The safety and stability of high-speed maglev trains traveling on viaducts in crosswinds critically depend on their aerodynamic characteristics.Therefore,this paper uses an improved delayed detached eddy simulation(IDDES)method to investigate the aerodynamic features of high-speed maglev trains with different marshaling lengths under crosswinds.The effects of marshaling lengths(varying from 3-car to 8-car groups)on the train’s aerodynamic performance,surface pressure,and the flow field surrounding the train were investigated using the three-dimensional unsteady compressible Navier-Stokes(N-S)equations.The results showed that the marshaling lengths had minimal influence on the aerodynamic performance of the head and middle cars.Conversely,the marshaling lengths are negatively correlated with the time-average side force coefficient(CS)and time-average lift force coefficient(Cl)of the tail car.Compared to the tail car of the 3-car groups,the CS and Cl fell by 27.77%and 18.29%,respectively,for the tail car of the 8-car groups.It is essential to pay more attention to the operational safety of the head car,as it exhibits the highest time average CS.Additionally,the mean pressure difference between the two sides of the tail car body increased with the marshaling lengths,and the side force direction on the tail car was opposite to that of the head and middle cars.Furthermore,the turbulent kinetic energy of the wake structure on the windward side quickly decreased as marshaling lengths increased. 展开更多

针对传统的人工更换带式输送机平托辊劳动强度大、安全性差、无法实现在不停机的状态下完成平托辊的更换等问题,文中以带式输送机实验台为研究对象,设计了一种带式输送机平托辊更换车。根据实际工况与功能要求确定平托辊更换方案,并对... 针对传统的人工更换带式输送机平托辊劳动强度大、安全性差、无法实现在不停机的状态下完成平托辊的更换等问题,文中以带式输送机实验台为研究对象,设计了一种带式输送机平托辊更换车。根据实际工况与功能要求确定平托辊更换方案,并对更换车核心部件进行结构设计,利用SolidWorks建立更换车三维模型。基于带式输送机实验台基本参数,对输送带举升机构进行受力分析,同时建立剪叉式升降机构的力学模型,根据虚功原理推导出伺服电缸推力与所受负载之间的表达式。采用Ansys Workbench对车架和剪叉式升降机构进行有限元仿真分析,仿真结果表明,更换车的强度与刚度均满足使用要求。采用ADAMS对剪叉式升降机构进行动力学仿真分析,得到升降平台位移、速度与加速度随时间变化的曲线图。结果表明,该机构在工作过程中运行平稳,验证了结构设计的合理性。 展开更多

针对固体火箭发动机推进剂/衬层界面,从性能失效、组分迁移、表征方法和在线监测四方面综述了界面贮存性能研究进展。首先,从界面脱粘和裂纹扩展两方面讨论界面贮存性能失效,提出有必要探索准确获取界面性能参数的方法和发动机层次的界... 针对固体火箭发动机推进剂/衬层界面,从性能失效、组分迁移、表征方法和在线监测四方面综述了界面贮存性能研究进展。首先,从界面脱粘和裂纹扩展两方面讨论界面贮存性能失效,提出有必要探索准确获取界面性能参数的方法和发动机层次的界面裂纹扩展试验方法,以完善发动机界面性能失效分析体系。其次,分析增塑剂和安定剂等不同组分的迁移机理,认为分子动力学模拟能降低组分迁移机理研究的成本。如能进一步与组分迁移抑制的工程经验相结合,建立完整的界面组分迁移模拟和评价体系,对组分迁移机理研究具有重要意义。再次,从宏观、细观和微观多个层次讨论了界面贮存性能的表征问题,提出应构建“宏观-细观-微观”的跨尺度综合表征体系,由浅入深,从不同尺度和不同特性全面表征界面贮存性能。最后,分析了实现界面贮存性能在线监测的主要技术途径,指出集成化、微型化和适应性是界面在线监测需要攻克的关键技术。 展开更多

基于MICU2探讨红花清肝十三味丸对急性肝损伤的保护作用

探讨红花清肝十三味丸(HHQG)对四氯化碳诱导急性肝损伤小鼠的保护效应及其分子机制,特别关注线粒体钙离子摄取蛋白2(Mitochondrial calcium uniporter 2,MICU2)与氧化应激的关联。通过HE/Masson组织学、血清学、肝组织氧化应激生化指标... 探讨红花清肝十三味丸(HHQG)对四氯化碳诱导急性肝损伤小鼠的保护效应及其分子机制,特别关注线粒体钙离子摄取蛋白2(Mitochondrial calcium uniporter 2,MICU2)与氧化应激的关联。通过HE/Masson组织学、血清学、肝组织氧化应激生化指标评估病理变化,基于不同时相(12~120 h)开展RNA-seq时序分析并以qPCR验证关键基因;采用蛋白质组学鉴定差异表达蛋白并进行功能富集与免疫组织化学时相验证;通过Pearson相关分析探讨MICU2与SOD/MDA关系;采用AutoDock Vina对HHQG关键活性成分(Apigenin、Emodin、Genistein、Kaempferol、Quercetin)与MICU2进行分子对接预测结合位点。结果表明,在12~72 h CCl4能够诱导明显的肝小叶结构破坏、炎症浸润及早期胶原沉积。此外,血清丙氨酸氨基转移酶(Alanine aminotransferse,ALT)和天冬氨酸氨基转移酶(Aspartate aminotransferase,AST)在12~72 h达高峰;HHQG可在早期显著减轻组织病理损伤、抑制炎症与胶原沉积,并使ALT、AST、超氧化物歧化酶(Superoxide dismutase,SOD)、丙二醛(Malondialdehyde,MDA)等指标部分或完全恢复。qPCR与蛋白免疫组化提示,MICU2在中期(48~72 h)显著上调,HHQG能抑制其异常上调。Pearson相关分析显示,在模型组各时间点,MICU2与SOD活性呈显著负相关(P<0.05),而与MDA含量呈显著正相关(P<0.05)。经HHQG干预后,随着MICU2表达被有效抑制,MICU2与SOD、MDA之间的相关性发生逆转,在各时间点MICU2与SOD呈正相关,与MDA呈负相关。蛋白组学结果也显示,HHQG对解毒、蛋白清除与抗氧化相关通路的调控。分子对接预测表明MICU2存在两个可结合口袋,5种活性成分均可与之结合(Apigenin得分最优),提示可能的直接作用位点。HHQG通过抑制炎症与胶原沉积、增强抗氧化能力并调控MICU2相关的线粒体钙稳态,从分子与功能层面缓解CCl4诱导的急性肝损伤。MICU2及其与HHQG活性成分的相互作用可能为其抗氧化防护作用的重要机制与潜在靶点。 展开更多

Dear Editor,The orientation of a membrane protein can provide important information about its interaction partners,function,and mechanism of action.In addition to traditional biochemical assays,new methods have been developed to reveal the orientation of membrane proteins.For example,computer programs have been developed to predict membrane protein topology but often have inaccuracies(Ott and Lingappa,2002;Schwacke et al.,2003).A widely used method is the fluorescence protease protection(FPP)assay(Lomize et al.,2006;Lorenz et al.,2008;White et al.,2015).However,it often fails to give good results for assays of plant membrane proteins.Here,we developed a simpler method based on release of a fluorescent protein tag(RFT)by the tobacco etch virus(TEV)protease to overcome the shortcomings of FPP and analyze the orientation of membrane proteins in plants. 展开更多
