(EBP)and its association with Dietary Approaches to Stop Hypertension(DASH)adherence among Chinese children and adolescents,providing evidence for d...Introduction:To investigate the prevalence of elevated blood pressure(EBP)and its association with Dietary Approaches to Stop Hypertension(DASH)adherence among Chinese children and adolescents,providing evidence for dietary interventions targeting pediatric hypertension.Methods:Data were derived from the“National Survey on Nutrition and Health of Children Aged 0–18 Years,”a key project under China’s 13th Five-Year Plan for National Science and Technology Infrastructure.DASH scores were calculated using standardized criteria,and multivariable logistic regression models were employed to evaluate associations between EBP and DASH adherence.Results:The prevalence of EBP in 7–17-year-olds was 15.4%.The median DASH score was 24 points,with the highest adherence tertile(T3)exhibiting a lower prevalence of EBP(14.9%)compared to T1(lowest adherence)(16.2%)and T2(moderate adherence)(15.2%).In the fully adjusted model(Model 2),adolescents aged 13–17 years in the T3 DASH adherence group showed reduced risk of EBP[odds ratio(OR)=0.78,95%confidence interval(CI)=0.60,0.98]relative to T1.Conclusion:Higher DASH adherence is associated with reduced EBP risk in adolescents aged 13–17 years,emphasizing the protective role of DASHrecommended foods in promoting cardiovascular health.展开更多
Modifying Ir by foreign metals with oxophilicity is a promising strategy to accelerate the hydrogen oxidation kinetics.However,uncontrolled enrichment and oxidative dissolution of metastable oxophilic dopants in conve...Modifying Ir by foreign metals with oxophilicity is a promising strategy to accelerate the hydrogen oxidation kinetics.However,uncontrolled enrichment and oxidative dissolution of metastable oxophilic dopants in conventional Ir-based alloys impair their activity and durability.Here,we address these challenges by atomically dispersing oxophilic Sn sites within Ir clusters to form dilute alloys.The Sn1-Ir pairs,confined within an atomic-scale lattice,prevent excessive*OH coverage caused by oxophilic site enrichment,while also reducing durability loss due to the dissolution of metastable dopants.Our analysis reveals that the Sn1-Ir pairs facilitate electron transfer between Sn1 and adjacent Ir sites,generating electron-rich Ir atoms and electron-poor Sn atoms.This modulation weakens*H and CO adsorption on Ir sites while enhancing OH adsorption on Sn sites.The resulting catalyst shows improved catalytic hydrogen oxidation performance in alkaline media,with mass activities 6.4 and 10.7 times higher than that of Ir/C and Pt/C,respectively.Under CO poisoning conditions,it retains 90.9%of its initial activity,outperforming both Ir/C and Pt/C.This work offers new perspectives on the design of dual-site catalysts for hydrogen oxidation catalysis.展开更多

文摘Objective:To investigate and explore the current status and correlation between psychological status and disease perception of adolescent patients with inflammatory bowel disease.Methods:Adolescent patients hospitalized in a tertiary hospital in Zhejiang Province and some adolescent patients attending the summer camp of Zhejiang Province Aizaiyanchang Foundation for Inflammatory Bowel Disease were selected as the study subjects,and a questionnaire survey was carried out on 148 adolescent patients with inflammatory bowel disease by using the Generalized Anxiety Disorder Scale(GAD-7),Patient Health Questionnaire(PHQ-9),and the Simplified Version of the Disease Perception Questionnaire(BIPQ).questionnaire survey.Results:The depression score of adolescent inflammatory bowel disease patients was(16.40±8.30),the anxiety score was(12.55±5.34),and the total illness perception score was(43.75±10.41).Disease perception of adolescent inflammatory bowel disease patients was at moderate to high level,in which adolescent inflammatory bowel disease patients had the strongest perception of disease control in cognitive dimension as(7.20±0.98)score,followed by strong perception of treatment effect in cognitive dimension as(6.95±2.23)score.According to Pearson correlation analysis it was concluded that their disease perception was positively correlated with anxiety and depression.Conclusion:The psychological condition of adolescent patients with inflammatory bowel disease is poor,and there are significant individual differences in the level of disease perception.Healthcare professionals should provide appropriate knowledge support and psychological interventions according to the different needs of adolescent patients.In addition,an online and offline communication platform should be established to better help adolescent patients with inflammatory bowel disease.

