OBJECTIVE: To investigate the effects of Huogu I formula on regulation of lipid metabolism in ste- roid-induced osteonecrosis of the femoral head (SONFH) rats and verify our hypothesis that Huogu I formula regulates lipid metabolism by down-regulating peroxisome proliferator-activated receptor gamma (PPARy) expression and activating Wnt signaling pathways. METHODS: Eighty-five rats were divided into four groups: control, model, Huogu 15 g/kg and Huogu 30 g/kg. Six weeks later, animals were anaesthe- tized, femora were dissected for histopathologicalexamination of the osteonecrotic changes and re- pair processes, micro computed tomography (Mi- cro-CT)-based micro-angiography was performed to assess vascularization. Serum lipid levels were detected by haematological examination. The ex- pressions of PPARy, Wnt3a, low density lipoprotein receptor-related protein 5 (LRP5) and 13-catenin were evaluated by immunohistochemistry, Western blot and quantitative real-time polymerase chain reaction analyses. RESULTS: The incidence of osteonecrosis, ratio of empty lacuna, adipose tissue area and adipocyte perimeter in the bone marrow were dramatically lower in the Huogu ~ formula treatment groups. By micro-CT quantification, Huogu ~ formula treat- ment dose-dependently increased vessel volume, vessel surface, percentage of vessel volume and vessel thickness of the femoral heads of SONFH rats. Levels of serum lipid in Huogu 15 g/kg and Huogu 30 g/kg groups reduced significantly. HuoguⅠformula treatment could suppress the ex- pression of PPARy and increase the expressions of Wnt3a, LRP5 and 13-catenin at both protein and mRNA levels. CONCLUSION: The results of our present study highlight the lipid-lowering potential of Huogu Ⅰ formula, and provide further evidence of the in- volvement of the PPARy inhibition and Wnt/LRPS/ 13-catenin signaling activation in the effects of Huogu Ⅰ formula. 展开更多

BACKGROUND Multidrug-resistant Gram-negative bacteria,exacerbated by excessive use of antimicrobials and immunosuppressants,are a major health threat.AIM To study the clinical efficacy and safety of colistin sulfate in the treatment of carbapenem-resistant Gram-negative bacilli-induced pneumonia,and to provide theoretical reference for clinical diagnosis and treatment.METHODS This retrospective analysis involved 54 patients with Gram-negative bacilli pneumonia admitted to intensive care unit of The General Hospital of the Northern Theater Command of the People's Liberation Army of China from August 2020 to June 2022.After bacteriological culture,the patients'airway secretions were collected to confirm the presence of Gram-negative bacilli.The patients were divided into the experimental and control groups according to the medication used.The research group consisted of 28 patients who received polymyxin sulfate combined with other drugs through intravenous,nebulization,or intravenous combined with nebulization,with a daily dosage of 1.5–3.0 million units.The control group consisted of 26 patients who received standard dosages of other antibiotics(including sulbactam sodium for injection,cefoperazone sodium sulbactam for injection,tigecycline,meropenem,or vaborbactam).RESULTS Of the 28 patients included in the research group,26 patients showed improvement,treatment was ineffective for two patients,and one patient died,with the treatment efficacy rate of 92.82%.Of the 26 patients in the control group,18 patients improved,treatment was ineffective for eight patients,and two patients died,with the treatment efficacy rate of 54.9%;significant difference was observed between the two groups(P<0.05).The levels of white blood cell(WBC),procalcitonin(PCT),and C-reactive protein(CRP)in both groups were significantly lower after treatment than before treatment(P<0.05),and the levels of WBC,PCT,and CRP in the research group were significantly lower than those in the control group(P<0.05).Compared with before treatment,there were no significant changes in aspartate aminotransferase,creatinine,and glomerular filtration rate in both groups,while total bilirubin and alanine aminotransferase decreased after treatment(P<0.05)with no difference between the groups.In patients with good clinical outcomes,the sequential organ failure assessment(SOFA)score was low when treated with inhaled polymyxin sulfate,and specific antibiotic treatment did not improve the outcome.Sepsis and septic shock as well as a low SOFA score were independent factors associated with good clinical outcomes.CONCLUSION Polymyxin sulfate has a significant effect on the treatment of patients with multiple drug-resistant Gram-negative bacilli pneumonia and other infections in the lungs and is safe and reliable.Moreover,the administration route of low-dose intravenous injection combined with nebulization shows better therapeutic effects and lower adverse reactions,providing new ideas for clinical administration. 展开更多

