Van der Waals(vdW)ferroelectric-semiconductor heterojunction provides reconfigurable band alignment based on optical/electrical-assisted polarization switching,which shows great potential to construct artificial visual neural systems.However,the mechanical exfoliation fabrication scheme for proof-of-concept demonstrations and fundamental studies is cumbersome and not scalable for practical application.Here,we present a synthetic strategy for the large-scale and high crystallinity growth of planar/verticalα-In_(2)Se_(3)/MoS_(2)heterojunctions by dynamically tuning the growth temperature.Furthermore,based on theα-In_(2)Se_(3)/MoS_(2)heterostructures,photo-synapse devices are designed and fabricated to simulate visual neural systems functions,including multistate storage,optical logic operation,potentiation and depression,paired-pulse facilitation(PPF),short-term memory(STM),long-term memory(LTM),and Learning-Forgetting-Relearning.By coupling the spatiotemporally relevant optical and electric information,the device can mimic the superior biological visual system’s light adaptation and Pavlovian conditioning.This work provides a strategy for dynamically tuning the orientation of ferroelectric-semiconductor heterojunction stacks and will give impetus to applying all-in-one sensing and memory-computing artificial vision systems. 展开更多

Ganoderma lingzhi is a new species of the prize medicinal mushroom Ganoderma(Agaricomycetes).Using angiotensin I-converting enzyme(ACE)as a target,a tripeptide Ser-Tyr-Pro(SYP)was discovered with preponderant ACE inhibitory activity with an 50%inhibiting concentration(IC_(50))value of 62.50μg/mL attribute to the formed salt bridge and hydrogen bonds between SYP and ACE.SYP even maintained superior bioactivity after intestinal digestion,and exerted no cytotoxicity,but presented incomplete bioavailability in blood of spontaneous hypertensive rats(SHRs).Furthermore,it performed antihypertensive effect in vivo by inhibiting the influx of Ca^(2+)through activating endothelial NO synthase(e NOS)/NO/guanosine 3',5'-cyclic monophosphate(c GMP)pathway,accompanied by attenuating angiotensin II(Ang II)/NADPH oxidase(NOX)/reactive oxygen species(ROS)pathway.This work not only discoverers a novel pharmacological ingredient from medicinal mushroom G.lingzhi for hypertension therapy,but also provides an insight into molecular mechanism of the ACE inhibitory peptide(ACEIP)on lowering blood pressure. 展开更多

Objective To reveal the effects and potential mechanisms by which synaptic vesicle glycoprotein 2A(SV2A)influences the distribution of amyloid precursor protein(APP)in the trans-Golgi network(TGN),endolysosomal system,and cell membranes and to reveal the effects of SV2A on APP amyloid degradation.Methods Colocalization analysis of APP with specific tagged proteins in the TGN,ensolysosomal system,and cell membrane was performed to explore the effects of SV2A on the intracellular transport of APP.APP,β-site amyloid precursor protein cleaving enzyme 1(BACE1)expressions,and APP cleavage products levels were investigated to observe the effects of SV2A on APP amyloidogenic processing.Results APP localization was reduced in the TGN,early endosomes,late endosomes,and lysosomes,whereas it was increased in the recycling endosomes and cell membrane of SV2A-overexpressed neurons.Moreover,Arl5b(ADP-ribosylation factor 5b),a protein responsible for transporting APP from the TGN to early endosomes,was upregulated by SV2A.SV2A overexpression also decreased APP transport from the cell membrane to early endosomes by downregulating APP endocytosis.In addition,products of APP amyloid degradation,including sAPPβ,Aβ1-42,and Aβ1-40,were decreased in SV2A-overexpressed cells.Conclusion These results demonstrated that SV2A promotes APP transport from the TGN to early endosomes by upregulating Arl5b and promoting APP transport from early endosomes to recycling endosomes-cell membrane pathway,which slows APP amyloid degradation. 展开更多

