METHODS: RNA interference was used to inhibit the expression of EDIL3 in human LECs in vitro. The morphology of cells was observed using an inverted microscope. Cell proliferation was assessed using Ed U kit. Cell migration was investigated using Transwell chamber and EMT of LECs was assessed using confocal microscope and Western blotting. The transforming growth factor β(TGFβ) pathway was investigated using Western blotting. RESULTS: The data showed that silencing EDIL3 expression changed LECs morphology and suppressed LECs proliferation(P〈0.05) and migration(P〈0.01). Furthermore, the result of Western blotting showed that EDIL3 depletion reduced the expression of α-smooth muscle actin(α-SMA)(P〈0.001) and vimentin(P〈0.01), while increased the expression of E-cadherin(P〈0.001). EDIL3 depletion could suppress the phosphorylation of Smad2(P〈0.01) and Smad3(P〈0.01) and the activation of exracellular signal regulated kinase(ERK)(P〈0.05). CONCLUSION: The findings indicate that EDIL3 might participate in the proliferation and EMT in LECs via TGFβ pathway and may be a potential therapeutic target for the treatment of posterior capsule opacification.展开更多
BACKGROUND Postpartum pulmonary arterial hypertension(PAH)complicated with hereditary hemorrhagic telangiectasia(HHT)is a rare condition.Diagnosing and treating PAH in patients with HHT can be challenging.To the best ...BACKGROUND Postpartum pulmonary arterial hypertension(PAH)complicated with hereditary hemorrhagic telangiectasia(HHT)is a rare condition.Diagnosing and treating PAH in patients with HHT can be challenging.To the best of our knowledge,no previous reports have investigated the efficacy of pulmonary vasodilators in improving hemodynamics in postpartum patients with this disease.CASE SUMMARY In this paper,we report a postpartum case of HHT combined with PAH,pre-senting with worsening dyspnea.Genetic testing revealed that the patient carried a heterozygous variant of activin receptor-like kinase 1.The patient received various treatments,including diuretics,anticoagulants,sildenafil,macitentan,inhalation of nitric oxide,and iloprost.Changes in PaO2/FiO2,pulmonary artery systolic pressure as assessed by echocardiography,and N-terminus pro-brain natriuretic peptide levels suggested that,except for iloprost inhalation,the other treatments appeared to have limited efficacy.CONCLUSION To our knowledge,this is the first report on efficacy of pulmonary vasodilators in postpartum patients with HHT and PAH.展开更多
For P91 steel weldment,performing post-weld heat treatment immediately after welding can enhance welding efficiency and reduce the risk of hydrogen-induced cracking.However,determining the post-weld cooling temperatur...For P91 steel weldment,performing post-weld heat treatment immediately after welding can enhance welding efficiency and reduce the risk of hydrogen-induced cracking.However,determining the post-weld cooling temperature(PWCT)is challenging due to potential impact on the mechanical properties of weldment after subsequent heat treatment.A systematic investigation is conducted to explore the effect of PWCT on the impact toughness of P91 steel welded joints.It has been demonstrated that the impact energy of the weld metal gradually increases from 40.2 to 49.5 J as the PWCT decreases from 300 to 100℃.Microstructural analysis reveals that PWCTs above 100℃ led to an increased dislocation density in the weld metal.Furthermore,in situ observations using a high-temperature confocal laser scanning microscope confirm that excessively high PWCTs result in the formation of untempered martensite after post-weld heat treatment.Such untempered martensites are identified as the primary cause of the reduced impact toughness in the weld metal.These findings underscore the importance of carefully controlling PWCT in welding procedures for P91 steel and similar grades.展开更多
An investigation on the tensile properties and strengthening mechanism of a dense 316L stainless steel(316LSS)material fabricated by laser powder bed fusion(L-PBF)has been conducted with varying heat treatment conditi...An investigation on the tensile properties and strengthening mechanism of a dense 316L stainless steel(316LSS)material fabricated by laser powder bed fusion(L-PBF)has been conducted with varying heat treatment conditions.Deformation mechanisms of as-built and heat-treated samples were elucidated through multiscale microscopy characterizations.The cellular structure characterized by high density dislocation provided a strong barrier to the dislocation propagation,enhancing the yield strength of L-PBF 316LSS.Additionally,the accumulation of free dislocations at grain boundaries triggered the initiation of deformation twins and synergistically interacted with the dislocation wall to establish a three-dimensional network pinning structure,thereby effectively improving the continuous work hardening capability of the as-built sample.In contrast,the absence of cellular structure in HT 1000 samples resulted in a 21.8%reduction in yield strength and a 12%increase in elongation,exhibiting typical strength-ductility trade-off.The absence of cellular structure facilitated the formation of more deformation twins and contributed to the manifestation of the dynamic Hall–Petch effect.It effectively extended the work hardening regime of L-PBF 316LSS,thereby delaying the necking and enhancing its plasticity.Importantly,the Hall–Petch constant has been modified by analyzing the dependence of resolved shear yield strength,which originated from the cellular structure,on the inverse square root of cell size(d−1/2).The modified Hall–Petch relationship accurately assessed the contribution of cellular structure to the yield strength of L-PBF 316LSS.The underlying strengthening mechanism of cellular structure was comprehensively revealed,and valuable insights for further optimization and enhancement of the mechanical properties of L-PBF 316LSS were offered.展开更多
