High-risk extranodal natural killer/T-cell lymphoma patients could benefit more from allogeneic hematopoietic stem cell transplantation as consolidation:A real-world multicenter analysis in China

Objective:Both allogeneic hematopoietic stem cell transplantation(allo-HSCT)and autologous HSCT(ASCT)are important therapies for extranodal natural killer/T-cell lymphoma(ENKTCL);however,no large-scale,multicenter study has compared the efficacy and safety between allo-HSCT and ASCT in these patients.Our multicenter,real-world study aimed to evaluate the outcomes of allo-HSCT vs.ASCT as consolidation in ENKTCL patients who had achieved a complete response(CR)or partial response(PR).Methods:This was a multicenter,retrospective study with nine hospitals in China,and 114 patients with ENKTCL were enrolled.Sixty patients received ASCT and 54 received allo-HSCT.The primary outcome was progression-free survival(PFS).In the sensitivity analysis,propensity score matching(PSM)analyses were conducted to adjust for baseline prognostic factors.Landmark analysis were conducted to minimize immortal-time bias.Results:Patients in the allo-HSCT group presented with more adverse prognostic factors.Allo-HSCT group showed a significantly better PFS and a lower disease progression rate compared with ASCT group in patients with Ann Arbor stageⅢ/Ⅳdisease(PFS:100%vs.82.0%,P=0.023;disease progression rate:0 vs.25.4%,P=0.024),those with intermediate/high prognostic index of natural killer lymphoma(PINK)scores(PFS:100%vs.84.4%,P=0.034;disease progression rate:0 vs.22.1%,P=0.034),those with intermediate/high international prognostic index(IPI)scores(PFS:100%vs.82.0%,P=0.038;disease progression rate:0 vs.25.4%,P=0.038),or those receiving HSCT at PR(PFS:100%vs.50%,P=0.046;disease progression rate:0 vs.50%,P=0.046)at the 1.5-4.0 follow-up.In multivariate analysis,receiving ASCT was significantly associated with a poorer PFS[hazard ratio(HR)=2.23,P=0.038]and overall survival(OS)(HR=2.45,P=0.045).In the sensitivity analysis,patients receiving allo-HSCT showed a significantly better PFS(70.3%vs.39.1%,P=0.039),OS(73.9%vs.42.0%,P=0.044),and a lower disease progression rate(22.6%vs.57.0%,P=0.017)compared with those receiving ASCT after propensity score matching.Conclusions:ENKTCL patients with high-risk characteristics could benefit more from allo-HSCT as consolidation. 展开更多

Background:Dihydrolipoamide S-acetyltransferase(DLAT)is a subunit of the pyruvate dehydrogenase complex(PDC),a rate-limiting enzyme complex,that can participate in either glycolysis or the tricarboxylic acid cycle(TCA).However,the pathogenesis is not fully understood.We aimed to perform a more systematic and comprehensive analysis of DLAT in the occurrence and progression of tumors,and to investigate its function in patients’prognosis and immunotherapy.Methods:The differential expression,diagnosis,prognosis,genetic and epigenetic alterations,tumor microenvironment,stemness,immune infiltration cells,function enrichment,single-cell analysis,and drug response across cancers were conducted based on multiple computational tools.Additionally,we validated its carcinogenic effect and possible mechanism in glioma cells.Results:We exhibited that DLAT expression was increased in most tumors,especially in glioma,and affected the survival of tumor patients.DLAT was related to RNA modification genes,DNA methylation,immune infiltration,and immune infiltration cells,including CD4+T cells,CD8+T cells,Tregs,and cancer-associatedfibroblasts.Single-cell analysis displayed that DLAT might regulate cancer by mediating angiogenesis,inflammation,and stemness.Enrichment analysis revealed that DLAT might take part in the cell cycle pathway.Increased expression of DLAT leads tumor cells to be more resistant to many kinds of compounds,including PI3Kβinhibitors,PKC inhibitors,HSP90 inhibitors,and MEK inhibitors.In addition,glioma cells with DLAT silence inhibited proliferation,migration,and invasion ability,and promoted cell apoptosis.Conclusion:We conducted a comprehensive analysis of DLAT in the occurrence and progression of tumors,and its possible functions and mechanisms.DLAT is a potential diagnostic,prognostic,and immunotherapeutic biomarker for cancer patients. 展开更多

