Heat stress hinders the growth and productivity of sweetpotato plants,predominantly through oxidative damage to cellular membranes.Therefore,the development of efficient approaches for mitigating heat-related impairme...Heat stress hinders the growth and productivity of sweetpotato plants,predominantly through oxidative damage to cellular membranes.Therefore,the development of efficient approaches for mitigating heat-related impairments is essential for the long-term production of sweetpotatoes.Melatonin has been recognised for its capacity to assist plants in dealing with abiotic stress conditions.This research aimed to investigate how different doses of exogenous melatonin influence heat damage in sweetpotato plants.Heat stress drastically affected shoot and root fresh weight by 31.8 and 44.5%,respectively.This reduction resulted in oxidative stress characterised by increased formation of hydrogen peroxide(H_(2)O_(2))by 804.4%,superoxide ion(O_(2)^(·-))by 211.5%and malondialdehyde(MDA)by 234.2%.Heat stress also reduced chlorophyll concentration,photosystemⅡefficiency(F_v/F_m)by 15.3%and gaseous exchange.However,pre-treatment with 100μmol L^(-1)melatonin increased growth and reduced oxidative damage to sweetpotato plants under heat stress.In particular,melatonin decreased H_(2)O_(2),O_(2)^(·-)and MDA by 64.8%,42.7%and 38.2%,respectively.Melatonin also mitigated the decline in chlorophyll levels and improved stomatal traits,gaseous exchange and F_(v)/F_(m)(13%).Results suggested that the favorable outcomes of melatonin treatment can be associated with elevated antioxidant enzyme activity and an increase in non-enzymatic antioxidants and osmo-protectants.Overall,these findings indicate that exogenous melatonin can improve heat stress tolerance in sweetpotatoes.This stu dy will assist re searchers in further investigating how melatonin makes sweetpotatoes more resistant to heat stress.展开更多
BACKGROUND Hepatic ischaemia-reperfusion injury(HIRI)is an unavoidable process in liver transplantation,where apoptosis plays a critical role.Human umbilical cord mesenchymal stem cell-derived exosomes(hucMSC-exos),wh...BACKGROUND Hepatic ischaemia-reperfusion injury(HIRI)is an unavoidable process in liver transplantation,where apoptosis plays a critical role.Human umbilical cord mesenchymal stem cell-derived exosomes(hucMSC-exos),which constitute a cellfree therapeutic approach,have garnered extensive attention in alleviating HIRI.However,the potential of hucMSC-exos in mitigating apoptosis and their underlying mechanisms remain largely unknown.AIM To investigate the effects of hucMSC-exos on apoptosis after HIRI and explore the underlying mechanisms.METHODS The therapeutic effects of hucMSC-exos on HIRI and hypoxia/reoxygenation injury in L02 cells were investigated.RNA sequencing was used to detect differentially expressed genes in L02 cells after hucMSC-exo treatment,and the expression of apoptosis markers in L02 cells was analyzed.MicroRNA(miRNA)sequencing was performed to analyse the miRNA expression profiles of hucMSCexos and L02 cells after hucMSC-exo treatment.Through a miRNA-mRNA integrated analysis,candidate miRNAs and their regulated target genes were identified.We subsequently studied the roles of these candidate miRNAs in mouse HIRI and L02 cell hypoxia/reoxygenation injury.RESULTSFluorescence confocal microscopy revealed that hucMSC-exos effectively homed to the liver and were taken up byhepatocytes, likely due to the presence of anti-very late antigen-4 and anti-lymphocyte function-associated antigen-1 on the surface of hucMSC-exos. HucMSC-exos alleviate hepatocyte damage by inhibiting apoptosis. Specifically,let-7i-5p within hucMSC-exos inhibited the expression of the factor-related apoptosis ligand protein in L02 cells,leading to the upregulation of B-cell lymphoma-2 and the downregulation of B-cell lymphoma-2-associated Xprotein and cysteinyl aspartate specific proteinase-3, thereby inhibiting L02 cell apoptosis and enhancing cellproliferation activity. The overexpression of let-7i-5p effectively enhanced the antiapoptotic effects of hucMSC-exosboth in vitro and in vivo.CONCLUSIONOur findings indicate that hucMSC-exos alleviate HIRI by inhibiting apoptosis. We demonstrated that hucMSCexostarget apoptosis in L02 cells and mediate the let-7i-5p/factor-related apoptosis ligand pathway, therebyameliorating HIRI. This study provides new insights into the role of hucMSC-exos in hepatocyte apoptosis andhighlights the potential of hucMSC-exos as a therapeutic strategy for HIRI.展开更多
Conductive biomaterials based on conductive polymers,carbon nanomaterials,or conductive inorganic nanomaterials demonstrate great potential in wound healing and skin tissue engineering,owing to the similar conductivit...Conductive biomaterials based on conductive polymers,carbon nanomaterials,or conductive inorganic nanomaterials demonstrate great potential in wound healing and skin tissue engineering,owing to the similar conductivity to human skin,good antioxidant and antibacterial activities,electrically controlled drug delivery,and photothermal effect.However,a review highlights the design and application of conductive biomaterials for wound healing and skin tissue engineering is lacking.In this review,the design and fabrication methods of conductive biomaterials with various structural forms including film,nanofiber,membrane,hydrogel,sponge,foam,and acellular dermal matrix for applications in wound healing and skin tissue engineering and the corresponding mechanism in promoting the healing process were summarized.The approaches that conductive biomaterials realize their great value in healing wounds via three main strategies(electrotherapy,wound dressing,and wound assessment)were reviewed.The application of conductive biomaterials as wound dressing when facing different wounds including acute wound and chronic wound(infected wound and diabetic wound)and for wound monitoring is discussed in detail.The challenges and perspectives in designing and developing multifunctional conductive biomaterials are proposed as well.展开更多
This paper proposes a new distributed formation flight protocol for unmanned aerial vehicles(UAVs)to perform coordinated circular tracking around a set of circles on a target sphere.Different from the previous results...This paper proposes a new distributed formation flight protocol for unmanned aerial vehicles(UAVs)to perform coordinated circular tracking around a set of circles on a target sphere.Different from the previous results limited in bidirectional networks and disturbance-free motions,this paper handles the circular formation flight control problem with both directed network and spatiotemporal disturbance with the knowledge of its upper bound.Distinguishing from the design of a common Lyapunov fiunction for bidirectional cases,we separately design the control for the circular tracking subsystem and the formation keeping subsystem with the circular tracking error as input.Then the whole control system is regarded as a cascade connection of these two subsystems,which is proved to be stable by input-tostate stability(ISS)theory.For the purpose of encountering the external disturbance,the backstepping technology is introduced to design the control inputs of each UAV pointing to North and Down along the special sphere(say,the circular tracking control algorithm)with the help of the switching function.Meanwhile,the distributed linear consensus protocol integrated with anther switching anti-interference item is developed to construct the control input of each UAV pointing to east along the special sphere(say,the formation keeping control law)for formation keeping.The validity of the proposed control law is proved both in the rigorous theory and through numerical simulations.展开更多