In this work,the synthesis of uniform zeolitic imidazolate framework-coated Mo-glycerate spheres and their subsequent conversion into hierarchical architecture containing bimetallic selenides heterostructures and nitr...In this work,the synthesis of uniform zeolitic imidazolate framework-coated Mo-glycerate spheres and their subsequent conversion into hierarchical architecture containing bimetallic selenides heterostructures and nitrogen-doped carbon shell are reported.Selenization temperature plays a significant role in determining the phases,morphology,and lithium-ion storage performance of the composite.Notably,the optimal electrode demonstrates an ultrahigh reversible capacity of 1298.2 mAh/g after 100 cycles at 0.2 A/g and an outstanding rate capability with the capacity still maintained 505.7 mAh/g after 300 cycles at 1.0 A/g,surpassing the calculated theoretical capacity according to individual component and most of the reported MoSe@C-or ZnSe@C-based anodes.Furthermore,ex-situ X-ray diffraction patterns reveal the combined conversion and alloying reaction mechanisms of the composite.展开更多
Microstructural evolution features have been systematically investigated for the weld metal of EH36 shipbuilding steel under an in situ confocal scanning laser microscope.The influence of cooling rate on microstructur...Microstructural evolution features have been systematically investigated for the weld metal of EH36 shipbuilding steel under an in situ confocal scanning laser microscope.The influence of cooling rate on microstructural changes during the transformation from austenite to ferrite has been clarified.It is found that ferrite side plates form preceding to acicular ferrites,although the starting temperature of respective component decreases as the cooling rate is raised.In particular,the growth rate of acicular ferrite is measured to increase significantly,rising from 30.4μm/s at a cooling rate of 3 K/s to 109.0μm/s at 15 K/s,driven primarily by an ever-increasing degree of undercooling.These findings highlight the critical role of cooling rate in dictating the sequence and growth rate of microstructural transformations,which is crucial for optimizing welding processes to obtain desired microstructures while avoiding the formation of deleterious components.展开更多
COMPERA 2.0 risk stratification has been demonstrated to be useful in patients with precapillary pulmonary hypertension(PH).However,its suitability for patients at risk for post-capillary PH or PH associated with left...COMPERA 2.0 risk stratification has been demonstrated to be useful in patients with precapillary pulmonary hypertension(PH).However,its suitability for patients at risk for post-capillary PH or PH associated with left heart disease(PH-LHD)is unclear.To investigate the use of COMPERA 2.0 in patients with severe aortic stenosis(SAS)undergoing transcatheter aortic valve replacement(TAVR),who are at risk for post-capillary PH,a total of 327 eligible SAS patients undergoing TAVR at our institution between September 2015 and November 2020 were included in the study.Patients were classified into four strata before and after TAVR using the COMPERA 2.0 risk score.The primary endpoint was all-cause mortality.Survival analysis was performed using Kaplan-Meier curves,log-rank test,and Cox proportional hazards regression model.The study cohort had a median(interquartile range)age of 76(70–80)years and a pulmonary arterial systolic pressure of 33(27–43)mmHg(1 mmHg=0.133 kPa)before TAVR.The overall mortality was 11.9%during 26(15–47)months of follow-up.Before TAVR,cumulative mortality was higher with an increase in the risk stratum level(log-rank,both P<0.001);each increase in the risk stratum level resulted in an increased risk of death(hazard ratio(HR)2.53,95%confidential interval(CI)1.54–4.18,P<0.001),which was independent of age,sex,estimated glomerular filtration rate(eGFR),hemoglobin,albumin,and valve type(HR 1.76,95%CI 1.01–3.07,P=0.047).Similar results were observed at 30 d after TAVR.COMPERA 2.0 can serve as a useful tool for risk stratification in patients with SAS undergoing TAVR,indicating its potential application in the management of PH-LHD.Further validation is needed in patients with confirmed post-capillary PH by right heart catheterization.展开更多
Metadata prefetching and data placement play a critical role in enhancing access performance for file systems operating over wide-area networks.However,developing effective strategies for metadata prefetching in envir...Metadata prefetching and data placement play a critical role in enhancing access performance for file systems operating over wide-area networks.However,developing effective strategies for metadata prefetching in environments with concurrent workloads and for data placement across distributed networks remains a significant challenge.This study introduces novel and efficient methodologies for metadata prefetching and data placement,leveraging fine-grained control of prefetching strategies and variable-sized data fragment writing to optimize the I/O bandwidth of distributed file systems.The proposed metadata prefetching technique employs dynamic workload analysis to identify dominant workload patterns and adaptively refines prefetching policies,thereby boosting metadata access efficiency under concurrent scenarios.Meanwhile,the data placement strategy improves write performance by storing data fragments locally within the nearest data center and transmitting only the fragment location metadata to the remote data center hosting the original file.Experimental evaluations using real-world system traces demonstrate that the proposed approaches reduce metadata access times by up to 33.5%and application data access times by 17.19%compared to state-of-the-art techniques.展开更多

