Objective:The effectiveness of chemotherapy is affected by tumor heterogeneity and drug resistance mechanisms;however,there are certain limitations.Electroacupuncture can regulate the tumor immune response and restore...Objective:The effectiveness of chemotherapy is affected by tumor heterogeneity and drug resistance mechanisms;however,there are certain limitations.Electroacupuncture can regulate the tumor immune response and restore bone marrow hematopoietic function,which is affected by chemotherapy.This study investigated the efficacy and mechanism of electroacupuncture combined with cisplatin in the treatment of non-small-cell lung cancer mice.Methods:To establish a mouse model of non-small-cell lung cancer,gene sequencing combined with bioinformatics analysis,flow cytometry,and liquid-phase chips was used to observe the expression of immune cells and related factors in the mouse tumor microenvironment.Flow cytometry was used to observe subpopulations of mouse bone marrow hematopoietic stem cells and progenitor cells.PAC1 receptor agonists were used to observe mouse tumor immunity and bone marrow hematopoiesis-related indicators.Results:The combination of electroacupuncture with high-and low-dose chemotherapy had a better tumor-suppressive effect.Electroacupuncture can affect the gene expression profile of immune cells,especially the expression levels of Ccr1,Cxcr5,Zbp1,and CamkIIα,and increases the levels of interferon-γ(IFN-γ)and interleukin(IL)-2 protein,upregulating the levels of cytokines Ccl4,Ccl3,and IL-6 in the tumor tissue.Additionally,electroacupuncture enhanced the infiltration of CD8+T cells,dendritic cells,and M1-type macrophages at the tumor site,and reduced the proportion of Th17 and Treg cells.Furthermore,electroacupuncture remodels the bone marrow hematopoietic microenvironment after chemotherapy by increasing the number of bone marrow hematopoietic stem cell subsets,leukocytes,and subpopulations in the peripheral blood.PAC1 receptor agonists have similar effects to those of electroacupuncture on hematopoietic protection and tumor immunity after chemotherapy.Conclusions:Electroacupuncture may improve chemotherapy-induced bone marrow suppression,reshape the tumor microenvironment immune response affected by chemotherapy,and change the tumor immune microenvironment to an anti-tumor mode by regulating tumor local immune-related cytokines.The PAC1 receptor may be a drug target for the treatment of myelosuppression and immunosuppression in patients with tumors.展开更多
This study aimed to identify the worst“bad actors”in mixtures of pollutants contributing to liver damage and shorter telomeres in the U.S.population,using weighted quantile sum(WQS)modeling with stratification by ra...This study aimed to identify the worst“bad actors”in mixtures of pollutants contributing to liver damage and shorter telomeres in the U.S.population,using weighted quantile sum(WQS)modeling with stratification by race/ethnicity.We conducted a comprehensive cross-sectional analysis of mixtures of pollutants in National Health and Nutrition Examination Survey datasets:(1)33,979 adults with blood levels of cadmium(Cd),lead(Pb),and mercury,including subsets with measurements of per-/polyfluoroalkyl substances(PFAS),and polychlorinated biphenyls(PCBs)/polychlorinated dibenzo-p-dioxins and dibenzofurans(PCDD/Fs);and(2)7360 adults with measurements of telomeres,Cd,and Pb.Multivariable-adjusted WQS regression examined associations between WQS mixture indices and liver injury(alanine aminotransferase(ALT)-elevation),advanced liverfibrosis(LF),and telomere length.WQSmetal indices were associated with advanced-LF in all racial/ethnic groups.The top contributor was Cd in the total population and in non-Hispanic Whites(NHW),while Pb was the top contributor in non-Hispanic Blacks(NHB).The WQSmetal-PCB-PCDD/F index was associated with ALT-elevation,with PCB126,Cd and Pb as main contributors;the odds ratio(OR)per decile was 1.50(95%CI,1.26–1.78),while the OR per decile of the WQSmetal-PFAS index was 1.03(95%CI,0.98–1.05),not significant.WQS_(metal indices)were associated with shorter telomeres.Cd was main contributor associated with advanced-LF in NHW,while Pb was the major bad actor in NHB,suggesting that NHB may be especially susceptible to Pb toxicity.Metals were associated with shorter telomeres.Metal and PCB/PCDD/F mixtures were associated with ALT-elevation.Heavy metals and organic chemicals may contribute to liver-related morbidity and healthcare disparities.展开更多
Boron-doped diamond(BDD)is a well-known anode material with a high pollutant degradation ability for electrochemical oxidation wastewater treatment.Nevertheless,the cost of production and mechanical strength of BDD me...Boron-doped diamond(BDD)is a well-known anode material with a high pollutant degradation ability for electrochemical oxidation wastewater treatment.Nevertheless,the cost of production and mechanical strength of BDD membranes remain unsatisfactory.Magnetic BDD particles derived from industrial waste may represent a promising alternative to BDD membranes,although the challenge remains in assembling these particles into a usable electrode.In this study,magnetic BDD particles were attracted to a Ti/RuO_(2)-IrO_(2)electrode using a magnet,thus constituting a novel 2.5-dimensional(2.5D)electrode.To ascertain the structure-activity relationship of the novel electrode,essential characterizations,multi-physics simulations,pollutant degradation and electrosynthesis experiments were conducted.The results indicate that an appropriate quantity of BDD particles(0.1 g/cm^(2))can enhance the number of active sites by approximately 20%.A strong synergistic effect was observed between the Ti/Ti/RuO_(2)-IrO_(2)and BDD particles in the degradation of various pollutants,including azo dye,p-benzoquinone,succinic acid and four kinds of real wastewaters,as well as glycerol conversion.The joint active sites on the interface between Ti/RuO_(2)-IrO_(2)and BDD particles,as well as the inner active sites on BDD particles,have been identified as crucial in the mineralization of pollutants and the generation of value-added products.The optimal amount of BDD particles(0.1 g/cm^(2))is sufficient to preserve the joint active sites and to maintain an adequate polarization on the BDD particles.Nevertheless,the hybrid feature of the 2.5D electrode is diminished when a greater quantity of BDD particles(0.3 g/cm^(2))is loaded.展开更多
This paper proposes a method to generate Bessel-like collimated beams with suppressed side lobes using the flexural vibration modes of a fixed boundary circular plate,which is excited by a longitudinally vibrating tra...This paper proposes a method to generate Bessel-like collimated beams with suppressed side lobes using the flexural vibration modes of a fixed boundary circular plate,which is excited by a longitudinally vibrating transducer in a ring excitation manner.The factors affecting the generation of Bessel-like collimated beams are investigated by theoretical analysis,numerical simulation and experimental methods.The results indicate that Bessel-like wave can be generated by a thin circular plate with fixed boundaries.The third-order mode of the circular plate can be modified to generate a collimated beam with suppressing side lobes when it is excited in a ring excitation manner and the excitation position lies between the first two nodal circles of the plate.As the excitation radius increases,the main lobe width of the resulting Bessel-like collimated beam decreases,the extent of the focusing region increases,and the amplitude of the side lobes initially increases and then decreases.Based on the simulation results,a prototype Bessel-like collimated beam generation system is made and measured experimentally.The experimental results are in good agreement with the numerical results.The Bessel-like collimated beam can be generated by the proposed system,which has potential application in the fields of long-range detection,imaging of highly attenuated materials,and airflow acceleration.展开更多