AIM:To develop the 17-item Asthenopia Survey Questionnaire(ASQ)-17 by Rasch analysis,and to generate a predictiveness score.METHODS:Totally 739 participants were recruited and 680 were involved in the result analysis in this prospective,cross-sectional study.Three rounds of Rasch analysis were used to analyze the psychometric characteristics of items and options.RESULTS:Phase 1 assessed the original ASQ-19,adjusted the item scoring mode to a four-point Likert response rating scale and combined the 18th and 19th items into a new item.Phase 2 deleted the 11th item.Phases 3 and 4 assessed the new ASQ-17.All the evaluation indexes of ASQ-17 were acceptable.The Infit and Outfit MnSq values of items were 0.67-1.48,the variance explained by the principal component and the unexplained variance explained by the first contrast were 53.90%-59.40%and 1.50-1.80 in three dimensions.The curve peaks of scores in each dimension were separated and in the same order.The PSR and PSI values were 2.80 and 0.89,respectively.The mean scores of dimensions A(9.5±4.1 vs 3.5±3.2),B(7.3±3.3 vs 2.5±2.7),C(4.3±2.2 vs 1.4±2.0)and total(21.1±8.1 vs 7.4±7.0)in asthenopia participants were significantly higher than those without asthenopia(all P<0.001).The area under the curve in two groups was 0.899(P<0.001).Youden’s index was up to the maximum value of 0.784 when the cut-off value was 12.5.CONCLUSION:ASQ-17 has stronger option sorting and suitability than ASQ-19.It is an effective assessment tool for asthenopia with an optimal cut-off threshold value of 12.5,which is suitable for diagnosis and curative effect evaluation. 展开更多

Wu-tou-tang was applied to treat rheumatoid arthritis,rheumatic arthritis,and joint pain for thousands of years.Aconiti Radix Cocta is the primary component of Wu-tou-tang,and Aconiti Radix and Pinelliae Rhizoma is one of the famous combination taboos in traditional Chinese medicine.A urinary metabonomics method based on the ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS) had been established to quantify the changes of the endogenous metabolites in the urine of adjuvant-induced arthritis rats treated by Wutou-tang and Wu-tou-tang and Pinelliae Rhizoma(WP) co-decoctions.There was a clear separation between Wu-tou-tang and WP co-decoction treated groups in the PCA model.16 potential biomarkers had been identified which correlated with how Pinelliae Rhizoma influenced the therapeutic effect made by Wu-tou-tang.Results showed that it could reduce the therapeutic effects or make some side effects when Pinelliae Rhizoma is added in Wu-tou-tang.This is the first time to use metabonomics to investigate the reason of combination taboo. 展开更多

OBJECTIVE: To investigate the effects of Ermiao Fang(EM) with medical guide Xixin(Herba Asari Mandshurici)(HAM) on bone marrow stem cell migration to a focal zone in osteoarthritis(OA) rats.METHODS: OA rats were induced by arthrectomy and assigned to sham-operated, model, EM, or EM plus HAM groups.All rats were injected with recombinant human granulocyte colony-stimulating factor 30μg·kg-1·d-1for7 days and treated with EMor EM plus HAM at 1.6 or 1.9 g·kg-1·d-1 for 3 or 6 weeks, respectively. Chondrocyte apoptosis and cartilage matrix components were tested by transferase-mediated deoxyuridine triphosphate-biotin nick end labeling assay and special staining. Levels of interleukin-1 beta(IL-1β) tumor necrosis factor alpha(TNF-α) nitric oxide(NO), and inducible nitric oxide synthase(iNOS) in serum were detected by enzyme-linked immunosorbent assay or radioimmunoassay. Matrix metalloproteinases(MMPs)-13,tissue inhibitors of metalloproteinases(TIMPs)-1,Bromodeoxyuridine(BrdU), cluster of differentiation 34(CD34), and stromal cell-derived factor 1(SDF-1) were measured by immunohistochemical assay.RESULTS:The EM and EM plus HAM groups had significantly less cartilage damage and synovium inflammation the model group. Moreover, the EM and EM plus HAM groups had less chondrocyte apoptosis and more proteoglycan and collagen content than the model group.The EM and EMplus HAM groups had obviously higher MMPs-13 and TIMPs-1 expression in the cartilage than the model group. Moreover, the two formula groups had less release of IL-1β, TNF-α, NO, and iNOS than model group. Importantly, the expressions of BrdU, CD34,and SDF-1 in cartilage were significantly higher in the EM and EM plus HAM-Medtreated rats than model group. Notably, the EM plus HAM treatment seemed to have the greatest effects.CONCLUSION: HAM improves the therapeutic effects of EM on OA rats by enhancing BMSC directional homing to the focal zone. 展开更多