METHODS:Two investigators independently searched ...AIM:To investigate the effects of emergent preopera-tive self-expandable metallic stent (SEMS) vs emer-gent surgery for acute left-sided malignant colonic obstruction. METHODS:Two investigators independently searched the MEDLINE, EMBASE and Cochrane Central Register of Controlled Trials, as well as references of included studies to identify randomized controlled trials (RCTs) that compared two or more surgical approaches for acute colonic obstruction. Summary risk ratios (RR) and 95% CI for colonic stenting and emergent surgery were calculated. RESULTS:Eight studies met the selection criteria, involving 444 patients, of whom 219 underwent SEMS and 225 underwent emergent surgery. Seven studies reported difference of the one-stage stoma rates between the two groups (RR, 0.60; 95% CI:0.48-0.76; P < 0.0001). Only three RCTs described the follow-up stoma rates, which showed no significant difference between the two groups (RR, 0.80; 95% CI:0.59-1.08; P = 0.14). Difference was not significant in the mortality between the two groups (RR, 0.91; 95% CI:0.50-1.66; P = 0.77), but there was significant difference (RR, 0.57; 95% CI:0.44-0.74; P < 0.0001) in the overall morbidity. There were no significant differences between the two groups in the anastomotic leak rate (RR, 0.60; 95% CI: 0.28-1.28; P = 0.19), occurrence of abscesses, including peristomal abscess, intraperitoneal abscess and parietal abscess (RR, 0.83; 95% CI:0.36-1.95; P = 0.68), and other abdominal complications (RR: 0.67; 95% CI: 0.40-1.12; P = 0.13). CONCLUSION:SEMS is not obviously more advantageous than emergent surgery for patients with acute left-sided malignant colonic obstruction.展开更多