Vent. to low phosphorus (P) conditions can be attributed to the large amount of root-exuded organic acids and the high efficiency of P extraction. However, microelement contents are influenced by low-P stress, and their effects on the photosynthetic capability of B. pa- pyrifera remain unknown. In this study, we investigated the effects of low-P treatment on net photosynthetic rate (PN); chlorophyll a fluorescence (ChlF) characteristics; and Fe, Mn, Cu, and Zn contents of B. papyrifera and Morus alba L. seedlings. Results show that B. papyrifera exhibited better photosynthetic capability under moderate P defi- ciency (0.125, 0.063, and 0.031 mmol/L P treatments), whereas the photosynthetic capability of M. alba decreased under moderate and severe P deficiency (0.016 and 0 mmol/L P treatments). Under moderate P deficiency, the decrease in Cu and Zn contents in B. papyrifera was lower than that in M. alba. Under severe P deficiency, a consid- erable decrease of photosynthetic capability in B. pa- pyrifera and M. alba was associated with low Cu and Zn contents. The PN of the two Moraceae species exhibited a better correlation with Cu and Zn contents than with Fe or Mn content. P deficiency could not only decrease cyclic photophorylation and photosynthetic efficiency, but could also affect the stability of thylakoid membrane structureand electron transport efficiency by influencing the con- tents of Cu or Zn, thereby affecting photosynthesis.展开更多
Promoting agricultural modernisation through technological change is an important strategy for China.China's carbon neutrality strategy is leading to systemic socio-economic changes that could exacerbate the uncer...Promoting agricultural modernisation through technological change is an important strategy for China.China's carbon neutrality strategy is leading to systemic socio-economic changes that could exacerbate the uncertainty of agricultural development.Therefore,applying a computable general equilibrium(CGE)model,this study characterises the agricultural sector in detail,introducing endogenous technological change proxied by research and development(R&D)to assess the impact of different technological change scenarios on agricultural development under the carbon neutrality target.The results show that allocating carbon revenue for R&D inputs can mitigate the significant negative impact of achieving carbon neutrality on knowledge capital and production in agricultural sectors.Overall,using carbon revenue only for R&D input in crop sectors has the optimal effect on increasing the agricultural sectors'knowledge capital,improving crop production and profit,reducing crop external dependence and promoting the synergistic reduction of carbon and pollutant emissions.However,this scenario has the largest negative impact on macro-economics and household welfare.In contrast,allocating carbon revenue to promote technological change in broader non-energy sectors or both crops and non-energy sectors can effectively mitigate negative socio-economic impacts,but the positive impact on agricultural development is minimal.These findings provide practical insights for the rational use of carbon revenue to expand agricultural R&D investment and ensure balanced agricultural and economic development under the carbon neutrality target.展开更多