基于微观孔隙结构差异性的致密砂岩储层评价——以塔里木盆地中秋1区块巴什基奇克组为例 被引量:1

为厘定孔隙型致密砂岩储层孔隙结构特征、建立并完善储层分类评价标准,以塔里木盆地中秋1区块巴什基奇克组致密砂岩气藏为例,采用普通薄片、铸体薄片、扫描电镜观察及压汞实验等多种测试手段,系统分析了致密砂岩储层的孔隙和喉道特征及... 为厘定孔隙型致密砂岩储层孔隙结构特征、建立并完善储层分类评价标准,以塔里木盆地中秋1区块巴什基奇克组致密砂岩气藏为例,采用普通薄片、铸体薄片、扫描电镜观察及压汞实验等多种测试手段,系统分析了致密砂岩储层的孔隙和喉道特征及井间差异性。结果表明:中秋101井、中秋102井和中秋2井岩石学特征差异不明显,但井间微观孔隙结构具有明显差异性。中秋101井面孔率最高,物性最好,孔喉半径分布以多峰偏粗态型为主,孔隙结构条件最好。中秋102井和中秋2井以双峰偏粗态型为主。研究区孔隙结构划分为4种类型,中秋101井孔隙结构类型以A类和C类为主;中秋102井以B类和C类为主,局部发育D类;中秋2井以C类为主,同时发育B类和D类。明确原生粒间孔对储层物性贡献最大,溶孔改善物性,微孔影响不大,原生粒间孔+溶孔+微孔型物性最好,4类孔隙结构物性依次变差,沉积作用是影响储层孔隙结构差异的基础,成岩作用是影响孔隙结构差异的主要因素。根据孔喉特征参数与物性的相关性,选取最大孔喉半径、平均孔喉半径、排驱压力和分选系数作为储层分类的主要评价指标,将研究区储层分为4类,Ⅰ类为优质储层,Ⅱ类为较好储层,Ⅲ类为中等储层,Ⅳ类为差储层,优质储层位于中秋101井区。 展开更多

为了进一步提升高熵氧化物负极材料的结构稳定性及电子/离子传输动力学,本研究以尖晶石型(Cr_(0.2)Fe_(0.2)Mn_(0.2)Ni_(0.2)Zn_(0.2))_(3)O_(4)高熵氧化物为研究基础,采用溶液燃烧法,通过S^(2-)阴离子掺杂策略调控其本征缺陷与微观结构... 为了进一步提升高熵氧化物负极材料的结构稳定性及电子/离子传输动力学,本研究以尖晶石型(Cr_(0.2)Fe_(0.2)Mn_(0.2)Ni_(0.2)Zn_(0.2))_(3)O_(4)高熵氧化物为研究基础,采用溶液燃烧法,通过S^(2-)阴离子掺杂策略调控其本征缺陷与微观结构,成功制备了具有可控氧空位和晶格畸变及介孔网状结构的尖晶石型(Cr_(0.2)Fe_(0.2)Mn_(0.2)Ni_(0.2)Zn_(0.2))_(3)O_(4-x)S_(x)(x=0, 0.15, 0.3, 0.6, 0.9)高熵氧化物锂离子电池负极材料。电化学测试结果表明,(Cr_(0.2)Fe_(0.2)Mn_(0.2)Ni_(0.2)Zn_(0.2))_(3)O_(3.7)S_(0.3)(S0.3)电极展示了优异的循环稳定性和倍率性能,在200 mA/g下循环150圈后可保持1513 mAh/g的可逆放电比容量,在1000 mA/g下循环350圈后仍具有310 mAh/g的放电比容量。其优异的电化学性能主要归因于:一方面,适量S^(2-)掺杂不仅增加构型熵,还能缓解晶格畸变并降低氧空位浓度,从而显著提升电极材料的循环稳定性;另一方面,晶格畸变、氧空位与晶粒尺寸的协同优化显著改善电子/离子传输(S0.3的电导率高达22.4 S/m,且D_(Li^(+))较大)并有效提升赝电容贡献率。本研究为兼具结构稳定性、高能量密度与长循环寿命的锂离子电池负极材料设计提供了新思路与实验基础。 展开更多