BACKGROUND There is currently a shortage of accurate,efficient,and precise predictive instruments for rectal neuroendocrine neoplasms(NENs).AIM To develop a predictive model for individuals with rectal NENs(R-NENs)usi...BACKGROUND There is currently a shortage of accurate,efficient,and precise predictive instruments for rectal neuroendocrine neoplasms(NENs).AIM To develop a predictive model for individuals with rectal NENs(R-NENs)using data from a large cohort.METHODS Data from patients with primary R-NENs were retrospectively collected from 17 large-scale referral medical centers in China.Random forest and Cox proportional hazard models were used to identify the risk factors for overall survival and progression-free survival,and two nomograms were constructed.RESULTS A total of 1408 patients with R-NENs were included.Tumor grade,T stage,tumor size,age,and a prognostic nutritional index were important risk factors for prognosis.The GATIS score was calculated based on these five indicators.For overall survival prediction,the respective C-indexes in the training set were 0.915(95%confidence interval:0.866-0.964)for overall survival prediction and 0.908(95%confidence interval:0.872-0.944)for progression-free survival prediction.According to decision curve analysis,net benefit of the GATIS score was higher than that of a single factor.The time-dependent area under the receiver operating characteristic curve showed that the predictive power of the GATIS score was higher than that of the TNM stage and pathological grade at all time periods.CONCLUSION The GATIS score had a good predictive effect on the prognosis of patients with R-NENs,with efficacy superior to that of the World Health Organization grade and TNM stage.展开更多