The effect of the second messengers cAMP and cyclic cGMP on Cx43 protein expression was also investigated. Enucleated human eyes (remnant after corneal transplantation) and bovine eyes were used. Tissue preparations of the anterior segments of the eyes have proceeded for immunohistochemical staining with polyclonal antibodies of Cx43 and Cx40. Isolated ciliary bodies of human and bovine eyes were incubated with cAMP analog 8-Bromo-cAMP or the cGMP analog 8-Bromo-cGMP, the expression of Cx43 protein in the tissues was then assessed by Western blot assay. Both in human and bovine ciliary bodies, strong immunoreactivity of Cx43, but not Cx40, was observed predominantly in the apical cytoplasmic portions of the pigment ciliary epithelial cells and non-pigmented ciliary epithelial cells. In human ciliary body both cAMP and cGMP up-regulated Cx43 expression, while in the bovine ciliary body, cGMP increased Cx43 expression but cAMP decreased it. Cx43 is the major component of human and bovine gap junctions in the ciliary epithelium. The regulation on the Cx43 expression by cAMP and cGMP in human and bovine ciliary bodies suggests the possibly different roles of these signal messengers in the intracellular communication.展开更多

The aim of this study is to clarify whether these DNA lesions participate in the progression of intrahepatic cholangiocarcinoma. METHODS: We investigated the relation of the formation of 8-nitroguanine and 8-oxodG and the expression of hypoxia-inducible factor-1α (HIF-1α) with tumor invasion in 37 patients with intra-hepatic cholangiocarcinoma. RESULTS: Immunohistochemical analyses revealed that 8-nitroguanine and 8-oxodG formation occurred to a much greater extent in cancerous tissues than in non-cancerous tissues. HIF-1α could be detected in cancerous tissues in all patients, suggesting low oxygen tension in the tumors. HIF-1α expression was correlated with inducible niltric oxide synthase (iNOS) expression (r = 0.369 and P = 0.025) and 8-oxodG formation (r = 0.398 and P = 0.015). Double immunofluorescence study revealed that iNOS and HIF-1α co-localized in cancerous tissues. Notably, the formation of 8-oxodG was correlated significantly with lymphatic invasion (r = 0.386 and P = 0.018). Moreover, 8- nitroguanine and 8-oxodG in non-cancerous tissues were associated significantly with neural invasion (P = 0.042 and P = 0.026, respectively). These results suggest that reciprocal activation between HIF-1α and iNOS mediates persistent DNA damage, which induces tumor invasiveness via mutations, resulting in poor prognosis. CONCLUSION: The formation of 8-nitroguanine and 8-oxodG plays an important role in multiple steps of genetic changes leading to tumor progression, including invasiveness.展开更多

Background:Arginine(Arg)is an essential amino acid(EAA)in poultry,an important substrate for protein synthesis and a precursor of several molecules.Supplementation of EAAs with low protein(LP)diet increases the utiliz...Background:Arginine(Arg)is an essential amino acid(EAA)in poultry,an important substrate for protein synthesis and a precursor of several molecules.Supplementation of EAAs with low protein(LP)diet increases the utilization efficiency of dietary crude protein(CP).However,if the EAA requirement is changed in hens fed a LP diet remains to be elucidated.The aim of the present study was to evaluate the optimal level of dietary Arg in the LP diet of hens.A total of 1350 Hy-Line Brown laying hens were randomly allocated to six dietary treatments:a basal diet(16%CP,positive control),or an isoenergetic LP diet(14%CP,0.80%Arg)supplemented 0,0.05%,0.10%,0.15%,and 0.20%L-Arg,corresponding to 0.80%,0.85%,0.90%,0.95%and 1.00%dietary Arg,respectively.Results:The feed efficiency was decreased(P<0.05)by 0.80%and 1.00%Arg-LP diets,compared to control.Within LP diets,dietary Arg level had significant quadratic effects(P<0.05)on laying rate,egg mass,and feed efficiency.Compared to control,the plasma CAT activity or T-AOC content were decreased by 0.80%(P<0.001).However,the hens offered 0.85%and 0.90%Arg-LP diets had higher CAT activity(P<0.001)than 0.80%Arg-LP diet.In contrast,1.00%Arg-LP group had the highest MDA and the lowest T-AOC content in plasma,liver,duodenal and jejunal mucosa(P<0.05).Compared to control,the villus height was decreased by 0.80%,0.95%and 1.00%Arg-LP diets,while the villus height to crypt depth(V/C)ratio was reduced by 0.95%and 1.00%Arg-LP diets in duodenum.Conclusion:The result demonstrates that LP diet(14%CP)deficient in Arg(0.80%Arg)result in augmented oxidative damage and impaired development of intestinal mucosa.According to the quadratic broken-line regression model,the optimal dietary arginine levels for Hy-Line Brown laying hens fed with low protein diet(14%CP)aged 33 to 40 weeks are 0.85%,0.86%,and 0.86%to obtained the maximum laying rate,egg mass,and feed efficiency,respectively.展开更多