Although the demand of ^(10)B separation has arisen in the 1930s,^(10)B/^(11)B are among the most difficult isotopes to separate due to the extremely similar relative atomic mass.Herein,we report an efficient separation of ^(10)B isotopologue by engineering amino-galloyl synergistic materials via a selective adsorption and isotope exchange reaction,achieving a record-high single-stage separation factor of 1.048 with ^(10)B abundance up to 21.42%.^(11)B MAS NMR results and DFT calculations reveal that the galloyl groups exhibit inherent high affinity for B(OH)4-,forming tetrahedral sp^(3) B-galloyl complexes.The relatively higher ^(10)B–O bond energy of ^(10)B-galloyl complexes facilitates the isotope exchange between11B in B-galloyl complexes and ^(10)B in B(OH)_(3).Flowthrough dynamic separation in fixed-bed demonstrates the feasibility and potential of large-scale deployment of this method in real-world,suggesting a promising avenue for the exploitation of more efficient enrichment of ^(10)B for the sustainable nuclear energy and biomedical research. 展开更多

Endosomal disruption by co-encapsulating gentamicin in lipid nanoparticles for efficient siRNA delivery and cancer therapy

Efficient siRNA delivery is highly desirable for disease treatment.However,the application of conventional nanoparticles is limited by the inability to escape from endo-lysosomes.Herein,we report a strategy using small-molecule drugs to enhance siRNA endo-lysosomal release,addressing this challenge.We encapsulated gentamicin(GM)into the marketed Onpattro■ formulation to establish LNP-siRNA/GM nanoparticles that promote siRNA endo-lysosomal escape through endosomal disruption,mechanistically exhibiting unique functionality and synergistic effects of LNP-siRNA/GM to improve cancer therapy.Besides,GM induced reactive oxygen species(ROS)and phospholipids accumulation in endolysosomes,as well as the physical characteristics of lipid nanoparticles(LNPs)were preserved.We also revealed that GM causes endo-lysosomal swelling and disrupts the endosomal membrane to enable siRNA release,as confirmed by Galectin 3 recruitment and acridine orange release.This approach achieved∼81%mRNA-EGFR silencing,which is more than LNP-siEGFR(∼56.23%)by enhancing siRNA endo-lysosomal escape efficiency.Meanwhile,LNP-siEGFR/GM exhibited significant biological activities in HepG2 cells,driven by the synergistic effects of siEGFR and GM with the VEGF and CXCL12 downregulation of,and ROS and phospholipids upregulation.Furthermore,tumor growth was notably suppressed after intravenous injection of LNP-siEGFR/GM in tumor-bearing nude mice.The combination of EGFR-siRNA and GM could also greatly inhibit angiogenesis,be antiproliferative,and induce tumor cells apoptosis.Therefore,this GM and siRNA co-delivery system would provide an efficient strategy for siRNA endosomal escape,significantly improving knockdown in various LNPs based siRNA delivery systems and efficiently enhancing cancer therapy. 展开更多

When dealing with expensive multiobjective optimization problems,majority of existing surrogate-assisted evolutionary algorithms(SAEAs)generate solutions in decision space and screen candidate solutions mostly by using designed surrogate models.The generated solutions exhibit excessive randomness,which tends to reduce the likelihood of generating good-quality solutions and cause a long evolution to the optima.To improve SAEAs greatly,this work proposes an evolutionary algorithm based on surrogate and inverse surrogate models by 1)Employing a surrogate model in lieu of expensive(true)function evaluations;and 2)Proposing and using an inverse surrogate model to generate new solutions.By using the same training data but with its inputs and outputs being reversed,the latter is simple to train.It is then used to generate new vectors in objective space,which are mapped into decision space to obtain their corresponding solutions.Using a particular example,this work shows its advantages over existing SAEAs.The results of comparing it with state-of-the-art algorithms on expensive optimization problems show that it is highly competitive in both solution performance and efficiency. 展开更多

为了研究阿霉素胶束中药物的存在形式及其与胶束释药行为的关系,以不同类型的普朗尼克胶束为载体材料制备了3种阿霉素胶束。普朗尼克载药胶束首先由阿霉素盐酸盐和脱氧胆酸钠通过静电相互作用形成复合物,然后通过亲疏水性作用封装到普... 为了研究阿霉素胶束中药物的存在形式及其与胶束释药行为的关系,以不同类型的普朗尼克胶束为载体材料制备了3种阿霉素胶束。普朗尼克载药胶束首先由阿霉素盐酸盐和脱氧胆酸钠通过静电相互作用形成复合物,然后通过亲疏水性作用封装到普朗尼克胶束的疏水段形成。以载药量和包封率为评价指标,动态光散射法测定其粒径大小及其分布、透析法测定载药胶束的体外释放行为。结果表明,制备的普朗尼克F127,F128,F68胶束的载药量分别为(0.15±0.11)%,(0.16±0.07)%和(0.17±0.1)%,包封率分别为(3.03±0.24)%,(3.11±0.15)%和(3.36±0.02)%。载药胶束的粒径分布均匀。在690 min内累计释放率均能达到70%以上,且无显著性差异。本文成功制备了均一、稳定的普朗尼克载药胶束,研究了不同普朗尼克载药胶束的药物释放行为,为药物输送系统提供理论依据和参考。 展开更多

