Drosophila melanogaster has been a popular model organism in the study of sleep and circadian rhythm.The Drosophila activity monitoring(DAM)system is one of the many tools developed for investigating sleep behavior in...Drosophila melanogaster has been a popular model organism in the study of sleep and circadian rhythm.The Drosophila activity monitoring(DAM)system is one of the many tools developed for investigating sleep behavior in fruit flies and has been acknowledged by researchers around the world for its simplicity and cost-effectiveness.Based on the simple activity data collected by the DAM system,a wide range of parameters can be generated for sleep and circadian studies.However,current programs that analyze DAM data cover a limited number of metrics and fail to provide individual data for the user to plot graphs and conduct analysis using other software.Therefore,we have developed SleepyFlyR,an R package that:(1)is simple and easy to use with a user-friendly user interface script;(2)provides a comprehensive analysis of sleep and activity parameters;(3)generates double-plotted graphs for sleep and activity patterns;(4)offers visualization of sleep and activity profiles across multiple days or within a single day;(5)calculates the changes of sleep and activity parameters between baseline and experiment;(6)stores both summary data and individual data in files with unique title.展开更多
Amyloid precursor protein(APP)can generate neurotoxic-amyloid 42(A 42)by proteolytic process,which plays a crucial role in the pathogenesis of Alzheimer’s disease(AD).Individuals with mild to moderate AD exhibit slee...Amyloid precursor protein(APP)can generate neurotoxic-amyloid 42(A 42)by proteolytic process,which plays a crucial role in the pathogenesis of Alzheimer’s disease(AD).Individuals with mild to moderate AD exhibit sleep disturbance,even before the onset of AD.The purpose of this study is to verify the effect of APP on sleep behavior by using an APP overexpressing Drosophila AD model.APP-overexpressed flies were grouped by age,and their sleep amounts were monitored.Our results demonstrated that APP overexpression had no impacts on sleep amounts in young(4-7 days after eclosion,4-7AE)flies.However,APP overexpression contributed to lower day and total sleep amounts in the middle-aged(11-14AE)flies.Moreover,old-aged(40AE)flies with overexpressing APP exhibited increased number of sleep bouts and decreased sleep time,indicating sleep fragmentation in these flies.Our results indicated that overexpression of APP in neurons has distinct effects on sleep behavior at different ages,but the specific mechanisms underlying the sleep regulation by APP are needed for further study.In addition,our data also suggest that sleep disturbance in AD animals can be caused by APP expression alterations,which provide a potential treating target for sleep intervention and therapy for AD patients.展开更多
[Objectives]This study was conducted to establish a method for the determination of aspartame in liquor by high performance liquid chromatography.[Methods]The liquor samples having volumes fixed with ultrapure water w...[Objectives]This study was conducted to establish a method for the determination of aspartame in liquor by high performance liquid chromatography.[Methods]The liquor samples having volumes fixed with ultrapure water were filtered by 0.22μm water phase,and then the content of aspartame in liquor was determined by high performance liquid chromatography using methanol(A)and ultrapure water(B)as mobile phase for isocratic elution.[Results]The results showed that a good linearity was obtained for the standard curve(R^(2)=99.98%)in the method,and its quantitative limit was 5.34 mg/kg.Its recovery was 91.26%,91.92%,and 90.55%,respectively.[Conclusions]The method has high sensitivity,high recovery and low quantitative limit,and it was a suitable method for the determination of aspartame in liquor.展开更多