Background:Bushen Zhuanggu Formula(BZF),derived from the classic Yougui Pills,has shown favorable clinical efficacy in treating advanced nontraumatic osteonecrosis of the femoral head(NONFH),particularly by promoting bone repair.However,its underlying mechanisms remain unclear.Objective:This study aimed to explore the mechanisms by which BZF promotes bone repair in advanced NONFH.Materials and methods:A total of 518 potential BZF targets were identified from the ETCMv2.0 database.Transcriptomic profiling of clinical cohorts revealed 485 differentially expressed genes in advanced NONFH patients compared to healthy controls.A drug target-disease gene interaction network was constructed to identify candidateBZF targets involved in NONFH pathogenesis.In vivo experiments were conducted to validate the effects of BZF in a rat model of advanced NONFH.Results:Network analysis identified key pathways associated with blood circulation obstruction,immune-inflammatory imbalance,and abnormal bone metabolism.Protein kinase C alpha(PKCA),Ras proto-oncogene(RAS),mitogen-activated protein kinase 3(ERK),ETS proto-oncogene 1(ETS1),and receptor activator of nuclear factor-kB ligand(RANKL)formed a signaling axis implicated in NONFH pathogenesis.BZF treatment alleviated joint inflammation,preserved trabecular bone morphology,reduced bone loss,and promoted bone repair.Mechanistically,BZF significantly downregulated the expression of PKCA,RAS,ERK,ETS1,and RANKL,improved blood circulation,and inhibited osteoclast activation while promoting osteoblast activation.Conclusion:BZF may promote bone repair in advanced NONFH by enhancing blood circulation and modulating the PKC-RAS-ERK-ETS1-RANKL signaling axis,thereby reversing dysregulated bone metabolism. 展开更多

Objective:The objective of this study was to investigate the effect of glycosides of Tripterygium wilfordii Hook.F(GTW)on bone erosion in collagen-induced arthritis(CIA)rats and osteoclastogenesis.Materials and Methods:The effects of GTW on bone destruction were assessed through hematoxylin and eosin analyses and tartrate-resistant acid phosphatase(TRAP)staining.In vitro,TRAP staining,F-actin,and quantitative polymerase chain reaction assays were used to evaluate the inhibitory effects of GTW on osteoclast differentiation.In addition,Western blot,immunohistochemistry,and immunofluorescence staining techniques were employed to explore the mechanisms of GTW by determining the expression of interleukin-8(IL-8),C-X-C motif chemokine receptor 2(CXCR2),nuclear factor of activated T cells 1(NFATc1),and p65.Results:GTW slowed the onset of arthritis and reduced arthritis scores.Our mechanistic studies demonstrated that GTW reduced the number of osteoclasts in rats with CIA and significantly suppressed receptor activator of nuclear factor kappa-B ligand-induced osteoclast differentiation,as evidenced by a decrease in TRAP-positive cells,alterations in F-actin rings,and modulation of osteoclast-specific gene expression.The inhibition of IL-8,CXCR2,NFATc1,and p65 activation by GTW was observed in both CIA rats and osteoclasts.Conversely,the introduction of IL-8 into the osteoclast culture system mitigated the effects of GTW on osteoclast differentiation.Conclusions:Our findings suggest that GTW suppressed osteoclastogenesis and bone loss by inhibiting the IL-8/CXCR2 signaling pathway.These results offer valuable insights into the potential therapeutic role of GTW in rheumatoid arthritis and lay the groundwork for future clinical applications. 展开更多

