目的微小RNA相关单核苷酸多态性(microRNA-related single nucleotide polymorphism,miR-SNP)与肿瘤的发生发展关系密切。本研究对中国人群中miR-SNP与结直肠癌(colorectal cancer,CRC)发生发展及化疗疗效的关系进行研究。方法首先,选...目的微小RNA相关单核苷酸多态性(microRNA-related single nucleotide polymorphism,miR-SNP)与肿瘤的发生发展关系密切。本研究对中国人群中miR-SNP与结直肠癌(colorectal cancer,CRC)发生发展及化疗疗效的关系进行研究。方法首先,选择较小等位基因频率>10%的miR-SNP;采用单碱基延伸结合基质辅助激光解吸离子化-飞行时间-质谱法(MALDI-TOF-MS)检测2013-05-10-2015-10-22无锡市第四人民医院收治的504例CRC患者和714名健康对照的miR-SNPs基因型。统计分析miR-SNP基因型与CRC临床病理参数及化疗疗效的关联性。结果rs61992671基因型在病例组和健康对照组中的分布差异有统计学意义,χ2=6.44,P=0.026;A/G型个体患CRC的风险为3.2%,比A/A型(5.5%)更低,OR=1.77。经性别和年龄调整后,rs12402181A等位基因携带者的肿瘤体积比G/G型更大,χ2=3.85,OR=1.67,P=0.047,矫正OR=1.68,矫正P=0.045;rs12402181杂合子患结肠癌的风险为54.4%,比纯合子(45.0%)高,χ2=3.95,P=0.047,矫正P=0.018。rs67106263基因型与CRC的肿瘤分化程度显著相关,χ2=8.46,P=0.020,矫正P=0.026;与G/G型相比,G/A型(OR=2.35,矫正OR=2.30)、A/A型(OR=2.78,矫正OR=2.69)和A等位基因携带者(OR=2.38,矫正OR=2.33)的肿瘤分化程度均明显更差。此外,在接受基于5-氟尿嘧啶化疗的CRC患者中,rs61992671基因型与化疗效果有关联,χ2=2.75,P=0.041,矫正P=0.046。结论 rs61992671与CRC的发生和5-氟尿嘧啶化疗疗效有关联;rs12402181和rs67106263与CRC的发展有关联。展开更多

Its expression and secretion are estrogen-regulated and are crucial for oocyte maturation. An in vitro xenoestro- gen screening model was established by measuring Vtg in- duction in cultured primary hepatocytes from crucian carp. Vtg production was detected by biotin-avidin sandwich ELISA method while Vtg and cytochrome P4501A1 (CYP1A1) mRNA induction were measured by semi- quan- titative PCR-primer dropping technique. Vtg and Vtg mRNA were dose-dependently induced by diethylstilbestrol (DES, 0.2—200 ng/mL) in hepatocytes of crucian carp. Co-treatment of the DES-induced hepatocytes with either 2,3,7,8-TCDD (TCDD, 0.1—4 pg/mL) or benzo[a]pyrene (B[a]P, 5—1000 ng/mL) resulted in a reduction of Vtg pro- duction and an increment of CYP1A1 mRNA expression both in a dose dependent manner, indicating the anti-estro- genic effects of the compounds. However, at lower tested concentrations, TCDD (0.1, 0.2 pg/mL), B[a]P (5 ng/mL) seemed to have a potentiating effect on Vtg expression and secretion, although by their own these compounds had no observable estrogenic effect on Vtg induction. Tamoxifen (a selective estrogen receptor modulators, 1 nmol/L—1 μmol/L), and β-naphtho-flavone (β-NF, an aryl hydrocarbon receptor inducing compounds, 2.5—1000 ng/mL) also were employed to study the possible interactions in DES-induced Vtg ex- pression. In co-treatment of the DES-induced hepatocytes with β-NF or tamoxifen, the decrease in Vtg production did parallel induction of CYP1A1 for β-NF, but tamoxifen inhib- ited Vtg induction did not parallel induced CYP1A1 expres- sion in all test concentrations. On the contrary, it was found that in co-treatment of the TCDD-induced hepatocytes with DES, TCDD induced CYP1A1 mRNA production was inhib- ited by DES also. To investigate the effect of dsRNA on silkworm cells, we trans- fected three kinds of synthetic dsRNAs of 435 bp(Ap1), 300 bp(Ap2) and 399 bp(AH) in length against the various regions of BmNPV’s DNA polymerase gene and DNA helicase gene, which are indispensable for viral replication in silkworm cells by TransMessengerTM transfection Reagent. Results indicated that in the experiment where silkworm cells were infected with wild-strain BmNPV of the three dsRNAs, Ap2 and AH can effectively suppress the replication of virus, but Ap1 had no effect on the inhibition of viral replication. Ap2 and AH can reduce the infective titer of BmNPV with a peak change of approximately 3—4 logs on day 4 post-infection. The results of reverse transcript polymerase chain reaction (RT-PCR) and DNA dot blotting also indicated that the ex- pression level of the two target genes and the quantity of viral DNA both distinctly decreased under the influence of Ap2 or AH. Furthermore, using fluorescence microscopy we analyzed the distribution patterns of dsRNA. Using fibroblast deriving from C57/BL6 ear tissue as nuclear donor, we produced cloned embryos by transferring the fibroblast nuclei into enucleated KM mouse oocytes (sin-gle nuclear transfer, SNT), transferring pronuclei from the SNT embryos into enucleated KM zygotes (nuclear into zy-gote, NZ), and 2-cell blastomere nuclei from SNT embryos into enucleated KM mouse oocytes (nuclear into oocytes, NO); tetraploid embryos (tetraploid embryos, TE) were ob-tained by fusing two blastomeres, one is from the SNT cloned embryos, and the other from normal 2-cell KM mouse em-bryos. In group SNT, the cloned embryos could not develop beyond 8-cell stage and the rate of 8-cell stage is only 0.3%; in group NO, the reconstructed embryos could develop to morula stage, the rate of 8-cell stage was significantly greater than that of SNT group (P < 0.05); in group NZ, the devel-opment rate was further improved, and the reconstructed embryos could develop into blastocyst stage, the rate of blastocyst was 1.9%; in group TE, as high as 62.3% of the reconstructed embryos could develop into blastocyst.