To study the shock initiation process of NTO-based polymer bonded explosive JEOL-1(32%octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine(HMX), 32% NTO, 28% Al and 8% binder system), the cylinder test, the gap experiments and numerical simulation were carried out. Firstly, we got the detonation velocity(7746 m/s) and the parameters of Jones-Wilkins-Lee(JWL) equation of state(EOS) for detonation product by cylinder test and numerical simulation. Secondly, the Hugoniot curve of unreacted explosive for JEOL-1 was obtained calculating the data of pressure and time at different Lagrangian positions. Then the JWL EOS of unreacted explosive was obtained by utilizing the Hugoniot curve as the reference curve. Finally, we got the pressure growth history of JEOL-1 under shock wave stimulation and the parameters of the ignition and growth reaction rate equation were obtained by the pressure-time curves measured by the shockinitiation gap experiment and numerical simulation. The determined trinomial ignition and growth model(IG model) parameters can be applied to subsequently simulation analysis and design of insensitive ammunition with NTO-based polymer bonded explosive.展开更多
Coxsackievirus A10(CVA10)is one of the major etiological agents of hand,foot,and mouth disease.There are no vaccine and antiviral drugs for controlling CVA10 infection.Reverse genetic tools for CVA10 will benefit its ...Coxsackievirus A10(CVA10)is one of the major etiological agents of hand,foot,and mouth disease.There are no vaccine and antiviral drugs for controlling CVA10 infection.Reverse genetic tools for CVA10 will benefit its mechanistic study and development of vaccines and antivirals.Here,two infectious clones for the prototype and a Myc-tagged CVA10 were constructed.Viable CVA10 viruses were harvested by transfecting the viral m RNA into human rhabdomyosarcoma(RD)cells.Rescued CVA10 was further confirmed by next generation sequencing and characterized experimentally.We also constructed the vectors for CVA10 subgenomic replicon with luciferase reporter and viral capsid with EGFP reporter,respectively.Co-transfection of the viral replicon RNA and capsid expresser in human embryonic kidney 293 T(HEK293 T)cells led to the production of single round infectious particles(SRIPs).Based on CVA10 replicon RNA,SRIPs with either the enterovirus A71(EVA71)capsid or the CVA10 capsid were generated.Infection by EVA71 SRIPs required SCARB2,while CVA10 SRIPs did not.Finally,we showed great improvement of the replicon activity and SRIPs production by insertion of a cis-active hammerhead ribozyme(HHRib)before the 50-untranslated region(UTR).In summary,reverse genetic tools for prototype strain of CVA10,including both the infectious clone and the SRIPs system,were successfully established.These tools will facilitate the basic and translational study of CVA10.展开更多

