BACKGROUND Adolescence is a period marked by physiological and psychological imbalances,which pose an increased risk for adolescents with major depressive disorder(MDD)to commit non-suicidal self-injury(NSSI).AIM To i...BACKGROUND Adolescence is a period marked by physiological and psychological imbalances,which pose an increased risk for adolescents with major depressive disorder(MDD)to commit non-suicidal self-injury(NSSI).AIM To investigate the moderating role of social support utilization in depression and NSSI among adolescents with MDD.METHODS This cross-sectional study enrolled 314 adolescents with MDD(258 with NSSI,56 without)from a Chinese tertiary psychiatric hospital(2021-2023).Participants completed validated scales,including the self-esteem scale,the Barratt impulsiveness scale,the self-rating depression scale,and the teenager social support rating scale.Logistic regression and hierarchical regression analyses were used to examine predictors of NSSI and the moderating effect of social support utilization.RESULTS Results showed that the NSSI group had higher depression levels,lower selfesteem,and greater impulsivity.While overall social support was higher in the NSSI group,social support utilization significantly moderated the depression-NSSI relationship.Specifically,higher utilization levels weakened the association between depression and NSSI(β=-0.001,P<0.05).CONCLUSION These findings suggest that effective utilization of social support,rather than its mere presence,is crucial in reducing NSSI risk among depressed adolescents.展开更多
Directed degradation of abundant renewable lignin into small aromatic compounds is crucial for lignin valorization but challenging.The degradation of lignin in natural environments typically involves multienzyme syner...Directed degradation of abundant renewable lignin into small aromatic compounds is crucial for lignin valorization but challenging.The degradation of lignin in natural environments typically involves multienzyme synergy.However,the proteinaceous characteristics of lignin-degrading enzymes restrict their accessibility to certain regions of intricate lignin,resulting in the multienzyme systems being unable to fully demonstrate their effectiveness.Herein,a de novo biomimetic enzyme-nanozyme hybrid system was constructed by combiningλ-MnO_(2) nanozyme with laccase CotA from Bacillus subtilis,aimed at facilitating lignin degradation under mild conditions.The lignin degradation rate of the CotA+λ-MnO_(2) hybrid system was determined to be 25.15%,which was much higher than those of the lignin degradation systems with only laccase CotA(15.32%)orλ-MnO_(2) nanozyme(14.90%).Notably,the proportion of aromatic chemicals in the products derived from the hybrid system reached as much as 48%,which was 41.2%and 118.2%higher than those of the CotA-andλ-MnO_(2)-catalyzed systems,respectively.Analysis of products mapping and lignin structure changes suggested that the higher proportion of aromatic compounds in the CotA+λ-MnO_(2)hybrid system was more likely to benefit from the laccase-mediated methoxylation.Moreover,electron paramagnetic resonance analysis indicated that the intensity and kind of free radicals such as·OH and·O_(2)^(-)are closely linked to the degradation rate and reaction type.This work is the inaugural application of an enzyme-nanozyme hybrid system for lignin degradation,demonstrating the potential of the synergistic interaction between enzyme and nanozyme in the directed degradation of lignin.展开更多
The river plume front between the diluted ocean water and salty ocean water in the Changjiang(Yangtze)River Delta(CRD)is well studied.Comparatively,less is known about the estuarine front in the CRD,which is formed be...The river plume front between the diluted ocean water and salty ocean water in the Changjiang(Yangtze)River Delta(CRD)is well studied.Comparatively,less is known about the estuarine front in the CRD,which is formed between the riverine freshwater and the diluted ocean water and has the highest magnitude of salinity gradient(SG)in the CRD.Estuarine fronts are of great significance to the riverine material transport in the estuary.Many biogeochemical processes are enhanced in estuarine fronts,which have brought about environmental problems.In this study,the seasonal variations of the estuarine fronts in the CRD were studied in wet(July)and dry(January)seasons in 2017,based on model simulations with high spatiotemporal resolutions using the Finite-Volume Community Ocean Model(FVCOM).The estuarine front included several sharp fronts with a SG>4(/500 m),and was bottom-trapped on the submerged delta front.Seasonal changes mainly occurred off the Jiangsu coast,where a significant estuarine front was formed in July.The estuarine fronts generated around the submerged delta topography were accompanied by the offshore extension of older estuarine fronts,which were diluted and evolved into plume fronts over a tidal cycle.The simulated estuarine fronts had a salinity range of 6 to 22 in the dry season and 6 to 14 in wet season 2017.The estuarine fronts hindered the residual current by altering its flow direction to the southeast.展开更多
Nickel-based superconductors have attracted great attention due to the finding of the Ruddlesden-Popper(R-P)bilayer nickelate La_(3)Ni_(2)O_(7)with superconducting critical temperature(T_(c))of 80 K at pressure above ...Nickel-based superconductors have attracted great attention due to the finding of the Ruddlesden-Popper(R-P)bilayer nickelate La_(3)Ni_(2)O_(7)with superconducting critical temperature(T_(c))of 80 K at pressure above 14 GPa.Recent efforts have been devoted to the study of La_(2)PrNi_(2)O_(7),while the detailed structure remains unclear.In this work,we explore the stability and physical properties of such an interesting system by using density functional theory and the U parameter simulation method implemented in VASP.The results show that the enthalpy of La_(2)PrNi_(2)O_(7)is slightly larger than its parent material bilayer R-P nickelate La_(3)Ni_(2)O_(7).The electronic structure analysis indicates that near the Fermi level,the eg orbit of Ni dominates and strongly hybridizes with the 2p orbit of O,thereby forming a significant van Hove singularity that is conducive to superconductivity.The Amam phase to the I4/mmm phase occurs,accompanied by an increase in the bandwidth of Ni 3d_(z)_(2) and an enhancement of the bonding-antibonding splitting(from about 0.5 eV to 1.5 eV),which leads to an increase in the density of states at the Fermi level.Our findings provide insights into the preparation and superconductivity of R-P bilayer nickelate.展开更多

