Screening for metronidazole-resistance associated gene fragments of H pylori by suppression subtractive hybridization 被引量:3

AIM: To screen for metronidazole (MTZ)-resistance associated gene fragments of H pylori by suppression subtractive hybridization (SSH). METHODS: Five MTZ-resistant (tester, T) and 1 MTZ- susceptible (driver, D) clinical H pylori isolates were selected. Genomic DNAs were prepared and submitted to Rsa I digestion. Then two different adaptors were ligated respectively to the 5'-end of two aliquots of the tester DNA fragments and SSH was made between the tester and driver DNAs. The specific inserts of tester strains were screened and MTZ-resistance related gene fragments were identified by dot blotting. RESULTS: Among the randomly selected 120 subtractive colonies, 37 DNA fragments had a different number of DNA copies (≥ 2 times) in resistant and susceptible strains and 17 of them had a significantly different number of DNA copies (≥ 3 times). Among the sequences obtained from the 17 DNA fragments, new sequences were found in 10 DNA fragments and duplicated sequences in 7 DNA fragments, representing respectively the sequences of depeptide ABC transporter periplasmic dipeptide-binding protein (dppA), permease protein (dppB), ribosomal protein S4 (rps4), ribonuclease Ⅲ (rnc), protease (pqqE), diaminopimelate epimerase (dapF), acetatekinase (ackA), H pylori plasmid pHP51 and Hpylori gene 1334. CONCLUSION: Gene fragments specific to MTZ-resistant H pylori strains can be screened by SSH and may be associated with MTZ-resistant Hpylori. 展开更多

A finite equilibrium current density arises in the anomalous Hall effect(AHE)as a result of time-reversal symmetry breaking,affecting both the differential current density and total current.In this paper,we illustrate the equilibrium current density in a ribbon-shaped system within the AHE regime,consisting of two sets of counterpropagating channels arranged in a zebra stripes pattern.While the middle channels are susceptible to scattering,the edge channels remain relatively robust.Despite this difference,all channels exhibit the same differential current density when subjected to a differential voltage across the two ends of the ribbon.When a differential voltage is applied to both sides of the ribbon,it results in a snaking pattern of differential current density forming across it.Furthermore,in a four-terminal device comprising the ribbon and two normal leads,it is found that Hall conductance is independent of ribbon width within certain scattering strengths due to the differences in robustness between middle and edge channels.These findings disclose the details of the AHE transport in a finite-sized system under weak scattering. 展开更多

Over the past few decades,angle-resolved photoemission spectroscopy(ARPES)has been one of the important tools to study electronic structure of crystals.In recent years,the spatial resolution of around 150 nm has been reached through tight focusing of the light spot(nano-ARPES).At present,the lower limit of the spot size of the light on the sample has been reached.Another way to further improve the spatial resolution is through using apertures to only let electrons from a small area of the sample pass.With both back-focal plane and image apertures,the size of the selected area can be as small as 20 nm.Yet,without aberration correction,the maximum opening angle at the sample for 20 nm spatial resolution is usually smaller than 3°,making this method not suitable for nano-ARPES.As shown in this paper,a conventional aberration corrector,which corrects chromatic and third-order spherical aberrations,is not enough either.Only when the fifth-order spherical aberration is also corrected,the opening angle at the sample is large enough for nano-ARPES.In this paper,the design of a time-of-fight PEEM/ARPES/nano-ARPES instrument,which is currently under development at the Quantum Science Center of Guangdong-Hong Kong-Macao Greater Bay Area,is presented.The main point of innovation is a fiveelectrode electron mirror corrector,which is used to correct simultaneously chromatic,third-order and fifth-order spherical aberrations,resulting in 1 nm spatial resolution with~230 mrad aperture angle in PEEM mode.This makes feasible the method of using apertures to improve the spatial resolution of the nano-ARPES mode.A new design of the magnetic prism array(MPA)is also presented,which preserves the rotational symmetry better than the existing designs. 展开更多

Helicobacter pylori eradication therapy for functional dyspepsia: Systematic review and meta-analysis 被引量:34

