Background and Aims:Despite advancements in diagnostic and therapeutic strategies,hepatocellular carcinoma(HCC)remains a leading cause of cancer-related mortality.Antioxidant-1(ATOX1)has been implicated in oncogenic p...Background and Aims:Despite advancements in diagnostic and therapeutic strategies,hepatocellular carcinoma(HCC)remains a leading cause of cancer-related mortality.Antioxidant-1(ATOX1)has been implicated in oncogenic processes across various cancer types;however,its specific role in HCC remains unclear.This study aimed to investigate the function of ATOX1 and its underlying molecular mechanisms in HCC.Methods:Immunohistochemical analysis was conducted to assess ATOX1 expression in HCC tissues.Cell Counting Kit-8,colony formation,Transwell migration,flow cytometry,and reactive oxygen species(ROS)assays were employed to evaluate the malignant behaviors of tumor cells.A xenograft mouse model was employed to assess the effects of ATOX1 knockdown on tumor growth in vivo.DCAC50 treatment was performed to inhibit the copper transport function of ATOX1.RNA sequencing was conducted to explore the potential molecular mechanisms of ATOX1 in HCC.Results:ATOX1 expression was significantly elevated in HCC tumor tissues.ATOX1 promoted cell proliferation,colony formation,and migration.Knockdown of ATOX1 suppressed tumor growth in vivo.Mechanistically,ATOX1 activated c-Myb,and thus enhanced the malignant phenotype of HCC cells via activation of the PI3K/AKT signaling pathway.Additionally,ATOX1 reduced intracellular copper accumulation and inhibited ROS production and apoptosis.Inhibition of ATOX1 by DCAC50 decreased cell proliferation while increasing ROS levels and apoptosis in HCC cells.Notably,acetylcysteine reversed the reduction in c-Myb expression induced by ATOX1 knockdown.Conclusions:ATOX1 may promote HCC carcinogenesis through the activation of the c-Myb/PI3K/AKT pathway and the inhibition of copper accumulation and oxidative stress.展开更多
The Trachinotus ovatus is a popular aquaculture species in China.There are no obvious morphological differences between male and female fish,even during maturity,prompting research studies on sex-related features of t...The Trachinotus ovatus is a popular aquaculture species in China.There are no obvious morphological differences between male and female fish,even during maturity,prompting research studies on sex-related features of this fish.To examine sex determination-and gonadal development-related genes,we conducted transcriptome analysis of the ovaries and testes of T.ovatus.A total of 345,972,132 high-quality clean reads were obtained from 12 libraries.In addition,28,137 gonad-expressed unigenes were obtained by mapping the clean reads to the T.ovatus reference genome.A total of 8,237 differentially expressed genes(DEGs)were identified between stage I ovaries and testes,including 3,235 testicular upregulated and 5,002 ovarian upregulated genes.Furthermore,13,448 DEGs were obtained between stage III ovarian and testicular libraries,including 7,576 testicular upregulated and 5,872 ovarian upregulated genes.The DEGs included some sex-determining genes such as sry,dmrt1,and amh.DEGs between ovarian and testicular libraries were significantly enriched with KEGG pathways that are involved in gonadal development,sex determination,and gonadal function.Then,10 DEGs(seven testicular upregulated and three ovarian upregulated unigenes)were selected for quantitative real-time PCR analysis.Four genes(zinc-binding protein A33-like,pro-opiomelanocortin-like,leucine-rich repeat and transmembrane domain-containing protein 2-like,and forkhead boxC1)showed a specific testicular expression pattern.The gonadally expressed as well as testicular and ovarian DEGs provide useful information for further research on the reproductive biology of T.ovatus.展开更多
The micro turbine(MT)is scheduled to address wind power uncertainties and thus lead to greater variations in the state of natural gas system(NGS).To meet this challenge,first,a dynamic model of a gas-electricity combi...The micro turbine(MT)is scheduled to address wind power uncertainties and thus lead to greater variations in the state of natural gas system(NGS).To meet this challenge,first,a dynamic model of a gas-electricity combined system with wind power is established,the dynamic model of the NGS is based on differential equations,so that it can easily connect with the MT model;and a droop control module is added to the Rowen's MT model,so that the improved model can adjust the output power according to the scheduling instructions.Then,a stability control method based on the variable regulation ratio gas pressure regulator is proposed,the essence of this method is to reduce the variation of natural gas storage in the gas pipeline,so as to ensure the stability of pressure.The simulation results show that the dynamic model is correct and the stability control method is effective.展开更多