Nephrotoxicity, especially acute kidney injury (AKI), is the most common and severe adverse reaction of cisplatin. Resveratrol and ginsenoside Rg1, two natural products, have been found to have renal protective effects. However, the effects and the mechanisms in cisplatin-induced AKI need further investigation. <strong>Methods:</strong> The mouse models of cisplatin-induced AKI and several treatment groups were established. Male C57BL/6 mice were divided into five groups: saline control group, cisplatin injury group, resveratrol treatment group, Rg1 treatment group, resveratrol and Rg1 combined treatment group. Serological analysis of serum urea nitrogen was aimed to reflect the function of kidney, and histological analysis of renal tissue sections was aimed to assess the damage of proximal convoluted tubules. The expression levels of autophagy-related proteins Beclin 1 and LC3 were detected by western blotting and qRT-PCR respectively. <strong>Results:</strong> The renal function was improved and renal damage was alleviated in Rg1 and resveratrol alone or combined treatment groups compared with the cisplatin injury group. For the mechanism, treatment with Rg1 and resveratrol alone or in combination decreased the expressions of Beclin 1 both at protein and mRNA levels, decreased LC3II/I protein levels, indicating that autophagy was inhibited by treatment with Rg1 and resveratrol alone or in combination. <strong>Conclusion:</strong> Resveratrol and Rg1 alleviated the kidney damage caused by cisplatin, and reduced autophagy was involved in the renoprotective effects of resveratrol and Rg1 against cisplatin-induced AKI. This study may provide new evidence to alleviate cisplatin-induced AKI.展开更多

Many monomer components are studied, the most often investigated are Rg3, and Rh2, etc. Reports have shown that ginsenosides can inhibit tumor cells by suppressing proliferation and metastasis, and promoting apoptosis. In addition, ginsenosides can enhance sensitivity to conventional chemotherapeutic drugs. Among its various post-translational modifications, phosphorylation is the most extensively studied. It is recognized that tau hyperphosphorylation is the root cause of neurodegeneration in Alzheimer's disease (AD); however, it is not clear how it causes neurodegeneration. Based on the findings that tau hyperphosphorylation leads to the escape of neurons from acute apoptosis and simultaneously impairs the function of neurons, we have proposed that the nature of AD neurodegeneration is the consequence of aborted apoptosis induced by tau phosphorylation. Therefore, proper manipulation of tau hyperphosphorylation could be promising for arresting AD neurodegeneration. In this review, the neuroprotective and neurodegenerative effects of tau hyperphosphorylation and our thoughts regarding their relationship are presented.展开更多

However, the mechanism underlying tau hyperphosphorylation is not fully unders...Hyperphosphorylated tau is the major protein component of neurofibrillary tangles in the brains of patients with Alzheimer’s disease(AD). However, the mechanism underlying tau hyperphosphorylation is not fully understood. Here, we demonstrated that exogenously expressed wild-type human tau40 was detectable in the phosphorylated form at multiple AD-associated sites in cytoplasmic and nuclear fractions from HEK293 cells.Among these sites, tau phosphorylated at Thr205 and Ser214 was almost exclusively found in the nuclear fraction at the conditions used in the present study. With the intracellular tau accumulation, the Ca2+concentration was significantly increased in both cytoplasmic and nuclear fractions. Further studies using site-specific mutagenesis and pharmacological treatment demonstrated that phosphorylation of tau at Thr205 increased nuclear Ca2+concentration with a simultaneous increase in the phosphorylation of Ca2+/calmodulin-dependent protein kinase IV(Ca MKIV) at Ser196. On the other hand, phosphorylation of tau at Ser214 did not significantly change the nuclear Ca2+/Ca MKIV signaling. Finally, expressing calmodulin-binding protein-4 that disrupts formation ofthe Ca2+/calmodulin complex abolished the okadaic acidinduced tau hyperphosphorylation in the nuclear fraction.We conclude that the intracellular accumulation of phosphorylated tau, as detected in the brains of AD patients, can trigger nuclear Ca2+/Ca MKIV signaling, which in turn aggravates tau hyperphosphorylation. Our findings provide new insights for tauopathies: hyperphosphorylation of intracellular tau and an increased Ca2+concentration may induce a self-perpetuating harmful loop to promote neurodegeneration.展开更多

