A Preliminary Study of Age and Sex of People with Hemophilia

Aim: This paper aims at providing more information of people with hemophilia for more targeted treatment. Methods: 1149 people with hemophilia are surveyed in 45 units, and then established by EpiData software. The number of people with hemophilia in 9 age intervals for the male and female is analyzed by Population Pyramid and Pareto Analysis. Results: It is found that the people with hemophilia are appearing in every age interval. Relative to the number of male patients, the number of female patients is extremely rare. Of 1134 male patients, relative frequencies for male ranked the first (23.0%), second (17.4%), third (17.2%) are 7 - 12, 2 - 3, 13 - 18 years old, respectively, while of 15 female patients, relative frequencies for female in these age intervals are only 13.3%, 13.3%, 6.7%, respectively. However, the most relative frequency of female patients is between 26 - 45 years old. Conclusion: There is a challenge of the aging hemophilia population. Most of male patients are between 7 - 12 and 2 - 3 years old. Most of female patients are between 26 - 45 years old. 展开更多

目的探究神经肽FF(neuropeptide FF,NPFF)通过激活Ras同源家族成员A(Ras homolog family member A,RhoA)/Rho相关卷曲螺旋蛋白激酶(Rho associated coiled-coil containing protein kinase,ROCK)/Yes相关蛋白(Yes-associated protein,Y... 目的探究神经肽FF(neuropeptide FF,NPFF)通过激活Ras同源家族成员A(Ras homolog family member A,RhoA)/Rho相关卷曲螺旋蛋白激酶(Rho associated coiled-coil containing protein kinase,ROCK)/Yes相关蛋白(Yes-associated protein,YAP)信号通路促进结直肠癌转移的分子机制。方法通过癌症基因组图谱(The Cancer Cenome Atlas,TCGA)公共数据库检索NPFF在结直肠癌组织中的表达及其与预后的关系。实时荧光定量聚合酶链式反应(real-time quantitative polymerase chain reaction,RT-qPCR)检测结直肠癌细胞和肠上皮正常细胞NCM460中NPFF的mRNA水平。通过小干扰RNA和质粒瞬时转染构建NPFF敲低和过表达的结直肠癌细胞株。蛋白质印迹法检测细胞中RhoA、YAP和磷酸化的YAP等蛋白水平。transwell实验检测敲低/过表达NPFF后结直肠癌细胞的体外迁移能力。细胞免疫荧光染色观察YAP的亚细胞定位情况。结果TCGA数据库和RT-qPCR结果显示,NPFF在结直肠癌组织和结直肠癌细胞DiFi、DLD-1、SW620、KM12C、HT29、SW480、HCT8和KM12SM中高表达(均P<0.05),并与患者不良预后相关。transwell迁移实验表明,SW480和KM12C细胞敲减NPFF后迁移能力减弱(均P<0.01),而DiFi和DLD-1细胞过表达NPFF后迁移能力增强(均P<0.01)。在NPFF过表达的DLD-1和DiFi细胞中添加ROCK 1/2抑制剂Y27632(5μmol/L,24 h)能够有效抵消NPFF诱导的细胞迁移能力(均P<0.01)。RhoA活性检测表明,在添加NPFF重组多肽(3μmol/L,30 min)后,SW480和DiFi细胞中的RhoA活化水平均增加(均P<0.05)。蛋白质印迹法显示,在添加NPFF重组多肽(3μmol/L,3 h、6 h和12h)后,SW480和DiFi细胞中的YAP发生明显的去磷酸化(均P<0.05)。免疫荧光染色结果显示,在DiFi细胞中添加NPFF重组多肽(3μmol/L,3 h)后肌动蛋白的荧光强度和YAP在核内定位均有所增加(均P<0.05)。核浆蛋白分离检测发现,在SW480和DiFi细胞中添加NPFF重组多肽(3μmol/L,3 h和6 h)后YAP由细胞质转入细胞核内。RT-qPCR分析表明,NPFF可上调SW480和DiFi细胞中YAP下游靶基因结缔组织生长因子(connective tissue growth factor,CTGF)、富含半胱氨酸蛋白61(cysteine rich angiogenic inducer 61,CYR61)、血管生成素样蛋白2(angiomotin like 2,AMOTL2)和锚蛋白重复结构域1(ankyrin repeat domain 1,ANKRD1)的表达(均P<0.05)。结论NPFF通过激活RhoA/ROCK/YAP信号通路促进结直肠癌的转移。 展开更多