文摘Cytotoxic lymphocytes are key players in the orchestration of immune response and elimination of defective cells. We have previously reported that natural killer (NK) cells enter target tumor ceils, leading to either target cell death or self-destruction within tumor cells. However, it has remained elusive as to the fate of NK cells after internalization and whether the heterotypic cell-in-cell process is different from that of the homotypic cell-in-cell event recently named entosis. Here, we show that NK cells undergo a cell-in-cell process with the ultimate fate of apoptosis within tumor cells and reveal that the internalization process requires the actin cytoskeletal regulator, ezrin. To visualize how NK cells enter into tumor cells, we carried out real-time dual color imaging analyses of NK cell internalization into tumor cells. Surprisingly, most NK cells commit to programmed cell death after their entry into tumor cells, which is distinctively different from entosis observed in the homotypic cell-in-cell process. The apoptotic cell death of the internalized NK cells was evident by activation of caspase 3 and DNA fragmentation. Furthermore, NK cell death after internalization is attenuated by the caspase inhibitor, Z-VAD-FMK, confirming apoptosis as the mode of NK cell death within tumor cells. To determine protein factors essential for the entry of NK cells into tumor cells, we car- ried out siRNA-based knockdown analysis and discovered a critical role of ezrin in NK cell internalization. Impor- tantly, PKA-mediated phosphorylation of ezrin promotes the NK cell internalization process. Our findings suggest a novel regulatory mechanism by which ezrin governs NK cell internalization into tumor cells.

文摘Objective:To describe the Chinese experience of natural orifice transluminal endoscopic surgery(NOTES)in urology.Methods:From December 2008 to May 2017,35 animal experiments and 305 clinical surgeries of NOTES or natural orifices specimen extractions(NOSE)were performed in China.The animal experiments included five kidney biopsies,24 nephrectomies and six partial nephrectomies.The clinical surgeries included 12 transvaginal NOSE(TV-NOSE),266 hybrid transvaginal NOTES(TV-NOTES)and 27 pure TV-NOTES.The TV-NOSE procedure was performed in five transumbilical laparoendoscopic single-site(U-LESS)nephrectomies,four suprapubic-assisted laparoendoscopic single-site surgery(SA-LESS)nephroureterectomies,and three laparoscopic radical cystectomies.The hybrid TV-NOTES procedure included 210 nephrectomies,31 adrenalectomies,eight nephroureterectomies,13 partial nephrectomies,and four heminephrectomies.The pure TV-NOTES procedure included five renal cyst decortications and 22 nephrectomies.Results:A total of 29 animal experiments were successfully performed.One partial nephrectomy was converted to standard laparoscopic surgery.Two kidney biopsies and two nephrectomies were unsuccessful.A total of 297 clinical surgeries were successfully performed.Six patients who underwent hybrid TV-NOTES were converted to open surgery.Two patients who underwent pure TV-NOTES were converted to SA-LESS.There were 22 major complications,16 occurred intraoperatively and six postoperatively.The mean visual analog score(VAS)of 48 h after the operation was 2.5 points in TV-NOSE,2.3 points in hybrid TV-NOTES and 1.7 points in pure TV-NOTES.The mean follow-up of 50.6(3.0-87.0)months showed that all patients were in good condition.The umbilicus scars were nearly invisible in TV-NOSE and hybrid TV-NOTES.The vaginal incision healed well.Conclusions:TV-NOSE and TV-NOTES are feasible,safe,and effective with little injury,low pain,fast recovery,and good cosmetic outcomes in properly selected patients.They are worth consideration for urological clinical practice.

文摘Global climate change is a threat to animals in nearly all biomes and ecosystems, especially for ectotherm whose life activities highly depend on environmental thermal regime. Population genetic diversity which is essential for adaptation to environmental change is a useful index for long-term species survival. In this paper, genetic diversity of eight Phrynocephalus forsythii population which distributed in Tarim Basin, China, were evaluated based on three mtDNA gene and its correlation with environment factors were investigated using RDA. Our result revealed that, the level of genetic diversity of P. forsythii populations was related to its location but there was no significant correlation between genetic distances and geographic distances in P. forsythii. However, we find that mtDNA of P. forsythii was subjected to selection pressure during evolution and population genetic diversity was significantly positively related to variation coefficient of rainfall(VCR) and altitude(AL), while significantly negatively related to longitude(N) and annual average temperature(AAT). Our result supported the previous prediction that excessive ambient heat is a threat to P. forsythii.

