Background Endoplasmic reticulum (ER) stress-related apoptosis is involved in the pathophysiology of many cardiovascular diseases, and Panax quinquefolium saponin (PQS) is able to inhibit excessive ER stress-related apoptosis of cardiomyocytes following hypoxia/reoxygenation and myocardial infarction. However, the pathway by which PQS inhibits the ER stress-related apoptosis is not well understood. To further investigate the protective effect of PQS against ER stress-related apoptosis, primary cultured eardiomyocytes were stimulated with thapsigargin (TG), which is widely used to model cellular ER stress, and it could induce apoptotic cell death in sufficient concentration. Methods Primary cultured cardiomyocytes from neonatal rats were exposed to TG (1 μmol/L) treatment for 24 h, following PQS pre-treatment (160 μg/mL) for 24 h or pre-treatment with small interfering RNA directed against protein kinase-like endoplasmic reticulum kinase (Si-PERK) for 6 h. The viability and apoptosis rate of cardiomyocytes were detected by cell counting kit-8 and flow cytometry respectively. ER stress-related protein expression, such as glucose-regulated protein 78 (GRP78), calreticulin, PERK, eukaryotic translation initiation factor 2α (elF2c0, activating transcription factor 4 (ATF4), and C/EBP homologous protein (CHOP) were assayed by western blotting. Results Both PQS pre-treatment and PERK knockdown remarkably inhibited the cardiomyocyte apoptosis induced by TG, increased cell viability, decreased phosphorylation of both PERK and eIF2α, and decreased protein levels of both ATF4 and CHOP. There was no statistically significant difference between PQS pre-treatment and PERK knockdown in the cardioprotective effect. Conclusions Our data indicate that the PERK-eIF2α-ATF4-CHOP pathway of ER stress is involved in the apoptosis induced by TG, and PQS might prevent TG-induced cardiomyocyte apoptosis through a mechanism involving the suppression of this pathway. These findings provide novel data regarding the molecular mechanisms by which PQS inhibits cardiomyocyte apoptosis. 展开更多

Recrystallization is a critical issue in ITER and future fusion devices as it strongly deteriorates the prop-erties and serving life of tungsten divertor armor.In this work,we investigated the influence of neon on the recrystallization behavior of tungsten and its underlying mechanism by both experimental anal-ysis and molecular dynamics simulation.Rolled tungsten was irradiated by neon ion with energies of 10 and 100 keV and fluences of 1×10^(18)-1×10^(20)m^(−2),followed by isochronal annealing at various temperatures(1473-2073 K)for 1 h.The results indicate that 10 keV neon ion irradiation slightly in-hibits tungsten recrystallization due to the shallow depth distribution of neon(35 nm),whereas 100 keV neon ion irradiation exerts a significant retarding effect on tungsten recrystallization when neon con-centration reaches∼102 appm.Grain boundaries can hardly move with neon concentration reaching 400 appm,above which the retarding effect is very strong and varies little.The retarded recrystallization is mainly attributed to the pinning effect of neon clusters/bubbles on the grain boundary movement dur-ing annealing.Large neon clusters are found to play a predominant role in the retarding effect and the un-clustered neon atoms show a very limited effect on the grain boundary movement.By comparing the neon effects on recrystallization in this work and helium effects reported in the literature,we found that helium exhibits a relatively stronger recrystallization retarding effect than neon.This is because helium is more easily to diffuse into grain boundaries and form larger clusters due to its high diffusivity and low self-trapping energy in tungsten,which can greatly impede the migration of grain boundaries.This work provides a fundamental insight into the neon-concentration dependent recrystallization retarding effect in tungsten and the corresponding underlying atomic-level mechanisms. 展开更多