Rose(Rosa chinensis),which is an economically valuable floral species worldwide,has three types,namely once-flowering(OF),occasional or re-blooming(OR),and recurrent or continuous flowering(CF).However,the mechanism u...Rose(Rosa chinensis),which is an economically valuable floral species worldwide,has three types,namely once-flowering(OF),occasional or re-blooming(OR),and recurrent or continuous flowering(CF).However,the mechanism underlying the effect of the age pathway on the duration of the CF or OF juvenile phase is largely unknown.In this study,we observed that the RcSPL1 transcript levels were substantially upregulated during the floral development period in CF and OF plants.Additionally,accumulation of RcSPL1 protein was controlled by rch-miR156.The ectopic expression of RcSPL1 in Arabidopsis thaliana accelerated the vegetative phase transition and flowering.Furthermore,the transient overexpression of RcSPL1 in rose plants accelerated flowering,whereas silencing of RcSPL1 had the opposite phenotype.Accordingly,the transcription levels of floral meristem identity genes(APETALA1,FRUITFULL,and LEAFY)were significantly affected by the changes in RcSPL1 expression.RcTAF15b protein,which is an autonomous pathway protein,was revealed to interact with RcSPL1.The silencing and overexpression of RcTAF15b in rose plants led to delayed and accelerated flowering,respectively.Collectively,the study findings imply that RcSPL1–RcTAF15b modulates the flowering time of rose plants.展开更多
Lignin is the most abundant naturally phenolic biomass,and the synthesis of high-performance renewable fuel from lignin has attracted significant attention.We propose the efficient synthesis of high-density fuels usin...Lignin is the most abundant naturally phenolic biomass,and the synthesis of high-performance renewable fuel from lignin has attracted significant attention.We propose the efficient synthesis of high-density fuels using simulated lignin cracked oil in tandem with hydroalkylation and deoxygenation reactions.First,we investigated the reaction pathway for the hydroalkylation of phenol,which competes with the hydrodeoxygenation form cyclohexane.And then,we investigated the effects of metal catalyst types,the loading amount of metallic,acid dosage,and reactant ratio on the reaction results.The phenol hydroalkylation and hydrodeoxygenation were balanced when 180℃ and 5 MPa H_(2)with the alkanes yield of 95%.By extending the substrate to other lignin-derived phenolics and simulated lignin cracked oil,we obtained the polycyclic alkane fuel with high density of 0.918 g·ml^(-1)and calorific value of41.2 MJ·L^(-1).Besides,the fuel has good low-temperature properties(viscosity of 9.3 mm^(2)·s^(-1)at 20℃ and freezing point below-55℃),which is expected to be used as jet fuel.This work provides a promising way for the easy and green production of high-density fuel directly from real lignin oil.展开更多
The stable operation of solid-state lithium metal batteries at low temperatures is plagued by severe restrictions from inferior electrolyte-electrode interface compatibility and increased energy barrier for Li^(+)migr...The stable operation of solid-state lithium metal batteries at low temperatures is plagued by severe restrictions from inferior electrolyte-electrode interface compatibility and increased energy barrier for Li^(+)migration.Herein,we prepare a dual-salt poly(tetrahydrofuran)-based electrolyte consisting of lithium hexafluorophosphate and lithium difluoro(oxalato)borate(LiDFOB).The Li-salt anions(DFOB−)not only accelerate the ring-opening polymerization of tetrahydrofuran,but also promote the formation of highly ion-conductive and sustainable interphases on Li metal anodes without sacrificing the Li^(+)conductivity of electrolytes,which is favorable for Li^(+)transport kinetics at low temperatures.Applications of this polymer electrolyte in Li||LiFePO_(4)cells show 82.3%capacity retention over 1000 cycles at 30℃and endow stable discharge capacity at−30℃.Remarkably,the Li||LiFePO4 cells retain 52%of their room-temperature capacity at−20℃and 0.1 C.This rational design of dual-salt polymer-based electrolytes may provide a new perspective for the stable operation of quasi-solid-state batteries at low temperatures.展开更多
The utilization of enteroviruses engineered with reporter genes serves as a valuable tool for advancing our understanding of enterovirus biology and its applications,enabling the development of effective therapeutic a...The utilization of enteroviruses engineered with reporter genes serves as a valuable tool for advancing our understanding of enterovirus biology and its applications,enabling the development of effective therapeutic and preventive strategies.In this study,our initial attempts to introduce a NanoLuc luciferase(NLuc)reporter gene into recombinant enteroviruses were unsuccessful in rescuing viable progenies.We hypothesized that the size of the inserted tag might be a determining factor in the rescue of the virus.Therefore,we inserted the 11-amino-acid HiBiT tag into the genomes of enterovirus A71(EV-A71),coxsackievirus A10(CVA10),coxsackievirus A7(CVA7),coxsackievirus A16(CVA16),namely EV-A71-HiBiT,CVA16-HiBiT,CVA10-HiBiT,CVA7-HiBiT,and observed that the HiBiT-tagged viruses exhibited remarkably high rescue efficiency.Notably,the HiBiT-tagged enteroviruses displayed comparable characteristics to the wild-type viruses.A direct comparison between CVA16-NLuc and CVA16-HiBiT recombinant viruses revealed that the tiny HiBiT insertion had minimal impact on virus infectivity and replication kinetics.Moreover,these HiBiT-tagged enteroviruses demonstrated high genetic stability in different cell lines over multiple passages.In addition,the HiBiT-tagged viruses were successfully tested in antiviral drug assays,and the sensitivity of the viruses to drugs was not affected by the HiBiT tag.Ultimately,our findings provide definitive evidence that the integration of HiBiT into enteroviruses presents a universal,convenient,and invaluable method for advancing research in the realm of enterovirus virology.Furthermore,HiBiT-tagged enteroviruses exhibit great potential for diverse applications,including the development of antivirals and the elucidation of viral infection mechanisms.展开更多
A convenient real-time monitoring of monomer concentration during living cationic ring-opening polymerizations of tetrahydrofuran(THF) initiated with methyl triflate(Me OTf) has been developed for kinetic investig...A convenient real-time monitoring of monomer concentration during living cationic ring-opening polymerizations of tetrahydrofuran(THF) initiated with methyl triflate(Me OTf) has been developed for kinetic investigation and determination of equilibrium monomer concentration([M]e) via in situ FTIR spectroscopy in combination with a diamond tipped attenuated total reflectance(ATR) immersion probe. The polymerization rate was first order with respect to monomer concentration and initiator concentration from the linear slope of ln([M]0-[M]e)/([M]-[M]e) versus polymerization time at different temperatures in various solvents. [M]e decreased linearly with initial monomer concentration while increased exponentially with increasing polymerization temperature. The equilibrium also strongly depends on solvent polarity and its interaction with monomer. The equilibrium polymerization time(te) decreased with increasing solvent polarity and decreased linearly with increasing [M]0 in three solvents with different slopes to the same point of bulk polymerization in the absence of solvent. Equation of Mn,e = 72.1/(0.14–0.04[M]e) has been established to provide a simple and effective approach for the prediction for the number-average molecular weight of poly THFs at equilibrium state(Mn,e) in the equilibrium living cationic ring-opening polymerization of THF at 0 °C.展开更多