Results indicated that the nttmber of lakes declined from 94 to 82 and the total surface area was reduced by 3.69 km2 during 1973-2010. The desert lake area reduced by different degrees in different periods, but this occurred most rapidly during 1973-1990. According to the statistics of lake area changes, lake area decreases mainly occurred in the lakes with areas less than 0.2 km2, while the areas of lakes greater than 0.9 km2 only fluctuated. The changes of lake areas were probably due to changes in the quantity of underground water supplies rather than the effects of local climate change or human factors.展开更多
Most large-scale evapotranspiration(ET)estimation methods require detailed information of land use,land cover,and/or soil type on top of various atmospheric measurements.The complementary relationship of evaporation(C...Most large-scale evapotranspiration(ET)estimation methods require detailed information of land use,land cover,and/or soil type on top of various atmospheric measurements.The complementary relationship of evaporation(CR)takes advantage of the inherent dynamic feedback mechanisms found in the soil−vegetation−atmosphere interface for its estimation of ET rates without the need of such biogeophysical data.ET estimates over the conterminous United States by a new,globally calibrated,static scaling(GCR-stat)of the generalized complementary relationship(GCR)of evaporation were compared to similar estimates of an existing,calibration-free version(GCR-dyn)of the GCR that employs a temporally varying dynamic scaling.Simplified annual water balances of 327 medium and 18 large watersheds served as ground-truth ET values.With long-term monthly mean forcing,GCR-stat(also utilizing precipitation measurements)outperforms GCR-dyn as the latter cannot fully take advantage of its dynamic scaling with such data of reduced temporal variability.However,in a continuous monthly simulation,GCR-dyn is on a par with GCR-stat,and especially excels in reproducing long-term tendencies in annual catchment ET rates even though it does not require precipitation information.The same GCR-dyn estimates were also compared to similar estimates of eight other popular ET products and they generally outperform all of them.For this reason,a dynamic scaling of the GCR is recommended over a static one for modeling long-term behavior of terrestrial ET.展开更多

After the hydrophilic functionalization, Co/polypyrrole (PPy) nanocomposites were prepared by in situ polymerization of pyrrole in aqueous dispersion of Co nanoparticles. The Co/ PPy nanocomposites show good electromagnetic properties with both magnetic loss and dielectric loss to electromag- netic wave. The electromagnetic wave absorbing band- width (reflection loss 〈-10 dB) for Co/PPy (20 wt%) is above 5.5 GHz at a thickness of 2 mm, and with a maximum reflection loss (around -20.02 dB) at 14.77 GHz. This magnetic nanoparticles/conducting polymer nanocomposites are great potential candidates for electromagnetic wave absorbent, because of their wide-absorbing frequency, strong absorption, good compatibility, low density, and controllable absorbing properties.展开更多
BACKGROUND Cuproptosis has recently been considered a novel form of programmed cell death.To date,long-chain non-coding RNAs(lncRNAs)crucial to the regulation of this process remain unelucidated.AIM To identify lncRNA...BACKGROUND Cuproptosis has recently been considered a novel form of programmed cell death.To date,long-chain non-coding RNAs(lncRNAs)crucial to the regulation of this process remain unelucidated.AIM To identify lncRNAs linked to cuproptosis in order to estimate patients'prognoses for hepatocellular carcinoma(HCC).METHODS Using RNA sequence data from The Cancer Genome Atlas Live Hepatocellular Carcinoma(TCGA-LIHC),a co-expression network of cuproptosis-related genes and lncRNAs was constructed.For HCC prognosis,we developed a cuproptosisrelated lncRNA signature(CupRLSig)using univariate Cox,lasso,and multivariate Cox regression analyses.Kaplan-Meier analysis was used to compare overall survival among high-and low-risk groups stratified by median CupRLSig risk score.Furthermore,comparisons of functional annotation,immune infiltration,somatic mutation,tumor mutation burden(TMB),and pharmacologic options were made between high-and low-risk groups.RESULTS Three hundred and forty-three patients with complete follow-up data were recruited in the analysis.Pearson correlation analysis identified 157 cuproptosis-related lncRNAs related to 14 cuproptosis genes.Next,we divided the TCGA-LIHC sample into a training set and a validation set.In univariate Cox regression analysis,27 LncRNAs with prognostic value were identified in the training set.After lasso regression,the multivariate Cox regression model determined the identified risk equation as follows:Risk score=(0.2659×PICSAR expression)+(0.4374×FOXD2-AS1 expression)+(-0.3467×AP001065.1 expression).The CupRLSig high-risk group was associated with poor overall survival(hazard ratio=1.162,95%CI=1.063-1.270;P<0.001)after the patients were divided into two groups depending upon their median risk score.Model accuracy was further supported by receiver operating characteristic and principal component analysis as well as the validation set.The area under the curve of 0.741 was found to be a better predictor of HCC prognosis as compared to other clinicopathological variables.Mutation analysis revealed that high-risk combinations with high TMB carried worse prognoses(median survival of 30 mo vs 102 mo of low-risk combinations with low TMB group).The low-risk group had more activated natural killer cells(NK cells,P=0.032 by Wilcoxon rank sum test)and fewer regulatory T cells(Tregs,P=0.021)infiltration than the high-risk group.This finding could explain why the low-risk group has a better prognosis.Interestingly,when checkpoint gene expression(CD276,CTLA-4,and PDCD-1)and tumor immune dysfunction and rejection(TIDE)scores are considered,highrisk patients may respond better to immunotherapy.Finally,most drugs commonly used in preclinical and clinical systemic therapy for HCC,such as 5-fluorouracil,gemcitabine,paclitaxel,imatinib,sunitinib,rapamycin,and XL-184(cabozantinib),were found to be more efficacious in the low-risk group;erlotinib,an exception,was more efficacious in the high-risk group.CONCLUSION The lncRNA signature,CupRLSig,constructed in this study is valuable in prognostic estimation of HCC.Importantly,CupRLSig also predicts the level of immune infiltration and potential efficacy of tumor immunotherapy.展开更多