Objective:To observe the effect of the early intervention with moxibustion at "Shènshū(肾俞BL23)" on learning and memory of Alzheimer’s disease(AD) rats modeled by ovariectomy and D-Galactose(D-Gal)in...Objective:To observe the effect of the early intervention with moxibustion at "Shènshū(肾俞BL23)" on learning and memory of Alzheimer’s disease(AD) rats modeled by ovariectomy and D-Galactose(D-Gal)injection and the effect on Ca2+/CaMKII/CREB signaling pathway so as to provide the evidence for the acupoint selection in the early intervention with acupuncture and moxibustion for AD.Methods:Sixty female SD rats at the age of 3 months were selected and randomly divided into a normal group,a sham-operation group,a model group,a "BL23" group,a "Wèishū"(胃俞BL21) group,and a non-acupoint group,10 rats in each group.Except in the normal group and the sham-operation group,bilateral ovaries were removed in the other 4 groups.3 days after the operation,D-Gal was injected intraperitoneally at a dose of 150 mg/kg/d for 90 days to establish AD rat model.In the sham-operation group,only the peripheral fat of ovarian was removed and 0.9% normal saline was injected intraperitoneally of the same dose for 90 days.In the "BL23" group,the "BL21" group and the non-acupoints group,since the 2 nd day after ovariectomy,moxibustion was applied at 9:00 a.m.every day in the rats.The rats were fixed on a fixed table,the mild moxibustion with self-prepared moxa stick was applied to bilateral "BL23""BL21" and the non-acupoint of each rat,the ignited end of moxa stick was 2 cm to 3 cm far from skin surface.The infrared thermometer was adopted to measure the temperature of the epidermis of each acupoint from time to time so as to maintain the local temperature to be(41 ± 0.5)℃.Mild moxibustion lasted for 10 min each time,consecutively for 5 days a week and at an interval of 2 days.The total consecutive intervention duration was 12 weeks.In the normal group and the shamoperation group,no moxibustion intervention was performed,but the rats were restricted in a same way during the experiment.At the end of modeling and intervention,water maze test was adopted to detect the learning and memory abilities of rats in each group.The western blotting was used to detect the expressions of p-CREB,p-CaMKII,CaMKII and CREB in hippocampus of rats in each group.Results:(1)Navigation test:since Day 72,compared with the sham-operation group,the escape latencies in all of the consecutive 5 days were longer in the model group(all P <0.05).Since Day 73,compared with the normal group,the escape latency in all of the consecutive 4 days was shorter in the "BL23"group(all P <0.05).(2) Times across platform:compared with the normal group,the number of times consecutively crossing the platform was lower in the model group(P <0.05).After moxibustion,compared with the model group,the number of times across platform was higher in the "BL23"group,indicating a statistical significance(P <0.05).(3)Moving track of water maze spatial probe:the "tendency"strategy was displayed in the normal group and the sham operation group.The "marginal" strategy,which is commonly seen in the AD rats,was displayed in the model group.The "random" strategy was displayed in the "BL23" group.The "marginal" strategy was presented in the "BL21" group and the non-acupoint group.(4) Expressions of hippocampal P-CAMKII,CAMKII,CREB and P-CREB in the rats:compared with the normal group,the phosphorylation degrees of CAMKII and CREB were significantly reduced(both P<0.05) in the model group.In 12 weeks of moxibustion,compared with the model group,the levels of P-CAMKII and P-CREB were increased in the "BL23" group and the "BL21" group(all P <0.05) and the levels in the "BL23" group were higher than that of the "BL21" group and the non-acupoint group(all P<0.05).Conclusion:Moxibustion at "BL23" achieves a better effect in improving the learning and memory of AD rats modeled by ovariectomy and D-Gal injection as compared with moxibustion at "BL21" and the non-acupoint.The effect mechanism is potentially related to the effective activation of Ca2+/CaMKII/CREB signaling pathway.展开更多

Therefore, they are valuable in regenerative medicine, human developmental biology and drug discovery. A number of hESC lines have been derived from the Chinese population, but limited of them are available for research purposes. Here we report the derivation and characterization of two hESC lines derived from human blastocysts of Chinese origin. These hESCs express alkaline phosphatase and hESC-specific markers, including Oct4, Nanog, SSEA-3, SSEA-4, TRA- 1-60 and TRA-1-81. They also have high levels of telomerase activity and normal karyotypes. These ceils can form embryoid body in vitro and can be differentiated into all three germ layers in vivo by teratoma formation. The newly established hESCs will be distributed for research purposes. The availability of hESC lines from the Chinese population will facilitate studies on the differences in hESCs from different ethnic groups.展开更多
A splicing mutation in VPS4B can cause dentin dysplasia type I(DD-I),a hereditary autosomal-dominant disorder characterized by rootless teeth,the etiology of which is genetically heterogeneous.In our study,dental foll...A splicing mutation in VPS4B can cause dentin dysplasia type I(DD-I),a hereditary autosomal-dominant disorder characterized by rootless teeth,the etiology of which is genetically heterogeneous.In our study,dental follicle cells(DFCs)were isolated and cultured from a patient with DD-I and compared with those from an age-matched,healthy control.In a previous study,this DD-I patient was confirmed to have a loss-of-function splicing mutation in VPS4B(IVS7+46C>G).The results from this study showed that the isolated DFCs were vimentin-positive and CK14-negative,indicating that the isolated cells were derived from the mesenchyme.DFCs harboring the VPS4B mutation had a significantly higher proliferation rate from day 3 to day 8 than control DFCs,indicating that VPS4B is involved in cell proliferation.The cells were then replenished with osteogenic medium to investigate how the VPS4B mutation affected osteogenic differentiation.Induction of osteogenesis,detected by alizarin red and alkaline phosphatase staining in vitro,was decreased in the DFCs from the DD-I patient compared to the control DFCs.Furthermore,we also found that the VPS4B mutation in the DD-I patient downregulated the expression of osteoblast-related genes,such as ALP,BSP,OCN,RUNX2,and their encoded proteins.These outcomes confirmed that the DD-I-associated VPS4B mutation could decrease the capacity of DFCs to differentiate during the mineralization process and may also impair physiological root formation and bone remodeling.This might provide valuable insights and implications for exploring the pathological mechanisms underlying DD-I root development.展开更多

