为缓解草莓脱毒苗生产中部分品种茎尖数量无法满足种苗生产需求的问题,以及筛选不同草莓品种用于叶片离体诱导最适宜的叶龄,该试验以桃熏、圣诞红、隋珠、黔莓1号和越秀为试验材料,研究了叶片离体诱导培养基:基本培养基(MS)+0.5 mg·... 为缓解草莓脱毒苗生产中部分品种茎尖数量无法满足种苗生产需求的问题,以及筛选不同草莓品种用于叶片离体诱导最适宜的叶龄,该试验以桃熏、圣诞红、隋珠、黔莓1号和越秀为试验材料,研究了叶片离体诱导培养基:基本培养基(MS)+0.5 mg·L^(-1)6-苄氨基嘌呤(6-BA)+1.5 mg·L^(-1)2,4-二氯苯甲酸(TDZ)+0.2 mg·L^(-1)萘乙酸(NAA)中,不同草莓品种和不同叶龄的再生频率的差异。与此同时,该试验还以隋珠叶片为外植体,研究不同刻伤方式对其愈伤组织诱导的影响。结果表明,圣诞红和桃熏在MS+0.5 mg·L^(-1)6-BA+1.5 mg·L^(-1)TDZ+0.2 mg·L^(-1)NAA培养基条件下不定芽诱导率较高;隋珠采用叶缘叶背混合刻伤的处理方式有利于提高其愈伤组织诱导率。以上结果表明,生产中部分栽培草莓可采用同种培养基配方进行叶片离体诱导,从而提高脱毒苗生产效率;不同刻伤方式对部分草莓品种的愈伤组织数量有一定的影响,可在试验中通过选择合适的刻伤方式提高叶片的愈伤组织诱导率。 展开更多

基于MRI影像组学对肝细胞癌术后复发的预测价值

目的:探讨基于磁共振成像(MRI)影像组学特征以及MRI征象构建联合模型,预测肝细胞癌(HCC)根治性切除术后复发的临床应用价值。方法:回顾性分析2016年5月至2020年5月淄博一四八医院收治的100例HCC根治性切除术患者的影像资料,所有患者术... 目的:探讨基于磁共振成像(MRI)影像组学特征以及MRI征象构建联合模型,预测肝细胞癌(HCC)根治性切除术后复发的临床应用价值。方法:回顾性分析2016年5月至2020年5月淄博一四八医院收治的100例HCC根治性切除术患者的影像资料,所有患者术前均行腹部增强MRI检查,术后至少随访两年。按照7∶3比例随机分为训练组(70例)和验证组(30例),根据术后随访结果训练组复发12例,无复发58例;验证组复发5例,无复发25例。使用3D-slicer软件对每例HCC患者术前MRI图像提取影像组学特征,计算观察者提取影像学特征的组内相关系数(ICC),选择最大相关最小冗余(mRMR)算法及lasso回归分析降维、筛选后建立影像组学标签。采用单因素及多因素logistic回归分析筛选MRI征象中预测复发的独立危险因素,并分别与平扫、动脉期、门脉期及肝胆期的影像组学标签构建影像组学模型;采用受试者工作特征(ROC)曲线评估各影像组学模型预测复发的诊断效能。结果:两位放射科医师对所有患者MRI图像中选取影像组学特征的ICC为0.903~0.957,一致性较好(ICC≥0.9)。与其他预测模型相比较,平扫影像组学模型在训练组和验证组中预测HCC根治性切除术后复发的ROC曲线下面积(AUC)最高分别为[0.951(95%CI:0.901~1.000)]和[0.968(95%CI:0.917~1.000)]。结论:基于MRI影像组学特征构建的联合模型对HCC根治性切除术后患者的复发具有较好的预测价值,其中平扫影像组学模型在无增强情况下以及欠发达地区对临床实施个性化治疗方案有一定指导作用。 展开更多

为了解河北省唐山市规模化奶牛场中牛病毒性腹泻病毒(BVDV)流行毒株的基因特征,本试验于唐山市2个规模化奶牛场采集了49份临床样品进行逆转录聚合酶链式反应(RT-PCR)检测,并对阳性样品进行病毒的分离,最终成功分离到1株非致细胞病变的B... 为了解河北省唐山市规模化奶牛场中牛病毒性腹泻病毒(BVDV)流行毒株的基因特征,本试验于唐山市2个规模化奶牛场采集了49份临床样品进行逆转录聚合酶链式反应(RT-PCR)检测,并对阳性样品进行病毒的分离,最终成功分离到1株非致细胞病变的BVDV毒株,命名为HB-1株。电镜观察结果显示,HB-1株具有典型的BVDV病毒粒子形态。对HB-1株全基因组序列进行扩增和测序,基因遗传进化分析结果显示,HB-1株属于BVDV-1m亚型。HB-1株结构蛋白E2全长序列糖基化位点和抗原表位预测结果显示,HB-1株E2蛋白含有4个保守的糖基化位点,其中抗原表位1高度保守。本试验为河北省进一步开展牛病毒性腹泻(BVD)流行病学调查、疫苗株筛选以及检测方法建立提供数据支撑。 展开更多