Unlike previous flare slamming analysis, our proposed method takes roll motion into account to calculate the impact angle and relative vertical velocity between ship sections on the bow flare and wave surface. We use the Wagner model to analyze the slamming impact forces and the slamming occurrence probability. Through numerical simulations, we investigate the maximum flare slamming pressures of a container ship for different speeds and wave conditions. To further clarify the mechanism of flare slamming phenomena in parametric rolling conditions, we also conduct real-time simulations to determine the relationship between slamming pressure and 3-DOF motions, namely roll, pitch, and heave.展开更多

A simple criterion was defined to help estimate the amount of depolarization in Nd:YAG amplifier stages. The degree of depolarization of the beam was 7.1% and the beam quality was measured to be M2= 3.69. Canine atopic dermatitis(CAD)is a prevalent genetically susceptible infammatory and pruritic allergic skin condition afecting not only the health of dogs but also the quality of life of their owners.Interleukin-31(IL-...Canine atopic dermatitis(CAD)is a prevalent genetically susceptible infammatory and pruritic allergic skin condition afecting not only the health of dogs but also the quality of life of their owners.Interleukin-31(IL-31)and interleukin-31 receptor alpha(IL-31RA)are essential for the development of pruritus in primates and mice.Hence,it is expected that inhibiting IL-31RA will be an efective approach to alleviate pruritus.The purpose of the study was to produce anti-canine IL-31RA polyclonal antibodies(anti-IL-31RA pAbs)and evaluate their efcacy in inhibiting house dust mite(HDM)-evoked pruritic responses.Dogs were immunized with antigens formed by IL-31RA recombinant short peptides coupled to BSA to produce anti-IL-31RA pAbs.The CAD model was developed by using HDM allergen stimulation,and the efects of IL-31RA pAbs on the reduction of pruritus in CAD model dogs were examined.The Canine Atopic Dermatitis Extent and Severity Index(CADESI)-4 and pruritus Visual Analog Scale(pVAS)were utilized to evaluate pruritic responses,and skin tissue samples were collected from the inguinal area for pathological assessment of skin infammatory cell infltration.The results showed that anti-IL-31RA pAbs with high titers(1:128,000)and specifcity were efectively produced.In the CAD model group,the severity of skin damage,pruritus score,infammatory cell infltration and level of infammatory factors were considerably elevated.Anti-IL-31RA pAbs relieved pruritic behavior and dermatitis in dogs compared to placebo-treated dogs.In conclusion,anti-IL-31RA pAbs efectively suppressed CAD in vivo and are anticipated to be an efective novel treatment for pruritic skin disorders such as CAD.展开更多

文摘Zebrafish is a powerful model for the investigation of hematopoiesis.In order to isolate novel mutants with hematopoietic defects, large-scale mutagenesis screening of zebrafish was performed.By scoring specific hematopoietic markers,52 mutants were identified and then classified into four types based on specific phenotypic traits.Each mutant represented a putative mutation of a gene regulating the relevant aspect of hematopoiesis,including early macrophage development,early granulopoiesis,embryonic myelopoiesis,and definitive erythropoiesis/lymphopoiesis.Our method should be applicable for other types of genetic screening in zebrafish.In addition,further study of the mutants we identified may help to unveil the molecular basis of hematopoiesis.

