Characterizing a new rat model of chronic pain after spine surgery

Chronic pain after spine surgery(CPSS)is a complex disorder characterized by multifactorial pathogenesis that occurs in 8%–40%of patients undergoing lumbar spine surgery.We aimed to develop a rat model that mimics clinical CPSS conditions by taking two sequential surgical procedures.Step 1:A plastic rod was inserted into the left L5 intervertebral foramen to produce a steady compression on the dorsal root ganglion(DRG)and the spinal nerve,a common cause of low back pain(LBP).Step 2:The rod was removed after 7 days when rats exhibited mechanical and heat hypersensitivity in the ipsilateral hindpaw,followed by a full L5 laminectomy to mimic spine decompression surgery in LBP patients.The retention of the rod induced a prolonged LBP-like behavior but was quickly resolved after rod removal without laminectomy.However,rats that received laminectomy after rod removal developed heightened mechanical and heat sensitivity in the hindpaw,impaired gait,and reduced spontaneous exploration activity,indicating CPSS.Patch clamp recording revealed a significant augmentation in the intrinsic excitability of smalldiameter DRG neurons in CPSS rats.Administration of Dermorphin[D-Arg2,Lys4](1–4)amide(DALDA,5mg/kg,i.p.),a peripherally acting mu-opioid receptor(MOR)-preferred agonist,attenuated pain hypersensitivity,capsaicin-induced[Ca^(2+)]i rising and the increased intrinsic excitability of DRG neurons from CPSS rats.Our findings suggest that this new model,which mirrors the nature of CPSS developed in patients,may be useful for future studies of the underlying mechanisms. 展开更多

The electrocatalytic oxidation of glycerol toward formic acid is one of the most promising pathways for transformation and utilization of glycerol.Herein,a series of well-defined Ni_(n)(SR)_(2n) nanoclusters(n=4,5,6;denoted as Ni NCs)were prepared for the electrocatalytic glycerol oxidation toward formic acid,in which Ni_(6)-PET-50CV afforded the most excellent electrocatalytic performance with a high formic acid selectivity of 93% and a high glycerol conversion of 86%.This was attributed to the lowest charge transfer impedance and the most rapid reaction kinetics.Combined electrochemical measurements and X-ray absorption fine structure measurements revealed that the structures of Ni NCs remained intact after CV scanning pretreatment and electrocatalysis via forming the Ni–O bond.Additionally,the kinetic studies and in-situ Fourier transformed infrared suggested a sequential oxidation mechanism,in which the main reaction steps of glycerol→glyceraldehyde→glyceric acid were very rapid to produce a high selectivity of formic acid even though the low glycerol conversion.This work presents an opportunity to study Ni NCs for the efficient electrocatalytic oxidation of biomass-derived polyhydroxyl platform molecules to produce value-added carboxylic acids. 展开更多

The Impacts of Major Chronic Diseases on Changes in Healthy Life Expectancy among Older Adults in China from 2011 to 2020

Objectives This study aimed to quantify the impact of major chronic diseases on changes in healthy life expectancy(HLE)from 2011 to 2020 in China using an age-specific disability weights(DW)estimation method.Methods HLE at age 60(HLE_(60))was used as the indicator of HLE in China.Cause-specific mortality rates were obtained from the cause-of-death database of the National Health Commission.Selfreported disease and disability status were derived from the China Health and Retirement Longitudinal Study.A total of 55,861 participants were included for DW estimation.Rates of disability,which was assessed using the Activities of Daily Living questionnaires,were estimated using data from 5,465 participants in 2011 and 9,910 participants in 2020.Age-specific DWs were calculated using a Bayesian logistic regression model.Changes in HLE_(60) were decomposed into mortality and disability effects by cause,based on the estimated DWs.Results HLE_(60) in China increased by 0.83 years from 2011 to 2020.Ischemic heart disease(IHD)contributed the most to the decline in HLE_(60),remaining the leading cause of reduction in terms of mortality effects.Diabetes showed the greatest impact on HLE_(60) due to disability,followed by stroke.The largest sex disparities in HLE_(60) were associated with disability from arthritis.Conclusion HLE_(60) in China improved from 2011 to 2020 and IHD remained the leading contributor to its decline,particularly through increased mortality.Disabilities related to diabetes,stroke,and arthritis had significant negative impacts.These findings highlight the need to strengthen integrated chronic disease prevention and rehabilitation services at community health centers. 展开更多