Previous studies have indicated that electrical stimulation of the cerebellar fastigial nucleus in rats may reduce brain infarct size, increase the expression of Ku70 in cerebral ischemia/ reperfusion area, and decrease the number of apoptotic neurons. However, the anti-apoptotic mechanism of Ku70 remains unclear. In this study, fastigial nucleus stimulation was given to rats 24, 48, and 72 hours before cerebral ischemia/reperfusion injury. Results from the electrical stim- ulation group revealed that rats exhibited a reduction in brain infarct size, a significant increase in the expression of KuT0 in cerebral ischemia/reperfusion regions, and a decreased number of terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL)-positive cells. Double immunofluorescence staining revealed no co-localization of Ku70 with TUNEL-positive cells. However, Ku70 partly co-localized with Bax protein in the cytoplasm of rats with cerebral ischemia/reperfusion injury. These findings suggest an involvement of Ku70 with Bax in the cy- toplasm of rats exposed to electrical stimulation of the cerebellar fastigial nucleus, and may thus provide an understanding into the anti-apoptotic activity of KuT0 in cerebral ischemia/reperfu- sion injury. 展开更多

目的利用美国FDA不良事件报告系统(FAERS)数据库探索玛巴洛沙韦在儿童人群中的相关不良事件,为儿童安全用药提供参考。方法检索FAERS数据库2018年第一季度至2024年第一季度儿童人群中以baloxavir marboxil或Xofluza®为主要怀疑药... 目的利用美国FDA不良事件报告系统(FAERS)数据库探索玛巴洛沙韦在儿童人群中的相关不良事件,为儿童安全用药提供参考。方法检索FAERS数据库2018年第一季度至2024年第一季度儿童人群中以baloxavir marboxil或Xofluza®为主要怀疑药物的不良事件报告,通过报告比值比法和贝叶斯置信度递进神经网络法进行数据分析。结果共筛选出以玛巴洛沙韦为主要怀疑药物的儿童不良事件报告424例,其中男性(42.69%)略多于女性(38.68%),5~11岁儿童占总数的71.93%,主要上报国家是美国(73.82%),严重不良事件占31.60%,主要上报时间集中在2019年(27.83%)和2020年(58.25%)。检出安全信号涉及系统器官分类7个,按信号频数分布,儿童人群中玛巴洛沙韦相关的不良事件依次为异常行为、谵妄、速发严重过敏反应等;按信号强度依次为热性谵妄、多形性红斑、谵妄等。还检出玛巴洛沙韦药品说明书未记载的不良事件信号,包括热性谵妄、脱水、意识状态改变和面部水肿。结论玛巴洛沙韦在儿童中存在精神类疾病和过敏反应等不良事件。因此,针对儿童患者使用玛巴洛沙韦时尤其注意精神系统症状如异常行为和热性谵妄的发生,保障儿童用药安全。 展开更多

Grain size is an important determinant of grain weight and yield in rice.Although several genes related to grain size have been identified,natural variations in these genes that affect grain size are poorly characterized.Here,we describe the grain length QTL GL10,encoding MADS56,which positively regulates grain length and grain weight.A natural allelic variation of NIL-gl10,containing an~1.0-kb deletion in the first exon that abolishes its transcription,results in shorter grain length,lower grain weight and delayed flowering in gl10 plants.The knockout of GL10 in the HJX74 background leads to grain phenotypes similar to that of NIL-gl10,while overexpression of GL10 results in increased grain length and weight and earlier heading date.GL10 regulates grain length by promoting greater longitudinal cell growth in the grain glume.Additionally,GL10 participates in the regulation of gibberellic acid(GA)signaling pathway genes in young panicle tissues.Analysis of GL10 haplotypes shows obvious divergence between the japonica and indica lineages.Our findings reveal an allelic variation of GL10 that may explain differences in grain length among modern cultivars and could be used to breed rice varieties with optimized grain shape. 展开更多