文摘Objective:The effectiveness of chemotherapy is affected by tumor heterogeneity and drug resistance mechanisms;however,there are certain limitations.Electroacupuncture can regulate the tumor immune response and restore bone marrow hematopoietic function,which is affected by chemotherapy.This study investigated the efficacy and mechanism of electroacupuncture combined with cisplatin in the treatment of non-small-cell lung cancer mice.Methods:To establish a mouse model of non-small-cell lung cancer,gene sequencing combined with bioinformatics analysis,flow cytometry,and liquid-phase chips was used to observe the expression of immune cells and related factors in the mouse tumor microenvironment.Flow cytometry was used to observe subpopulations of mouse bone marrow hematopoietic stem cells and progenitor cells.PAC1 receptor agonists were used to observe mouse tumor immunity and bone marrow hematopoiesis-related indicators.Results:The combination of electroacupuncture with high-and low-dose chemotherapy had a better tumor-suppressive effect.Electroacupuncture can affect the gene expression profile of immune cells,especially the expression levels of Ccr1,Cxcr5,Zbp1,and CamkIIα,and increases the levels of interferon-γ(IFN-γ)and interleukin(IL)-2 protein,upregulating the levels of cytokines Ccl4,Ccl3,and IL-6 in the tumor tissue.Additionally,electroacupuncture enhanced the infiltration of CD8+T cells,dendritic cells,and M1-type macrophages at the tumor site,and reduced the proportion of Th17 and Treg cells.Furthermore,electroacupuncture remodels the bone marrow hematopoietic microenvironment after chemotherapy by increasing the number of bone marrow hematopoietic stem cell subsets,leukocytes,and subpopulations in the peripheral blood.PAC1 receptor agonists have similar effects to those of electroacupuncture on hematopoietic protection and tumor immunity after chemotherapy.Conclusions:Electroacupuncture may improve chemotherapy-induced bone marrow suppression,reshape the tumor microenvironment immune response affected by chemotherapy,and change the tumor immune microenvironment to an anti-tumor mode by regulating tumor local immune-related cytokines.The PAC1 receptor may be a drug target for the treatment of myelosuppression and immunosuppression in patients with tumors.

文摘This study aimed to identify the worst“bad actors”in mixtures of pollutants contributing to liver damage and shorter telomeres in the U.S.population,using weighted quantile sum(WQS)modeling with stratification by race/ethnicity.We conducted a comprehensive cross-sectional analysis of mixtures of pollutants in National Health and Nutrition Examination Survey datasets:(1)33,979 adults with blood levels of cadmium(Cd),lead(Pb),and mercury,including subsets with measurements of per-/polyfluoroalkyl substances(PFAS),and polychlorinated biphenyls(PCBs)/polychlorinated dibenzo-p-dioxins and dibenzofurans(PCDD/Fs);and(2)7360 adults with measurements of telomeres,Cd,and Pb.Multivariable-adjusted WQS regression examined associations between WQS mixture indices and liver injury(alanine aminotransferase(ALT)-elevation),advanced liverfibrosis(LF),and telomere length.WQSmetal indices were associated with advanced-LF in all racial/ethnic groups.The top contributor was Cd in the total population and in non-Hispanic Whites(NHW),while Pb was the top contributor in non-Hispanic Blacks(NHB).The WQSmetal-PCB-PCDD/F index was associated with ALT-elevation,with PCB126,Cd and Pb as main contributors;the odds ratio(OR)per decile was 1.50(95%CI,1.26–1.78),while the OR per decile of the WQSmetal-PFAS index was 1.03(95%CI,0.98–1.05),not significant.WQS_(metal indices)were associated with shorter telomeres.Cd was main contributor associated with advanced-LF in NHW,while Pb was the major bad actor in NHB,suggesting that NHB may be especially susceptible to Pb toxicity.Metals were associated with shorter telomeres.Metal and PCB/PCDD/F mixtures were associated with ALT-elevation.Heavy metals and organic chemicals may contribute to liver-related morbidity and healthcare disparities.

文摘Boron-doped diamond(BDD)is a well-known anode material with a high pollutant degradation ability for electrochemical oxidation wastewater treatment.Nevertheless,the cost of production and mechanical strength of BDD membranes remain unsatisfactory.Magnetic BDD particles derived from industrial waste may represent a promising alternative to BDD membranes,although the challenge remains in assembling these particles into a usable electrode.In this study,magnetic BDD particles were attracted to a Ti/RuO_(2)-IrO_(2)electrode using a magnet,thus constituting a novel 2.5-dimensional(2.5D)electrode.To ascertain the structure-activity relationship of the novel electrode,essential characterizations,multi-physics simulations,pollutant degradation and electrosynthesis experiments were conducted.The results indicate that an appropriate quantity of BDD particles(0.1 g/cm^(2))can enhance the number of active sites by approximately 20%.A strong synergistic effect was observed between the Ti/Ti/RuO_(2)-IrO_(2)and BDD particles in the degradation of various pollutants,including azo dye,p-benzoquinone,succinic acid and four kinds of real wastewaters,as well as glycerol conversion.The joint active sites on the interface between Ti/RuO_(2)-IrO_(2)and BDD particles,as well as the inner active sites on BDD particles,have been identified as crucial in the mineralization of pollutants and the generation of value-added products.The optimal amount of BDD particles(0.1 g/cm^(2))is sufficient to preserve the joint active sites and to maintain an adequate polarization on the BDD particles.Nevertheless,the hybrid feature of the 2.5D electrode is diminished when a greater quantity of BDD particles(0.3 g/cm^(2))is loaded.