目的探讨血小板反应蛋白1(THBS1)在调控结直肠癌(CRC)进展中的作用。方法使用GEPIA2、TGGA、GTEx数据库分析THBS1在CRC中的表达与免疫浸润的关系。根据不同转染方式,将HCT8细胞分为转染阴性对照小干扰RNA(siRNA)的阴性对照组(siNC组)、... 目的探讨血小板反应蛋白1(THBS1)在调控结直肠癌(CRC)进展中的作用。方法使用GEPIA2、TGGA、GTEx数据库分析THBS1在CRC中的表达与免疫浸润的关系。根据不同转染方式,将HCT8细胞分为转染阴性对照小干扰RNA(siRNA)的阴性对照组(siNC组)、转染THBS1 siRNA-1的敲低组1(siTHBS1-1组)和转染THBS1 siRNA-2的敲低组2(siTHBS1-2组)。采用实时荧光定量聚合酶链反应和蛋白质印迹法检测THBS1表达情况。使用Transwell实验和5-乙炔基-2′-脱氧尿苷实验验证THBS1对HCT8细胞迁移、侵袭和增殖能力的影响。使用多重免疫荧光染色验证THBS1在CRC患者的肿瘤组织和癌旁组织中的表达。结果GEPIA2数据库分析结果显示,CRC组织THBS1信使RNA(mRNA)的中位表达水平低于正常组织。TIMER2.0在线平台分析结果显示,结肠腺癌、食管癌、膀胱尿路上皮癌患者癌组织THBS1基因表达水平低于癌旁组织,差异有统计学意义(P<0.05),直肠腺癌患者癌组织THBS1基因表达水平有低于癌旁组织的趋势。同时使用GEPIA2网站联合GTEx数据库分析结果显示,肾上腺皮质癌、卵巢癌、结肠腺癌和直肠腺癌患者癌组织THBS1 mRNA表达水平低于癌旁组织,差异均有统计学意义(P<0.05)。TIMER2.0在线平台分析结果显示,结肠腺癌患者THBS1 mRNA表达水平与CD8^(+)T细胞、巨噬细胞/单核细胞、中性粒细胞、髓样树突状细胞的浸润水平呈正相关(P<0.05)。siTHBS1-1组和siTHBS1-2组迁移和侵袭的细胞数量多于siNC组,细胞增殖率高于siNC组,差异均有统计学意义(P<0.05)。CRC癌组织中组织学评分低于癌旁组织,差异有统计学意义(P=0.023)。结论THBS1在CRC中低表达,敲低THBS1可以促进CRC细胞的迁移、侵袭和增殖。 展开更多

结直肠癌是全球高发的恶性肿瘤。其传统治疗面临器官功能丧失与生活质量下降等挑战。内镜黏膜下剥离术(endoscopic submucosal dissection,ESD)作为一种根治性微创治疗手段,适用于早期结直肠癌,通过黏膜下层精准剥离实现病变整块切除,... 结直肠癌是全球高发的恶性肿瘤。其传统治疗面临器官功能丧失与生活质量下降等挑战。内镜黏膜下剥离术(endoscopic submucosal dissection,ESD)作为一种根治性微创治疗手段,适用于早期结直肠癌,通过黏膜下层精准剥离实现病变整块切除,显著降低局部复发率,已逐步替代根治性手术成为关键治疗选择。本文系统综述了ESD在结直肠癌领域的最新进展,涵盖其适应证与禁忌证的演变、临床应用疗效与并发症风险、在溃疡性结肠炎相关肿瘤性病变治疗中的效果和器械创新(特别是机器人辅助系统)对技术革新的推动作用。总体而言,ESD正推动结直肠癌治疗模式向保留器官功能的微创根治性治疗转变,其技术标准化与持续进步是优化临床预后的关键。 展开更多

目的:从生物层面探讨延安市宫颈癌发病主次要危险因素,为延安地区预防及控制宫颈癌的发病提供科学依据。方法:选取2017年01月至2020年09月在我市两家三甲医院诊断为宫颈癌的患者200例为病例组,在延安市六个县区中选择与病例组年龄、住... 目的:从生物层面探讨延安市宫颈癌发病主次要危险因素,为延安地区预防及控制宫颈癌的发病提供科学依据。方法:选取2017年01月至2020年09月在我市两家三甲医院诊断为宫颈癌的患者200例为病例组,在延安市六个县区中选择与病例组年龄、住所匹配的200例样本为对照组,进行病例对照研究。内容包括:自编的《宫颈癌危险因素调查量表》,含病原微生物感染情况、月经史、生育史及家族史。结果:病例组与对照组在一级亲属癌症史、是否绝经、怀孕次数≥4次、生育头胎年龄<19岁、是否在家分娩、梅毒螺旋体的感染、高危型HPV的感染方面的差异有统计学意义(χ^(2)=11.559、9.814、4.968、28.281、27.951、8.578、273.125,P<0.05)。多因素回归分析显示一级亲属有癌症史(OR=4.924,95%CI=1.930~12.558)、在家分娩(OR=4.221,95%CI=2.030~8.777)、高危型HPV感染(OR=119.762,95%CI=55.205~259.816)是宫颈癌发病的生物危险因素。结论:延安市妇女宫颈癌生物性的发病因素复杂,主要是高危型HPV感染,可以通过宫颈癌的健康教育、提高宫颈癌的筛查率、HPV疫苗的接种率等措施减少宫颈癌的发病风险。 展开更多