AIM: To evaluate whether Helicobacter pylori (H. pylori) eradication therapy benefits patients with functional dyspepsia (FD).METHODS: Randomized controlled trials (RCTs) investigating the efficacy and safety of H. pylori eradication therapy for patients with functional dyspepsia published in English (up to May 2015) were identified by searching PubMed, EMBASE, and The Cochrane Library. Pooled estimates were measured using the fixed or random effect model. Overall effect was expressed as a pooled risk ratio (RR) or a standard mean difference (SMD). All data were analyzed with Review Manager 5.3 and Stata 12.0.RESULTS: This systematic review included 25 RCTs with a total of 5555 patients with FD. Twenty-three of these studies were used to evaluate the benefits of H. pylori eradication therapy for symptom improvement; the pooled RR was 1.23 (95%CI: 1.12-1.36, P &#x0003c; 0.0001). H. pylori eradication therapy demonstrated symptom improvement during long-term follow-up at &#x02265; 1 year (RR = 1.24; 95%CI: 1.12-1.37, P &#x0003c; 0.0001) but not during short-term follow-up at &#x0003c; 1 year (RR = 1.26; 95%CI: 0.83-1.92, P = 0.27). Seven studies showed no benefit of H. pylori eradication therapy on quality of life with an SMD of -0.01 (95%CI: -0.11 to 0.08, P = 0.80). Six studies demonstrated that H. pylori eradication therapy reduced the development of peptic ulcer disease compared to no eradication therapy (RR = 0.35; 95%CI: 0.18-0.68, P = 0.002). Eight studies showed that H. pylori eradication therapy increased the likelihood of treatment-related side effects compared to no eradication therapy (RR = 2.02; 95%CI: 1.12-3.65, P = 0.02). Ten studies demonstrated that patients who received H. pylori eradication therapy were more likely to obtain histologic resolution of chronic gastritis compared to those who did not receive eradication therapy (RR = 7.13; 95%CI: 3.68-13.81, P &#x0003c; 0.00001).CONCLUSION: The decision to eradicate H. pylori in patients with functional dyspepsia requires individual assessment. 展开更多

AIM To evaluate the outcomes of furazolidone-and amoxicillin-based quadruple therapy for treatment of Helicobacter pylori(H. pylori) infection and identify predictors of failed eradication.METHODS Patients with H. pylori infection treated with furazolidone, amoxicillin, bismuth, and proton pump inhibitor therapy(January 2015 to December 2015) who received the ^(13)C-urea breath test > 4 wk after treatment were evaluated. Demographic and clinical data including prior H. pylori treatment attempts, medication adherence, alcohol and cigarette consumption during therapy, and treatment-related adverse events were recorded by reviewing medical records and telephone surveys. H. pylori eradication rates for overall and subgroups were evaluated. Multivariate analysis was performed to identify independent predictors of failed H. pylori eradication.RESULTS Of the 992 patients treated and retested for H. pylori infection, the overall eradication rate was 94.5% [95% confidence interval(CI): 94.1%-95.9%]. H. pylori eradication rate of primary therapy was 95.0%(95%CI: 93.5%-96.5%), while that of rescue therapy was 91.3%(95%CI: 86.8%-95.8%). Among the 859 patients who completed the study protocol, 144(17%) reported treatment-related adverse events including 24(3%) leading to premature discontinuation. On multivariate analysis, poor medication adherence [adjusted odds ratio(AOR) = 6.7, 95%CI: 2.8-15.8], two or more previous H. pylori treatments(AOR = 7.4, 95%CI: 2.2-24.9), alcohol consumption during therapy(AOR = 4.4, 95%CI: 1.5-12.3), and possibly smoking during therapy(AOR = 1.9, 95%CI: 0.9-4.3) were associated with failed H. pylori eradication. CONCLUSION Furazolidone-and amoxicillin-based quadruple therapy for H. pylori infection in an area with a high prevalence of clarithromycin resistance demonstrated high eradication rates as primary and rescue therapies with a favorable safety profile. Patient education targeting abstinence from alcohol during therapy and strict medication adherence may further optimize H. pylori eradication. 展开更多