Specifically,let-7i-5p within hucMSC-exos inhibited the expression of the factor-related apoptosis ligand protein in L02 cells,leading to the upregulation of B-cell lymphoma-2 and the downregulation of B-cell lymphoma-2-associated Xprotein and cysteinyl aspartate specific proteinase-3, thereby inhibiting L02 cell apoptosis and enhancing cellproliferation activity. The overexpression of let-7i-5p effectively enhanced the antiapoptotic effects of hucMSC-exosboth in vitro and in vivo.CONCLUSIONOur findings indicate that hucMSC-exos alleviate HIRI by inhibiting apoptosis. We demonstrated that hucMSCexostarget apoptosis in L02 cells and mediate the let-7i-5p/factor-related apoptosis ligand pathway, therebyameliorating HIRI. This study provides new insights into the role of hucMSC-exos in hepatocyte apoptosis andhighlights the potential of hucMSC-exos as a therapeutic strategy for HIRI.

文摘Conductive biomaterials based on conductive polymers,carbon nanomaterials,or conductive inorganic nanomaterials demonstrate great potential in wound healing and skin tissue engineering,owing to the similar conductivity to human skin,good antioxidant and antibacterial activities,electrically controlled drug delivery,and photothermal effect.However,a review highlights the design and application of conductive biomaterials for wound healing and skin tissue engineering is lacking.In this review,the design and fabrication methods of conductive biomaterials with various structural forms including film,nanofiber,membrane,hydrogel,sponge,foam,and acellular dermal matrix for applications in wound healing and skin tissue engineering and the corresponding mechanism in promoting the healing process were summarized.The approaches that conductive biomaterials realize their great value in healing wounds via three main strategies(electrotherapy,wound dressing,and wound assessment)were reviewed.The application of conductive biomaterials as wound dressing when facing different wounds including acute wound and chronic wound(infected wound and diabetic wound)and for wound monitoring is discussed in detail.The challenges and perspectives in designing and developing multifunctional conductive biomaterials are proposed as well.