This study compares the changes of DNA repair in brain tissue dominated by the middle cerebral artery after electroacupuncture at the acupoints of Renzhong (DU 26), Neiguan (PC 6), Quchi (LI 11) and Zusanli (ST 36) in rats. In the apurinic/apyrimidinic endonuclease DNA basic group reparative excision experiments, the apurinic/apyrimidinic endonuclease activity of brain tissue increased slightly after electroacupuncture in rats. In the DNA polymerase β (Pol β)experiments, the Pol β ac-tivity of brain tissue increased after electroacupuncture, especially at DU 26 and PC 6. In the DNA ligase experiments, the expression of DNA ligase 1 in brain tissue increased significantly after electroacupuncture. These findings demonstrate increased activity of apurinic/apyrimidinic en-donuclease, Pol β and DNA ligase 1 after electroacupuncture at DU 26 and PC 6. Also, DNA repair in brain tissue supplied by the middle cerebral artery is promoted after electroacupuncture at DU 26 and PC 6, which are more effective than the LI 11 and ST 36 acupoints . 展开更多

Objective:To explore the rules of acupuncture point selection in thetreatment of rheumatoid arthritis by applying data mining and modern Statistical software.Methods:We retrieved literature from China hownet(CNKI),VIP database(VIP),Wanfang,PubMed,MEDLINE,Embase database from the founding of the database to February 1,2020.The main information of meridians and acupoints were entered into Excel,SPSS22.0 was used to extract data and do clustering analysis.Results:Quchi,Zusanli,Hegu,Yanglingquan,Xiyan were often chosen for electropuncture treatment of rheumatoid arthritis.Most of the acupoints are located in the large intestine channel of hand yangming bladder meridian or bladder channel of foot Taiyang,Shaoyang gallbladder channel of foot,stomachmeridian of foot Yangming,the energizer meridian and concentrated in the upper and lower extremities.The frequency of use of five Shu points,crossing point,source point,lowerconfluent and eight meeting points acupoints is higher.Cluster analysis can divide the included acupoints into 5 clusters.Conclusion:Quchi,Zusanli,Hegu,Yanglingquan,Xiyan are often chosen for electropuncture treatment of rheumatoid arthritis.Most of the selected acupoints aremostly distributed in the acupoints which are located in the large intestine channel of hand yangming bladder meridian or bladder channel of foot Taiyang,Shaoyang gallbladder channel of foot,Stomach meridian of foot Yangming,and the energizer meridian. 展开更多

Impact of ambient temperature on postoperative cognitive dysfunction in elderly patients surgery:A seasonal comparison in tropical regions

BACKGROUND To investigate whether seasonal differences in ambient temperature affect the incidence of early postoperative cognitive dysfunction(POCD)among elderly patients undergoing laparoscopic surgery in tropical regions.Additionally,it explored the perioperative risk factors associated with early POCD following abdominal laparoscopic surgery.AIM To investigate the influence of seasonal differences in ambient temperature on POCD of elderly patients METHODS A total of 125 patients aged≥65 years from Hainan Province,China,who underwent laparoscopic surgery under general anesthesia with tracheal intubation,were enrolled. All patients completed the Mini-Mental State Examination one day before surgery and onpostoperative days 1, 3, and 7. A decline of ≥ 2 points from baseline was considered indicative of cognitivedysfunction. Serum levels of S100 calcium binding protein B and neuron-specific enolase were measured usingenzyme-linked immunosorbent assay at three time points: Preoperatively, immediately after extubation, and 24hours postoperatively. Perioperative clinical data were collected to identify potential risk factors for POCD.Propensity score matching (PSM) was performed (1:1, caliper = 0.03), resulting in 41 matched patient pairs betweenwinter and summer groups.RESULTSAfter PSM, baseline characteristics including age, gender, body mass index, education level, comorbidities, andsurgical variables were well balanced between groups. There were no significant differences in the incidence ofPOCD on postoperative days 1, 3, and 7 between patients undergoing laparoscopic surgery in winter vs summer.However, multivariable logistic regression revealed that surgical duration (day 1, P value = 0.049), advanced ageand elevated creatinine (day 3, P value = 0.044, P value = 0.008), and hypoalbuminemia (day 3, P value = 0.042;day7, P value = 0.015) were independently associated with early POCD.CONCLUSIONAmbient temperature differences between winter and summer in tropical regions did not significantly affect theincidence of early POCD in elderly patients undergoing laparoscopic surgery. Nonetheless, age, longer surgicalduration, elevated creatinine, and hypoalbuminemia emerged as key risk factors. These findings underscore theimportance of perioperative optimization to reduce the risk of POCD in elderly patients, regardless of seasonaltemperature variations. 展开更多

