Electroreduction of CO_(2) into CH_(4) under acidic conditions is a promising strategy for CO_(2) utilization,which allows for high CO_(2) conversion efficiency.However,the selectivity of CH_(4) is low because the hyd...Electroreduction of CO_(2) into CH_(4) under acidic conditions is a promising strategy for CO_(2) utilization,which allows for high CO_(2) conversion efficiency.However,the selectivity of CH_(4) is low because the hydrogen evolution reaction is enhanced under acidic conditions.Here,we report that the CO_(2) can be efficiently reduced into CH_(4) over a Cu catalyst by modifying with a glutamic acid molecule under acidic conditions.The CH_(4) Faradaic efficiency can reach 62.9% with a current density of 450 mA cm^(-2).Meanwhile,a single-pass carbon efficiency of 48.1% toward CH_(4) is achieved.Experiments revealed that the glutamic acid molecule can enhance the concentration of Kt on the surface of Cu,which can suppress the HER and promote CO_(2) reduction,resulting in high selectivity of CH_(4) under acidic conditions.展开更多
目的探讨孕晚期超声血流参数联合外周血生长阻滞特异性蛋白6(growth arrest specific protein 6,Gas6)预测胎儿生长受限(fetal growth restriction,FGR)的临床价值。方法选择2022年1—12月在医院建卡的孕晚期孕妇285例为研究对象,所有...目的探讨孕晚期超声血流参数联合外周血生长阻滞特异性蛋白6(growth arrest specific protein 6,Gas6)预测胎儿生长受限(fetal growth restriction,FGR)的临床价值。方法选择2022年1—12月在医院建卡的孕晚期孕妇285例为研究对象,所有孕妇均进行超声检查与外周血Gas6检查,随访至妊娠结束,根据是否发生FGR分为发生组与未发生组,对比2组超声血流参数[胎儿脐动脉及子宫动脉的收缩期与舒张期流速比(systolic/diastolic velocity ratio,S/D)、阻力指数(resistance index,RI)、搏动指数(pulsatility index,PI)]、外周血Gas6水平及临床资料,Logistic回归分析孕晚期孕妇发生FGR的影响因素。绘制受试者工作特征(receiver operating characteristic,ROC)曲线,以曲线下面积(area under the curve,AUC)评估预测价值。结果随访至妊娠结束,285例孕晚期孕妇发生FGR24例,发生率为8.42%,剩余261例均未发生FGR。发生组剖宫产例数占比高于未发生组(P<0.05),发生组新生儿体重低于未发生组(P<0.05)。发生组胎儿脐动脉的S/D(2.72±0.38)、RI(0.85±0.17)、PI(1.08±0.32)高于未发生组[(2.10±0.30)、(0.59±0.13)、0.75±0.24](P<0.05),发生组子宫动脉的S/D(3.41±0.48)、RI(0.60±0.15)、PI(1.22±0.27)高于未发生组[(2.33±0.39)、(0.37±0.08)、0.83±0.19](P<0.05)。发生组外周血Gas6水平(12.67±2.41)ng/L高于未发生组(9.25±1.78)ng/L(P<0.05)。子宫动脉PI(OR=3.518,95%CI:1.547~8.004)、胎儿脐动脉RI(OR=3.762,95%CI:1.654~8.558)、胎儿脐动脉S/D(OR=2.907,95%CI:1.278~6.612)、外周血Gas6(OR=3.043,95%CI:1.338~6.923)为孕晚期孕妇发生FGR的影响因素(P<0.05)。子宫动脉PI、胎儿脐动脉RI、胎儿脐动脉S/D、外周血Gas6及四者联合预测孕晚期孕妇发生FGR的AUC值分别为0.792、0.803、0.823、0.815、0.899(P<0.05),且四者联合的AUC值更高(P<0.05)。结论孕晚期超声血流参数(子宫动脉PI、胎儿脐动脉RI、胎儿脐动脉S/D)、外周血Gas6在预测FGR中具有重要价值,且四者联合具有更高的预测价值。展开更多
AIM: To detect the expression of DPC4 in malignant and non-malignant specimens of human pancreas,and observe the inhibition of retroviral pLXSN containing DPC4 on pancreatic carcinoma cells in vitro.METHODS: The expre...AIM: To detect the expression of DPC4 in malignant and non-malignant specimens of human pancreas,and observe the inhibition of retroviral pLXSN containing DPC4 on pancreatic carcinoma cells in vitro.METHODS: The expression of DPC4 was determined in 40 pancreatic adenocarcinoma and 36 non-malignant pancreatic specimens by reverse-transcriptase polymerase chain reaction (RT-PCR) and immunohisto-chemistry.Furthermore,we constructed retroviral vectors containing DPC4,which then infected the pancreatic carcinoma cell line BxPC-3.Cell growth in vitro after being infected was observed,and the vascular endothelial growth factor (VEGF) mRNA level in the daughter cells was determined by semi-quantitative PCR assay.RESULTS: The RT-PCR assay showed a positive rate of DPC4 mRNA in 100% (36/36) of normal specimens,compared to 40% (16/40) in adenocarcinoma specimens.The regional and intense positive cases of DPC4 expression in adenocarcinoma detected by immunohistochemistry were 10 and four,whereas it was all positive expression in normal tissues.There was a significant difference of DPC4 expression between them.The stable expression of DPC4 in the pancreatic carcinoma cells BxPC-3 could be resumed by retroviral vector pLXSN transfection,and could inhibit cell growth in vitro.Rather,DPC4 could decrease VEGF mRNA transcription levels.CONCLUSION: The deletion of DPC4 expression in pancreatic carcinoma suggests that loss of DPC4 may be involved in the development of pancreatic carcinoma.The retroviral vector pLXSN containing DPC4 can inhibit the proliferation of pancreatic carcinoma cells,and down-regulate the level of VEGF.展开更多
The approval of using monoclonal antibodies as a targeted therapy in the management of patients with B cell lymphoma has led to new treatment options for this group of patients. Production ofmonoclonal antibodies by t...The approval of using monoclonal antibodies as a targeted therapy in the management of patients with B cell lymphoma has led to new treatment options for this group of patients. Production ofmonoclonal antibodies by the traditional hybridoma technology is costly, and the resulting murine antibodies often have the disadvantage of triggering human anti-mouse antibody (HAMA) response. Therefore recombinant Fab antibodies generated by the phage display technology can be a suitable alternative in managing B cell lymphoma. In this study, we extracted total RNA from spleen cells of BALB/c mice immunized with human B lymphoma cells, and used RT-PCR to amplify cDNAs coding for the κ light chains and Fd fragments of heavy chains. After appropriate restriction digests, these cDNA fragments were successively inserted into the phagemid vector pComb3H-SS to construct an immunized Fab phage display library. The diversity of the constructed library was approximately 1.94× 10^7. Following five rounds of biopanning, soluble Fab antibodies were produced from positive clones identified by ELISA. From eight positive clones, FabC06, FabC21, FabC43 and FabC59 were selected for sequence analysis. At the level of amino acid sequences, the variable heavy domains (VH) and variable light domains (VL) were found to share 88-92% and 89-94% homology with sequences coded by the corresponding murine germline genes respectively. Furthermore, reactivity with membrane proteins of the B cell lymphoma was demonstrated by immunohistochemistry and western blotting. These immunized Fab antibodies may provide a valuable tool for further study of B cell lymphoma and could also contribute to the improvement of disease therapy.展开更多