文摘This paper proposes a method to generate Bessel-like collimated beams with suppressed side lobes using the flexural vibration modes of a fixed boundary circular plate,which is excited by a longitudinally vibrating transducer in a ring excitation manner.The factors affecting the generation of Bessel-like collimated beams are investigated by theoretical analysis,numerical simulation and experimental methods.The results indicate that Bessel-like wave can be generated by a thin circular plate with fixed boundaries.The third-order mode of the circular plate can be modified to generate a collimated beam with suppressing side lobes when it is excited in a ring excitation manner and the excitation position lies between the first two nodal circles of the plate.As the excitation radius increases,the main lobe width of the resulting Bessel-like collimated beam decreases,the extent of the focusing region increases,and the amplitude of the side lobes initially increases and then decreases.Based on the simulation results,a prototype Bessel-like collimated beam generation system is made and measured experimentally.The experimental results are in good agreement with the numerical results.The Bessel-like collimated beam can be generated by the proposed system,which has potential application in the fields of long-range detection,imaging of highly attenuated materials,and airflow acceleration.
文摘The aim of the study was to assess the distribution of connexin43 (Cx43) and connexin40 (Cx40) in human and bovine ciliary bodies. The effect of the second messengers cAMP and cyclic cGMP on Cx43 protein expression was also investigated. Enucleated human eyes (remnant after corneal transplantation) and bovine eyes were used. Tissue preparations of the anterior segments of the eyes have proceeded for immunohistochemical staining with polyclonal antibodies of Cx43 and Cx40. Isolated ciliary bodies of human and bovine eyes were incubated with cAMP analog 8-Bromo-cAMP or the cGMP analog 8-Bromo-cGMP, the expression of Cx43 protein in the tissues was then assessed by Western blot assay. Both in human and bovine ciliary bodies, strong immunoreactivity of Cx43, but not Cx40, was observed predominantly in the apical cytoplasmic portions of the pigment ciliary epithelial cells and non-pigmented ciliary epithelial cells. In human ciliary body both cAMP and cGMP up-regulated Cx43 expression, while in the bovine ciliary body, cGMP increased Cx43 expression but cAMP decreased it. Cx43 is the major component of human and bovine gap junctions in the ciliary epithelium. The regulation on the Cx43 expression by cAMP and cGMP in human and bovine ciliary bodies suggests the possibly different roles of these signal messengers in the intracellular communication.

文摘AIM: Nitrative and oxidative DNA damage such as 8-nitroguanine and 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG) formation has been implicated in initiation and/ or promotion of inflammation-mediated carcinogenesis. The aim of this study is to clarify whether these DNA lesions participate in the progression of intrahepatic cholangiocarcinoma. METHODS: We investigated the relation of the formation of 8-nitroguanine and 8-oxodG and the expression of hypoxia-inducible factor-1α (HIF-1α) with tumor invasion in 37 patients with intra-hepatic cholangiocarcinoma. RESULTS: Immunohistochemical analyses revealed that 8-nitroguanine and 8-oxodG formation occurred to a much greater extent in cancerous tissues than in non-cancerous tissues. HIF-1α could be detected in cancerous tissues in all patients, suggesting low oxygen tension in the tumors. HIF-1α expression was correlated with inducible niltric oxide synthase (iNOS) expression (r = 0.369 and P = 0.025) and 8-oxodG formation (r = 0.398 and P = 0.015). Double immunofluorescence study revealed that iNOS and HIF-1α co-localized in cancerous tissues. Notably, the formation of 8-oxodG was correlated significantly with lymphatic invasion (r = 0.386 and P = 0.018). Moreover, 8- nitroguanine and 8-oxodG in non-cancerous tissues were associated significantly with neural invasion (P = 0.042 and P = 0.026, respectively). These results suggest that reciprocal activation between HIF-1α and iNOS mediates persistent DNA damage, which induces tumor invasiveness via mutations, resulting in poor prognosis. CONCLUSION: The formation of 8-nitroguanine and 8-oxodG plays an important role in multiple steps of genetic changes leading to tumor progression, including invasiveness.

文摘Time-frequency-based methods are proven to be effective for parameter estimation of linear frequency modulation (LFM) signals. The smoothed pseudo Winger-Ville distribution (SPWVD) is used for the parameter estimation of multi-LFM signals, and a method of the SPWVD binarization by a dynamic threshold based on the Otsu algorithm is proposed. The proposed method is effective in the demand for the estimation of different parameters and the unknown signal-to-noise ratio (SNR) circumstance. The performance of this method is confirmed by numerical simulation.

文摘Background:Arginine(Arg)is an essential amino acid(EAA)in poultry,an important substrate for protein synthesis and a precursor of several molecules.Supplementation of EAAs with low protein(LP)diet increases the utilization efficiency of dietary crude protein(CP).However,if the EAA requirement is changed in hens fed a LP diet remains to be elucidated.The aim of the present study was to evaluate the optimal level of dietary Arg in the LP diet of hens.A total of 1350 Hy-Line Brown laying hens were randomly allocated to six dietary treatments:a basal diet(16%CP,positive control),or an isoenergetic LP diet(14%CP,0.80%Arg)supplemented 0,0.05%,0.10%,0.15%,and 0.20%L-Arg,corresponding to 0.80%,0.85%,0.90%,0.95%and 1.00%dietary Arg,respectively.Results:The feed efficiency was decreased(P<0.05)by 0.80%and 1.00%Arg-LP diets,compared to control.Within LP diets,dietary Arg level had significant quadratic effects(P<0.05)on laying rate,egg mass,and feed efficiency.Compared to control,the plasma CAT activity or T-AOC content were decreased by 0.80%(P<0.001).However,the hens offered 0.85%and 0.90%Arg-LP diets had higher CAT activity(P<0.001)than 0.80%Arg-LP diet.In contrast,1.00%Arg-LP group had the highest MDA and the lowest T-AOC content in plasma,liver,duodenal and jejunal mucosa(P<0.05).Compared to control,the villus height was decreased by 0.80%,0.95%and 1.00%Arg-LP diets,while the villus height to crypt depth(V/C)ratio was reduced by 0.95%and 1.00%Arg-LP diets in duodenum.Conclusion:The result demonstrates that LP diet(14%CP)deficient in Arg(0.80%Arg)result in augmented oxidative damage and impaired development of intestinal mucosa.According to the quadratic broken-line regression model,the optimal dietary arginine levels for Hy-Line Brown laying hens fed with low protein diet(14%CP)aged 33 to 40 weeks are 0.85%,0.86%,and 0.86%to obtained the maximum laying rate,egg mass,and feed efficiency,respectively.