Colorectal cancer causes the third most common type of malignant tumors with high morbidity and mortality.Chemotherapy is currently one of the most effective and common treatments for colorectal cancer.However,the poor water solubility of some chemotherapeutics,untargeted drug delivery,and the undesirable systemic side effects of conventional treatment remain the major issues for colorectal cancer chemotherapy.Fortunately,drug delivery systems(DDS)based on biomaterials have been widely investigated and found to be capable of resolving those issues with good performance.Therefore,the main goal of this review is to summarize and discuss the progress and potential advantages of different DDS for colorectal cancer chemotherapy.We not only reviewed the nanocarriers used to improve the solubility of chemotherapeutics,including liposomes,micelles,and nanoparticles,but also discussed targeted DDS based on specific ligand-receptor recognition and tumor microenvironmental stimulus responses.Furthermore,locally administered systems based on hydrogels and microspheres,which have been shown to increase drug accumulation at the tumor site while decreasing systemic toxicity,were also emphasized.DDS provides a good option for improving the efficacy of chemotherapy in the treatment of colorectal cancer. 展开更多

Background: Food is an important environmental factor that affects animals' energy metabolism and food shortage has significant effects on animals' behavior, physiology and biochemistry. However, to date few studies have focused on the thermogenesis and its effects on the body condition of birds. In this study, we examined the effects of food restriction on the body mass, basal metabolic rate(BMR) and body composition, and several physiological, biochemical and molecular markers potentially related to thermogenesis, in the Chinese Bulbul(Pycnonotus sinensis).Methods: Birds in the control group were provided with food ad libitum whereas those in the food restriction group were provided with one-half of the usual quantity of food for 12 days. Oxygen consumption was measured using an open-circuit respirometry system. Mitochondrial state 4 respiration and cytochrome c oxidase(COX) activity in the liver and pectoral muscle were measured with a Clark electrode. Avian uncoupling protein(avUCP) mRNA expression was determined in pectorals muscle with quantitative Real-time PCR.Results:Chinese Bulbuls in food restriction group decreased in body mass,BMR and internal organ(heart,kidneys,small intestine and total digestive tract)mass compared with the control group over the 12-day period of food restriction.Bulbuls in the food restriction group also had lower levels of state-4 respiration,COX activity in the liver and muscle,and mitochondrial avUCP gene expression in muscle compared to the control group.BMR was positively correlated with body mass,state 4 respiration in the liver and COX activity in the muscle.Conclusions:Our data indicate that Chinese Bulbuls not only sustain food shortage through simple passive mechanisms,such as reducing body and organ mass and energy expenditure,but also by reducing energetic metabolism in the liver and muscle. 展开更多

In this study,the hydrogel network was reinforced by covalent-like hydrogen bonding,and the strong binding ability of boron-nitrogen coordination served as the main driving force.Among them,acrylamide(AM)and 3-acrylamidophenylboronic acid(AAPBA)were the main body,and the numerous hydroxyl groups in the trehalose(Treh)molecule and other polymer groups formed strong hydrogen bonding interactions to improve the mechanical properties of the PAM/PAAPBA/Treh(PAAT)hydrogel and ensured the simplicity of the synthesis process.The hydrogel possessed high strain at break(1239%),stress(64.7 kPa),low hysteresis(100%to 500%strain,corresponding to dissipation energy from 1.37 to 7.80 kJ/m^(3)),and outstanding cycling stability(retained more than 90%of maximum stress after 200 ten-sile cycles).By integrating carbon nanotubes(CNTs)into PAAT hydrogel(PAATC),the PAATC hydrogel with excellent strain response performance was successfully constructed.The PAATC conductive hydro-gel exhibited high sensitivity(gauge factor(GF)=10.58 and sensitivity(S)=0.304 kPa^(-1)),wide strain response range(0.5%-1000%),fast response time(450 ms),and short recovery time(350 ms),excellent fatigue resistance,and strain response stability.Furthermore,the PAATC-based triboelectric nanogener-ator(TENG)displayed outstanding energy harvesting performance,which shows its potential for appli-cation in self-powered electronic devices. 展开更多

Multiple myeloma(MM)is the second most common hematological tumor characterized by the proliferation of monoclonal plasma cells.Melphalan(MEL)is commonly used in the treatment of MM and is especially essential for patients undergoing autologous stem cell transplantation(ASCT).Although many drugs for MM have been developed in recent years,chemotherapy followed by ASCT remains the optimal option.Melphalan,the backbone of the conditioning regimen,brings severe toxicities at a high dose.Nanodrug delivery systems enable drugs to be highly effective and have low toxicity.In this study,methoxy poly(ethylene glycol)-poly(D,L-lactide)copolymer(MPEG-PDLLA)was chosen to encapsulate melphalan,and the characteristics,effectiveness,and safety of MEL/MPEG-PDLLA in vitro and in vivo were investigated.MEL/MPEG-PDLLA showed slow release and was easily engulfed by MM cells despite a result of the antitumor assay comparable to that of free melphalan in vitro.The in vivo results showed that MEL/MPEG-PDLLA could significantly alleviate tumor burden and prolong survival time without increasing the toxicity to vital organs.In addition,MEL/MPEG-PDLLA could significantly reduce the damage to the intestinal mucosa caused by melphalan.In conclusion,MEL/MPEG-PDLLA shows improved antitumor activity and has the potential to alleviate pains of MM patients undergoing ASCT. 展开更多