AIM To determine whether fructo-oligosaccharide(FOS) affects visceral sensitivity, inflammation, and production of intestinal short-chain fatty acids(SCFA) in an irritable bowel syndrome(IBS) mouse model.METHODS Mice were randomly assigned to daily oral gavage of saline solution with or without FOS(8 g/kg body weight) for 14 d. Mice were further assigned to receive either daily one-hour water avoidance stress(WAS) or sham-WAS for the first 10 d. After 2 wk, visceral sensitivity was measured by abdominal withdrawal reflex in response to colorectal distension and mucosal inflammation was evaluated. Gas chromatography, real-time reverse transcription PCR, and immunohistochemistry assays were used to quantify cecal concentrations of SCFA, intestinal cytokine expression, and number of intestinal mast cells per high-power field(HPF), respectively.RESULTS Mice subjected to WAS exhibited visceral hypersensitivity and low-grade inflammation. Among mice subjected to WAS, FOS increased visceral hypersensitivity and led to higher cecal concentrations of acetic acid(2.49 ± 0.63 mmol/L vs 1.49 ± 0.72 mmol/L, P < 0.05), propionic acid(0.48 ± 0.09 mmol/L vs 0.36 ± 0.05 mmol/L, P < 0.01), butyric acid(0.28 ± 0.09 mmol/L vs 0.19 ± 0.003 mmol/L, P < 0.05), as well as total SCFA(3.62 ± 0.87 mmol/L vs 2.27 ± 0.75 mmol/L, P < 0.01) compared to saline administration. FOS also increased ileal interleukin(IL)-23 mR NA(4.71 ± 4.16 vs 1.00 ± 0.99, P < 0.05) and colonic IL-1β mR NA(2.15 ± 1.68 vs 0.88 ± 0.53, P < 0.05) expressions as well as increased mean mast cell counts in the ileum(12.3 ± 2.6 per HPF vs 8.3 ± 3.6 per HPF, P < 0.05) and colon(6.3 ± 3.2 per HPF vs 3.4 ± 1.2 per HPF, P < 0.05) compared to saline administration in mice subjected to WAS. No difference in visceral sensitivity, intestinal inflammation, or cecal SCFA levels was detected with or without FOS administration in mice subjected to sham-WAS.CONCLUSION FOS administration intensifies visceral hypersensitivity and gut inflammation in stress-induced IBS mice, but not in the control mice, and is also associated with increased intestinal SCFA production. 展开更多

AIM: To validate 4-sample lactose hydrogen breath testing(4SLHBT) compared to standard 13-sample LHBT in the clinical setting.METHODS: Irritable bowel syndrome patients with diarrhea(IBS-D) and healthy volunteers(HVs) were enrolled and received a 10 g, 20 g, or 40 g doselactose hydrogen breath test(LHBT) in a randomized, double-blinded, controlled trial. The lactase gene promoter region was sequenced. Breath samples and symptoms were acquired at baseline and every 15 min for 3 h(13 measurements). The detection rates of lactose malabsorption(LM) and lactose intolerance(LI) for a 4SLHBT that acquired four measurements at 0, 90, 120, and 180 min from the same data set were compared with the results of standard LHBT.RESULTS: Sixty IBS-D patients and 60 HVs were studied. The genotype in all participants was C/C-13910. LM and LI detection rates increased with lactose dose from 10 g, 20 g to 40 g in both groups(P < 0.001). 4SLHBT showed excellent diagnostic concordance with standard LHBT(97%-100%, Kappa 0.815-0.942) with high sensitivity(90%-100%) and specificity(100%) at all three lactose doses in both groups.CONCLUSION: Reducing the number of measurements from 13 to 4 samples did not significantly impact on the accuracy of LHBT in health and IBS-D. 4SLHBT is a valid test for assessment of LM and LI in clinical practice. 展开更多

