Li-Fraumeni syndrome(LFS),a rare autosomal-dominant inheritance condition,is associated with a family cancer history as well as pathogenic/likely-pathogenic TP53 germline variants(P/LP TP53 GV).The current clinical me...Li-Fraumeni syndrome(LFS),a rare autosomal-dominant inheritance condition,is associated with a family cancer history as well as pathogenic/likely-pathogenic TP53 germline variants(P/LP TP53 GV).The current clinical methods for detecting LFS are limited.Here,we retrospectively investigate P/LP TP53 GV among Chinese cancer patients by next-generation sequencing and evaluate its relationship with a family cancer history.A total of 270 out of 19,226 cancer patients have TP53 GV,including 53 patients with P/LP TP53 GV.Patients with P/LP TP53 GV are mainly found in male with glioma,lung cancer or sarcoma.The median age of diagnosis for P/LP TP53 GV patients is significantly lower than that of non-P/LP TP53 GV patients(31-years vs.53-years;P<0.01).One LFS patient and 3 Li-Fraumeni-like syndrome(LFL)patients are among the 26 followed-up P/LP TP53 GV patients.Among 25 types of P/LP TP53 GV,the highest variant frequencies occurred at codon 175 and 248.p.M237 I,p.R158 H,p.C238 Y and p.C275 R,are firstly identified among the Chinese LFS/LFL patients.This study reports the(P/LP)TP53 GV characteristics of Chinese pan-cancer patients.These findings suggest analyzing the P/LP TP53 GV in cancer patients is an effective strategy for identifying cancer predisposition syndrome.展开更多

Chinese hamster lung fibroblast (V79) cells transfected with a human aquaporin-4-mCherry fusion protein gene were used to detect anti-aquaporin-4 antibody in neuromyelitis optica patient sera by immunofluorescence. Anti-aquaporin-4 autoantibody was stably detected by immunofluorescence in neuromyelitis optica patient sera exclusively. The sensitivity of the assay for neuromyelitis optica was 90% and the specificity for neuromyelitis optica was 100%. The anti-aquaporin-4 antibody titers in sera were tested with serial dilutions until the signal disappeared. A positive correlation was detected between Expanded Disability Status Scale scores and serum anti-aquaporin-4 antibody titers. The anti-aquaporin-4 antibody assay is highly sensitive and specific in the sera of Chinese neuromyelitis optica patients. Detection of aquaporin-4 autoantibody is important for the diagnosis and treatment of neuromyelitis optica.展开更多

Transport stress is commonly suffered by animals with gastrointestinal dysfunction a common symptom.Currently,the mechanisms of transport stress-induced intestine impairment are largely unknown.The aim of this study w...Transport stress is commonly suffered by animals with gastrointestinal dysfunction a common symptom.Currently,the mechanisms of transport stress-induced intestine impairment are largely unknown.The aim of this study was to investigate the effects of transport stress on the expression of neuronal nitric oxide synthase(nNOS)and the distribution of nNOS-positive neurons of the intestines in rats and to explore the neuroendocrine mechanism of transport stress.In this study,Sprague Dawley rats(n=6)were subjected on a constant temperature shaker for 1(S1d)or 3 d(S3d).Rats exhibited increased serum glucose and diminished total number of leukocytes,in which lymphocytes level was also decreased in the S1d group(P<0.05).Also,normal intestinal morphology was disrupted in the S1d rats,and the thickness of muscle layers was decreased in duodenum,jejunum and colon of S3d rats.In addition,it was found that nNOS expression,as well as the number of nNOSpositive neurons in the myenteric plexus were downregulated in duodenum,jejunum and colon of S3d rats compared with that of unstressed rats(P<0.05).These data reveals that transport stress induced intestinal damage and uncovers potential action mechanisms that nNOS-positive neurons and nNOS expression might be involved in modulating this process.展开更多