Previous studies at home and abroad have mainly focused on single dam-break, and little attention has been paid so far to the dam-break of cascade reservoirs. Multi-source flooding, which can lead to three-dimensional turbulent phenomena and superposition effects, is the main difference between the dam-break of cascade and single reservoirs. Detailed descriptions of the coupled numerical simulation of multi-source flooding have little been reported, and the initial wet riverbed is rarely considered in current models. Therefore, in this paper, a method based on the three-dimensional (Formula presented.) turbulence model coupled with the volume-of-fluid method is proposed to simulate the dam-break flooding of cascade reservoirs. The upstream river, reservoir, and downstream river are connected by the internal boundary method, and the initial conditions, including river flow and reservoir water, are determined according to the results of the numerical simulation. Coupled numerical simulation of different dam-break flooding is then achieved. The present work solves the challenges presented by the enhancement and superposition of natural river flow, upstream flooding, and downstream flooding. The complex relationships between indicators and water conditions cause fuzzy and gray uncertainties in evaluation of water quality. Compared to conventional single-factor evaluation methods, the combination evaluation method can consider these two uncertainties to produce more objective and reasonable evaluation results. In this paper, we propose a combination evaluation method with two main parts:(1) the use of fuzzy comprehensive evaluation and gray correlation analysis as submodels with which to consider the fuzzy and gray uncertainties and(2) the establishment of a combination model based on minimum bias squares. In addition, using this method, we evaluate the water quality of a ditch in a typical rice–wheat system of Yixing city in the Taihu Lake Basin during three rainfall events. The results show that the ditch water quality is not good and we found the chemical oxygen demand to be the key indicator that affects water quality most significantly. The proposed combination evaluation method is more accurate and practical than single-factor evaluation methods in that it considers the uncertainties of fuzziness and grayness. 展开更多

Channel engineering stability with underground goafs is a complex three-dimensional problem, especially when considering channel leakage, and is influenced by a number of processes, such as seepage, fluid structure interaction (FSI), modeling, and selection of geological mechanical parameters. In this study, stability finite element analysis by one-way FSI was performed by establishing an integrated 3D engineering geological model. The extended Fourier amplitude sensitivity test was used to quantitatively assess the first-order and total sensitivities of the engineering model to critical geological mechanical parameters. Results illustrate that the channel engineering deformation is under a reasonable range and the elastic modulus is the highest total sensitivity parameter for the channel tilt and curvature at 0.7395 and 0.7525, respectively. Moreover, the most observable coupling effects for the curvature and horizontal strain are cohesion (0.1933) and density (0.7410), respectively. © 2017, Tianjin University and Springer-Verlag GmbH Germany. 展开更多

[ Method] Using RAPD molecular marker technique, eight effective primers were screened from 35 RAPD random primers for amplification. UPGMA clus- ter analysis was performed using NTSYSpc. ver. 2.02 software. [ Result] A total of 86 loci were amplified using the screened primers, including 84 polymorphic loci, indicating an average polymorphic percentage (PPB) of 97.67%. The similarity coefficients of 30 populations of R. laevigata Michx. ranged from O. 11 to O. 58, and these populations were clustered into groups A, B and C. [ Conclusion ] R. laevigata Michx. exhibits multiple genetic polymorphism. R. laevigata Michx. populations from Guizhou Province have closer genetic relationships. 展开更多

Dynamic manipulation of enzymatic activity is a challenging task for applications in chemical and pharmaceutical industries due to the difficult modification and variable conformation of various enzymes.Here, we report a new strategy for reversible dynamic modulation of enzymatic activity by near-infrared light-induced photothermal conversion based on polyphenol-functionalized liquid metal nanodroplets(LM). The metal-phenolic nanocoating not only provides colloidal stability of LM nanodroplets but also generates nanointerfaces for the assembly of various enzymes on the LM nanodroplets. Upon near infrared(NIR) irradiation, the localized microenvironmental heating through photothermal effect of the LM nanodroplets allows tailoring the enzymatic activity without affecting the bulk temperature. A library of functional enzymes, including proteinase K, glucoamylase, glucose oxidase, and Bst DNA polymerase, is integrated to perform a reversible control and enhanced activities even after five times of cycles, demonstrating great potential in bacterial fermentation, bacteriostasis, and target gene amplification. 展开更多
