Stroke,a major cerebrovascular disease,has high morbidity and mortality.Effective methods to reduce the risk and improve the prognosis are lacking.Currently,uric acid(UA)is associated with the pathological mechanism,p...Stroke,a major cerebrovascular disease,has high morbidity and mortality.Effective methods to reduce the risk and improve the prognosis are lacking.Currently,uric acid(UA)is associated with the pathological mechanism,prognosis,and therapy of stroke.UA plays pro/anti-oxidative and pro-inflammatory roles in vivo.The specific role of UA in stroke,which may have both neuroprotective and damaging effects,remains unclear.There is a U-shaped association between serum uric acid(SUA)levels and ischemic stroke(IS).UA therapy provides neuroprotection during reperfusion therapy for acute ischemic stroke(AIS).Urate-lowering therapy(ULT)plays a protective role in IS with hyperuricemia or gout.SUA levels are associated with the cerebrovascular injury mechanism,risk,and outcomes of hemorrhagic stroke.In this review,we summarize the current research on the role of UA in stroke,providing potential targets for its prediction and treatment.展开更多
Objective:To investigate the relationship between Jiaotai pill(JTP),its main component berberine(BBR),and the serotonin(5-HT)system in regulating islet hormone secretion and alleviating pancreatic b-cell dysfunction d...Objective:To investigate the relationship between Jiaotai pill(JTP),its main component berberine(BBR),and the serotonin(5-HT)system in regulating islet hormone secretion and alleviating pancreatic b-cell dysfunction during type 2 diabetes mellitus(T2DM)progression.Methods:T2DM rat model was established using a high-fat diet and streptozotocin injection.JTP,BBR,and Metformin were intragastrically administered for 35 days.The analyzed indices included blood glucose,blood lipids,islet hormones,and proteins related to 5-HT synthesis,secretion,and transport.Additionally,an in vitro model of glucose injury in islet cells was established to study the effects of JTP and BBR on islet hormone secretion following tryptophan hydroxylase 1(TPH1)inhibition.Results:JTP and BBR significantly improved blood glucose and lipid levels and islet morphology in T2DM rats.Both models exhibited reduced islet 5-HT levels and impaired islet hormone secretion.However,the administration of JTP and BBR reversed these effects.Furthermore,JTP and BBR upregulated the expression of TPH1(P=.0194,P=.0413)transglutaminase 2(TGase2;P=.0492,P=.0349),serotonin transporter(SERT,P=.0090),and 5-hydroxytryptamine 1F receptor(5-HT1FR)in the islet 5-HT pathway(P=.0194).In the cell model,the regulatory effects of JTP and BBR on islet hormone levels were significantly weakened after TPH1 inhibition(P=.001),suggesting that JTP and BBR influence islet hormone secretion through the pancreatic 5-HT system.Conclusion:The islet 5-HT system is correlated with islet hormone secretion dysfunction in T2DM.JTP and BBR can improve islet hormone secretion by activating the TPH1/TGase2/SERT/5-HT1FR pathway in the islet 5-HT system in T2DM rats.展开更多

To quickly detect and count the number of bayberry trees,this paper improves the YOLO-v4 model and proposes an optimal YOLO-v4 method for detecting bayberry trees based on UAV images.We used the Leaky_ReLU activation ...To quickly detect and count the number of bayberry trees,this paper improves the YOLO-v4 model and proposes an optimal YOLO-v4 method for detecting bayberry trees based on UAV images.We used the Leaky_ReLU activation function to accelerate the model extraction speed and used the DIoU NMS to retain the most accurate prediction boxes.In order to increase the recall rate of the object detection and construct the optimal YOLO-v4 model,the K-Means clustering method was embedded into DIoU NMS.We trained the model using UAV images of bayberry trees,it was determined that the optimal YOLO-v4 model threshold was 0.25,which had the best extraction effect.The optimal YOLO-v4 model had a detection accuracy of up to 97.78%and a recall rate of up to 98.16%on the dataset.The optimal YOLO-v4 model was compared with YOLO-v4,YOLO-v4 tiny,the YOLO-v3 model,and the Faster R-CNN model.With guaranteed accuracy,the recall rate was higher,up to 97.45%,and the detection of bayberry trees was better in different contexts.The result shows that the optimal YOLO-v4 model can accurately achieve the rapid detection and statistics of the number of bayberry trees in large-area orchards.展开更多
A combination of the viscoelastic properties of hyaluronic acid(HA)and the elastic properties of star shaped 8-arm poly(ethylene glycol)(8-arm PEG)was used to design in-situ forming hydrogels.Hydrogels were prepared b...A combination of the viscoelastic properties of hyaluronic acid(HA)and the elastic properties of star shaped 8-arm poly(ethylene glycol)(8-arm PEG)was used to design in-situ forming hydrogels.Hydrogels were prepared by the enzymatic crosslinking of a partially tyramine modified 8-arm PEG and a tyramine conjugated HA using horseradish peroxidase in the presence of hydrogen peroxide.Hydrogels of the homopolymer conjugates and mixtures thereof were rapidly formed within seconds under physiological conditions at low polymer and enzyme concentrations.Elastic hydrogels with high gel content(≥95%)and high storage moduli(up to 22.4 kPa)were obtained.An in vitro study in the presence of hyaluronidase(100 U/mL)revealed that with increasing PEG content the degradation time of the hybrid hydrogels increased up to several weeks,whereas hydrogels composed of only hyaluronic acid degraded within 2 weeks.Human mesenchymal stem cells(hMSCs)incorporated in the hybrid hydrogels remained viable as shown by a PrestoBlue and a live-dead assay,confirming the biocompatibility of the constructs.The production of an extracellular matrix by re-differentiation of encapsulated human chondrocytes was followed over a period of 28 days.Gene expression indicated that these highly elastic hydrogels induced an enhanced production of collagen type II.At low PEG-TA/HA-TA ratios a higher expression of SOX 9 and ACAN was observed.These results indicate that by modulating the ratio of PEG/HA,injectable hydrogels can be prepared applicable as scaffolds for tissue regeneration applications.展开更多
