Thinned young citrus fruits(TYCF)are a by-product rich in bioactive compounds.We prepared a TYCF extract and purified it using AB-8 macroporous resin to obtain a purified extract(PE).The purification increased the tot...Thinned young citrus fruits(TYCF)are a by-product rich in bioactive compounds.We prepared a TYCF extract and purified it using AB-8 macroporous resin to obtain a purified extract(PE).The purification increased the total phenolic content by 2.1-fold and the total flavonoid content by 11.1-fold.After purification,DPPH and ABTS radical-scavenging activities were significantly enhanced.The main flavonoids identified in PE were hesperidin and narirutin.Furthermore,PE significantly reduced LPS-induced NO production and decreased proinflammatory cytokines(e.g.,TNF-αand IL-1β)in RAW 264.7 cells.Metabolomics analysis revealed that lysoPE(18:0),D-sphingosine,glycerophospho-N-palmitoyl-ethanolamine,1-stearoyl-sn-glycero-3-phospho-1D-myo-inositol,lysoPC(16:1),9-HODE,monomethylphosphate,2-oxohenicosanoic acid,10,12-hexadecadienal,2-eicosapentaenoylglycerol,2-arachidonoylglycerol and 2-deoxyguanosine were dysregulated in the LPS-treated group,whereas PE partly reversed these LPS-induced alterations.Molecular docking indicated favorable dock-ing scores for major PE components against TNF-αand IL-1β,suggesting potential interactions.In H_(2)O_(2)-treated PC12 cells,PE improved cell viability,reduced MDA and ROS levels,and showed preliminary anti-senescent effects as indicated by SA-β-gal staining,suggesting neuroprotective potential under oxidative stress.Thus,TYCF may be a valuable resource for developing health-promoting foods and therapeutic applications.展开更多