Our results are:(1) The current was mainly a(clockwise) rotating flow, displaying reciprocating flow characteristics,and vertically the current directions were the same throughout the vertical profile.(2) The horizontal current speed was strongest during August(summer) with an average speed of 51.8 cm/s. The average current speeds during spring tides were highest in August and weakest in September, with speeds of 59.9 and 42.8 cm/s,respectively.(3) Considerable differences exist in average current speeds in different layers and seasons. The highest average current speeds were found in the middle–upper layers in August and in the middle–lower layers in September and October.(4) The residual current speed was highest in August, when the speed was 12.5–47.1 cm/s,whereas the vertical average current speed was 34.3 cm/s. The depth-averaged residual current speeds in September and October were only 50% of that in August, and the residual current direction gradually rotated in a counter-clockwise direction from the lower to surface layers.(5) Typhoon waves had a significant influence on the currents, and even affected the middle and lower water layers at depths of >70.0 m. Our results showed that the currents are controlled by the dynamic interplay of the Taiwan Warm Current, incursion of the Kuroshio Current onto the continental shelf, and monsoonal changes.展开更多

Results obtained showed a number of observations: 1) The annual mean temperature of Shandong Peninsula showed a significant increasing trend, with a distinct abrupt change point detected around 1990, during the past 5 decades. The warming of the Peninsula over the last 50 years was due mainly to the significant increase of annual minimum temperature. The annual maximum temperature demonstrated a mixed trend of decreasing and increasing, but was statistically insignificant, and no abrupt change was detected;2) The annual precipitation exhibited a decreasing trend during the past 5 decades, with an abrupt change detected around 1980 at most stations;but there was an earlier transition point at 1966, at a few stations. The reduction in precipitation, from June to September, was responsible mainly for the decrease of annual precipitation. Besides, the proportion of the June-September precipitation in the year declined slightly over the last 50 years;3) In comparison, the temperature evolution in Shandong Peninsula was basically consistent with most parts of China, but warmed at a faster rate over the same period;the decreasing trend of precipitation was more significant compared with the other climate zones of China. Within the Peninsula, the abrupt change of temperature and precipitation in the Southeast was earlier than that in the Northwest;the reduction of precipitation was larger in the Southeast while the increase of temperature was more significant in the Northwest. This research was of great importance to understand the climate change and its environmental effects in the coastal zone.展开更多

To hunt for genetic variants underlying human height variation, we performed a genome wide CNV association study for human height in 618 Chinese unrelated subjects using Affymetrix 500K array set. After adjusting for age and sex, we found that four CNVs at 6p21.3, 8p23.3-23.2, 9p23 and 16p12.1 were associated with human height (with borderline significant p value: 0.013, 0.011, 0.024, 0.049; respectively). However, after multiple tests correction, none of them was associated with human height. We observed that the gain of copy number (more than 2 copies) at 8p23.3-23.2 was associated with lower height (normal copy number vs. gain of copy number: 161.2 cm vs. 153.7 cm, p = 0.011), which accounted for 0.9% of height variation. Loss of copy number (less than 2 copies) at 6p21.3 was associated with 0.8% lower height (loss of copy number vs. normal copy number: 154.5 cm vs. 161.1 cm, p = 0.013). Since no important genes influencing height located in CNVs at loci of 8p23.3-23.2 and 6p21.3, the two CNVs may cause the structural rear- rangements of neighbored important candidate genes, thus regulates the variation of height. Our results expand our knowledge of the genetic factors underlying height variation and the biological regulation of human height.展开更多

The composition of the system and the functions of the modules are described, then the three-dimensional graphic engine based on Open Scene Graph (OSG) and the way of scene organization based on hierarchical tree are introduced, moreover, the methods of fault simulation and detection instrument simulation based on algorithm component library are presented. 文摘BACKGROUND Adolescence is a period marked by physiological and psychological imbalances,which pose an increased risk for adolescents with major depressive disorder(MDD)to commit non-suicidal self-injury(NSSI).AIM To investigate the moderating role of social support utilization in depression and NSSI among adolescents with MDD.METHODS This cross-sectional study enrolled 314 adolescents with MDD(258 with NSSI,56 without)from a Chinese tertiary psychiatric hospital(2021-2023).Participants completed validated scales,including the self-esteem scale,the Barratt impulsiveness scale,the self-rating depression scale,and the teenager social support rating scale.Logistic regression and hierarchical regression analyses were used to examine predictors of NSSI and the moderating effect of social support utilization.RESULTS Results showed that the NSSI group had higher depression levels,lower selfesteem,and greater impulsivity.While overall social support was higher in the NSSI group,social support utilization significantly moderated the depression-NSSI relationship.Specifically,higher utilization levels weakened the association between depression and NSSI(β=-0.001,P<0.05).CONCLUSION These findings suggest that effective utilization of social support,rather than its mere presence,is crucial in reducing NSSI risk among depressed adolescents.
文摘Directed degradation of abundant renewable lignin into small aromatic compounds is crucial for lignin valorization but challenging.The degradation of lignin in natural environments typically involves multienzyme synergy.However,the proteinaceous characteristics of lignin-degrading enzymes restrict their accessibility to certain regions of intricate lignin,resulting in the multienzyme systems being unable to fully demonstrate their effectiveness.Herein,a de novo biomimetic enzyme-nanozyme hybrid system was constructed by combiningλ-MnO_(2) nanozyme with laccase CotA from Bacillus subtilis,aimed at facilitating lignin degradation under mild conditions.The lignin degradation rate of the CotA+λ-MnO_(2) hybrid system was determined to be 25.15%,which was much higher than those of the lignin degradation systems with only laccase CotA(15.32%)orλ-MnO_(2) nanozyme(14.90%).Notably,the proportion of aromatic chemicals in the products derived from the hybrid system reached as much as 48%,which was 41.2%and 118.2%higher than those of the CotA-andλ-MnO_(2)-catalyzed systems,respectively.Analysis of products mapping and lignin structure changes suggested that the higher proportion of aromatic compounds in the CotA+λ-MnO_(2)hybrid system was more likely to benefit from the laccase-mediated methoxylation.Moreover,electron paramagnetic resonance analysis indicated that the intensity and kind of free radicals such as·OH and·O_(2)^(-)are closely linked to the degradation rate and reaction type.This work is the inaugural application of an enzyme-nanozyme hybrid system for lignin degradation,demonstrating the potential of the synergistic interaction between enzyme and nanozyme in the directed degradation of lignin.