Objective To observe the effects of moxibustion at Huantiao(GB30)acupoint on nerve repair,regeneration,and function in rats with sciatic nerve injury(SNI),and explore the possible mechanism of SNI improvement via moxibustion.Methods A total of 70 specific pathogen-free(SPF)grade male Sprague-Dawley(SD)rats were randomly assigned to control group(n=10)and model group(n=60).Following replication of SNI to model group rats,60 SNI model rats were randomly allocated to SNI groups of 1 d,3 d,and 7 d and moxibustion groups of 1 d,3 d,and 7 d with 10 rats in each group.Moxibustion groups were given moxibustion at the Huantiao(GB30)acupoint on the affected side with a 5 cm distance from the skin under isoflurane respiratory anesthesia and treated once a day for 20 min for 1 d,3 d,and 7 d,respectively.Control and SNI groups were anesthetized with isoflurane daily for 20 min.Open field tests and thermal pain threshold tests were conducted,and the general condition of rats was observed in each group pre-modeling and on treatment day 1,3,and 7.At the end of the treatment,immunofluorescence was used to detect the axonal growth rate,axonal growth density,and Schwann cells(SCs)proliferation in the middle 1-mm cross-section of the crush injury segment in rats.The gastrocnemius muscles on both sides of the rats were taken and weighed to calculate the wet weight ratio of the gastrocnemius muscles on both sides to observe the muscle atrophy of the rats,and hematoxylin-eosin(HE)staining was used to observe the pathomorphological changes of the gastrocnemius muscles on the affected side.Quantitative real-time polymerase chain reaction(qPCR)was used to detect the expression levels of nerve growth factor(NGF),interferon(IFN),macrophage migration inhibitory factor(MIF),interleukin(IL)-4,and transforming growth factor(TGF)-βin the sciatic nerve tissue of the rats.Results After modeling,rats in both moxibustion and SNI groups showed typical signs of pain behaviors(bending and curling of the hind soles of the affected side,licking claws,and lameness)and decreased activity compared with control group.The main benefits of moxibustion were evident from day 3:compared with SNI group,rats in moxibustion group had marked relief of pain behavior,increased activity levels and movement,and a lower response to thermal pain.At the same time,moxibustion significantly promoted the repair of SNI,as evidenced by the significantly better axonal growth rate,growth density,and SCs proliferation density in the crush injury segment compared with SNI group(P<0.01).Moxibustion also regulated the local microenvironment of the injury,up-regulated the pro-nerve repair factors NGF,IL-4,and TGF-β(P<0.05),and down-regulated the pro-inflammatory factors IFN-γ(P<0.01)and MIF(P<0.05).By day 7,the histomorphology of the gastrocnemius muscle in moxibustion group was improved,as indicated by enlarged muscle fibers,elevated regular myocyte morphology and wet weight ratio of the affected and unaffected sides(P<0.05),as well as a sustained high expression levels of NGF,IL-4,and TGF-β(P<0.05,P<0.05,and P<0.01,respectively),and a maintenance of low level of IFN-γ(P<0.01).Concurrently,the MIF level was not significantly different from SNI group(P>0.05).Conclusion Moxibustion at the Huantiao(GB30)acupoint effectively improves motor function and promotes recovery of sensory function and nerve regeneration in SNI rats,which may be related to the regulation of local inflammatory response,the promotion of nerve growth factor expression,the improvement of regenerative microenvironment,and the acceleration of SCs proliferation and axonal growth rate in damaged nerves. 展开更多

