To investigate the mechanism of glycosides of cistanches improving cognitive performance dysfunction in SAMP8 mice by AMPK/mTOR signaling pathway.Male 7-month-old SAMP8 mice were randomly divided into 5 groups:model group,GCs group,GCs+3-MA group,GCs+compound C group,Vehicle group.Morris water maze was used to detect the ability of learning and memory.Western blot was used to detect the protein expression of SYN,PSD-95,BDNF,LC3,AMPK and m-TOR.The escape latency of GCs group,GCs+3-MA group and GCs+compound C group were significantly shortened(P<0.05).Compared with GCs group,the escape latency and percent of time spent in the target quadrant of GCs+compound C group were significantly increased(P<0.05). 展开更多

It was found that the extract of total flavones from Dracoce phalum moldavica L.(TFD)could exert neuroprotective effect on moase Parkinson's disease(PD)model,but the underlying mechanisms remained unclear.The aim of this study was to detect the mechanisms of TFD neuroprotective effect on PD mice.The PD model was established by injection with MPTP to mice.Using behavioral experiment and ELISA test as well as immunohistochemistry and Western blot analysis,the following parameters were examined:①the falling latency and climbing time,②the content of serum MDA as well as activities of SOD and GSH-Px,③the count of dopaminergic neurons with expression of TH in the substantia nigra of model mice,and④the expression levels of TH,caspase-3,Bcl-2 and Bax in the midbrain tissue. 展开更多

目的探讨蒙药香青兰总黄酮化合物(total flavones of Dracocephalummoldavica L.,TFDM)对MPTP诱导帕金森(Parkinson’s disease,PD)模型小鼠的神经保护作用。方法建立MPTP诱导帕金森病模型,通过转棒实验、爬杆实验评价TFDM对PD小鼠运动... 目的探讨蒙药香青兰总黄酮化合物(total flavones of Dracocephalummoldavica L.,TFDM)对MPTP诱导帕金森(Parkinson’s disease,PD)模型小鼠的神经保护作用。方法建立MPTP诱导帕金森病模型,通过转棒实验、爬杆实验评价TFDM对PD小鼠运动功能的影响;通过ELISA检测TFDM对氧化应激因子超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)表达的情况;通过免疫组织化学和Western blot检测TFDM对酪氨酸羟化酶(tyrosine hydroxylase,TH)和凋亡相关蛋白Caspase-3、Bcl-2、Bax表达的影响。结果行为学评价结果显示,TFDM治疗显著增加小鼠的下落潜伏期(P<0.05),明显降低小鼠的爬杆时间(P<0.05);ELISA结果显示,TFDM治疗显著降低MDA的含量(P<0.01),同时SOD和GSH-Px的表达明显升高(P<0.05);免疫组织化学和Western bolt结果均显示,TFDM治疗组的TH、Bcl-2、Bcl-2/Bax蛋白表达水平明显上调(P<0.05),同时Caspase-3、Bax表达水平明显下降(P<0.05)。结论TFDM对PD模型小鼠具有改善运动功能障碍、减轻氧化应激损伤和减少多巴胺能神经元丢失的作用,并可能通过抑制Caspase-3蛋白活性、上调Bcl-2和下调Bax蛋白的表达发挥神经保护作用。 展开更多

目的:探讨香青兰总黄酮(total flavones of Dracocephalum moldavica L.,TFDM)对帕金森病(Parkinson’s disease,PD)模型小鼠中脑黑质内多巴胺转运蛋白表达的效果。方法:C517BL/6J小鼠腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methyl-... 目的:探讨香青兰总黄酮(total flavones of Dracocephalum moldavica L.,TFDM)对帕金森病(Parkinson’s disease,PD)模型小鼠中脑黑质内多巴胺转运蛋白表达的效果。方法:C517BL/6J小鼠腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPTP)制备PD动物模型,多种行为学实验方法评价动物运动功能;尼氏染色观测黑质区多巴胺能神经元的数量;RT-PCR和Western blot检测黑质酪氨酸羟化酶(tyrosine hydroxylase,TH)、囊泡单胺转运蛋白2(Vesicular monoamine transporter 2,VMAT2)和多巴胺转运蛋白(Dopamine transporter,DAT)的表达情况。结果:行为学检测表明,TFDM组显著增加小鼠的运动距离(P<0.05),显著增加小鼠的下落潜伏期(P<0.05),明显减少小鼠的爬杆时间(P<0.05);MPTP腹腔注射明显减少了黑质内多巴胺能神经元的数量,TFDM显著的提高了PD模型小鼠黑质内多巴胺能神经元数量;RT-PCR结果显示,TFDM组的TH、VMAT2和DAT的mRNA表达均显著增高(P<0.05);TFDM组TH、VMAT2和DAT蛋白表达均明显上调(P<0.05)。结论:TFDM可能通过上调VMAT2和DAT蛋白的表达,减少DA能神经元丢失,进而改善PD模型小鼠运动功能障碍。 展开更多

目的:研究中药香青兰总黄酮(TFD)改善帕金森病模型小鼠神经损伤的作用机制。方法:120只小鼠被随机分为正常对照组20只和观察组100只,观察组小鼠腹腔注射30 mg/(kg·d)MPTP以制备PD小鼠模型,对照组小鼠腹腔注射等体积生理盐水,连续7 ... 目的:研究中药香青兰总黄酮(TFD)改善帕金森病模型小鼠神经损伤的作用机制。方法:120只小鼠被随机分为正常对照组20只和观察组100只,观察组小鼠腹腔注射30 mg/(kg·d)MPTP以制备PD小鼠模型,对照组小鼠腹腔注射等体积生理盐水,连续7 d后再将观察组小鼠随机分为5组,分别为模型组和TFD治疗组25、50、75、100 mg/(kg·d):。治疗组按照小鼠的体重灌胃给药,正常对照组和模型组小鼠给予等容量的生理盐水,连续1个月。通过行为学实验评价小鼠运动功能,通过免疫荧光组织化学技术检测小鼠中脑黑质区组织细胞酪氨酸羟化酶(TH)和选择性自噬接头蛋白(P62)阳性染色的细胞数,采用荧光定量PCR和Western-blot技术检测小鼠黑质区组织的微管相关蛋白1轻链3β(LC3B)、P62、自噬效应蛋白(Beclin1)和TH mRNA和蛋白表达水平。结果:TFD治疗显著增加PD模型小鼠的悬挂运动评分、运动距离和下落潜伏期(P<0.05);TFD治疗增加PD模型小鼠黑质区组织表达P62和TH的阳性染色细胞数,TFD治疗增加PD模型小鼠黑质区组织表达PINK1、Parkin、P62、和TH蛋白的阳性染色细胞数(P<0.05)。结论:TFD可能通过上调LC3B、P62、Beclin1改善自噬,促进多巴胺能神经元受损线粒体及其他细胞器的清除,从而对MPTP诱导的PD模型小鼠产生神经保护作用。 展开更多