文摘Lake area information in the Badain Jaran Desert in 1973, 1990, 2000, and 2010 was obtained by visual interpretation and water index analysis of remote sensing images, based on the spatial and temporal characteristics of lake area changes during 37 years. Results indicated that the nttmber of lakes declined from 94 to 82 and the total surface area was reduced by 3.69 km2 during 1973-2010. The desert lake area reduced by different degrees in different periods, but this occurred most rapidly during 1973-1990. According to the statistics of lake area changes, lake area decreases mainly occurred in the lakes with areas less than 0.2 km2, while the areas of lakes greater than 0.9 km2 only fluctuated. The changes of lake areas were probably due to changes in the quantity of underground water supplies rather than the effects of local climate change or human factors.

文摘Most large-scale evapotranspiration(ET)estimation methods require detailed information of land use,land cover,and/or soil type on top of various atmospheric measurements.The complementary relationship of evaporation(CR)takes advantage of the inherent dynamic feedback mechanisms found in the soil−vegetation−atmosphere interface for its estimation of ET rates without the need of such biogeophysical data.ET estimates over the conterminous United States by a new,globally calibrated,static scaling(GCR-stat)of the generalized complementary relationship(GCR)of evaporation were compared to similar estimates of an existing,calibration-free version(GCR-dyn)of the GCR that employs a temporally varying dynamic scaling.Simplified annual water balances of 327 medium and 18 large watersheds served as ground-truth ET values.With long-term monthly mean forcing,GCR-stat(also utilizing precipitation measurements)outperforms GCR-dyn as the latter cannot fully take advantage of its dynamic scaling with such data of reduced temporal variability.However,in a continuous monthly simulation,GCR-dyn is on a par with GCR-stat,and especially excels in reproducing long-term tendencies in annual catchment ET rates even though it does not require precipitation information.The same GCR-dyn estimates were also compared to similar estimates of eight other popular ET products and they generally outperform all of them.For this reason,a dynamic scaling of the GCR is recommended over a static one for modeling long-term behavior of terrestrial ET.

文摘In this work, cobalt nanoparticles were syn- thesized by chemical reduction procedure. After the hydrophilic functionalization, Co/polypyrrole (PPy) nanocomposites were prepared by in situ polymerization of pyrrole in aqueous dispersion of Co nanoparticles. The Co/ PPy nanocomposites show good electromagnetic properties with both magnetic loss and dielectric loss to electromag- netic wave. The electromagnetic wave absorbing band- width (reflection loss 〈-10 dB) for Co/PPy (20 wt%) is above 5.5 GHz at a thickness of 2 mm, and with a maximum reflection loss (around -20.02 dB) at 14.77 GHz. This magnetic nanoparticles/conducting polymer nanocomposites are great potential candidates for electromagnetic wave absorbent, because of their wide-absorbing frequency, strong absorption, good compatibility, low density, and controllable absorbing properties.

文摘BACKGROUND Cuproptosis has recently been considered a novel form of programmed cell death.To date,long-chain non-coding RNAs(lncRNAs)crucial to the regulation of this process remain unelucidated.AIM To identify lncRNAs linked to cuproptosis in order to estimate patients'prognoses for hepatocellular carcinoma(HCC).METHODS Using RNA sequence data from The Cancer Genome Atlas Live Hepatocellular Carcinoma(TCGA-LIHC),a co-expression network of cuproptosis-related genes and lncRNAs was constructed.For HCC prognosis,we developed a cuproptosisrelated lncRNA signature(CupRLSig)using univariate Cox,lasso,and multivariate Cox regression analyses.Kaplan-Meier analysis was used to compare overall survival among high-and low-risk groups stratified by median CupRLSig risk score.Furthermore,comparisons of functional annotation,immune infiltration,somatic mutation,tumor mutation burden(TMB),and pharmacologic options were made between high-and low-risk groups.RESULTS Three hundred and forty-three patients with complete follow-up data were recruited in the analysis.Pearson correlation analysis identified 157 cuproptosis-related lncRNAs related to 14 cuproptosis genes.Next,we divided the TCGA-LIHC sample into a training set and a validation set.In univariate Cox regression analysis,27 LncRNAs with prognostic value were identified in the training set.After lasso regression,the multivariate Cox regression model determined the identified risk equation as follows:Risk score=(0.2659×PICSAR expression)+(0.4374×FOXD2-AS1 expression)+(-0.3467×AP001065.1 expression).The CupRLSig high-risk group was associated with poor overall survival(hazard ratio=1.162,95%CI=1.063-1.270;P<0.001)after the patients were divided into two groups depending upon their median risk score.Model accuracy was further supported by receiver operating characteristic and principal component analysis as well as the validation set.The area under the curve of 0.741 was found to be a better predictor of HCC prognosis as compared to other clinicopathological variables.Mutation analysis revealed that high-risk combinations with high TMB carried worse prognoses(median survival of 30 mo vs 102 mo of low-risk combinations with low TMB group).The low-risk group had more activated natural killer cells(NK cells,P=0.032 by Wilcoxon rank sum test)and fewer regulatory T cells(Tregs,P=0.021)infiltration than the high-risk group.This finding could explain why the low-risk group has a better prognosis.Interestingly,when checkpoint gene expression(CD276,CTLA-4,and PDCD-1)and tumor immune dysfunction and rejection(TIDE)scores are considered,highrisk patients may respond better to immunotherapy.Finally,most drugs commonly used in preclinical and clinical systemic therapy for HCC,such as 5-fluorouracil,gemcitabine,paclitaxel,imatinib,sunitinib,rapamycin,and XL-184(cabozantinib),were found to be more efficacious in the low-risk group;erlotinib,an exception,was more efficacious in the high-risk group.CONCLUSION The lncRNA signature,CupRLSig,constructed in this study is valuable in prognostic estimation of HCC.Importantly,CupRLSig also predicts the level of immune infiltration and potential efficacy of tumor immunotherapy.