Objective:To describe the contemporary trends in total,inpatient,and outpatient expenditure on major subtypes of cancer in different classifications of hospitals in China's Mainland.Methods:Home page of Inpatient Medical Records(HIMRs)and Hospital Annual Reports(HARs)were used to estimate hospital care expenditure on cancer.Inpatient payments and their share of cancer were calculated with the top-down method.Kriging spatial interpolation methods were used at the county level and summed at the province level.Outpatient expenditure was estimated with inpatient expenditure and the ratios of outpatient to inpatient payments in specialized cancer hospitals,stratified by province.Total expenditure on cancer was the sum of both payments.Log-linear regression was applied to estimate annual percentage change(APC)of expenditure.Results:Total expenses for cancer of Chinese residents reached up to 304.84 billion Chinese Yuan(CNY)in2017,accounting for 5.8%of the total health expenses(THE).After adjusting for consumer price index(CPI),medical expenses for cancer have increased from 63.30 billion CNY in 2008 to 249.56 billion CNY in 2017[APC:15.2%,95%confidence interval(95%CI):13.4%-17.0%].The APC was slightly higher than THE around 2013,while was lower after 2013.During 2008-2017,the ratio of inpatient to outpatient costs for cancer decreased from4.3:1 to 3.8:1.The inpatient payments for cancer mainly happened in grade 3 general hospitals,East China,and among lung,colorectal,and stomach cancer;while the fastest increase was found in West China,and among thyroid,prostate,and colorectal cancer.Conclusions:During 2008-2017,the rapid growth trend of medical expenses for cancer has been effectively controlled with the continuous deepening of medical reform and improvements of residents’health care.More attention should be paid to potential increases of medical costs caused by technological progress and demand release.Socialized and multi-channel insurance financing modes should be explored in the future. 展开更多

Myocardial ischemia(MI)causes somatic referred pain and sympathetic hyperactivity,and the role of sensory inputs from referred areas in cardiac function and sympathetic hyperactivity remain unclear.Here,in a rat model,we showed that MI not only led to referred mechanical hypersensitivity on the forelimbs and upper back,but also elicited sympathetic sprouting in the skin of the referred area and C8–T6 dorsal root ganglia,and increased cardiac sympathetic tone,indicating sympathetic-sensory coupling.Moreover,intensifying referred hyperalgesic inputs with noxious mechanical,thermal,and electro-stimulation(ES)of the forearm augmented sympathetic hyperactivity and regulated cardiac function,whereas deafferentation of the left brachial plexus diminished sympathoexcitation.Intradermal injection of the α_(2) adrenoceptor(α_(2)AR)antagonist yohimbine and agonist dexmedetomidine in the forearm attenuated the cardiac adjustment by ES.Overall,these findings suggest that sensory inputs from the referred pain area contribute to cardiac functional adjustment via peripheral α_(2)AR-mediated sympathetic-sensory coupling. 展开更多

In Wireless Sensor Network(WSN),because battery and energy supply are constraints,sleep scheduling is always needed to save energy while maintaining connectivity for packet delivery.Traditional schemes have to ensure high duty cycling to ensure enough percentage of active nodes and then derogate the energy efficiency.This paper proposes an RFID based non-preemptive random sleep scheduling scheme with stable low duty cycle.It employs delay tolerant network routing protocol to tackle the frequent disconnections.A low-power RFID based non-preemptive wakeup signal is used to confirm the availability of next-hop before sending packet.It eliminates energy consumption of repeated retransmission of the delayed packets.Moreover,the received wakeup signal is postponed to take effect until the sleep period is finished,and the waken node then responds to the sending node to start the packet delivery.The scheme can keep stable duty cycle and then ensure energy saving effect compared with other sleeping scheduling methods. 展开更多

