In frozen semen technology, about 50% of sperm died because of cryoinjury or cryodamage during the process of cryopreservation and thawing of the semen. These cryo-damages can be minimized by use of various cryoprotective agents or cold shock absorbers in the frozen semen technology. Of which, egg yolk (EY) is most primarily important in the extender preparation for various mammalian species for longer period and it works against the cold shock during the cryopreservation process due to the presence of low density lipoproteins (LDL). Further, EY contains substances other than LDL that affect the sperm quality parameters especially reduced motility and inhibit the respiration of sperm;therefore, there was heavy demand on replacement of the EY with the particular responsible substances (LDL) in the semen extender. Later on, various investigators tried to extract the LDL from the EY of hen and finally succeeded to extract the LDL from the EY of hen for semen preservation of various species. The concentration of LDL used in various species is varied and this may be due to the composition and concentration of phospholipids, cholesterol and its proportion in the sperm membrane. In bovine species, the concentration of LDL was standardized as 8% (w/v) on dry matter basis. This is equal to 20% EY used in conventional semen extender. As it is explained that the 20% EY contains 68% LDL (13.6 g) and on dry matter basis, it is approximately 60% (8.16 g). This calculation indicates 20% EY contains 8% LDL on dry matter basis. The LDL protects the sperm by various mechanisms to maintain the integrity of sperm membrane, which is explained in the present review. It was concluded that the investigation is still to be carried out to find out the exact roles of apoproteins and lipids of LDL and to indentify and isolate the detrimental substances presented in the whole EY.展开更多

Methods: A total of 160 semen samples were ...Objective: To assess the deleterious effects of foot and mouth disease vaccination on antioxidant profiles as well as oxidative stress in the semen of breeding mithun bulls. Methods: A total of 160 semen samples were collected from 8 adult healthy mithun bulls with good body condition score (5-6), which were maintained at semen collection centre, ICAR-National Research Centre on Mithun, Medziphema, Nagaland, India, twice a week, 4 weeks before vaccination (pre-vaccination stage) and 12 weeks after vaccination (post-vaccination stage) to understand and know the influence of vaccine stress on seminal antioxidant as well as oxidative stress profiles in mithun. Vaccine was injected at the end of week 4 and semen samples were collected & analysed up to week 16 of the investigation period. Results: Data analysis showed that foot and mouth disease vaccination has significantly (P<0.05) altered the seminal antioxidant, biochemical and oxidative stress profiles up to week 10th of vaccine administration. However, the immunised bulls were recovered in physical health status, semen production and its antioxidant profiles gradually. Conclusions: The results of the present study suggest that the semen sample collection as well as preservation should to be suspended up to week 10th of post immunization to get antioxidant and biochemical profiles in normal level in the semen to preserve normal conception rate in artificial breeding programme by using such semen in mithun species.展开更多
Objective:To optimize the concentration of PDC-109 protein in semen for higher cryopreservability and fertility by sequestration or neutralization of PDC-109 by its antibodies(anti-PDC-109 antibodies)in bubaline speci...Objective:To optimize the concentration of PDC-109 protein in semen for higher cryopreservability and fertility by sequestration or neutralization of PDC-109 by its antibodies(anti-PDC-109 antibodies)in bubaline species.Methods:PDC-109 protein was purified by applying two-step chromatography procedures.Purified protein was injected in rabbits to raise antibodies.These raised anti-PDC-109 antibodies were used in neutralization or sequestration of PDC-109 in in-vitro model.Ejaculates were collected from buffaloes and splited for four groups.Group 1 received egg yolk Tris glycerol extender,without anti-PDC-109 antibodies,while group 2 to 4 received anti-PDC-109 antibodies 266μg/mL,80μg/mL,and 26μg/mL in Tris-fructose-citrate buffer,respectively.Semen quality parameters viz.,forward progressive motility,viability,total morphological abnormality,acrosomal integrity,plasma membrane integrity,cryoinjury and in-vitro zona binding index were evaluated.Results:Semen quality parameters of neat semen were within the normal range of bubaline species.Sperm motility,livability,acrosomal integrity,plasma membrane integrity,and cholesterol content of sperm were decreased and total sperm abnormality was increased significantly in post-thaw semen compared to those in pre-freeze and fresh semen(P<0.05).Semen in group 2 had higher sperm motility,livability,acrosomal integrity,plasma membrane integrity,and cholesterol content of sperm and lower total sperm abnormality significantly compared to those in group 1,3 and 4 at pre-freeze and post-thaw stages(P<0.05).Conclusions:Sequestration or neutralization of PDC-109 by its antibodies significantly improves pre-freeze,and post-thaw semen quality parameters and in-vitro zona binding index with simultaneously reducing cryoinjury or cryodamage in the sperm of bubaline species.展开更多

With the wider growth of web-based documents,the necessity of automatic document clustering and text summarization is increased.Here,document summarization that is extracting the essential task with appropriate inform...With the wider growth of web-based documents,the necessity of automatic document clustering and text summarization is increased.Here,document summarization that is extracting the essential task with appropriate information,removal of unnecessary data and providing the data in a cohesive and coherent manner is determined to be a most confronting task.In this research,a novel intelligent model for document clustering is designed with graph model and Fuzzy based association rule generation(gFAR).Initially,the graph model is used to map the relationship among the data(multi-source)followed by the establishment of document clustering with the generation of association rule using the fuzzy concept.This method shows benefit in redundancy elimination by mapping the relevant document using graph model and reduces the time consumption and improves the accuracy using the association rule generation with fuzzy.This framework is provided in an interpretable way for document clustering.It iteratively reduces the error rate during relationship mapping among the data(clusters)with the assistance of weighted document content.Also,this model represents the significance of data features with class discrimination.It is also helpful in measuring the significance of the features during the data clustering process.The simulation is done with MATLAB 2016b environment and evaluated with the empirical standards like Relative Risk Patterns(RRP),ROUGE score,and Discrimination Information Measure(DMI)respectively.Here,DailyMail and DUC 2004 dataset is used to extract the empirical results.The proposed gFAR model gives better trade-off while compared with various prevailing approaches.展开更多