Influenza A viruses(IAV)are responsible for seasonal flu epidemics,which can lead to high morbidity and mortality each year.Like other viruses,influenza virus can hijack host cellular machinery for its replication.Hos...Influenza A viruses(IAV)are responsible for seasonal flu epidemics,which can lead to high morbidity and mortality each year.Like other viruses,influenza virus can hijack host cellular machinery for its replication.Host cells have evolved diverse cellular defense to resist the invasion of viruses.As the main components of promyelocytic leukemia protein nuclear bodies(PML-NBs),PML can inhibit the replication of many medically important viruses including IAV.However,the mechanism of PML against IAV is unclear.In the present study,we found PML was induced in response to IAV infection and ectopic expression of PML could inhibit IAV replication,whereas knockdown of endogenous PML expression could enhance IAV replication.Further studies showed that PML increased the expression of FBXW7 by inhibiting its K48-linked ubiquitination and enhanced the interaction between FBXW7 and SHP2,which negatively regulated IAV replication during infection.Moreover,PML stabilized RIG-I to promote the production of typeⅠIFN.Collectively,these data indicated that PML inhibited IAV replication by enhancing FBXW7 expression in the antiviral immunity against influenza virus and extended the mechanism of PML in antiviral immunity.展开更多

The hydrogels are dually crosslinked single network with the PAM grafted vinyl...A facile method to fabricate tough and highly stretchable polyacrylamide (PAM) nanocomposite physical hydrogel (NCP gel) was proposed. The hydrogels are dually crosslinked single network with the PAM grafted vinyl hybrid silica nanoparticles (VSNPs) as the analogous covalent crosslinking points and the reversible hydrogen bonds among the PAM chains as the physical crosslinking points. In order to further elucidate the toughening mechanism of the PAM NCP gel, especially to understand the role of the dual crosslinking points, the PAM hybrid hydrogels (H gels) and a series of poly(acrylamide-co-dimethylacrylamide) (P(AM-co-DMAA)) NCP gels were designed and fabricated. Their mechanical properties were compared with those of the PAM NCP gels. The PAM H gels are prepared by simply mixing the PAM chains with bare silica nanoparticles (SNPs). Relative to the poor mechanical properties of the PAM H gel, the PAM NCP gel is remarkably tough and stretchable and also generates large number of micro-cracks to stop notch propagation, indicating the important role of PAM grafted VSNPs in toughening the NCP gel. In the P(AM-co-DMAA) NCP gels, the P(AM-co- DMAA) chains are grafted on VSNPs and the polydimethylacrylamide (PDMAA) only forms very weak hydrogen bonds between themselves. It is found that mechanical properties of the PAM NCP gel, such as the tensile strength and the elongation at break, are enhanced significantly, but those of the P(AM-co-DMAA) NCP gels decreased rapidly with decreasing AM content. This result reveals the role of the hydrogen bonds among the grafted polymer chains as the physical crosslinking points in toughening the NCP gel.展开更多

A total of 48 and 64 QTLs were identified in 2004 and 2005, respectively. The contribution of each QTL to the phenotypic variation ranged from 4.0% to 43.7%. Most QTLs co-localized, forming 29 QTL clusters on the chromosomes with three major ones detected in both years, which were mapped on chromosomes 1, 7 and 9, respectively. The two QTL clusters for amino acid content, qAa1 and qAa7, influenced almost all the traits with the allele from Zhenshan 97B, and the third QTL cluster for amino acid content, qAa9, increased the lysine content with the allele from Delong 208. A wide coincidence was found between the QTL detected under this study and the loci involved in amino acid metabolism pathways in nitrogen assimilation and transport, or protein biosynthesis. The results would facilitate the identification of candidate genes and could be used in marker-assisted selection for the favorable allele in rice quality improvement.展开更多

文摘An investigation on the tensile properties and strengthening mechanism of a dense 316L stainless steel(316LSS)material fabricated by laser powder bed fusion(L-PBF)has been conducted with varying heat treatment conditions.Deformation mechanisms of as-built and heat-treated samples were elucidated through multiscale microscopy characterizations.The cellular structure characterized by high density dislocation provided a strong barrier to the dislocation propagation,enhancing the yield strength of L-PBF 316LSS.Additionally,the accumulation of free dislocations at grain boundaries triggered the initiation of deformation twins and synergistically interacted with the dislocation wall to establish a three-dimensional network pinning structure,thereby effectively improving the continuous work hardening capability of the as-built sample.In contrast,the absence of cellular structure in HT 1000 samples resulted in a 21.8%reduction in yield strength and a 12%increase in elongation,exhibiting typical strength-ductility trade-off.The absence of cellular structure facilitated the formation of more deformation twins and contributed to the manifestation of the dynamic Hall–Petch effect.It effectively extended the work hardening regime of L-PBF 316LSS,thereby delaying the necking and enhancing its plasticity.Importantly,the Hall–Petch constant has been modified by analyzing the dependence of resolved shear yield strength,which originated from the cellular structure,on the inverse square root of cell size(d−1/2).The modified Hall–Petch relationship accurately assessed the contribution of cellular structure to the yield strength of L-PBF 316LSS.The underlying strengthening mechanism of cellular structure was comprehensively revealed,and valuable insights for further optimization and enhancement of the mechanical properties of L-PBF 316LSS were offered.

文摘In this work,the synthesis of uniform zeolitic imidazolate framework-coated Mo-glycerate spheres and their subsequent conversion into hierarchical architecture containing bimetallic selenides heterostructures and nitrogen-doped carbon shell are reported.Selenization temperature plays a significant role in determining the phases,morphology,and lithium-ion storage performance of the composite.Notably,the optimal electrode demonstrates an ultrahigh reversible capacity of 1298.2 mAh/g after 100 cycles at 0.2 A/g and an outstanding rate capability with the capacity still maintained 505.7 mAh/g after 300 cycles at 1.0 A/g,surpassing the calculated theoretical capacity according to individual component and most of the reported MoSe@C-or ZnSe@C-based anodes.Furthermore,ex-situ X-ray diffraction patterns reveal the combined conversion and alloying reaction mechanisms of the composite.