文摘This paper proposes a new distributed formation flight protocol for unmanned aerial vehicles(UAVs)to perform coordinated circular tracking around a set of circles on a target sphere.Different from the previous results limited in bidirectional networks and disturbance-free motions,this paper handles the circular formation flight control problem with both directed network and spatiotemporal disturbance with the knowledge of its upper bound.Distinguishing from the design of a common Lyapunov fiunction for bidirectional cases,we separately design the control for the circular tracking subsystem and the formation keeping subsystem with the circular tracking error as input.Then the whole control system is regarded as a cascade connection of these two subsystems,which is proved to be stable by input-tostate stability(ISS)theory.For the purpose of encountering the external disturbance,the backstepping technology is introduced to design the control inputs of each UAV pointing to North and Down along the special sphere(say,the circular tracking control algorithm)with the help of the switching function.Meanwhile,the distributed linear consensus protocol integrated with anther switching anti-interference item is developed to construct the control input of each UAV pointing to east along the special sphere(say,the formation keeping control law)for formation keeping.The validity of the proposed control law is proved both in the rigorous theory and through numerical simulations.

文摘The strong adaptability of Broussonetia papyrifera (L.) Vent. to low phosphorus (P) conditions can be attributed to the large amount of root-exuded organic acids and the high efficiency of P extraction. However, microelement contents are influenced by low-P stress, and their effects on the photosynthetic capability of B. pa- pyrifera remain unknown. In this study, we investigated the effects of low-P treatment on net photosynthetic rate (PN); chlorophyll a fluorescence (ChlF) characteristics; and Fe, Mn, Cu, and Zn contents of B. papyrifera and Morus alba L. seedlings. Results show that B. papyrifera exhibited better photosynthetic capability under moderate P defi- ciency (0.125, 0.063, and 0.031 mmol/L P treatments), whereas the photosynthetic capability of M. alba decreased under moderate and severe P deficiency (0.016 and 0 mmol/L P treatments). Under moderate P deficiency, the decrease in Cu and Zn contents in B. papyrifera was lower than that in M. alba. Under severe P deficiency, a consid- erable decrease of photosynthetic capability in B. pa- pyrifera and M. alba was associated with low Cu and Zn contents. The PN of the two Moraceae species exhibited a better correlation with Cu and Zn contents than with Fe or Mn content. P deficiency could not only decrease cyclic photophorylation and photosynthetic efficiency, but could also affect the stability of thylakoid membrane structureand electron transport efficiency by influencing the con- tents of Cu or Zn, thereby affecting photosynthesis.

文摘Promoting agricultural modernisation through technological change is an important strategy for China.China's carbon neutrality strategy is leading to systemic socio-economic changes that could exacerbate the uncertainty of agricultural development.Therefore,applying a computable general equilibrium(CGE)model,this study characterises the agricultural sector in detail,introducing endogenous technological change proxied by research and development(R&D)to assess the impact of different technological change scenarios on agricultural development under the carbon neutrality target.The results show that allocating carbon revenue for R&D inputs can mitigate the significant negative impact of achieving carbon neutrality on knowledge capital and production in agricultural sectors.Overall,using carbon revenue only for R&D input in crop sectors has the optimal effect on increasing the agricultural sectors'knowledge capital,improving crop production and profit,reducing crop external dependence and promoting the synergistic reduction of carbon and pollutant emissions.However,this scenario has the largest negative impact on macro-economics and household welfare.In contrast,allocating carbon revenue to promote technological change in broader non-energy sectors or both crops and non-energy sectors can effectively mitigate negative socio-economic impacts,but the positive impact on agricultural development is minimal.These findings provide practical insights for the rational use of carbon revenue to expand agricultural R&D investment and ensure balanced agricultural and economic development under the carbon neutrality target.

文摘3-nitro-1,2,4-tri-azol-5-one(NTO) is a high energy insensitive explosive. To study the shock initiation process of NTO-based polymer bonded explosive JEOL-1(32%octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine(HMX), 32% NTO, 28% Al and 8% binder system), the cylinder test, the gap experiments and numerical simulation were carried out. Firstly, we got the detonation velocity(7746 m/s) and the parameters of Jones-Wilkins-Lee(JWL) equation of state(EOS) for detonation product by cylinder test and numerical simulation. Secondly, the Hugoniot curve of unreacted explosive for JEOL-1 was obtained calculating the data of pressure and time at different Lagrangian positions. Then the JWL EOS of unreacted explosive was obtained by utilizing the Hugoniot curve as the reference curve. Finally, we got the pressure growth history of JEOL-1 under shock wave stimulation and the parameters of the ignition and growth reaction rate equation were obtained by the pressure-time curves measured by the shockinitiation gap experiment and numerical simulation. The determined trinomial ignition and growth model(IG model) parameters can be applied to subsequently simulation analysis and design of insensitive ammunition with NTO-based polymer bonded explosive.