以某型号电压转换器为实例,分析零件的结构形状,选择合适的成型材料。基于响应面分析法,以塑件的翘曲变形量为目标,选取熔体温度、充填压力、充填持续时间为优化变量,设计响应面试验,建立响应面方案。将试验数据进行二次回归方程拟合,... 以某型号电压转换器为实例,分析零件的结构形状,选择合适的成型材料。基于响应面分析法,以塑件的翘曲变形量为目标,选取熔体温度、充填压力、充填持续时间为优化变量,设计响应面试验,建立响应面方案。将试验数据进行二次回归方程拟合,并对试验数据进行响应面分析,得到塑件的翘曲变形量与各影响因素的变化趋势,通过方差分析对响应面分析的可靠性进行证明,得到最小翘曲变形量下的优化工艺参数。优化后的工艺参数为:熔体温度245℃,充填压力96%,充填持续时间为9.64 s。优化结果表明:优化后塑件的翘曲变形量为1.545 mm,相比优化前降低9.6%,达到目标要求。优化模型的误差率仅为1.17%,表明响应面优化模型具有较高的准确率和可靠性,为其他类似模具的设计与优化提供借鉴与指导。 展开更多

红火蚁(Solenopsis invicta)是极具危险性的外来入侵物种,研究海岛在城市化发展中红火蚁发生程度的空间格局及驱动因子,对保护海岛生态安全具有重要意义。本研究以福建省东部的海坛岛为例,运用核密度、空间自相关模型分析红火蚁发生程... 红火蚁(Solenopsis invicta)是极具危险性的外来入侵物种,研究海岛在城市化发展中红火蚁发生程度的空间格局及驱动因子,对保护海岛生态安全具有重要意义。本研究以福建省东部的海坛岛为例,运用核密度、空间自相关模型分析红火蚁发生程度空间格局,进一步运用地理探测器揭示环境因子(8种)、社会经济因子(10种)以及两类因子交互作用对红火蚁发生程度的影响。结果显示:研究区红火蚁发生程度空间密度表现为不均匀聚集特征,农田发生程度最为严重(高-高聚集)、园林绿化用地次之(高-低聚集和低-低聚集)、居民区周边最轻,且发生区域呈现正向空间自相关关系。18种影响因子对红火蚁发生程度空间分异解释度的q值范围为0.014–0.278。从整体上看,两类因子对红火蚁发生程度的影响存在差异,且社会经济因子q值平均数高于环境因子。对红火蚁发生程度影响最大的经济因子是农村人口数量(q=0.278),其次为乡镇面积(q=0.268)。在环境因子中,土壤类型(q=0.172)和年均降水量(q=0.149)的影响力较强。环境因子与社会经济因子两两叠加作用都将正向增强红火蚁的发生程度,其中,乡镇面积与邻近景区距离、年均降水量与邻近景区距离两组因子交互作用对红火蚁发生程度的影响力最大(q=0.466)。本研究结果表明农业耕作、城镇化建设中绿化苗木引种、道路交通运输对海岛型城市红火蚁发生程度的空间格局起到关键性作用。因此,在防控中不仅要加强对调运苗木和往来货物的检疫,还应采取因地适宜的防控措施,有效抑制红火蚁种群数量和扩散速度。 展开更多