BACKGROUND Gastric signet ring cell carcinoma(GSRC)is a distinctive type of gastric cancer.It is a mucus-secreting adenocarcinoma that may progress to distant metastasis at an early stage.Because of poor differentiation,aggressive invasion,rapid progre-ssion,and other high-risk characteristics,early surgical intervention should be prioritized.AIM To explore the clinical efficacy of fluorouracil(5-FU)combined with paclitaxel and oxaliplatin for the treatment of advanced GSRC.METHODS A total of 85 patients with advanced GSRC were selected between January 2020 and June 2021 and randomly divided into a control group(n=42,receiving standard chemotherapy)and a treatment group(n=43,receiving monotherapy with oxaliplatin,5-FU,and paclitaxel).Patients in the treatment group received a 135 mg/m2 infusion of paclitaxel for 3 hours,a 400 mg/m2 infusion of calcium folate(or 200 mg/m2 of levocalcium folate)for 2 hours,and an 85 mg/m2 infusion of oxaliplatin for 2 hours.This was followed by a continuous intravenous infusion of 2200-2400 mg/m25-FU for 46 hours using a portable pump.RESULTS The treatment group showed a median survival time of 11.7 months and an objective response rate(ORR)of 32.5%,significantly higher than the control group(P<0.05).Serum carcinoembryonic antigen(CEA),carbohydrate antigen 19-9(CA19-9),and albumin levels were correlated with treatment effectiveness in advanced GSRC(P<0.01),but total serum protein was not correlated(P>0.05).Safety and survival were assessed in all patients.Short-term efficacy was evaluated in 66 patients,with a disease control rate of 89.4%and an ORR of 48.5%.Median progression-free survival was 7.0 months(95%confidence interval[CI]:6.85-7.15),and median overall survival was 10.6 months(95%CI:9.86-11.3).Primary grade III/IV adverse events included neutropenia(22.1%)and peripheral neurotoxicity(10.3%).CONCLUSION This treatment regimen is more effective for patients with advanced GSRC.Serum levels of CEA,CA19-9,and albumin predicted chemotherapy efficacy,while total protein concentration correlated minimally and insigni-ficantly. 展开更多

Previous studies have shown that somatic sensation by acupuncture and visceral nociceptive stimulation can converge in the nucleus tractus solitarii where neurons integrate signals impact- ing on the function of organs. To explore the role of the nucleus tractus solitarii in the protective mechanism of pre-moxibustion on gastric mucosa, nucleus tractus solitarii were damaged in rats and pre-moxibustion treatment at the Zusanli (ST36) point followed. The gastric mucosa was then damaged by the anhydrous ethanol lavage method. Morphological observations, enzyme linked immunosorbent assays, and western immunoblot analyses showed that gastric mucosa surface lesion and the infiltration of inflammatory cells were significantly ameliorated after pre-moxibustion treatment. Furthermore, the gastric mucosal damage index and somatostatin level were reduced, and epidermal growth factor content in the gastric mucosa and heat-shock protein-70 expression were increased. These results were reversed by damage to the nucleus tractus solitarii. These findings suggest that moxibustion pretreatment at the Zusanli point is protective against acute gastric mucosa injury, and nucleus tractus solitarii damage inhibits these responses. Therefore, the nucleus tractus solitarii may be an important area for regulating the signal transduction of the protective effect of pre-moxibustion on gastric mucosa. 展开更多