文摘Coxsackievirus A10(CVA10)is one of the major etiological agents of hand,foot,and mouth disease.There are no vaccine and antiviral drugs for controlling CVA10 infection.Reverse genetic tools for CVA10 will benefit its mechanistic study and development of vaccines and antivirals.Here,two infectious clones for the prototype and a Myc-tagged CVA10 were constructed.Viable CVA10 viruses were harvested by transfecting the viral m RNA into human rhabdomyosarcoma(RD)cells.Rescued CVA10 was further confirmed by next generation sequencing and characterized experimentally.We also constructed the vectors for CVA10 subgenomic replicon with luciferase reporter and viral capsid with EGFP reporter,respectively.Co-transfection of the viral replicon RNA and capsid expresser in human embryonic kidney 293 T(HEK293 T)cells led to the production of single round infectious particles(SRIPs).Based on CVA10 replicon RNA,SRIPs with either the enterovirus A71(EVA71)capsid or the CVA10 capsid were generated.Infection by EVA71 SRIPs required SCARB2,while CVA10 SRIPs did not.Finally,we showed great improvement of the replicon activity and SRIPs production by insertion of a cis-active hammerhead ribozyme(HHRib)before the 50-untranslated region(UTR).In summary,reverse genetic tools for prototype strain of CVA10,including both the infectious clone and the SRIPs system,were successfully established.These tools will facilitate the basic and translational study of CVA10.

文摘An experimental picosecond time-resolved X-ray ferromagnetic resonance(TR-XFMR)apparatus with a time resolution of 13 ps(RMS)or 31 ps(FWHM)was constructed and demonstrated in the 07U and 08U1A soft X-ray beamlines at the Shanghai Synchrotron Radiation Facility(SSRF)using pump-probe detection and X-ray magnetic circular dichroism(XMCD)spectroscopy.Element and time-resolved ferromagnetic resonance was excited by continuous microwave phase-locking of the bunch clock within the photon beam during synchrotron radiation and was characterized by detecting the magnetic circular dichroism signals of the elements of interest in the magnetic films.Using this equipment,we measured the amplitude of the element-specific moment precession during ferromagnetic resonance(FMR)at 2 GHz in a single Ni81Fe19layer.

文摘Rose(Rosa chinensis),which is an economically valuable floral species worldwide,has three types,namely once-flowering(OF),occasional or re-blooming(OR),and recurrent or continuous flowering(CF).However,the mechanism underlying the effect of the age pathway on the duration of the CF or OF juvenile phase is largely unknown.In this study,we observed that the RcSPL1 transcript levels were substantially upregulated during the floral development period in CF and OF plants.Additionally,accumulation of RcSPL1 protein was controlled by rch-miR156.The ectopic expression of RcSPL1 in Arabidopsis thaliana accelerated the vegetative phase transition and flowering.Furthermore,the transient overexpression of RcSPL1 in rose plants accelerated flowering,whereas silencing of RcSPL1 had the opposite phenotype.Accordingly,the transcription levels of floral meristem identity genes(APETALA1,FRUITFULL,and LEAFY)were significantly affected by the changes in RcSPL1 expression.RcTAF15b protein,which is an autonomous pathway protein,was revealed to interact with RcSPL1.The silencing and overexpression of RcTAF15b in rose plants led to delayed and accelerated flowering,respectively.Collectively,the study findings imply that RcSPL1–RcTAF15b modulates the flowering time of rose plants.

文摘Lignin is the most abundant naturally phenolic biomass,and the synthesis of high-performance renewable fuel from lignin has attracted significant attention.We propose the efficient synthesis of high-density fuels using simulated lignin cracked oil in tandem with hydroalkylation and deoxygenation reactions.First,we investigated the reaction pathway for the hydroalkylation of phenol,which competes with the hydrodeoxygenation form cyclohexane.And then,we investigated the effects of metal catalyst types,the loading amount of metallic,acid dosage,and reactant ratio on the reaction results.The phenol hydroalkylation and hydrodeoxygenation were balanced when 180℃ and 5 MPa H_(2)with the alkanes yield of 95%.By extending the substrate to other lignin-derived phenolics and simulated lignin cracked oil,we obtained the polycyclic alkane fuel with high density of 0.918 g·ml^(-1)and calorific value of41.2 MJ·L^(-1).Besides,the fuel has good low-temperature properties(viscosity of 9.3 mm^(2)·s^(-1)at 20℃ and freezing point below-55℃),which is expected to be used as jet fuel.This work provides a promising way for the easy and green production of high-density fuel directly from real lignin oil.