文摘The river plume front between the diluted ocean water and salty ocean water in the Changjiang(Yangtze)River Delta(CRD)is well studied.Comparatively,less is known about the estuarine front in the CRD,which is formed between the riverine freshwater and the diluted ocean water and has the highest magnitude of salinity gradient(SG)in the CRD.Estuarine fronts are of great significance to the riverine material transport in the estuary.Many biogeochemical processes are enhanced in estuarine fronts,which have brought about environmental problems.In this study,the seasonal variations of the estuarine fronts in the CRD were studied in wet(July)and dry(January)seasons in 2017,based on model simulations with high spatiotemporal resolutions using the Finite-Volume Community Ocean Model(FVCOM).The estuarine front included several sharp fronts with a SG>4(/500 m),and was bottom-trapped on the submerged delta front.Seasonal changes mainly occurred off the Jiangsu coast,where a significant estuarine front was formed in July.The estuarine fronts generated around the submerged delta topography were accompanied by the offshore extension of older estuarine fronts,which were diluted and evolved into plume fronts over a tidal cycle.The simulated estuarine fronts had a salinity range of 6 to 22 in the dry season and 6 to 14 in wet season 2017.The estuarine fronts hindered the residual current by altering its flow direction to the southeast.

文摘Nickel-based superconductors have attracted great attention due to the finding of the Ruddlesden-Popper(R-P)bilayer nickelate La_(3)Ni_(2)O_(7)with superconducting critical temperature(T_(c))of 80 K at pressure above 14 GPa.Recent efforts have been devoted to the study of La_(2)PrNi_(2)O_(7),while the detailed structure remains unclear.In this work,we explore the stability and physical properties of such an interesting system by using density functional theory and the U parameter simulation method implemented in VASP.The results show that the enthalpy of La_(2)PrNi_(2)O_(7)is slightly larger than its parent material bilayer R-P nickelate La_(3)Ni_(2)O_(7).The electronic structure analysis indicates that near the Fermi level,the eg orbit of Ni dominates and strongly hybridizes with the 2p orbit of O,thereby forming a significant van Hove singularity that is conducive to superconductivity.The Amam phase to the I4/mmm phase occurs,accompanied by an increase in the bandwidth of Ni 3d_(z)_(2) and an enhancement of the bonding-antibonding splitting(from about 0.5 eV to 1.5 eV),which leads to an increase in the density of states at the Fermi level.Our findings provide insights into the preparation and superconductivity of R-P bilayer nickelate.
文摘<strong>Background:</strong> Cisplatin, a chemotherapeutic agent, is widely used in the treatment of malignant tumors. Nephrotoxicity, especially acute kidney injury (AKI), is the most common and severe adverse reaction of cisplatin. Resveratrol and ginsenoside Rg1, two natural products, have been found to have renal protective effects. However, the effects and the mechanisms in cisplatin-induced AKI need further investigation. <strong>Methods:</strong> The mouse models of cisplatin-induced AKI and several treatment groups were established. Male C57BL/6 mice were divided into five groups: saline control group, cisplatin injury group, resveratrol treatment group, Rg1 treatment group, resveratrol and Rg1 combined treatment group. Serological analysis of serum urea nitrogen was aimed to reflect the function of kidney, and histological analysis of renal tissue sections was aimed to assess the damage of proximal convoluted tubules. The expression levels of autophagy-related proteins Beclin 1 and LC3 were detected by western blotting and qRT-PCR respectively. <strong>Results:</strong> The renal function was improved and renal damage was alleviated in Rg1 and resveratrol alone or combined treatment groups compared with the cisplatin injury group. For the mechanism, treatment with Rg1 and resveratrol alone or in combination decreased the expressions of Beclin 1 both at protein and mRNA levels, decreased LC3II/I protein levels, indicating that autophagy was inhibited by treatment with Rg1 and resveratrol alone or in combination. <strong>Conclusion:</strong> Resveratrol and Rg1 alleviated the kidney damage caused by cisplatin, and reduced autophagy was involved in the renoprotective effects of resveratrol and Rg1 against cisplatin-induced AKI. This study may provide new evidence to alleviate cisplatin-induced AKI.
文摘Tumor is a serious disease that threatens human health and has a high mortality. Chemotherapy is the most commonly used treatment, but it has a lot of side effects due to its toxicity. It has been found that ginsenosides exert an effective antitumor role. Ginsenosides are a class of triterpenoid saponins primarily found in the plant genus Panax. Many monomer components are studied, the most often investigated are Rg3, and Rh2, etc. Reports have shown that ginsenosides can inhibit tumor cells by suppressing proliferation and metastasis, and promoting apoptosis. In addition, ginsenosides can enhance sensitivity to conventional chemotherapeutic drugs. In this review, the recent articles about anti-tumor of ginsenosides were reviewed to promote the further development of anti-tumor therapy.
文摘Background: Adult hemophagocytic syndrome is a critical condition that is often difficult to diagnose and results in a bad prognosis because of the lack of effective and unified treatment. Aim: To investigate the clinical, diagnosis and treatment of hemophagocytic syndrome. Case Presentation: A 22-year-old female patient received a splenectomy, and the spleen was sent to the King Med Center of Medical Inspection. The results confirmed the diagnosis of hemophagocytic syndrome related to EB (Epstein-Barr) virus infection. Conclusion: Early diagnosis and early intervention are important factors for affecting the progression of the disease and improving its prognosis.
文摘Pulsed power technology,whereas the electrical energy stored in a relative long period is released in much shorter timescale,is an efficient method to create high energy density physics(HEDP)conditions in laboratory.Around the beginning of this century,China Academy of Engineering Physics(CAEP)began to build some experimental facilities for HEDP investigations,among which the Primary Test Stand(PTS),a multi-module pulsed power facility with a nominal current of 10 MA and a current rising time~90 ns,is an important achievement on the roadmap of the electro-magnetically driven inertial confinement fusion(ICF)researches.PTS is the first pulsed power facility beyond 10 TW in China.Therefore,all the technologies have to be demonstrated,and all the engineering issues have to be overcome.In this article,the research outline,key technologies and the preliminary HEDP experiments are reviewed.Prospects on HEDP research on PTS and pulsed power development for the next step are also discussed.
文摘Since abnormal post-translational modifications or gene mutations of tau have been detected in over twenty neurodegenerative disorders, tau has attracted widespread interest as a target protein. Among its various post-translational modifications, phosphorylation is the most extensively studied. It is recognized that tau hyperphosphorylation is the root cause of neurodegeneration in Alzheimer's disease (AD); however, it is not clear how it causes neurodegeneration. Based on the findings that tau hyperphosphorylation leads to the escape of neurons from acute apoptosis and simultaneously impairs the function of neurons, we have proposed that the nature of AD neurodegeneration is the consequence of aborted apoptosis induced by tau phosphorylation. Therefore, proper manipulation of tau hyperphosphorylation could be promising for arresting AD neurodegeneration. In this review, the neuroprotective and neurodegenerative effects of tau hyperphosphorylation and our thoughts regarding their relationship are presented.

