A rapid and reproducible method to develop transgenic plants with enhanced transformation efficiency using Agrobacterium has been developed for the elite indica rice variety BPT 5204. Different rice calli aged from 3 to 30 d were co-cultivated with pre-incubated Agrobacterium suspension culture (LBA4404: pSB1, pCAMBIA1301) and incubated in dark for 3 d. Based on the transient GUS gene expression analysis, 6-day-old young calli showed high transformation frequency followed by 21-day-old ones. Thus, both 6-and 21-day-old calli were used for assessing the stable transformation efficiency. It was observed that the 6-day-old young transformed calli showed about 2-fold higher regeneration frequency when compared with 21-day-old calli. The transformation efficiency was enhanced for young calli to 5.9% compared with 0.8% of the 21-day-old calli. Molecular and genetic analysis of transgenic plants (To) revealed the presence of 1-2 copies of T-DNA integration in transformants and it follows Mendalian ratio in T1 transgenic plants. From the present study, it was concluded that the development of transgenic rice plants in less duration with high regeneration and transformation efficiency was achieved in BPT 5204 by using 6-day-old young calli as explants. 展开更多

Genotype × environmental interactions for analyzing adaptability and stability in different clones of Dalbergia sissoo Roxb.

In total 36 superior clones of Dalbergia sissoo Roxb., screened from 300 selections conducted in natural and growing range of India and Nepal, were multiplied using single nodal cuttings and estab- lished to evaluate genotypexenvironmental interactions for adaptability and stability at the age of 30 months in three geographical locations in the state of Punjab, India. Clone 124 had maximum adaptability and stability (bi = 1.04) to perform exceedingly well over the locations. Clones 36 and 1 were stable with mean regression coefficient of 0.84 and 1.22, respectively. Nonetheless, clone 4 1 performed exceedingly well for all the characters to attain maximum population mean, and the perform- ance varied substantially across the locations. Therefore, clone 41 was considered as productive but non-adaptive clone. Though some of the clones were sensitive to sites, 14 clones for height, 16 for collar diameter, 12 for DBH and 7 for volume were relatively un-sensitive with higher regression coefficient. Nonetheless, clone 124 was the most Stable with average bi value of 1.04 and productive, which could play an important role in future breeding and commercial deployment of stable and produc- tive planting stock of Dalbergia sissoo. 展开更多

Objective:To screen antimicrobial potence of some pipefish species collected from Tuticorin coastal environment.Methods:Antimicrobial activity of pipefishes in methanol extract was investigated against 10 bacterial and 10 fungal human pathogenic strains.Results:Among the tested strains,in Centriscus scutatus,pipefish showed maximum zone of inhibition against Vibrio cholerae(8 mm)and minimum in the sample of Hippichthys cyanospilos against Klebseilla pneumoniae(2 mm).In positive control,maximum zone of inhibition was recorded in Vibrio cholerae(9 mm)and minimum in Klebseilla pneumoniae,and Salmonella paratyphi(5 mm).Chemical investigation indicated the presence of peptides as evidenced by ninhydrin positive spots on thin layer chromatography and presence of peptide.In SDS PAGE,in Centriscus scutatus,four bands were detected in the gel that represented the presence of proteins in the range nearly 25.8-75 kDa.In Hippichthys cyanospilos,five bands were detected in the gel that represented the presence of proteins in the range nearly 20.5-78 kDa.The result of FT-IR spectrum revealed that the pipe fishes extracts compriseed to have peptide derivatives as their predominant chemical groups.Conclusions:It can be conclude that this present investigation suggests the tested pipe fishes will be a potential source of natural bioactive compounds. 展开更多