文摘Microstructural evolution features have been systematically investigated for the weld metal of EH36 shipbuilding steel under an in situ confocal scanning laser microscope.The influence of cooling rate on microstructural changes during the transformation from austenite to ferrite has been clarified.It is found that ferrite side plates form preceding to acicular ferrites,although the starting temperature of respective component decreases as the cooling rate is raised.In particular,the growth rate of acicular ferrite is measured to increase significantly,rising from 30.4μm/s at a cooling rate of 3 K/s to 109.0μm/s at 15 K/s,driven primarily by an ever-increasing degree of undercooling.These findings highlight the critical role of cooling rate in dictating the sequence and growth rate of microstructural transformations,which is crucial for optimizing welding processes to obtain desired microstructures while avoiding the formation of deleterious components.

文摘COMPERA 2.0 risk stratification has been demonstrated to be useful in patients with precapillary pulmonary hypertension(PH).However,its suitability for patients at risk for post-capillary PH or PH associated with left heart disease(PH-LHD)is unclear.To investigate the use of COMPERA 2.0 in patients with severe aortic stenosis(SAS)undergoing transcatheter aortic valve replacement(TAVR),who are at risk for post-capillary PH,a total of 327 eligible SAS patients undergoing TAVR at our institution between September 2015 and November 2020 were included in the study.Patients were classified into four strata before and after TAVR using the COMPERA 2.0 risk score.The primary endpoint was all-cause mortality.Survival analysis was performed using Kaplan-Meier curves,log-rank test,and Cox proportional hazards regression model.The study cohort had a median(interquartile range)age of 76(70–80)years and a pulmonary arterial systolic pressure of 33(27–43)mmHg(1 mmHg=0.133 kPa)before TAVR.The overall mortality was 11.9%during 26(15–47)months of follow-up.Before TAVR,cumulative mortality was higher with an increase in the risk stratum level(log-rank,both P<0.001);each increase in the risk stratum level resulted in an increased risk of death(hazard ratio(HR)2.53,95%confidential interval(CI)1.54–4.18,P<0.001),which was independent of age,sex,estimated glomerular filtration rate(eGFR),hemoglobin,albumin,and valve type(HR 1.76,95%CI 1.01–3.07,P=0.047).Similar results were observed at 30 d after TAVR.COMPERA 2.0 can serve as a useful tool for risk stratification in patients with SAS undergoing TAVR,indicating its potential application in the management of PH-LHD.Further validation is needed in patients with confirmed post-capillary PH by right heart catheterization.