文摘Hyperphosphorylated tau is the major protein component of neurofibrillary tangles in the brains of patients with Alzheimer’s disease(AD). However, the mechanism underlying tau hyperphosphorylation is not fully understood. Here, we demonstrated that exogenously expressed wild-type human tau40 was detectable in the phosphorylated form at multiple AD-associated sites in cytoplasmic and nuclear fractions from HEK293 cells.Among these sites, tau phosphorylated at Thr205 and Ser214 was almost exclusively found in the nuclear fraction at the conditions used in the present study. With the intracellular tau accumulation, the Ca2+concentration was significantly increased in both cytoplasmic and nuclear fractions. Further studies using site-specific mutagenesis and pharmacological treatment demonstrated that phosphorylation of tau at Thr205 increased nuclear Ca2+concentration with a simultaneous increase in the phosphorylation of Ca2+/calmodulin-dependent protein kinase IV(Ca MKIV) at Ser196. On the other hand, phosphorylation of tau at Ser214 did not significantly change the nuclear Ca2+/Ca MKIV signaling. Finally, expressing calmodulin-binding protein-4 that disrupts formation ofthe Ca2+/calmodulin complex abolished the okadaic acidinduced tau hyperphosphorylation in the nuclear fraction.We conclude that the intracellular accumulation of phosphorylated tau, as detected in the brains of AD patients, can trigger nuclear Ca2+/Ca MKIV signaling, which in turn aggravates tau hyperphosphorylation. Our findings provide new insights for tauopathies: hyperphosphorylation of intracellular tau and an increased Ca2+concentration may induce a self-perpetuating harmful loop to promote neurodegeneration.

文摘The activity levels of long-lived radionuclides in minerals have received more and more concern for the public health. The inductively coupled plasma mass spectrometry was used to measure the content of uranium and thorium in 60 mineral samples collected from 16 mines of seven provinces in China. The contents of uranium and thorium ranged0.17 ± 0.04 lg g^(-1)to 15.3 ± 2.39 lg g^(-1), and 0.19 ± 0.04 lg g^(-1)to 19.6 ± 7.56 lg g^(-1), respectively. The highest levels of U and Th contents were found in aluminum ore,whereas the lowest was found in antimony and copper ores.