为了建立快速、准确的检测小反刍兽疫病毒抗体的方法,试验从GenBank中下载小反刍兽疫病毒结构蛋白H基因序列,并对其进行分析设计,通过基因合成方法得到相应片段,进一步构建重组质粒,将目的片段插入酵母基因组中,利用酵母表达系统表达目... 为了建立快速、准确的检测小反刍兽疫病毒抗体的方法,试验从GenBank中下载小反刍兽疫病毒结构蛋白H基因序列,并对其进行分析设计,通过基因合成方法得到相应片段,进一步构建重组质粒,将目的片段插入酵母基因组中,利用酵母表达系统表达目的蛋白并纯化,以纯化后的H蛋白作为包被抗原,建立检测小反刍兽疫病毒抗体的间接ELISA方法,并用该方法对40份阴阳性血清和临床189份样品进行检测,将检测结果分别与中和试验和商品化试剂盒的检测结果进行比较。结果表明:成功构建并表达小反刍兽疫病毒H蛋白。基于H蛋白建立的间接ELISA抗体检测方法对40份阴阳性血清的检测结果与中和试验比较,总符合率为95%(38/40);对临床189份样品的检测结果与商品化试剂盒比较,总符合率为93.12%(176/189)。说明该方法与中和试验及商品化试剂盒检测结果有较好符合度,可以用于小反刍兽疫临床血清抗体的检测及流行病学调查。 展开更多

目的分析腹腔镜辅助经腹直肠癌切除术与经自然腔道取标本术(natural orifice specimen extraction surgery,NOSES)对低位直肠癌患者胃肠功能及肿瘤标志物水平的影响。方法选取2019年2月-2022年2月在中国人民解放军联勤保障部队第960医... 目的分析腹腔镜辅助经腹直肠癌切除术与经自然腔道取标本术(natural orifice specimen extraction surgery,NOSES)对低位直肠癌患者胃肠功能及肿瘤标志物水平的影响。方法选取2019年2月-2022年2月在中国人民解放军联勤保障部队第960医院淄博医疗区治疗的80例低位直肠癌患者作为研究对象,随机分为对照组和研究组,各40例。对照组采用腹腔镜辅助经腹直肠癌切除术进行治疗,研究组进行采用腹腔镜NOSES术治疗。比较2组手术相关指标、胃肠功能、肿瘤标志物水平及并发症发生情况。结果与对照组相比,研究组手术时间较短、术中出血量较少,且术后排便频率、肛门排气及胃肠功能恢复时间均降低(P<0.05);与术前相比,2组癌胚抗原(carcinoembryonic antigen,CEA)、糖类抗原242(carbohydrate antigen 242,CA242)、糖类抗原724(Sugar-class antigen 724,CA724)表达水平降低,且研究组降低幅度较对照组大(P<0.05);与对照组相比,研究组并发症发生率较低(P<0.05)。结论腹腔镜NOSES术可有效降低低位直肠癌患者的肿瘤标志物水平,改善患者胃肠功能,减少术中出血量,缩短手术时间,降低并发症的发生,临床治疗效果优于腹腔镜辅助经腹直肠癌切除术。 展开更多

We study the ultrafast solvation dynamics of protein-precipitant complexes.Protein subtilisin carlsberg(SC)was mixed with several polyethylene glycol(PEG)precipitants for protein crystallization.Picosecond-resolved emission spectra from single intrinsic tryptophan residue(Trp-113)are recorded to construct solvation correlation functions.For precipitant concentrations with various crystallization effects,we observe drastically different solvation relaxation processes.These differences in solvation dynamics are correlated with the local protein structural integrity and water-network stability upon interaction with the precipitants.The solvation dynamics at the protein surface is proposed as a new perspective to study precipitant-protein interactions. 展开更多