Objective:To observe the effect and mechanism of Xiangsha Liujunzi decoction(XSLJZD)drug serum on gastric antrum smooth muscle cells(SMCs)in rats with functional dyspepsia(FD).Methods:Gastric antrum SMCs from rats with FD were isolated,cultured,and then divided into six groups as follows:control,model,domperidone,low-dose XSLJZD(LXSLJZD),medium-dose XSLJZD(MXSLJZD),and high-dose XSLJZD(HXSLJZD).Each group was administered the corresponding drug serum for intervention.Drug serum intervention conditions and proliferative activity of SMCs were tested by cholecystokinin octapeptide.Ghrelin,gastrin,somatostatin,and substance P(SP)levels were measured by ELISA.Somatostatin and SP mRNA expression was measured by real-time PCR.Results:A concentration of 10%drug serum for 24 h was decided to be the best intervention condition for later study.The mean optical density value in the model group was lower than that in the control group(P紏.001).Optical density values in the domperidone and HXSLJZD groups were higher than those in the model group(P?.025,P?.032,respectively).Gastrin,SP,and ghrelin levels in the model group were lower(P?.007,P?.037,P?.005,respectively),but somatostatin levels were higher,compared with those in the control group(P?.031).Gastrin,SP,and ghrelin levels in the domperidone,MXSLJZD,and HXSLJZD groups were higher than those in the model group(all P<.05).Somatostatin levels in the four drug-treated groups were lower than those in the model group(P?.002,P?.007,P?.001,P?.009,respectively).SP mRNA levels in the model group were lower than those in the control,domperidone,MXSLJZD,and HXSLJZD groups(P?.037 P?.016,P?.025,P?.002,respectively).Somatostatin mRNA levels in the model group were higher than those in the control and MXSLJZD groups(P紏.042,P紏.035).Conclusions:XSLJZD and domperidone drug serum effectively promote proliferative activity of gastric antrum SMCs in an FD model.The mechanism of this activity may be regulated by gastrointestinal hormones. 展开更多

In addition, the oxide spacer layer between the metal (e.g., AI, Ag and Au) substrate and the perovskite film plays an important role. According to the design specifications,the construction of extended piles involves traversing the tunnel’s upper region and extending to the underlying rock layer.To address this challenge,a subterranean transfer structure spanning multiple subway tunnels was proposed.Deliberating on the function of piles in the transfer structure as springs with axial and bending stiffness,and taking into account the force balance and deformation coordination conditions of beams and plates within the transfer structure,we established a simplified mechanical model that incorporates soil stratification by combining it with the Winkler elastic foundation beam model.The resolved established simplifiedmechanicalmodel employed finite difference technology and the Newton-Simpsonmethod,elucidating the mechanical mechanism of the transfer structure.The research findings suggest that the load carried by the upper structural columns can be transferred to the pile foundation beneath the beams through the transfer structure,subsequently reaching the deep soil layer and ensuring minimal impact on adjacent tunnels.The established simplified analysis method can be used for stress analysis of the transfer structure,concurrently considering soil stratification,pile foundation behavior,and plate action.The pile length,pile section size,and beam section size within the transfer structure should account for the characteristics of the upper load,ensuring an even distribution of the beam bending moment. 展开更多

A type of Si-based blocked impurity band photoelectric detector with a planar architecture is designed and demonstrated by a modified silicon semiconductor processing technique.In this route,multiple ion implantation is utilized to ensure the uniform distribution of the P elements in silicon,and rapid thermal annealing treatment is used to activate the P atoms and reduce damages caused by ion-implantation.The fabricated prototype device exhibits an excellent photoelectric response performance.With a direct current(DC)bias voltage of-2.3 V,the device detectivity to blackbody irradiation is as high as 5×10^(13)cm·Hz^(1/2)/W,which corresponds to a device responsivity of nearly 4.6 A/W,showing their potential applications in infrared detection,infrared astrophysics,and extraterrestrial life science.In particular,the developed device preparation process is compatible with that for the CMOS-circuit,which greatly reduces the manufacturing cost. 展开更多