文摘Hierarchically porous carbon materials are promising for energy storage,separation and catalysis.It is desirable but fairly challenging to simultaneously create ultrahigh surface areas,large pore volumes and high N contents in these materials.Herein,we demonstrate a facile acid-base enabled in situ molecular foaming and activation strategy for the synthesis of hierarchically macro-/meso-/microporous N-doped carbon foams(HPNCFs).The key design for the synthesis is the selection of histidine(His)and potassium bicarbonate(PBC)to allow the formation of 3D foam structures by in situ foaming,the PBC/His acid-base reaction to enable a molecular mixing and subsequent a uniform chemical activation,and the stable imidazole moiety in His to sustain high N contents after carbonization.The formation mechanism of the HPNCFs is studied in detail.The prepared HPNCFs possess 3D macroporous frameworks with thin well-graphitized carbon walls,ultrahigh surface areas(up to 3200 m^2 g^-1),large pore volumes(up to 2.0 cm^3 g^-1),high micropore volumes(up to 0.67 cm^3 g^-1),narrowly distributed micropores and mesopores and high N contents(up to 14.6 wt%)with pyrrolic N as the predominant N site.The HPNCFs are promising for supercapacitors with high specific capacitances(185-240 F g^-1),good rate capability and excellent stability.They are also excellent for CO2 capture with a high adsorption capacity(~4.13 mmol g^-1),a large isosteric heat of adsorption(26.5 kJ mol^-1)and an excellent CO2/N2 selectivity(~24).

文摘Dracomolphesin A-E(1-5),five 3,4-seco-phenylpropanoids featuring an aromatic ring opened framework,were isolated from the aerial parts of Dracocephalum moldavica.The structures with absolute configurations were determined by spectroscopic methods coupled with Mosher method.Notably,these compounds represented an example of aromatic ring cleavage products of phenylpropanoids.The possible biosynthetic pathway of these compounds was proposed.Compounds 1,2,4 and 5 were demonstrated to be Nrf2 pathway activators.

文摘4-Hydroxyphenylpyruvate dioxygenase(HPPD)is an important target for both drug and pesticide discovery.As a typical Fe(II)-dependent dioxygenase,HPPD catalyzes the complicated transformation of 4-hydroxyphenylpyruvic acid(HPPA)to homogentisic acid(HGA).The binding mode of HPPA in the catalytic pocket of HPPD is a focus of research interests.Recently,we reported the crystal structure of Arabidopsis thaliana HPPD(At HPPD)complexed with HPPA and a cobalt ion,which was supposed to mimic the pre-reactive structure of At HPPD-HPPA-Fe(II).Unexpectedly,the present study shows that the restored At HPPD-HPPA-Fe(II)complex is still nonreactive toward the bound dioxygen.QM/MM and QM calculations reveal that the HPPA resists the electrophilic attacking of the bound dioxygen by the trim of its phenyl ring,and the residue Phe381 plays a key role in orienting the phenyl ring.Kinetic study on the F381 A mutant reveals that the HPPD-HPPA complex observed in the crystal structure should be an intermediate of the substrate transportation instead of the pre-reactive complex.More importantly,the binding mode of the HPPA in this complex is shared with several well-known HPPD inhibitors,suggesting that these inhibitors resist the association of dioxygen(and exert their inhibitory roles)in the same way as the HPPA.The present study provides insights into the inhibition mechanism of HPPD inhibitors.

文摘The gut microbiota has gained attention because of its importance in facilitating host survival and evolution.However,it is unclear whether gut microbial communities are determined by the host(heritable factor) or environment(environmental factor).In this study,we investigated the gut microbial communities and potential functional signatures of two sympatric species distributed along an elevation gradient,the toadheaded lizards Phrynocephalus axillaris and P.forsythii.Our results indicated that at high elevations,the gut microbial communities of P.axillaris and P.forsythii did not significantly differ,and the phylogenetic relationships of gut microbial communities contradicted their hosts.At low altitudes,the two lizards could be distinguished based on their significantly different gut microbial communities.Compared to low-altitude populations,Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis showed that at higher altitudes,energy metabolism,such as carbohydrate,lipid,and amino acids metabolism were higher in both lizards.While a larger number of pathogenic bacteria were found in the lowaltitude population of P.forsythii.This suggests that the convergence of gut microbiota of two lizards at highaltitude stem from environmental factors,as they were exposed to the same environmental stress,whereas the divergence at low-altitude stemmed from heritable factors,as they were exposed to different environmental stresses.These results provide a new perspective regarding whether heritable or environmental factors dominate the gut microbiota during exposure to environmental stress.