文摘Metadata prefetching and data placement play a critical role in enhancing access performance for file systems operating over wide-area networks.However,developing effective strategies for metadata prefetching in environments with concurrent workloads and for data placement across distributed networks remains a significant challenge.This study introduces novel and efficient methodologies for metadata prefetching and data placement,leveraging fine-grained control of prefetching strategies and variable-sized data fragment writing to optimize the I/O bandwidth of distributed file systems.The proposed metadata prefetching technique employs dynamic workload analysis to identify dominant workload patterns and adaptively refines prefetching policies,thereby boosting metadata access efficiency under concurrent scenarios.Meanwhile,the data placement strategy improves write performance by storing data fragments locally within the nearest data center and transmitting only the fragment location metadata to the remote data center hosting the original file.Experimental evaluations using real-world system traces demonstrate that the proposed approaches reduce metadata access times by up to 33.5%and application data access times by 17.19%compared to state-of-the-art techniques.
文摘AIM:To investigate the effects of emergent preopera-tive self-expandable metallic stent (SEMS) vs emer-gent surgery for acute left-sided malignant colonic obstruction. METHODS:Two investigators independently searched the MEDLINE, EMBASE and Cochrane Central Register of Controlled Trials, as well as references of included studies to identify randomized controlled trials (RCTs) that compared two or more surgical approaches for acute colonic obstruction. Summary risk ratios (RR) and 95% CI for colonic stenting and emergent surgery were calculated. RESULTS:Eight studies met the selection criteria, involving 444 patients, of whom 219 underwent SEMS and 225 underwent emergent surgery. Seven studies reported difference of the one-stage stoma rates between the two groups (RR, 0.60; 95% CI:0.48-0.76; P < 0.0001). Only three RCTs described the follow-up stoma rates, which showed no significant difference between the two groups (RR, 0.80; 95% CI:0.59-1.08; P = 0.14). Difference was not significant in the mortality between the two groups (RR, 0.91; 95% CI:0.50-1.66; P = 0.77), but there was significant difference (RR, 0.57; 95% CI:0.44-0.74; P < 0.0001) in the overall morbidity. There were no significant differences between the two groups in the anastomotic leak rate (RR, 0.60; 95% CI: 0.28-1.28; P = 0.19), occurrence of abscesses, including peristomal abscess, intraperitoneal abscess and parietal abscess (RR, 0.83; 95% CI:0.36-1.95; P = 0.68), and other abdominal complications (RR: 0.67; 95% CI: 0.40-1.12; P = 0.13). CONCLUSION:SEMS is not obviously more advantageous than emergent surgery for patients with acute left-sided malignant colonic obstruction.

文摘The complexity of oral ulcerations poses considerable diagnostic and therapeutic challenges to oral specialists.The expert consensus was conducted to summarize the diagnostic work-up for difficult and complicated oral ulcers,based on factors such as detailed clinical medical history inquiry,histopathological examination,and ulceration-related systemic diseases screening.Not only it can provide a standardized procedure of oral ulceration,but also it can improve the diagnostic efficiency,in order to avoid misdiagnosis and missed diagnosis.

文摘BACKGROUND There is currently a shortage of accurate,efficient,and precise predictive instruments for rectal neuroendocrine neoplasms(NENs).AIM To develop a predictive model for individuals with rectal NENs(R-NENs)using data from a large cohort.METHODS Data from patients with primary R-NENs were retrospectively collected from 17 large-scale referral medical centers in China.Random forest and Cox proportional hazard models were used to identify the risk factors for overall survival and progression-free survival,and two nomograms were constructed.RESULTS A total of 1408 patients with R-NENs were included.Tumor grade,T stage,tumor size,age,and a prognostic nutritional index were important risk factors for prognosis.The GATIS score was calculated based on these five indicators.For overall survival prediction,the respective C-indexes in the training set were 0.915(95%confidence interval:0.866-0.964)for overall survival prediction and 0.908(95%confidence interval:0.872-0.944)for progression-free survival prediction.According to decision curve analysis,net benefit of the GATIS score was higher than that of a single factor.The time-dependent area under the receiver operating characteristic curve showed that the predictive power of the GATIS score was higher than that of the TNM stage and pathological grade at all time periods.CONCLUSION The GATIS score had a good predictive effect on the prognosis of patients with R-NENs,with efficacy superior to that of the World Health Organization grade and TNM stage.

文摘Influenza A viruses(IAV)are responsible for seasonal flu epidemics,which can lead to high morbidity and mortality each year.Like other viruses,influenza virus can hijack host cellular machinery for its replication.Host cells have evolved diverse cellular defense to resist the invasion of viruses.As the main components of promyelocytic leukemia protein nuclear bodies(PML-NBs),PML can inhibit the replication of many medically important viruses including IAV.However,the mechanism of PML against IAV is unclear.In the present study,we found PML was induced in response to IAV infection and ectopic expression of PML could inhibit IAV replication,whereas knockdown of endogenous PML expression could enhance IAV replication.Further studies showed that PML increased the expression of FBXW7 by inhibiting its K48-linked ubiquitination and enhanced the interaction between FBXW7 and SHP2,which negatively regulated IAV replication during infection.Moreover,PML stabilized RIG-I to promote the production of typeⅠIFN.Collectively,these data indicated that PML inhibited IAV replication by enhancing FBXW7 expression in the antiviral immunity against influenza virus and extended the mechanism of PML in antiviral immunity.