Objective:To study the effect of Alpinia officinarum Hance(A.officinarum) 80% alcohol extract on the primary dysmenorrhea.Methods:A.officinarum 80% alcohol extract were enriched by macroporous adsorption resins.Female mice of primary dysmenorrhea model were established by oxytocin induction; the effects of A.officinarum 80% alcohol extract on primary dysmenorrhea were observed by body twist method; and the homogenate level of prostaglandin F_(2α)(PGF_(2α)),prostaglandin E_2(PGE_2) and Ca^(2+) in the uterus were observed in oxytocin-induced female mice.Results:The writhing frequency of primary dysmenorrhea mice was significantly decreased after treatment of A.officinarum 80% alcohol extract and the level of PGF_(2α),PGE_2 and Ca^(2+) in mice uterus was significantly decreased(P<0.05,P<0.01) in groups of mice treated with middle and high dosage of A.officinarum 80% alcohol extract compared with that of model group.Conclusions:These findings suggest that A.Officinarum 80% alcohol extract can significantly relieve primary dysmenorrhea. 展开更多

In construction,it is important to check whether workers wear safety helmets in real time.We proposed using an unmanned aerial vehicle(UAV)to monitor construction workers in real time.As the small target of aerial photography poses challenges to safety-helmet-wearing detection,we proposed an improved YOLOv4 model to detect the helmet-wearing condition in aerial photography:(1)By increasing the dimension of the effective feature layer of the backbone network,the model’s receptive field is reduced,and the utilization rate of fine-grained features is improved.(2)By introducing the cross stage partial(CSP)structure into path aggregation network(PANet),the calculation amount of themodel is reduced,and the aggregation efficiency of effective features at different scales is improved.(3)The complexity of the YOLOv4 model is reduced by introducing group convolution and the pruning PANet multi-scale detection mode for de-redundancy.Experimental results show that the improved YOLOv4 model achieved the highest performance in the UAV helmet detection task,that the mean average precision(mAP)increased from83.67%of the original YOLOv4 model to 91.03%,and that the parameter amount of the model is reduced by 24.7%.The results prove that the improved YOLOv4 model can effectively respond to the requirements of real-time detection of helmet wearing by UAV aerial photography. 展开更多

The present paper proposes a detection method for building exterior wall cracks since manual detection methods have high risk and low efficiency.The proposed method is based on Unmanned Aerial Vehicle(UAV)and computer vision technology.First,a crack dataset of 1920 images was established using UAV to collect the images of a residential building exterior wall under different lighting conditions.Second,the average crack detection precisions of different methods including the Single Shot MultiBox Detector,You Only Look Once v3,You Only Look Once v4,Faster Regional Convolutional Neural Network(R-CNN)and Mask R-CNN methods were compared.Then,the Mask R-CNN method with the best performance and average precision of 0.34 was selected.Finally,based on the characteristics of cracks,the utilization ratio of Mask R-CNN to the underlying features was improved so that the average precision of 0.9 was achieved.It was found that the positioning accuracy and mask coverage rate of the proposed Mask R-CNN method are greatly improved.Also,it will be shown that using UAV is safer than manual detection because manual parameter setting is not required.In addition,the proposed detection method is expected to greatly reduce the cost and risk of manual detection of building exterior wall cracks and realize the efficient identification and accurate labeling of building exterior wall cracks. 展开更多

The Influenza A(H1N1)pdm09 virus caused a global pandemic in 2009 and has circulated seasonally ever since.As the continual genetic evolution of hemagglutinin in this virus leads to antigenic drift,rapid identification of antigenic variants and characterization of the antigenic evolution are needed.In this study,we developed PREDAC-H1pdm,a model to predict antigenic relationships between H1N1pdm viruses and identify antigenic clusters for post-2009 pandemic H1N1 strains.Our model performed well in predicting antigenic variants,which was helpful in influenza surveillance.By mapping the antigenic clusters for H1N1pdm,we found that substitutions on the Sa epitope were common for H1N1pdm,whereas for the former seasonal H1N1,substitutions on the Sb epitope were more common in antigenic evolution.Additionally,the localized epidemic pattern of H1N1pdm was more obvious than that of the former seasonal H1N1,which could make vaccine recommendation more sophisticated.Overall,the antigenic relationship prediction model we developed provides a rapid determination method for identifying antigenic variants,and the further analysis of evolutionary and epidemic characteristics can facilitate vaccine recommendations and influenza surveillance for H1N1pdm. 展开更多