Objective:To determine the effect of gut microbiota on a rat model of post-traumatic stress disorder(PTSD)and explore the correlation of gut microbiota with behavior and neurotransmitters.Methods:We established a single prolonged stress(SPS)model to examine the pathogenesis of PTSD on rat behavior,gut microbiota,and neurotransmitter levels.Rats were separated into control and model groups,and neurotransmitter levels were measured using enzyme linked immunosorbent assay.Then,16 S rRNA sequencing was used to compare the gut microbiota between the control and model groups.Results:Compared with those in the control group,freezing time significantly increased,while number of standing upright,crossing frequency,time spent in the central arena,and total distance traveled were significantly reduced in the model group after exposure to SPS(all P<.05).Meanwhile,serotonin,or 5-hydroxytryptamine,levels in the brain in the model group were significantly lower than those the control group(P紏.0332).In addition,changes were observed in the gut microbiota diversity and relative abundances of bacterial phyla,orders,families,and genera in the model group.Especially,changes in Firmicutes,Bacteroidetes,Cyanobacteria,and Proteobacteria levels were most pronounced after SPS exposure.Correlation analysis showed that the strongest positive correlation was found between Bacteroidaceae and 5-HT(P?.0009).Moreover,RF32 abundance was the most negatively related to 5-HT(P?.0009),crossing frequency(P?.0007),and total distance(P?.0003).Conclusion:Our results suggest that SPS model rats showed differences in behavior,neurotransmitter levels,and gut microbiota with control rats.Moreover,Firmicutes,Bacteroidetes,Cyanobacteria,and Proteobacteria were most relevant to the exhibited fear-like and anxiety-like behaviors and significant serotonin content reduction in SPS model rats. 展开更多

PbZrxTi1-xO3(PZT)fims are fabricated on F-doped tin oxide(FTO)substrates using chemical solutions containing PVP polymer and rapid thermal annealing processing.The dependence of the layered PZT multilayer formation and their optical properties on the Zr content x are examined.It is found that all the PZT films are crystallized and exhibit 110-preferred orientation.When x varies in the region of 0-0.8,the PZT films display lamellar structures,and a high reflection band occurs in each optical reflectance spectrum curve.Especially,those PZT fikms with Zr/Ti atomic ratio of 35/65-65/35 show clearly layered cross-sectional morphologies arranged alternatively by porous and dense PZT layers,and have a peak optical reflectivity of>70%and a band width of>45 nm.To obtain the optimal Bragg reflection performance of the PZT multilayers,the Zr content should be selected in the range of 0.35-0.65. 展开更多

Objective:Microcirculatory disturbance is pathologically critical to acute pancreatitis(AP),which can be effectively alleviated by traditional Chinese medicine(TCM)formulas that activate blood flow.However,there has been no evidence-based research to date.Therefore,a well-designed systematic review and meta-analysis is necessary to elucidate the therapeutic transformative benefit of improving microcirculation during AP.This study aims to confirm the therapeutic efficacy of TCM formulas and explore the potential mechanisms underlying their effects on AP treatment.Methods:Studies from eight databases including Pubmed,Embase,Web of Science,Cochrane Library,CNKI,CBM,Wanfang,and Chinese VIP,were screened for the eligible randomized controlled trials(RCTs).The APACHEⅡscore and effectiveness rate were set as primary outcomes,while mortality rate,complications,total hospital stays,serum amylase recovery time,the time until the disappearance of abdominal pain,microcirculation indicators,and inflammation indicators were chosen as secondary outcomes.A systematic review and meta-analysis were subsequently conducted.Network pharmacology analysis was performed to analyze potential bioactive components with relevant targets of the core herbs included in the TCM formulas for activating blood flow.Results:A total of 51 RCTs(n=3721)were included.Compared with conventional western medical treatments alone,TCM groups were associated with lower APACHEⅡscore(SMD=1.36,95%CI:2.01 to 0.71,P=0.000)and higher effectiveness rate(RR:1.22,95%CI:1.18 to 1.26,P=0.000).Furthermore,the formulas for activating blood flow demonstrated significant efficacy in improving both microcirculation and inflammation indicators.Additionally,six core Chinese herbal medicines including Rhei Radix et Rhizoma with the highest frequency,Aurantii Fructus Immaturus,Paeoniae Radix Rubra,Bupleuri Radix,Salviae Miltiorrhizae Radix et Rhizoma,and Corydalis Rhizoma were filtered out from the adopted TCM formulas.Finally,166 shared targets between the six herbs and AP were identified.KEGG analysis indicated that lipid and atherosclerosis pathway is highly related to microcirculation.Conclusion:TCM formulas for activating blood flow significantly improve microcirculation and alleviate AP.Further high-quality,well-designed RCTs and deep mechanism exploration are required. 展开更多