文摘Ti3C2Tx nanosheet,the first synthesized MXene with high capacity performance and charge/discharge rate,has attracted increasingly attention in renewable energy storage applications.By performing systematic density functional theory calculations,the theoretical capacity of the intrinsic structure of single-and multi-layered Ti3C2T2(T=F or O)corresponding to M(M=Li and Na)atoms are investigated.Theoretical volumetric capacity and gravimetric capacity are obtained,which are related to the stacking degree.The optimal ratios of capacity to structure are determined under different stacking degrees for understanding the influence of surface functional groups on energy storage performance.Its performance can be tuned by performing surface modification and increasing the interlayer distance.In addition,the reason for theoretical capacity differences of M atoms is analyzed,which is attributed to difference in interaction between the M-ions and substrate and the difference in electrostatic exclusion between adsorbed M-ions.These results provide an insight into the understanding of the method of efficiently increasing the energy storage performance,which will be useful for designing and using high performance electrode materials.

文摘Natural polysaccharides named PEP-0.1-1,PEP-0-1 and PEP-0-2 from edible mushroom species Pleurotus eryngii were obtained in the present study.Results showed that molecular weights of these polysaccharides were 3235,2041 and 23933 Da,respectively.Further,structural characterization revealed that PEP-0.1-1 had a→4-α-D-Glcp-1→backbone and contained→4)-α-D-Glcp and→4)-β-D-Glcp reducing end groups.PEP-0-1 backbone contained→4-α-D-Glcp-1→and→6-α-3-O-Me-D-Galp-1→,and the side chains containedα-D-Glcp,β-D-Manp-1→andα-D-Glcp-3→.However,PEP-0-2 backbone consisted of→4-α-DGlcp-1→and→6-α-3-O-Me-D-Galp-(1→6)-α-D-Galp-1→while the side chains containedα-D-Glcp andβ-D-Manp-1→.Biological activity analysis was then carried out and found that all these polysaccharides could significantly suppress the relative mRNA expression of toll-like receptor 4,nitric oxide(NO),tumor necrosis factor-α,interleukin(IL)-1βand IL-6 in lipopolysaccharide(LPS)-induced inflammation of RAW264.7 cells,as well as the over secretion of the above cell cytokines.Moreover,Western blotting analysis revealed that all these purified fractions displayed significant inhibition effects on the expression of c-Jun N-terminal kinases protein induced by LPS in mitogen activated protein kinase pathway,along with the relieving on the inhibition effect of LPS on IκB-αprotein expression.In summary,the information generated by the present study could provide a theoretical basis for the exploration of novel healthy food materials from edible mushroom with antiinflammation activities.

文摘Since the research of flare slamming prediction is seldom when parametric rolling happens, we present an efficient approximation method for flare slamming analysis of large container ships in parametric rolling conditions. We adopt a 6-DOF weakly nonlinear time domain model to predict the ship motions of parametric rolling conditions. Unlike previous flare slamming analysis, our proposed method takes roll motion into account to calculate the impact angle and relative vertical velocity between ship sections on the bow flare and wave surface. We use the Wagner model to analyze the slamming impact forces and the slamming occurrence probability. Through numerical simulations, we investigate the maximum flare slamming pressures of a container ship for different speeds and wave conditions. To further clarify the mechanism of flare slamming phenomena in parametric rolling conditions, we also conduct real-time simulations to determine the relationship between slamming pressure and 3-DOF motions, namely roll, pitch, and heave.

文摘Pulse-burst 1064-nm picosecond azimuthal polarization beam amplification up to an average power of 16.32 W using side-pumped Nd: YAG amplifiers has been demonstrated. The maximum envelop energy as much as 16.32 mJ, corresponding to a power amplification factor of 299.5%. A simple criterion was defined to help estimate the amount of depolarization in Nd:YAG amplifier stages. The degree of depolarization of the beam was 7.1% and the beam quality was measured to be M2= 3.69. The reason for the azimuthal polarization depolarization and beam quality degradation were explained theoretically and experimentally during the amplification process.

文摘Canine atopic dermatitis(CAD)is a prevalent genetically susceptible infammatory and pruritic allergic skin condition afecting not only the health of dogs but also the quality of life of their owners.Interleukin-31(IL-31)and interleukin-31 receptor alpha(IL-31RA)are essential for the development of pruritus in primates and mice.Hence,it is expected that inhibiting IL-31RA will be an efective approach to alleviate pruritus.The purpose of the study was to produce anti-canine IL-31RA polyclonal antibodies(anti-IL-31RA pAbs)and evaluate their efcacy in inhibiting house dust mite(HDM)-evoked pruritic responses.Dogs were immunized with antigens formed by IL-31RA recombinant short peptides coupled to BSA to produce anti-IL-31RA pAbs.The CAD model was developed by using HDM allergen stimulation,and the efects of IL-31RA pAbs on the reduction of pruritus in CAD model dogs were examined.The Canine Atopic Dermatitis Extent and Severity Index(CADESI)-4 and pruritus Visual Analog Scale(pVAS)were utilized to evaluate pruritic responses,and skin tissue samples were collected from the inguinal area for pathological assessment of skin infammatory cell infltration.The results showed that anti-IL-31RA pAbs with high titers(1:128,000)and specifcity were efectively produced.In the CAD model group,the severity of skin damage,pruritus score,infammatory cell infltration and level of infammatory factors were considerably elevated.Anti-IL-31RA pAbs relieved pruritic behavior and dermatitis in dogs compared to placebo-treated dogs.In conclusion,anti-IL-31RA pAbs efectively suppressed CAD in vivo and are anticipated to be an efective novel treatment for pruritic skin disorders such as CAD.