Objective: To observe the effects of remote ischemia on cognitive function and neuronal pathological damage in rats with cognitive impairment induced by bilateral common carotid artery occlusion(BCAO).Methods:Male SD rats were selected to establish the cognitive impairment model induced by cerebral ischemia reperfusion caused by BCAO.The tests included three groups of rats:a sham group,a model group with vascular cognitive impairment (VCI) , and a remote ischemic conditioning (RIC) group (VCI + RIC group). From 24 h after operation, both hind limbs of rats in VCI + RIC group were treated with RIC. After 28 d, Morris water maze test and HE staining was used to observe the pathological changes of white matter and hippocampus in each group.Results: After 3 d mice in VCI group began to improve gradually. The recovery of rats in the VCI + RIC group was relatively slow,but they started to recover rapidly 2 d after the operation.Morris water maze test showed that the escape latency of rats in VCI group and VCI+RIC group was longer than that in the sham group, and the score of VCI+RIC group was better than that of the VCI group, but there was a significant difference between the two groups(P<0.05).The space exploration experiment was performed at 7 d and 28 d after the operation;the VCI+RIC group outperformed the VCI group in both trials;the difference between the two groups was statistically significant (P<0.05).In the target quadrant exploration time, the difference between the VCI group (33.5±11.3 s) and the VCI+RIC group (41.2±9.7 s) was statistically significant (P<0.05).Results from the hematoxylin and eosin(HE)staining showed that compared with VCI group, cortical cells in VCI + RIC group had loose stroma, thinner nerve fibers, fewer broken cells, and slightly shrunken cells. Compared with VCI group, neurons in VCI + RIC group had a little vacuolar degenera-tion and slightly shrunken cell volume.Conclusion:Cerebral ischemia-reperfusion injury can cause learning and memory impairment in rats, leading to VCI. RIC can significantly improve VCI and play a neuroprotective role. 展开更多

Background:To compare the safety and effectiveness of direct mechanical thrombectomy and bridging therapy for stroke with acute anterior circulation large vessel occlusion within 4.5 hours of onset.Methods:Retrospectively collected from 66 patients with acute ischemic stroke admitted to the Department of Neurology of Tongliao Hospital and Xuanwu Hospital from August 2019 to November 2021 within 4.5 hours.According to the different recanalization methods,30 patients were assigned to the direct thrombectomy treatment group,and 36 patients in the bridging treatment group(i.e.,the intravenous thrombolysis bridging mechanical thrombectomy treatment group).The primary outcome measure was the neurological outcome at the onset of 90d.Secondary outcome measures were intraoperative vascular recanalization and reperfusion,and the US National Institute of Health Stroke Scale score at 24 hours after surgery.The primary safety indicators are intracranial hemorrhage,including symptomatic intracranial hemorrhage and non-symptomatic intracranial hemorrhage,and 90d mortality.Results:The direct thrombectomy group had lower body mass index,hypertension and baseline Alberta early computed tomography score than the bridging treatment group,and longer time from onset to visit than the bridging group(206.5(119.5,256.25)min vs.150.5(25.205,212.75)min),the above difference were statistically significant(P<0.05).There were no significant differences in successful vascular reperfusion(93%vs.89%),24 hours postoperative National Institute of Health Stroke Scale score(11(5,18)vs.11(5,20)),intracranial hemorrhage(11%vs.14%),symptomatic intracranial hemorrhage(7%vs.17%),90d mRS0 to 2 points(43%vs.36%)and 90d mortality(23%vs.22%)(P>0.05).Conclusion:Similar clinical efficacy and safety of direct mechanical thrombectomy and bridging therapy for acute anterior circulation large vessel occlusive stroke within 4.5 hours of onset,direct thrombectomy can be used as an alternative scheme for acute anterior circulation intracranial large artery occlusive stroke. 展开更多

Background:The occurrence of early neurological deterioration following intravenous thrombolysis(IVT)is considered a particularly ominous clinical event and is strongly correlated with poor outcomes.Initiating tirofiban within 24 h after IVT has been suggested as a better treatment option to achieve long-term functional outcomes.However,the rationality of this remedy is a controversial.The purpose of the study was to evaluate the safety and efficacy of early intravenous tirofiban administration after IVT in patients with acute ischemic stroke(AIS).Methods:Databases including PubMed,EMBASE,Cochrane Library,and Web of Science were searched for clinical trials on early tirofiban implementation after IVT in patients with AIS from inception to September 2022.Odds ratios(ORs)were generated for dichotomous variants via meta-analysis using STATA 17.0 MP.Results:Five clinical trials with 725 patients were eligible.The study outcomes demonstrated that early tirofiban administration after IVT was not associated with symptomatic intracranial hemorrhage(OR,0.78;95%confidence interval(CI),0.22–2.74;P=0.70),asymptomatic intracranial hemorrhage(OR,1.11;95%CI,0.52–2.37;P=0.80),systemic bleeding(OR,0.97;95%CI,0.42–2.23;P=0.94),and death(OR,1.05;95%CI,0.47–2.31;P=0.91),but may reduce the incidence of early neurological deterioration(OR,0.09;95%CI,0.02–0.50;P=0.01),and was significantly associated with 90-day excellent(modified Rankin scale score 0–1)(OR,2.01;95%CI,1.35–3.02;P=0.00)and favorable(modified Rankin scale score 0–2)(OR,2.30;95%CI,1.63–3.23;P=0.00)functional outcomes.Conclusion:The early intravenous administration of tirofiban after IVT in patients with AIS may be a safe and effective treatment strategy that improves long-term neurological functional outcomes without increasing the risk of adverse events. 展开更多