文摘Objective:To observe the effect of the early intervention with moxibustion at "Shènshū(肾俞BL23)" on learning and memory of Alzheimer’s disease(AD) rats modeled by ovariectomy and D-Galactose(D-Gal)injection and the effect on Ca2+/CaMKII/CREB signaling pathway so as to provide the evidence for the acupoint selection in the early intervention with acupuncture and moxibustion for AD.Methods:Sixty female SD rats at the age of 3 months were selected and randomly divided into a normal group,a sham-operation group,a model group,a "BL23" group,a "Wèishū"(胃俞BL21) group,and a non-acupoint group,10 rats in each group.Except in the normal group and the sham-operation group,bilateral ovaries were removed in the other 4 groups.3 days after the operation,D-Gal was injected intraperitoneally at a dose of 150 mg/kg/d for 90 days to establish AD rat model.In the sham-operation group,only the peripheral fat of ovarian was removed and 0.9% normal saline was injected intraperitoneally of the same dose for 90 days.In the "BL23" group,the "BL21" group and the non-acupoints group,since the 2 nd day after ovariectomy,moxibustion was applied at 9:00 a.m.every day in the rats.The rats were fixed on a fixed table,the mild moxibustion with self-prepared moxa stick was applied to bilateral "BL23""BL21" and the non-acupoint of each rat,the ignited end of moxa stick was 2 cm to 3 cm far from skin surface.The infrared thermometer was adopted to measure the temperature of the epidermis of each acupoint from time to time so as to maintain the local temperature to be(41 ± 0.5)℃.Mild moxibustion lasted for 10 min each time,consecutively for 5 days a week and at an interval of 2 days.The total consecutive intervention duration was 12 weeks.In the normal group and the shamoperation group,no moxibustion intervention was performed,but the rats were restricted in a same way during the experiment.At the end of modeling and intervention,water maze test was adopted to detect the learning and memory abilities of rats in each group.The western blotting was used to detect the expressions of p-CREB,p-CaMKII,CaMKII and CREB in hippocampus of rats in each group.Results:(1)Navigation test:since Day 72,compared with the sham-operation group,the escape latencies in all of the consecutive 5 days were longer in the model group(all P <0.05).Since Day 73,compared with the normal group,the escape latency in all of the consecutive 4 days was shorter in the "BL23"group(all P <0.05).(2) Times across platform:compared with the normal group,the number of times consecutively crossing the platform was lower in the model group(P <0.05).After moxibustion,compared with the model group,the number of times across platform was higher in the "BL23"group,indicating a statistical significance(P <0.05).(3)Moving track of water maze spatial probe:the "tendency"strategy was displayed in the normal group and the sham operation group.The "marginal" strategy,which is commonly seen in the AD rats,was displayed in the model group.The "random" strategy was displayed in the "BL23" group.The "marginal" strategy was presented in the "BL21" group and the non-acupoint group.(4) Expressions of hippocampal P-CAMKII,CAMKII,CREB and P-CREB in the rats:compared with the normal group,the phosphorylation degrees of CAMKII and CREB were significantly reduced(both P<0.05) in the model group.In 12 weeks of moxibustion,compared with the model group,the levels of P-CAMKII and P-CREB were increased in the "BL23" group and the "BL21" group(all P <0.05) and the levels in the "BL23" group were higher than that of the "BL21" group and the non-acupoint group(all P<0.05).Conclusion:Moxibustion at "BL23" achieves a better effect in improving the learning and memory of AD rats modeled by ovariectomy and D-Gal injection as compared with moxibustion at "BL21" and the non-acupoint.The effect mechanism is potentially related to the effective activation of Ca2+/CaMKII/CREB signaling pathway.

文摘Human embryonic stem cells (hESCs) can self-renew indefinitely and differentiate into all cell types in the human body. Therefore, they are valuable in regenerative medicine, human developmental biology and drug discovery. A number of hESC lines have been derived from the Chinese population, but limited of them are available for research purposes. Here we report the derivation and characterization of two hESC lines derived from human blastocysts of Chinese origin. These hESCs express alkaline phosphatase and hESC-specific markers, including Oct4, Nanog, SSEA-3, SSEA-4, TRA- 1-60 and TRA-1-81. They also have high levels of telomerase activity and normal karyotypes. These ceils can form embryoid body in vitro and can be differentiated into all three germ layers in vivo by teratoma formation. The newly established hESCs will be distributed for research purposes. The availability of hESC lines from the Chinese population will facilitate studies on the differences in hESCs from different ethnic groups.

文摘A splicing mutation in VPS4B can cause dentin dysplasia type I(DD-I),a hereditary autosomal-dominant disorder characterized by rootless teeth,the etiology of which is genetically heterogeneous.In our study,dental follicle cells(DFCs)were isolated and cultured from a patient with DD-I and compared with those from an age-matched,healthy control.In a previous study,this DD-I patient was confirmed to have a loss-of-function splicing mutation in VPS4B(IVS7+46C>G).The results from this study showed that the isolated DFCs were vimentin-positive and CK14-negative,indicating that the isolated cells were derived from the mesenchyme.DFCs harboring the VPS4B mutation had a significantly higher proliferation rate from day 3 to day 8 than control DFCs,indicating that VPS4B is involved in cell proliferation.The cells were then replenished with osteogenic medium to investigate how the VPS4B mutation affected osteogenic differentiation.Induction of osteogenesis,detected by alizarin red and alkaline phosphatase staining in vitro,was decreased in the DFCs from the DD-I patient compared to the control DFCs.Furthermore,we also found that the VPS4B mutation in the DD-I patient downregulated the expression of osteoblast-related genes,such as ALP,BSP,OCN,RUNX2,and their encoded proteins.These outcomes confirmed that the DD-I-associated VPS4B mutation could decrease the capacity of DFCs to differentiate during the mineralization process and may also impair physiological root formation and bone remodeling.This might provide valuable insights and implications for exploring the pathological mechanisms underlying DD-I root development.