文摘The stable operation of solid-state lithium metal batteries at low temperatures is plagued by severe restrictions from inferior electrolyte-electrode interface compatibility and increased energy barrier for Li^(+)migration.Herein,we prepare a dual-salt poly(tetrahydrofuran)-based electrolyte consisting of lithium hexafluorophosphate and lithium difluoro(oxalato)borate(LiDFOB).The Li-salt anions(DFOB−)not only accelerate the ring-opening polymerization of tetrahydrofuran,but also promote the formation of highly ion-conductive and sustainable interphases on Li metal anodes without sacrificing the Li^(+)conductivity of electrolytes,which is favorable for Li^(+)transport kinetics at low temperatures.Applications of this polymer electrolyte in Li||LiFePO_(4)cells show 82.3%capacity retention over 1000 cycles at 30℃and endow stable discharge capacity at−30℃.Remarkably,the Li||LiFePO4 cells retain 52%of their room-temperature capacity at−20℃and 0.1 C.This rational design of dual-salt polymer-based electrolytes may provide a new perspective for the stable operation of quasi-solid-state batteries at low temperatures.

文摘The utilization of enteroviruses engineered with reporter genes serves as a valuable tool for advancing our understanding of enterovirus biology and its applications,enabling the development of effective therapeutic and preventive strategies.In this study,our initial attempts to introduce a NanoLuc luciferase(NLuc)reporter gene into recombinant enteroviruses were unsuccessful in rescuing viable progenies.We hypothesized that the size of the inserted tag might be a determining factor in the rescue of the virus.Therefore,we inserted the 11-amino-acid HiBiT tag into the genomes of enterovirus A71(EV-A71),coxsackievirus A10(CVA10),coxsackievirus A7(CVA7),coxsackievirus A16(CVA16),namely EV-A71-HiBiT,CVA16-HiBiT,CVA10-HiBiT,CVA7-HiBiT,and observed that the HiBiT-tagged viruses exhibited remarkably high rescue efficiency.Notably,the HiBiT-tagged enteroviruses displayed comparable characteristics to the wild-type viruses.A direct comparison between CVA16-NLuc and CVA16-HiBiT recombinant viruses revealed that the tiny HiBiT insertion had minimal impact on virus infectivity and replication kinetics.Moreover,these HiBiT-tagged enteroviruses demonstrated high genetic stability in different cell lines over multiple passages.In addition,the HiBiT-tagged viruses were successfully tested in antiviral drug assays,and the sensitivity of the viruses to drugs was not affected by the HiBiT tag.Ultimately,our findings provide definitive evidence that the integration of HiBiT into enteroviruses presents a universal,convenient,and invaluable method for advancing research in the realm of enterovirus virology.Furthermore,HiBiT-tagged enteroviruses exhibit great potential for diverse applications,including the development of antivirals and the elucidation of viral infection mechanisms.

文摘A convenient real-time monitoring of monomer concentration during living cationic ring-opening polymerizations of tetrahydrofuran(THF) initiated with methyl triflate(Me OTf) has been developed for kinetic investigation and determination of equilibrium monomer concentration([M]e) via in situ FTIR spectroscopy in combination with a diamond tipped attenuated total reflectance(ATR) immersion probe. The polymerization rate was first order with respect to monomer concentration and initiator concentration from the linear slope of ln([M]0-[M]e)/([M]-[M]e) versus polymerization time at different temperatures in various solvents. [M]e decreased linearly with initial monomer concentration while increased exponentially with increasing polymerization temperature. The equilibrium also strongly depends on solvent polarity and its interaction with monomer. The equilibrium polymerization time(te) decreased with increasing solvent polarity and decreased linearly with increasing [M]0 in three solvents with different slopes to the same point of bulk polymerization in the absence of solvent. Equation of Mn,e = 72.1/(0.14–0.04[M]e) has been established to provide a simple and effective approach for the prediction for the number-average molecular weight of poly THFs at equilibrium state(Mn,e) in the equilibrium living cationic ring-opening polymerization of THF at 0 °C.