A smart contract honeypot is a special type of smart contract.This type of contract seems to have obvious vulnerabilities in contract design.If a user transfers a certain amount of funds to the contract,then the user can withdraw the funds in the contract.However,once users try to take advantage of this seemingly obvious vulnerability,they will fall into a real trap.Consequently,the user’s investment in the contract cannot be retrieved.The honeypot induces other accounts to launch funds,which seriously threatens the security of property on the blockchain.Detection methods for honeypots are available.However,studying the manner by which to defend existing honeypots is insufficient to fight against honeypots.The new honeypots that may appear in the future from the perspective of an attacker must also be predicted.Therefore,we propose a type of adversarial honeypot.The code and behavioral features of honeypots are obtained through a comparative analysis of the 158,568 nonhoneypots and 352 honeypots.To build an adversarial honeypot,we try to separately hide these features and make the honeypot bypass the existing detection technology.We construct 18 instances on the basis of the proposed adversarial honeypot and use an open-source honeypot detection tool to detect these instances.The experimental result shows that the proposed honeypot can bypass the detection tool with a 100%ratio.Therefore,this type of honeypot should be given attention,and defensive measures should be proposed as soon as possible. 展开更多

BACKGROUND Gastric cancer,characterized by a multifactorial etiology and high heterogeneity,continues to confound researchers in terms of its pathogenesis.Curcumin,a natural anticancer agent,exhibits therapeutic promise in gastric cancer.Its effects include promoting cell apoptosis,curtailing tumor angiogenesis,and enhancing sensitivity to radiation and chemotherapy.Long noncoding RNAs(lncRNAs)have garnered significant attention as biomarkers for early screening,diagnosis,treatment,and drug response because of their remarkable specificity and sensitivity.Recent investigations have revealed an association between aberrant lncRNA expression and early diagnosis,clinical staging,metastasis,drug sensitivity,and prognosis in gastric cancer.A profound understanding of the intricate mechanisms through which lncRNAs influence gastric cancer develop-ment can provide novel insights for precision treatment and tailored management of patients with gastric cancer.This study aimed to unravel the potential of curcumin in suppressing the malignant behavior of gastric cancer cells by upregu-lating specific lncRNAs and modulating gastric cancer onset and progression.AIM To identify lncRNAs associated with curcumin treatment and investigate the role of lncRNA AC022424.2 in the effects of curcumin on gastric cancer cell apoptosis,proliferation,and invasion.Furthermore,these findings were validated in clinical samples.METHODS The study employed CCK-8 assays to assess the impact of curcumin on gastric cancer cell proliferation,flow cytometry to investigate its effects on apoptosis,and scratch and Transwell assays to evaluate its influence on the migration and invasion of BGC-823 and MGC-803 cells.Western blotting was used to gauge changes in the protein expression levels of CDK6,CDK4,Bax,Bcl-2,caspase-3,P65,and the PI3K/Akt/mTOR pathway in gastric cancer cell lines after curcumin treatment.Differential expression of lncRNAs before and after curcumin treatment was assessed using lncRNA sequencing and validated using quantitative reverse transcription polymerase chain reaction(qRT-PCR)in BGC-823 and MGC-803 cells.AC022424.2-1 knockdown BGC-823 and MGC-803 cells were generated to scrutinize the impact of lncRNA AC022424.2 on apoptosis,proliferation,migration,and invasion of gastric cancer cells.Western blotting was performed to ascertain changes in the expression of proteins implicated in the PI3K/Akt/mTOR and NF-κB signaling pathways.RT-PCR was employed to measure lncRNA AC022424.2 expression in clinical gastric cancer tissues and to correlate its expression with clinical pathological characteristics.RESULTS Curcumin induced apoptosis and hindered proliferation,migration,and invasion of gastric cancer cells in a dose-and time-dependent manner.LncRNA AC022424.2 was upregulated after curcumin treatment,and its knockdown enhanced cancer cell aggressiveness.LncRNA AC022424.2 may have affected cancer cells via the PI3K/Akt/mTOR and NF-κB signaling pathways.LncRNA AC022424.2 downregulation was correlated with lymph node metastasis,making it a potential diagnostic and prognostic marker.CONCLUSION Curcumin has potential anticancer effects on gastric cancer cells by regulating lncRNA AC022424.2.This lncRNA plays a significant role in cancer cell behavior and may have clinical implications in diagnosis and prognosis evaluation.The results of this study enhance our understanding of gastric cancer development and precision treatment. 展开更多