文摘Current characteristics and vertical variations during summer and autumn in the southern East China Sea were investigated by measuring current profile, tide, wind, and wave data for 90 d from July 28 to October 25, 2015. Our results are:(1) The current was mainly a(clockwise) rotating flow, displaying reciprocating flow characteristics,and vertically the current directions were the same throughout the vertical profile.(2) The horizontal current speed was strongest during August(summer) with an average speed of 51.8 cm/s. The average current speeds during spring tides were highest in August and weakest in September, with speeds of 59.9 and 42.8 cm/s,respectively.(3) Considerable differences exist in average current speeds in different layers and seasons. The highest average current speeds were found in the middle–upper layers in August and in the middle–lower layers in September and October.(4) The residual current speed was highest in August, when the speed was 12.5–47.1 cm/s,whereas the vertical average current speed was 34.3 cm/s. The depth-averaged residual current speeds in September and October were only 50% of that in August, and the residual current direction gradually rotated in a counter-clockwise direction from the lower to surface layers.(5) Typhoon waves had a significant influence on the currents, and even affected the middle and lower water layers at depths of >70.0 m. Our results showed that the currents are controlled by the dynamic interplay of the Taiwan Warm Current, incursion of the Kuroshio Current onto the continental shelf, and monsoonal changes.

文摘The research of human facial age estimation(AE)has attracted increasing attention for its wide applications.Up to date,a number of models have been constructed or employed to perform AE.Although the goal of AE can be achieved by either classification or regression,the latter based methods generally yield more promising results because the continuity and gradualness of human aging can naturally be preserved in age regression.However,the neighbor-similarity and ordinality of age labels are not taken into account yet.To overcome this issue,the cumulative attribute(CA)coding was introduced.Although such age label relationships can be parameterized via CA coding,the potential relationships behind age features are not incorporated to estimate age.To this end,in this paper we propose to perform AE by encoding the potential age feature relationships with CA coding via an implicit modeling strategy.Besides that,we further extend our model to gender-aware AE by taking into account gender variance in aging process.Finally,we experimentally validate the superiority of the proposed methodology.
文摘The climate change in Shandong Peninsula, China was analyzed in this paper by the non-parametric Mann-Kendall test, Accumulated Difference Curve and Order Cluster Analysis methods, based upon the datas of annual mean, maximum and minimum temperature and annual precipitation, precipitation from June to September over the past 50 years. Results obtained showed a number of observations: 1) The annual mean temperature of Shandong Peninsula showed a significant increasing trend, with a distinct abrupt change point detected around 1990, during the past 5 decades. The warming of the Peninsula over the last 50 years was due mainly to the significant increase of annual minimum temperature. The annual maximum temperature demonstrated a mixed trend of decreasing and increasing, but was statistically insignificant, and no abrupt change was detected;2) The annual precipitation exhibited a decreasing trend during the past 5 decades, with an abrupt change detected around 1980 at most stations;but there was an earlier transition point at 1966, at a few stations. The reduction in precipitation, from June to September, was responsible mainly for the decrease of annual precipitation. Besides, the proportion of the June-September precipitation in the year declined slightly over the last 50 years;3) In comparison, the temperature evolution in Shandong Peninsula was basically consistent with most parts of China, but warmed at a faster rate over the same period;the decreasing trend of precipitation was more significant compared with the other climate zones of China. Within the Peninsula, the abrupt change of temperature and precipitation in the Southeast was earlier than that in the Northwest;the reduction of precipitation was larger in the Southeast while the increase of temperature was more significant in the Northwest. This research was of great importance to understand the climate change and its environmental effects in the coastal zone.

文摘Copy number variation (CNV) is a type of genetic variation which may have important roles in phenotypic variability and disease susceptibility. To hunt for genetic variants underlying human height variation, we performed a genome wide CNV association study for human height in 618 Chinese unrelated subjects using Affymetrix 500K array set. After adjusting for age and sex, we found that four CNVs at 6p21.3, 8p23.3-23.2, 9p23 and 16p12.1 were associated with human height (with borderline significant p value: 0.013, 0.011, 0.024, 0.049; respectively). However, after multiple tests correction, none of them was associated with human height. We observed that the gain of copy number (more than 2 copies) at 8p23.3-23.2 was associated with lower height (normal copy number vs. gain of copy number: 161.2 cm vs. 153.7 cm, p = 0.011), which accounted for 0.9% of height variation. Loss of copy number (less than 2 copies) at 6p21.3 was associated with 0.8% lower height (loss of copy number vs. normal copy number: 154.5 cm vs. 161.1 cm, p = 0.013). Since no important genes influencing height located in CNVs at loci of 8p23.3-23.2 and 6p21.3, the two CNVs may cause the structural rear- rangements of neighbored important candidate genes, thus regulates the variation of height. Our results expand our knowledge of the genetic factors underlying height variation and the biological regulation of human height.
文摘In order to meet the training needs of Partial Discharge (PD) detection personnel, a PD detection training simulator is developed. The composition of the system and the functions of the modules are described, then the three-dimensional graphic engine based on Open Scene Graph (OSG) and the way of scene organization based on hierarchical tree are introduced, moreover, the methods of fault simulation and detection instrument simulation based on algorithm component library are presented. Application shows that the system can provide various detection schemes, and it is an ideal training tool for the detection personnel.
