韭菜子等补肾中药促进雷帕霉素诱导的急性退化胸腺再生

目的:探讨以韭菜子为代表的多种补肾中药对小鼠胸腺急性退化后再生过程的影响。方法:8周龄BALB/c雄性小鼠48只,随机分为正常对照组、模型对照组、阴性对照组、阳性对照组、蛇床子组、补骨脂组、覆盆子组和韭菜子组,每组6只。除正常对照... 目的:探讨以韭菜子为代表的多种补肾中药对小鼠胸腺急性退化后再生过程的影响。方法:8周龄BALB/c雄性小鼠48只,随机分为正常对照组、模型对照组、阴性对照组、阳性对照组、蛇床子组、补骨脂组、覆盆子组和韭菜子组,每组6只。除正常对照组外,各组小鼠接受雷帕霉素(1 mg·kg^(-1)·d^(-1))连续腹腔注射5 d并饥饿处理14 h诱导胸腺急性退化。造模成功后,阴性对照组腹腔注射等量等渗氯化钠溶液,阳性对照组腹腔注射二甲双胍(300 mg·kg^(-1)·d^(-1)),其他各组分别灌胃给予蛇床子(0.78 g·kg^(-1)·d^(-1))、补骨脂(0.39 g·kg^(-1)·d^(-1))、覆盆子(0.78 g·kg^(-1)·d^(-1))、韭菜子(0.39 g·kg^(-1)·d^(-1))醇提物,每天1次,持续给药5 d。模型对照组在造模完成后、其余各组在不同药物处理后进行生物学动态观察,末次给药2 h后用抓力测定仪进行抓力测定;苏木精-伊红(HE)染色观察胸腺病理变化;多重免疫荧光法观察胸腺上皮细胞结构及分布;流式细胞术分析胸腺及外周血T细胞亚群比例;定量聚合酶链反应(PCR)检测小鼠脾脏单个核细胞基因组DNA中T细胞受体切除环(TREC)水平评估胸腺输出功能;定量逆转录PCR检测胸腺组织中胸腺衰老相关及功能相关基因的表达水平;免疫组织化学法验证其中关键因子细胞周期依赖性蛋白激酶抑制因子1A(p21)和肿瘤蛋白53(p53)的表达水平。结果:雷帕霉素可诱导小鼠胸腺萎缩,四肢抓力显著降低(P<0.01)。与阴性对照组比较,补骨脂组、覆盆子组及韭菜子组小鼠四肢抓力均增强(均P<0.05),且各给药组小鼠脾脏TREC水平均降低(均P<0.05),其中韭菜子组的胸腺功能及组织结构恢复程度最接近正常状态。进一步研究发现,与阴性对照组比较,韭菜子组胸腺CD4单阳性T细胞(CD3^(+)TCR-β+CD4^(+)CD8^(-))亚群比例增加(P<0.01),外周血CD3^(+)CD28^(+)T细胞及CD3^(+)CD8^(+)CD28^(+)T细胞比例增加(均P<0.01),且韭菜子组胸腺细胞中IL-1α、IL-6、p21、p53的信使RNA表达均减少(均P<0.05),免疫组织化学结果进一步验证了p21及p53蛋白表达减少。此外,韭菜子可增强正常小鼠的四肢抓力(P<0.01),抑制胸腺输出,并改变T细胞亚群分布,而对胸腺质量及Foxn1、SIRT1、p21、CXCL2和PTMα表达无显著影响。结论:韭菜子等补肾中药均能加速雷帕霉素诱导的小鼠胸腺急性退化后的再生过程,以韭菜子功能最显著,提示在胸腺受损或退化的病理状态下,其具有较强的促进胸腺再生和功能恢复的作用。 展开更多

Objective:Angelica sinensis,a traditional Chinese herbal medicine,boasts rich history and diverse pharmacological effects,influencing the hematopoietic system,circulatory system,and immune system.This study conducted a bibliometric analysis of angelica sinensis literature over the past 15 years to identify research hotspots and gaps.Methods:Utilizing the Web of Science Core Collection(WoSCC),data was analyzed through VOSviewer and Excel,with polynomial regression assessing annual publication trends.Through co-citation analysis,keyword co-occurrence and bibliographic coupling,this study uncovered distinct knowledge structures and dynamics within agelica research,offering profound understanding of current research priorities and trends.Results:The study revealed a significant annual increase in publications on angelica sinensis.Over the past 15 years,1,588 articles by 7,960 authors from 53 countries and 1,355 organizations have been published.China demonstrated notable productivity and influence in this field.Angelica polysaccharide and Z-ligustilide emerged as the most studied active ingredients.Angelica sinensis demonstrated protective effects against apoptosis and oxidative stress-induced cell damage,with its anti-inflammatory properties becoming a focal point of recent research.The classical prescription“Danggui Buxue decoction”was extensively studied for its hematopoietic,anti-tumor effects,and ability to stabilize human iron balance and ameliorate depression.Furthermore,research on angelica sinensis has spanned multiple disciplinary areas,including pharmacology,medicinal chemistry,complementary medicine and plant sciences.Conclusions:As a pivotal gynecological drug,angelica sinensis has been the focus of extensive research endeavors over the past 15 years,yielding comprehensive fundamental studies and diverse clinical applications in hematopoiesis,immunity,and other related fields. 展开更多