文摘The intestinal mucosa is a highly compartmentalized structure that forms a directbarrier between the host intestine and the environment, and its dysfunction couldresult in a serious disease. As T cells, which are important components of themucosal immune system, interact with gut microbiota and maintain intestinalhomeostasis, they may be involved in the process of intestinal barrier dysfunction.P2X7 receptor (P2X7R), a member of the P2X receptors family, mediates the effectsof extracellular adenosine triphosphate and is expressed by most innate or adaptiveimmune cells, including T cells. Current evidence has demonstrated thatP2X7R is involved in inflammation and mediates the survival and differentiationof T lymphocytes, indicating its potential role in the regulation of T cell function.In this review, we summarize the available research about the regulatory role andmechanism of P2X7R on the intestinal mucosa-derived T cells in the setting ofintestinal barrier dysfunction.

文摘A facile method to fabricate tough and highly stretchable polyacrylamide (PAM) nanocomposite physical hydrogel (NCP gel) was proposed. The hydrogels are dually crosslinked single network with the PAM grafted vinyl hybrid silica nanoparticles (VSNPs) as the analogous covalent crosslinking points and the reversible hydrogen bonds among the PAM chains as the physical crosslinking points. In order to further elucidate the toughening mechanism of the PAM NCP gel, especially to understand the role of the dual crosslinking points, the PAM hybrid hydrogels (H gels) and a series of poly(acrylamide-co-dimethylacrylamide) (P(AM-co-DMAA)) NCP gels were designed and fabricated. Their mechanical properties were compared with those of the PAM NCP gels. The PAM H gels are prepared by simply mixing the PAM chains with bare silica nanoparticles (SNPs). Relative to the poor mechanical properties of the PAM H gel, the PAM NCP gel is remarkably tough and stretchable and also generates large number of micro-cracks to stop notch propagation, indicating the important role of PAM grafted VSNPs in toughening the NCP gel. In the P(AM-co-DMAA) NCP gels, the P(AM-co- DMAA) chains are grafted on VSNPs and the polydimethylacrylamide (PDMAA) only forms very weak hydrogen bonds between themselves. It is found that mechanical properties of the PAM NCP gel, such as the tensile strength and the elongation at break, are enhanced significantly, but those of the P(AM-co-DMAA) NCP gels decreased rapidly with decreasing AM content. This result reveals the role of the hydrogen bonds among the grafted polymer chains as the physical crosslinking points in toughening the NCP gel.

文摘The phenotypes of protein and amino acid contents were measured in an F9 recombinant inbred line population derived from a cross between Zhenshan 97B and Delong 208. A total of 48 and 64 QTLs were identified in 2004 and 2005, respectively. The contribution of each QTL to the phenotypic variation ranged from 4.0% to 43.7%. Most QTLs co-localized, forming 29 QTL clusters on the chromosomes with three major ones detected in both years, which were mapped on chromosomes 1, 7 and 9, respectively. The two QTL clusters for amino acid content, qAa1 and qAa7, influenced almost all the traits with the allele from Zhenshan 97B, and the third QTL cluster for amino acid content, qAa9, increased the lysine content with the allele from Delong 208. A wide coincidence was found between the QTL detected under this study and the loci involved in amino acid metabolism pathways in nitrogen assimilation and transport, or protein biosynthesis. The results would facilitate the identification of candidate genes and could be used in marker-assisted selection for the favorable allele in rice quality improvement.