文摘Speciation has never been directly observed in nature because it is a lengthy phenomenon.Although rare,ring species are an optimal natural example of speciation and can be identified through the assessment of the geographical conditions of their potential habitat.Phrynocephalus forsythii is endemic to the Tarim Basin,which compnses the Taklamakan Desert and surrounded by mountains on three sides.This study aimed to determine whether P.forsythii had a ring-species-like divergence pattern through the characterization of the genetic features of 17 populations covering the major distribution of this species.Species distribution modelling revealed that P.forsythii had a continuous circular distribution around the Tarim Basin.Gene flow was observed in most adjacent populations except for two terminal populations of the ring,which exhibit the highest differentiation.Genetic distance and geographic distance were significantly correlated,indicating that the observed differentiation resulted from genetic variation gradually accumulating during population dispersion.Although our results do not definitively indicate that P.forsythii is a ring species,our results indicate a ring-shaped diversification This phenomenon elucidates the potential mechanism underlying speciation in the presence of gene flow,providing insight into this evolutionary process.

文摘Objective:The aim of the study was to observe the therapeutic effect of FOLFOX4 regimen plus Fufangchangtai decoction on postoperative colorectal cancers.Methods:Thirty postoperative colorectal cancer patients were allocated into control and experiment groups respectively.Patients in experiment group were given Fufangchangtai decoction combined with FOLFOX4 regimen.Patients in control group were given FOLFOX4 regimen alone.Efficacy was evaluated after 2 periods of treatment.Results:The improvement rate of symptoms were 86.6% in experiment group compared to 53.3% in control group.KPS was stable in experiment group,and decreased in control group.QOL was increased in experiment group,and stable in control group after the treatment.For impact of immunity parameters,there were enhancements of CD3+ and CD4+ in experiment group,while they did not change in control group.In experiment group,WBC reduction was slighter than that of control group.The differences were not remarkable in PLT reduction,alimentary response,and toxicity of liver and kidney and nervous system.Conclusion:The clinical observation showed that Fufangchangtai decoction plus FOLFOX4 regimen could effectively enhance KPS,improve the symptoms,the quality of life and the immunity state,and down-regulate the side effects.In conclusion,Fufangchangtai decoction can decrease the toxicity so as to increase the treatment effect.

文摘Tree species diversity is vital for maintaining ecosystem functions,yet our ability to map the distribution of tree diversity is limited due to difficulties in traditional field-based approaches.Recent developments in spaceborne remote sensing provide unprecedented opportunities to map and monitor tree diversity more efficiently.Here we built partial least squares regression models using the multispectral surface reflectance acquired by Sentinel-2 satellites and the inventory data from 74 subtropical forest plots to predict canopy tree diversity in a national natural reserve in eastern China.In particular,we evaluated the underappreciated roles of the practical definition of forest canopy and phenological variation in predicting tree diversity by testing three different definitions of canopy trees and comparing models built using satellite imagery of different seasons.Our best models explained 42%–63%variations in observed diversities in cross-validation tests,with higher explanation power for diversity indices that are more sensitive to abundant species.The models built using imageries from early spring and late autumn showed consistently better fits than those built using data from other seasons,highlighting the significant role of transitional phenology in remotely sensing plant diversity.Our results suggested that the cumulative diameter(60%–80%)of the biggest trees is a better way to define the canopy layer than using the subjective fixeddiameter-threshold(5–12 cm)or the cumulative basal area(90%–95%)of the biggest trees.Remarkably,these approaches resulted in contrasting diversity maps that call attention to canopy structure in remote sensing of tree diversity.This study demonstrates the potential of mapping and monitoring tree diversity using the Sentinal-2 data in species-rich forests.

文摘An electrochemical immunosensor for sensitive detection of thyroid stimulating hormone(TSH) has been developed by using an inkjet printed microchip and based on a double signal amplification strategy using magnetic beads(MBs), alkaline phosphatase(ALP) and p-aminophenyl phosphate(pAPP) reaction.Differential pulse voltammetry(DPV), cyclic voltammogram(CV) and amperometric i-t curve(i-t) were employed to characterize the immunosensor. High sensitivity and good selectivity were observed. The detection linear range was from 0.01 μIU/mL to 10 μIU/mL, in which the peak currents increased along with the concentration. The detection limit was 0.005 μIU/mL at S/N = 3. The immunosensor was also applied for TSH detection in human serum with recoveries from 98.0% to 101.8% and relative standard deviations from 1.3% to 3.1%, demonstrating potential value in clinical diagnosis.

文摘We propose a new injection scheme that can generate electron beams with simultaneously a few permille energy spread, submillimeter milliradian emittance, and more than a 100 pC charge in laser wakefield accelerators. In this scheme, a relatively loosely focused laser pulse drivesthe plasma wakefield, and a tightly focused laser pulse with similar intensity triggers an interference ring pattern that creates onion-like multisheaths in the plasma wakefield. Owing to the change in wavefront curvature after the focal position of the tightly focused laser, the innermostsheath of the wakefield expands, which slows down the effective phase velocity of the wakefield and triggers injection of plasma electrons.Both quasicylindrical and fully three-dimensional particle-in-cell simulations confirm the generation of beams with the above mentionedproperties.