Method: Patients' nursing paths for ESD were established based on a systematic...Objective: To investigate the effect of the nursing path on postoperative complications after endoscopic submucosal dissection (ESD). Method: Patients' nursing paths for ESD were established based on a systematic analysis of relevant literature, previous clinical procedures and consultation with chief physicians from a gastroenterology department. Forty patients were divided into the observation or control group and were assigned to follow the nursing path or the conventional path after an ESD, respectively. Next, the incidence of postoperative complications of these two groups was measured, primarily assessing the occurrences of bleeding, perforation, infection, and anxiety. Results: The incidence of complications after ESD in the observation group was remarkably lower than that of the control group (P 〈 0.05). Conclusion: The nursing path for ESD has a positive role in reducing certain frequent complications that occur after ESD, particularly infection and anxiety; in addition, the nursing path optimizes nursing care in patients with gastrointestinal stromal tumor.展开更多

The loci of 60% of the patients could be positioned using a combined positron emission tomography/CT imaging modality. Hyper- and hypometabolism foci were examined as part of this study. Patients who had abnormal metabolism in positron emission tomography/CT imaging were divided into intermittent-phase group and the seizure-phase group. The intermittent-phase group was further divided into a single-focus group and a multiple-foci group according to the number of seizure foci detected by imaging. Following gamma knife treatment, seizure frequency was significantly lower in the intermittent-phase group and the seizure-phase group. Wieser's classification reached Grade I or II in nearly 40% of patients. Seizure frequency was significantly lower following treatment, but Wieser's classification score was significantly higher in the seizure-phase group compared with the intermittent-phase group. Seizure frequency was significantly lower following treatment in the single-focus group, but Wieser's classification score was significantly higher in the single-focus group as compared with the multiple-foci group.展开更多

Chlamydia trachomatis outer membrane protein 2 (Ctomp2) is a major immunogen in chlamydial infections and a highly genus-conserved structural protein of all Chlamydia species. To purify the protein and to prepare mo...Chlamydia trachomatis outer membrane protein 2 (Ctomp2) is a major immunogen in chlamydial infections and a highly genus-conserved structural protein of all Chlamydia species. To purify the protein and to prepare monoclonal antibodies (mAbs) against it, the recombinant protein was induced by IPTG, which was confirmed by SDS-PAGE and purified by means of a Ni^2+ -charged resin colunm. The denatured protein was refolded in the GSH-GSSH buffer gradually and identified by Westem blotting. Then the BALB/c mice were immunized with the recombinant protein to prepare the mAb against Ctomp2. The obtained mAbs were characterized. Genital specimens were tested with indirect ELISA mostly made of the mAb and cell culture in 84 patients with genital symptoms. The results showed that high-level expression of the recombinant protein was achieved, which existed as inclusion body and amounted to 38 % of total bacterium protein. A mAb against Ctomp2 was obtained. It belongs to IgG 2b. The titers were as high as 1:40 000. The Western blotting showed that the mAb only reacted with the recombinant protein. It had no crossing reactions against E. coli, N. gonorhoea, M. hominis, U. urealyticum and M. penetrans. It had high specifity. In comparison with gold standard test-ceil culture, the sensitivities, specificities, positive predictive values and negative predictive values of indirect ELISA were 95.24%, 100%, 100% and 98.44%, respectively. The above-mentioned research work contributed not only to the further study of the structure and function of this protein, but also to the establishment of the method for its clinical application, for it had not been reported before.展开更多

It had no crossing reactions against E. coli, N. gonorhoea, M. hominis, U. urealyticum and M. penetrans. It had high specifity. In comparison with gold standard test-ceil culture, the sensitivities, specificities, positive predictive values and negative predictive values of indirect ELISA were 95.24%, 100%, 100% and 98.44%, respectively. The above-mentioned research work contributed not only to the further study of the structure and function of this protein, but also to the establishment of the method for its clinical application, for it had not been reported before.