Deciphering the neuronal response to injury in the spinal cord is essential for exploring treatment strategies for spinal cord injury(SCI).However,this subject has been neglected in part because appropriate tools are lacking.Emerging in vivo imaging and labeling methods offer great potential for observing dynamic neural processes in the central nervous system in conditions of health and disease.This review first discusses in vivo imaging of the mouse spinal cord with a focus on the latest imaging techniques,and then analyzes the dynamic biological response of spinal cord sensory and motor neurons to SCI.We then summarize and compare the techniques behind these studies and clarify the advantages of in vivo imaging compared with traditional neuroscience examinations.Finally,we identify the challenges and possible solutions for spinal cord neuron imaging. 展开更多

Digital assets have boomed over the past few years with the emergence of Non-fungible Tokens(NFTs).To be specific,the total trading volume of digital assets reached an astounding$55.5 billion in 2022.Nevertheless,numerous security concerns have been raised by the rapid expansion of the NFT ecosystem.NFT holders are exposed to a plethora of scams and traps,putting their digital assets at risk of being lost.However,academic research on NFT security is scarce,and the security issues have aroused rare attention.In this study,the NFT ecological process is comprehensively explored.This process falls into five different stages encompassing the entire lifecycle of NFTs.Subsequently,the security issues regarding the respective stage are elaborated and analyzed in depth.A matrix model is proposed as a novel contribution to the categorization of NFT security issues.Diverse data are collected from social networks,the Ethereum blockchain,and NFT markets to substantiate our claims regarding the severity of security concerns in the NFT ecosystem.From this comprehensive dataset,nine key NFT security issues are identified from the matrix model and then subjected to qualitative and quantitative analysis.This study aims to shed light on the severity of NFT ecosystem security issues.The findings stress the need for increased attention and proactive measures to safeguard the NFT ecosystem. 展开更多

Non-alcoholic fatty liver disease(NAFLD)has become the world’s most common chronic liver disease and is considered one of the main causes of occult liver cirrhosis.To date,major guidelines in China and abroad have not recommended any drugs with precise efficacy and safety for NAFLD,especially for NAFLD-related liver fibrosis or cirrhosis,which lack effective drugs.Traditional Chinese medicine has significant curative effects and advantages in preventing and treating NAFLD.We retrospectively analyzed a case of liver cirrhosis due to NAFLD that was treated solely with traditional Chinese medicine for nearly one year at the Diagnosis and Treatment Center of Advantage Specific Diseases for Fatty Liver in our hospital to provide a reference for the prevention and treatment of non-alcoholic liver fibrosis and liver cirrhosis by traditional Chinese medicine. 展开更多

Analyzing the impact of radio interference(RI)variation during foul weather conditions is an area that has received limited study.This paper provides a statistical analysis of RI measurements obtained from a long-term observation station close to the world’s first commercially operating 1000 kV UHV AC double-circuit transmission line in China.During six months of observations,the impact of RI was studied on the line during fog,drizzle,and light snow and rain.It was found that RI increases linearly with the natural logarithm of the precipitation intensity.The Levenberg-Marquardt algorithm(LMA)is employed to fit the RI value with the precipitation intensity.The reasonable distribution of RI in different foul weather is verified by one-sample K-S test.This test is seen as beneficial for further RI prediction based on statistical weather mode. 展开更多

The high current of hybrid DC circuit breaker(HCB)in the process of breaking generates strong transient magnetic field(MF),which may interfere with the normal operation of driver control units(DCUs).Therefore,the analysis for transient MF disturbance is of great significance.Due to the large space scale of HCB,with the large number of power electronic devices,the numerical calculation method has the disadvantages of large consumption of resources and slow calculation speed in solving the transient MF inside the HCB.In this study,the current generation mechanism of a 500 kV HCB is analysed,and the equivalent current path is obtained by considering the skin effect.Combined with the temporal and spatial distribution of transient current,the multi-process transient equivalent model of transient MF calculation is established,and the model has the advantages of high precision and high speed.Then,the breaking experiment of HCB is carried out,and the transient MF near the DCU is measured.The experiment results verify the validity of the model.Furthermore,the disturbance of transient MF at the DCU of the transfer branch during breaking 25 kA current is analysed. 展开更多