文摘Doxorubicin(DOx)is the most effective chemotherapeutic for breast cancer,but it is usually associated with severe cardiotoxicity.Further investigation to alleviate its side effects is essential.The present study investigated the mechanism of the cross-organ communication between tumors and the heart and potential intervention targets.Morphological bubble-like protrusions were observed in both adult murine ventricular cardiomyocytes(AMvCs)and human induced pluripotent stem cell-derived cardiomyocytes(hiPSC-CMs)cocultured with breast cancer cells(BCCs),along with elevated expression of pyroptosis-related proteins.Exosomes(EXOs)from DOX-treated BCCs aggravated DOX-induced cardiotoxicity(DOXIC)in an orthotopic mouse model of breast cancer.Blocking miRNAs by knocking down Rab27a or inhibiting the release of EXOs in cancer tissue by Dicer enzyme knockout attenuated this additional injury effect.Exosomal miRNA sequencing revealed that miR-216a-5p is especially upregulated in EXOs from DOX-induced BCCs.Mechanistically,miR-216a-5p was upregulated by enhanced transcription mediated by DOXinduced AMP-dependent transcription factor 3(ATF3)and packaged into EXOs by splicing factor 3b subunit 4(SF3B4)in BCCs.Itchy E3 ubiquitin-protein ligase(ITCH)was identified as a novel downstream target mRNA of miR-216a-5p.ITCH negatively mediated thioredoxin-interacting protein(TXNIP)ubiquitination to activate the NOD-,LRR-and pyrin domain-containing protein 3(NLRP3)inflammasome pathway,ultimately leading to cardiomyocyte pyroptosis.Our findings revealed novel cross-organ pathogenic communication between breast cancer and the heart through the exosomal miR-216a-5p-mediated ITCH/TXNIP/NLRP3 pathway,which drives cardiomyocyte pyroptosis.These findings suggest that targeting myocardial miR-216a-5p or blocking harmful EXOs from breast cancer is a potential therapeutic strategy for alleviating DOXIC.

文摘Introduction:To investigate the prevalence of elevated blood pressure(EBP)and its association with Dietary Approaches to Stop Hypertension(DASH)adherence among Chinese children and adolescents,providing evidence for dietary interventions targeting pediatric hypertension.Methods:Data were derived from the“National Survey on Nutrition and Health of Children Aged 0–18 Years,”a key project under China’s 13th Five-Year Plan for National Science and Technology Infrastructure.DASH scores were calculated using standardized criteria,and multivariable logistic regression models were employed to evaluate associations between EBP and DASH adherence.Results:The prevalence of EBP in 7–17-year-olds was 15.4%.The median DASH score was 24 points,with the highest adherence tertile(T3)exhibiting a lower prevalence of EBP(14.9%)compared to T1(lowest adherence)(16.2%)and T2(moderate adherence)(15.2%).In the fully adjusted model(Model 2),adolescents aged 13–17 years in the T3 DASH adherence group showed reduced risk of EBP[odds ratio(OR)=0.78,95%confidence interval(CI)=0.60,0.98]relative to T1.Conclusion:Higher DASH adherence is associated with reduced EBP risk in adolescents aged 13–17 years,emphasizing the protective role of DASHrecommended foods in promoting cardiovascular health.

文摘Modifying Ir by foreign metals with oxophilicity is a promising strategy to accelerate the hydrogen oxidation kinetics.However,uncontrolled enrichment and oxidative dissolution of metastable oxophilic dopants in conventional Ir-based alloys impair their activity and durability.Here,we address these challenges by atomically dispersing oxophilic Sn sites within Ir clusters to form dilute alloys.The Sn1-Ir pairs,confined within an atomic-scale lattice,prevent excessive*OH coverage caused by oxophilic site enrichment,while also reducing durability loss due to the dissolution of metastable dopants.Our analysis reveals that the Sn1-Ir pairs facilitate electron transfer between Sn1 and adjacent Ir sites,generating electron-rich Ir atoms and electron-poor Sn atoms.This modulation weakens*H and CO adsorption on Ir sites while enhancing OH adsorption on Sn sites.The resulting catalyst shows improved catalytic hydrogen oxidation performance in alkaline media,with mass activities 6.4 and 10.7 times higher than that of Ir/C and Pt/C,respectively.Under CO poisoning conditions,it retains 90.9%of its initial activity,outperforming both Ir/C and Pt/C.This work offers new perspectives on the design of dual-site catalysts for hydrogen oxidation catalysis.