The realization of controllable polarity photoresponse within a single device is a crucial advancement for simulating biological bipolar vision cells to drive the development of next-generation optoelectronic technologies.Nevertheless,current polarity photodetectors face significant challenges in fully suppressing symmetric photocurrent cancellation and optimizing carrier transport efficiency.Here,we propose a graphene-intercalated MoS_(2)/MoTe_(2)heterojunction,featuring a tailorable built-in electric field and a high efficiency transport channel.Spatially resolved photocurrent reveals that the controllable polarity photoresponse originates from the bias-dependent equivalent built-in electric field of MoS_(2)/MLG/MoTe_(2)heterojunction.The controllable polarity photoresponse realizes a large-area uniform“heart-shaped”photocurrent region.In enhanced polarity photoresponse mode,the photodetector exhibits broadband detection capabilities from visible(638 nm)to infrared(1550 nm)light,achieving a high responsivity of 18.1 A/W and an excellent detectivity of 2.8×10^(12)Jones,as well as fast response times of 94/119μs.Furthermore,precise imaging with a resolution better than 0.5 mm was successfully demonstrated,highlighting its polarity photoresponse for practical imaging applications.This work provides a new paradigm for controllable polarity photoresponse programmed by intercalated low-dimensional material structures,paving the way for next-generation intelligent sensing chips. 展开更多

Synthetic control of metal oxide nanocrystals with precise size and morphology is of great importance for promising optoelectronic applications due to their unique size-and shape-dependent optical/optoelectronic properties.Nevertheless,the understanding of the mechanism for size and morphology control of metal oxide nanocrystals are less studied.Here,we demonstrate that acetic acid,the byproduct of the initial chemical reaction of precursors,plays a dominant role in determining the morphology of indium oxide(In_(2)O_(3))nanocrystals by influencing the nucleation of the nanocrystals formation.Sufficient acetic acid would induce anisotropic growth,leading to the generation of nanoflowers,while limited existence of acetic acid results in sphere-shaped nanocrystals.Furthermore,the effects of geometries of In_(2)O_(3) nanocrystals on their plasmonic properties are studied.The resulting plasmonic In_(2)O_(3) nanocrystals show size-tunable plasmon resonance peaks in the near-infrared to mid-infrared regime and outstanding air/thermal stability.Our work shall give an in-depth understanding of the mechanism for geometry control of nanocrystals and offer more opportunities in potential optoelectronic and photothermal applications based on plasmonic metal oxide nanocrystals. 展开更多

Background: Functional dyspepsia (FD) is a common tipper gastrointestinal disorder worldwide, but the current treatments for FD are still unsatisfactory. The aims of this study were to investigate the efficacy and safety of Qi-Zhi-Wei-Tong granules in patients with postprandial distress syndrome (PDS)-predominant FD. Methods: The study was conducted as a randomized, double-blinded, multicenter, placebo-controlled design in 197 patients with PDS. All participants received placebo treatment for 1 week. Patients whose total symptom score decreased by 〈50% after the placebo treatment were recruited into the 4-week treatment period, in which they were randomly assigned to be treated with either Qi-Zhi-Wei-Tong granules or placebo. The patients were then followed for 2 weeks without any treatment. Dyspeptic symptoms were scored at weeks 2 and 4 during the random treatment period and 2 weeks after the treatment. Anxiety and depression symptoms were also scored and compared. Results: (1) The total effective rates in the Qi-Zhi-Wei-Tong granules group at weeks 2 and 4 during the random treatment period and 2 weeks after treatment were all significantly higher than those in the placebo group (38.82% vs. 8.75%, P 〈 0.001; 69.14% vs. 16.25%, P 〈 0.001' 77.65% vs. 21.25%, P 〈 0.001 ). (2) The total dyspeptic symptoms scores in the Qi-Zhi-Wei-Tong granules group at weeks 2 and 4 and 2 weeks after treatment were significantly lower than those in the placebo group. (3) The severity and frequency of each dyspeptic symptom at weeks 2 and 4 and the follow-up period were all significantly lower than those in the placebo group. (4) The anxiety scores in the Qi-Zhi-Wei-Tong granules group were significantly lower than those in the placebo group. (5) Qi-Zhi-Wei-Tong granules did not have more adverse eflbcts than the placebo.