Nanoparticles show greatly potential for therapeutic studies due to the sophisticated surface which can mimic the functionality of protein. Herein, we design the bis(pyrene)-Lys-Leu-Val-Phe-PheGly-poly ethylene glycol(BP-KLVFFGPEG) based nanoparticles, which capture Aβthrough recognition and co-assembly of KLVFF. The resulting co-assemblies show different morphologies, such as large aggregates, nanoparticles and nanofibers corresponding to the different length chain of PEG, leading to different capture efficiencies. Human adenoviruses(HAdVs)are highly contagious pathogens with various genotypes implicated in acute respiratory disease(ARD)and linked to fatality,especially in immunosuppressed patients,young children,and military recruits.Currently,no vaccines or specific drugs are approved for clinical use.The hosts of adenoviruses are strictly species-specific,which strongly limits the development of vaccines and drugs against HAdVs.In this study,immunocompetent BALB/c mice were challenged with different doses of human adenovirus type 5(HAdV-5)via tail intravenous injection(i.v.).All mice challenged with a high dose of HAdV-5(3.21010 TCID50/kg)died within 3–5 days,while those receiving a low dose of HAdV-5(8109 or 4109 TCID50/kg)survived.Interestingly,among the mice receiving a medium dose of HAdV-5(1.61010 TCID50/kg),60%(n¼3/5)of male mice died,while all female mice survived.This suggests that male mice may be more susceptible to HAdV-5 infection than female mice,consistent with clinical findings in children.HAdV-5 DNA was mainly distributed in the liver,followed by the spleen and lung.Pathological changes were observed in the lung,liver,and spleen,with severity increasing in correlation with the virus challenge dosage.Transcriptome and qPCR analyses of the liver indicated that the down-regulated expression of the H2-Aa,H2-Ea-ps,CD74,and H2-Eb1 genes in male mice,as well as the AHR gene in female mice,may contribute to the observed higher mortality rates in male mice.Therefore,this effective,feasible,and costefficient mouse model could serve as a candidate for evaluating HAdV vaccines and anti-adenovirus therapeutics. 展开更多

Among their natural scenery and folk custom, there are various tourist resources could be developed. Developing sport resource which is in the Yunnan section of the Silk Road will be important for boosting Yunnan' s tourism. After opening up its tourism, the sport tourism could be transformed into economic and industrial superiority, and then improves the whole economic development of Yunnan areas, achieving a coordinated development for Yunnan' s tourism and sport industry. The article analyses and studies the present situation of development which connected with the sport resource in the Yunnan section of the Silk Road. Chimeric antigen receptor T(CAR-T)cells have reshaped the treatment landscape of hematological malignancies,offering a potentially curative option for patients.Despite these major milestones in the field of immuno-oncology,growing experience with CAR-T cells has also highlighted several limitations of this strategy.The production process of CAR-T cells is complex,time-consuming,and costly,thus leading to poor drug accessibility.The potential carcinogenic risk of viral transfection systems remains a matter of controversy.Treatment-related side effects,such as cytokine release syndrome,can be life-threatening.And the biggest challenge is the inadequate efficacy related to poor infiltration and retention of CAR-T cells in tumor tissues and impaired T cell activation caused by the immunosuppressive tumor microenvironment(TME).Innovative strategies are urgently needed to address these problems,and nanomedicine offers good solutions to these challenges.In this review,we provide a comprehensive summary of recent advancements in the application of nanomaterials to enhance CAR-T cell therapy.We examine the role of innovative nanoparticle-based delivery systems in the production of CAR-T cells,with a particular focus on polymeric delivery systems and lipid nanoparticles(LNPs).Furthermore,we explore various strategies for delivering immune stimulators,which significantly enhance the efficacy of CAR-T cells by modulating T cell viability and functionality or by reprogramming the immunosuppressive TME.In addition,we discuss several novel therapeutic approaches aimed at mitigating the adverse effects associated with CAR-T therapies.Finally,we offer an integrated perspective on the future challenges and opportunities facing CAR-T therapies. 展开更多