Evaluating the permeability and irreducible water saturation of tight sandstone reservoirs is challenging.This study uses distribution functions to fit measured NMR T_(2)distributions of tight sandstone reservoirs and extract parameters for characterizing pore size distribution.These parameters are then used to establish prediction models for permeability and irreducible water saturation of reservoirs.Results of comparing the fit of the T_(2)distributions by the Gauss and Weibull distribution functions show that the fitting accuracy with the Weibull distribution function is higher.The physical meaning of the statistical parameters of the Weibull distribution function is defined to establish nonlinear prediction models of permeability and irreducible water saturation using the radial basis function(RBF)method.Correlation coefficients between the predicted values by the established models and the measured values of the tight sandstone core samples are 0.944 for permeability and 0.851 for irreducible water saturation,which highlight the effectiveness of the prediction models. 展开更多

Objective Nasal-type natural killer/T-cell lymphoma of Waldeyer's ring (WR-NK/TL) has different clinico- pathological characteristics from those of other subtypes of NK/T lymphoma; thus, the optimal treatment remains unclear. To find a more effective treatment model for WR-NK/TL, we conducted a single-center study of concurrent radiochemotherapy. Methods Forty-five patients with newly diagnosed stage IE to liE WR-NKTL were randomly divided into two groups. The 23 cases in the concurrent radiochemotherapy group were treated with three-dimensional conformal radiotherapy (48-52 Gy) and 2 courses of DICE (dexamethasone, ifosfamide, cisplatin, and etoposide) synchronous chemotherapy. The 22 cases in the radiotherapy group only received three-dimen- sional conformal radiotherapy (50-54 Gy). The primary end points were overall survival (OS), progression- free survival (PFS), and toxicity. Results The 1-, 3-, and 4-year OS and PFS rates were 95.5%, 65.6%, and 45.9%, and 86.4%, 56.0%, and 46.7% in the radiotherapy group, and 100%, 88.5%, and 88.5%, and 100%, 82.0%, and 73.8% in the concurrent radiochemotherapy group, respectively. The OS (P = 0.0477) and PFS rates (P = 0.0488) were higher in the concurrent radiochemotherapy group than in the radiotherapy group. The overall re- sponse rate was 100% in both the radiotherapy group [complete response (CR), 18 cases] and concurrent radiochemotherapy group (CR, 22 cases). The concurrent radiochemotherapy group had more severe side effects, especially grade 3 + 4 events, such as leukopenia, anorexia, and stomatitis. However, side effects benefiting from excellent oral care were endurable. Conclusion Radiotherapy plus concurrent DICE chemotherapy may be an effective and safe compre- hensive treatment for patients with WR-NKTL. 展开更多

Two-dimensional(2D)nuclear magnetic resonance(NMR)inversion operates with massive echo train data and is an ill-posed problem.It is very important to select a suitable inversion method for the 2D NMR data processing.In this study,we propose a fast,robust,and effective method for 2D NMR inversion that improves the computational efficiency of the inversion process by avoiding estimation of some unneeded regularization parameters.Firstly,a method that combines window averaging(WA)and singular value decomposition(SVD)is used to compress the echo train data and obtain the singular values of the kernel matrix.Subsequently,an optimum regularization parameter in a fast manner using the signal-to-noise ratio(SNR)of the echo train data and the maximum singular value of the kernel matrix are determined.Finally,we use the Butler-Reeds-Dawson(BRD)method and the selected optimum regularization parameter to invert the compressed data to achieve a fast 2D NMR inversion.The numerical simulation results indicate that the proposed method not only achieves satisfactory 2D NMR spectra rapidly from the echo train data of different SNRs but also is insensitive to the number of the final compressed data points. 展开更多