AIM:To investigate the occurrence of eye asthenopia in Chinese adults and pinpoint the factors contributing to it using a 17-item Asthenopia Survey Questionnaire(ASQ-17).METHODS:A total of 2509 Chinese adults aged 18 and above from 30 regions in China participated in a crosssectional online survey in February 2020.The survey utilized the ASQ-17,which had been proven reliable and validated for assessing asthenopia-related symptoms experienced in the past two weeks among the Chinese population.Data on demographics and living conditions,including age,gender,humidity,air quality in their residential areas,frequency of heightened anxiety or depression,daily duration of near vision activity,sleep duration,sleep quality,and history of eye surgery,were collected.Principal component analysis and multivariate logistic regression were employed to identify independent factors associated with asthenopia.RESULTS:Out of the 2502 participants,with an average age of 31±8y included in the analysis,asthenopia was prevalent in 35.2%of cases.Multivariate analysis revealed that the most influential risk factor was poor sleep and mental well-being,which encompassed shorter daily sleep duration,lower sleep quality,and more frequent feelings of heightened anxiety or depression[odds ratio(OR):2.07,95%confidence interval(CI):1.88–2.29,P<0.001].This was followed by each additional 2h of daily near vision activity relative to 4h(OR:1.33,95%CI:1.21–1.45,P<0.001),and lower humidity and worse air quality in the residential area(OR:1.10,95%CI:1.02–1.21,P=0.019).CONCLUSION:Asthenopia is a common issue among Chinese adults,and preventative measures should focus on improving sleep and mental well-being.Further research targeting physiological exposure,different age groups or longitudinal studies to establish causality are needed to explore the role of sleep and mental status as an influencing factor. 展开更多

Nowadays,a stack of heavily doped polysilicon(poly-Si)and tunnel oxide(SiO_(x))is widely employed to improve the passivation performance in n-type tunnel oxide passivated contact(TOPCon)silicon solar cells.In this case,it is critical to develop an in-line advanced fabrication process capable of producing high-quality tunnel SiO_(x).Herein,an in-line ozone-gas oxidation(OGO)process to prepare the tunnel SiO_(x) is proposed to be applied in n-type TOPCon solar cell fabrication,which has obtained better performance compared with previously reported in-line plasma-assisted N2O oxidation(PANO)process.In order to explore the underlying mechanism,the electrical properties of the OGO and PANO tunnel SiO_(x) are analyzed by deep-level transient spectroscopy technology.Notably,continuous interface states in the band gap are detected for OGO tunnel SiO_(x),with the interface state densities(D_(it))of 1.2×10^(12)–3.6×10^(12) cm^(-2) eV^(-1) distributed in Ev+(0.15–0.40)eV,which is significantly lower than PANO tunnel SiO_(x).Furthermore,X-ray photoelectron spectroscopy analysis indicate that the percentage of SiO_(2)(Si^(4+))in OGO tunnel SiO_(x) is higher than which in PANO tunnel SiO_(x).Therefore,we ascribe the lower D_(it) to the good inhibitory effects on the formation of low-valent silicon oxides during the OGO process.In a nutshell,OGO tunnel SiO_(x) has a great potential to be applied in n-type TOPCon silicon solar cell,which may be available for global photovoltaics industry. 展开更多

The antioxidant activity of SPH could retain under acidic environment, but rapidly reduced under alkaline conditions. Addition of D-galactose, D-xylose, and D-fructose at 65</span><span style="font-family:Verdana;">&deg;</span><span style="font-family:Verdana;">C could increase the antioxidant activity of SPH, but no such effect was not observed at this temperature. With the increase of storage time, the antioxidant activity of SPH gradually decreased. Moreover, pepsin digestion treatment slightly reduced the antioxidant activity of SPH, and further trypsin and mixed enzyme (trypsin + chymotrypsin) digestion significantly reduced this activity (p</span><span style="font-family:Verdana;"> < </span><span style="font-family:Verdana;">0.05). This research also studied the relationship between serum concentration of TGFβ1 and childhood asthma. Method: From June 2022 to December 2023, 121 children had physical examination in affiliated Hospital of Youjiang Medical University for Nationalities were selected as control group and 118 children suffered from asthma in affiliated Hospital of Youjiang Medical University for Nationalities during the same period were selected as asthma group. Result: There was no correlation between rs1800469, rs1800470, rs2241712, rs2241715, rs4803455 and asthma in Baise, Guangxi children. Linkage disequilibrium analysis showed that there were strong linkage disequilibrium among rs1800469, rs1800470, rs2241712, rs2241715 and rs4803455. Their haplotypes had no significant correlation with childhood asthma. The serum concentration of TGFβ1 in asthma group was lower than that in control group (p β1 had no significant relationship with the genotypes of rs1800469, rs1800470, rs2241712, rs2241715 and rs4803455. 展开更多