The advancement of the extraction of uranium(Ⅵ)(U(Ⅵ))from seawater holds significant strategic importance for the sustainable progression of nuclear energy.In this study,the photocatalyst heterojunction of graphene oxide/graphitic carbon nitride(GO/g-C_(3)N_(4))was encapsulated by the spinning of fiber to obtain a graphene oxide/graphitic carbon nitride/polyamide oxime(GO/g-C_(3)N_(4)/PAO)film with adsorption and photoreduction ability toward U(Ⅵ).The enhanced adsorption capacity of the GO/g-C_(3)N_(4)/PAO nanofiber was achieved through the synergistic effects of the aminoxime group in PAO and the photocatalytic performance of GO/g-C_(3)N_(4).The GO/g-C_(3)N_(4)/PAO composite exhibited an adsorption capacity of 190.7 mg g−1(pH=6,t=600 min,C0=99.9 mg L^(−1),and m/v=0.5 g L^(−1))under light conditions.GO/g-C_(3)N_(4)/PAO showed good recycling reproducibility following five cycles of adsorption.After 30 days of adsorption in natural seawater,the adsorption capacity of GO/g-C_(3)N_(4)/PAO for U(Ⅵ)was 10.39 mg g−1,representing a 38.6%increase compared to its performance in the absence of light.The favorable adsorption characteristics exhibited by the GO/g-C_(3)N_(4)/PAO composite suggest its potential as a viable option for the extraction of uranium from seawater. 展开更多

Gastric cancer is the fifth most common cancer and the third leading cause of cancer death worldwide,posing a severe threat to human health.Surgical resection remains the most preferred option for gastric cancer treatment.However,for advanced gastric cancer,the curative effect of surgical resection is usually limited by the local recurrence,peritoneal carcinomatosis,or distal metastasis.Intraoperative chemotherapy is an attractive in situ adjuvant treatment strategy to reduce the recurrence and metastasis after surgical resection.Here,we designed a 5-fluorouracil(5-FU)and cis-platinum(DDP)co-delivery system based on a biodegradable temperature-sensitive hydrogel(PDLLA-PEG-PDLLA,PLEL)for intraoperative adjuvant combination chemotherapy of gastric cancer.This 5-FU+DDP/PLEL hydrogel system characterized by a special sol-gel phase transition in response to physiological temperature and presented sustained drug release in vitro and in vivo.A strong synergistic cell proliferation inhibition and apoptosis promotion of 5-FU+DDP/PLEL were observed against gastric cancer MKN45-luc cells.After intraperitoneal injection,the dual-drug loaded hydrogel formulation showed superior anti-tumor effects than the single-drug carrying hydrogels and combination of free 5-FU and DDP on the gastric cancer peritoneal carcinomatosis model.The use of hydrogel for dual-drug delivery had benefited to fewer side effects as well.What’s more,we established a mouse model for postsurgical residual tumors and peritoneal carcinomatosis of gastric cancer,in which the intraoperative administration of 5-FU+DDP/PLEL also remarkably inhibited the local recurrence of the orthotopic tumors and the growth of the abdominal metastatic tumors,resulting in an extended lifetime.Hence,this developed dual-drug loaded hydrogel system has great potential in the intraoperative chemotherapy of gastric cancer,that suggests a clinically-relevant and valuable option for postsurgical management of gastric cancer. 展开更多

Efficient capture of uranium(VI)(U(VI))from seawater is of great significance to the sustainable development of nuclear energy and environmental protection,which is also a serious challenge at present.In this study,hollow Zn/Co zeolitic imidazolate framework(H-ZIF)was decorated on polyacrylamide/sodium alginate(PAM/SA)hydrogel by chelating and covalently crosslinking,and a new type of PAM/SA/H-ZIF hydrogel was synthesized.The combination of PAM/SA and H-ZIF gives PAM/SA/H-ZIF hydrogel excellent mechanical properties,good stability,and abundant surface functional groups,which is beneficial to improving the adsorption properties.The extraction amount of U(VI)by PAM/SA/H-ZIF is 171.14 mg·g^(-1)at C_(0)=99.52 mg·L^(-1)and pH=5.0.The adsorption equilibrium is reached in 120 min and the adsorption process fits well with Langmuir isotherm model and pseudo-second-order rate equation.The PAM/SA/H-ZIF also showed good recyclability and stability after 10 cycles of adsorption-desorption.More importantly,the rate of uranium adsorption is 0.196 mg·g^(-1)·day^(-1)after 30 days,which implies that the PAM/SA/H-ZIF could serve as a potential adsorbent for the development of uranium capture from seawater. 展开更多