随着城市居民绿色低碳出行思想的提高,网约车合乘出行方式应运而生.但由于合乘模式涉及到的行驶路线问题,乘客与乘客、乘客与驾驶员之间容易产生分歧,并且网约车合乘出行模式的相关成本不明确等诸多问题,网约车合乘模式没有被大范围推... 随着城市居民绿色低碳出行思想的提高,网约车合乘出行方式应运而生.但由于合乘模式涉及到的行驶路线问题,乘客与乘客、乘客与驾驶员之间容易产生分歧,并且网约车合乘出行模式的相关成本不明确等诸多问题,网约车合乘模式没有被大范围推广和应用.针对网约车合乘出行模式存在的问题,研究并构建了网约车合乘路径优化模型,模型中考虑了车辆等待时间成本、行驶距离成本、收益、容量约束以及时间窗约束等.针对网约车合乘模型的特点,并基于遗传算法思想,研究设计了满足合乘模型约束条件的求解遗传算法.并使用Matlab软件运行算法程序对算例进行求解,运行44.08 s得到最大利润6906.2971元及车辆详细行驶路线,实验表明,通过构建的网约车合乘模型和设计的遗传算法,可以得到合乘路径近似最优解,证明了模型和算法的可行性和有效性. 展开更多

Objective To supply a scientific basis for standardizing the normal reference value of forced vital capacity(FVC)of Chinese younger women.Methods We studied the relationship between the normal reference value of 21767 samples of FVC of younger women and eight geographical factors in 157 areas in China.Results It was found that the correlation between geographical factors and the normal reference value of FVC of younger women was quite significant(F=5.884,P=0.000).By adopting the method of mathematical regression analysis,one regression equation was inferred:=3.146+0.00006919X1+0.01315X4-0.006966X6+0.09524X8±0.254.In the above equation,is the normal reference value of younger women’FVC(L);X1 is the altitude(m);X4 is the annual mean air temperature(℃);X6 is the annual mean relative humidity(%);X8 is the annual mean wind speed(m/s);0.254 is the value of the residual standard deviation.Conclusion If geographical values are obtained in a certain area,the normal reference value of FVC of younger women in this area can be obtained by using the regression equation.Furthermore,according to the geographical factors,China can be divided into eight regions:Northeast China Region,North China Region,Shanxi-Shaanxi-Inner Mongolia Region,middle and lower reaches of the Yangzte River Region,Southeast China Region,Northwest China Region,Southwest China Region,and Qinghai-Tibet Plateau Region. 展开更多

Background Hand,foot,and mouth disease caused by enterovirus 71(EV71)infection is prevalent in the Asia-Pacific region in recent years.Currently,no drug is available for the prevention and treatment of EV71 infection.IMB-0523,a N-phenylbenzamide derivative,inhibits hepatitis B virus replication by upregulating the expression of APOBEC3G.In the present study,the effect of IMB-0523 on EV71 replication and related mechanism were investigated.Methods The cytotoxicity of IMB-0523 was determined by cell counting kit.Quantitative real-time polymerase chain reaction and Western blot assay were used to detect the effect of IMB-0523 on EV71 replication and related mechanism.Cytopathic effect assay was used to investigate the effect of IMB-0523 on different EV71 strains,coxsackievirus A16,and coxsackieviruses of group B.Results The results showed that IMB-0523 could dose-dependently inhibit EV71 replication.Preliminary mechanism studies showed that IMB-0523 could activate STAT3 signaling to upregulate the expression of interferon-stimulated genes to play an antiviral role.In addition,IMB-0523 inhibited the replication of different EV71 strains,coxsackievirus A16,and coxsackieviruses of group B.Conclusions IMB-0523 inhibits EV71 replication by activating the STAT3 signaling pathway to upregulate interferon-stimulated gene expression.IMB-0523 has broad-spectrum antiviral potential and may be used as a lead compound for the development of broad spectrum antiviral drugs. 展开更多

The antiviral effect of jiadifenoic acids C against coxsackievirus B3

Coxsackievirus B type 3(CVB3)is one of the major causative pathogens associated with viral meningitis and myocarditis,which are widespread in the human population and especially prevalent in neonates and children.These infections can result in dilated cardiomyopathy(DCM)and other severe clinical complications.There are no vaccines or drugs approved for the prevention or therapy of CVB3-induced diseases.During screening for anti-CVB3 candidates in our previous studies,we found that jiadifenoic acids C exhibited strong antiviral activities against CVB3 as well as other strains of Coxsackie B viruses(CVBs).The present studies were carried out to evaluate the antiviral activities of jiadifenoic acids C.Results showed that jiadifenoic acids C could reduce CVB3 RNA and proteins synthesis in a dose-dependent manner.Jiadifenoic acids C also had a similar antiviral effect on the pleconaril-resistant variant of CVB3.We further examined the impact of jiadifenoic acids C on the synthesis of viral structural and non-structural proteins,finding that jiadifenoic acids C could reduce VP1 and 3D protein production.A time-course study with Vero cells showed that jiadifenoic acids C displayed significant antiviral activities at 0–6 h after CVB3 inoculation,indicating that jiadifenoic acids C functioned at an early step of CVB3 replication.However,jiadifenoic acids C had no prophylactic effect against CVB3.Taken together,we show that jiadifenoic acids C exhibit strong antiviral activities against all strains of CVB,including the pleconaril-resistant variant.Our study could provide a significant lead for anti-CVB3 drug development. 展开更多