UHVDC converter valves during operation may experience overvoltage,which come from the AC or DC systems to which they are connected.Therefore,building an equivalent circuit model(ECM)for the converter valve to analyze the interlayer transient voltage distribution characteristics has important engineering significance for safe and reasonable voltage equalization methods and improving the stability of the DC system.This paper proposes a two-port equivalent circuit model for ±1100 kV converter valve based on the structure of the valve and parameter extraction methods presented.In terms of lumped parameters,integrated ECMs for valve layers are built through impedance-frequency characteristic analysis;in terms of parasitic capacitance parameters,port equivalent parasitic capacitance parameters are obtained by terminal capacitance method and iterative equivalence methods proposed in this paper.By combining integrated ECMs of valve layers and port equivalent parasitic capacitances,the two-port ECM is obtained.Simulations are carried out to test the effectiveness of the twoport ECM.Using the ECM,the voltage transmission characteristics and their influencing factors are analyzed,depending on which corresponding voltage equalization method is proposed in this paper,and the effect of this method is verified through simulation. 展开更多

In traditional hybrid DC circuit breakers(HCBs)schemes,complex equipment is usually required to ensure current commutation from mechanical to semiconductor switches,which cause not only additional construction costs and volume but also additional losses and maintenance work.Different from this condition,a low-loss and compact HCB scheme that does not use separate current commutation equipment is proposed in this study.By rationally using the charge and discharge of the snubber capacitor,the self-commutated semiconductor switch(SCS)module can integrate both the current commutation and shutdown functions,thereby greatly reducing cost and volume.The working process is discussed and analyzed in detail,and then a 10-kA prototype is developed and tested,which verifies the feasibility and effectiveness of the proposed scheme.Index Terms-HVDC circuit breakers,hybrid DC circuit breakers(HCBs),self-commutated semiconductor switch(SCS). 展开更多

The current interruption test based on an LC resonance circuit for hybrid DC circuit breakers(HVDC CBs)is widely employed to characterize the current interruption capability of CBs.In order to ensure a high-fidelity replica of the fault current in a high voltage application,this paper first proposes an equivalent model of the test circuit,where not only parasitic resistances but also the threshold voltages and on-state resistances of various semiconductor devices are considered.Moreover,the analytical formula of the test current is derived by including the working principle of the HVDC CB.Secondly,the parameter extraction method,which combines finite element analysis and measurements by an impedance analyzer,is given in this paper.The extracted result implies that,in current interruption transients,equivalent resistances of 500 kV CB are as large as 535 mS,which have a significant influence on current waveforms.Thirdly,the 34 kV/25 kA current interruption test for the 500 kV CB is conducted.The measured results are proved to be consistent with the analytical results obtained from the proposed model,and the relative error is less than 2%. 展开更多

High-frequency voltage oscillation occurs at the initial stage of DC short-circuit fault in the modular multilevel converter(MMC)-based high-voltage direct-current systems,which directly threatens the reliable operation of the secondary equipment in DC circuit breakers(DCCBs).Therefore,this paper focusses on the voltage oscillation characteristic at DCCB terminals,considered the stray parameters of connecting lines,the parasitic parameters of main equipment and the impedance of grounding grid,established equivalent circuit models of MMC-HVDC converter station at the initial stage of pole-topole and pole-to-ground fault,and obtained the analytical formula for the voltage oscillation frequency.Then,the comparison with the DC short-circuit test data of�200 kV MMC-HVDC converter station verified the validity of the models and revealed the mechanism of voltage oscillation.Finally,the influence of the parameters of the converter station on the oscillation characteristics was discussed.It was proposed to install the DCCB at the MMC valve's terminal near the DC side,and the oscillation frequency is about 10–60 kHz,which is more than 5 times lower than that at the DC reactor's terminal near the DC pole bus,and the disturbance of oscillation to DCCB was alleviated. 展开更多
