Molecular characterization of Echinococcus granulosus in paraffin-embedded human tissues from Southwest Iran

Objective:To investigate Echinococcus(E.)granulosus genotypes as the causative agents of hydatidosis in humans in the southwest of Iran(Khuzestan province).Methods:In this study,isolates of 80 archived human paraffin embedded hydatid cysts were collected from pathology laboratories in Ahvaz city,Khuzestan province.DNA was extracted and examined by nested-PCR of ribosomal DNA(rDNA)internal transcribed spacer 1(ITS1),and PCR-RFLP.In addition,the sequences of fragments of genes coding for Cox space1 and NADH dehydrogenase 1(ND1)were also examined.Results:Of the 80 paraffin samples,44(55.0%)were from the liver,27(33.8%)from the lung,and the rest from other organs.The amplified hydatid genomic DNA showed that the cysts were E.granulosus strains.The results of PCR-RFLP and sequencing analysis revealed the presence of G1 genotype(sheep strain)in all human isolates.Furthermore,no camel strain(G6)was detected among all samples in the regions studied.Conclusions:The molecular findings indicate that the predominant genotype involved in E.granulosus transmission in southwest of Iran is the common sheep strain(G1),which occurs in human populations.These results may have important implications for hydatid disease control in the studied areas. 展开更多

Detection of acute and chronic toxoplasmosis amongst multi-transfused thalassemia patients in southwest of Iran

Objective: Since pre-transfusion screening for Toxoplasma gondii is not performed on blood packs, thalassemia patients are susceptible to acquiring toxoplasmosis;thus, the aim of this study was to evaluate the seroprevalence status of T. gondii in individuals who suffer from thalassemia in comparison to healthy persons in the southwest of Iran. Methods: In this case-control study, 117 thalassemia patients and 205 healthy persons participated. All samples were tested for the presence of specific IgG and IgM antibodies against T. gondii using ELISA technique. Data were analyzed using Chi-square test. Results: Seroprevalence of anti-T. gondii IgG was detected in 30.76% (36/117) of patients and 20% (41/205) of healthy individuals (P=0.04), also anti-T. gondii IgM in these groups was detected 1.70% (2/117) and 0.48% (1/205), respectively (P=0.3). In present study, nine related risk factors with toxoplasmosis were evaluated and data analysis showed that only contact with cat was significantly correlated with IgG seroprevalence (P=0.02). Conclusions: Current research suggests thalassemia patients are more prone than normal persons to acquiring T. gondii infection (P=0.04, OR:1.77). Due to limited studies in this high risk group, further studies are recommended. 展开更多

Iran is considered as one of the malaria-endemic countries in the globe.It is estimated that approximately 80%of malaria infections happen only in two provinces,Sistan-Baluchestan and Hormozgan provinces(in southeast and south of Iran,respectively)[1].Based on formal reports,Iran is in the pre-elimination phase of the infection and elimination of malaria is the final goal,which is targeted by 2020[2].The total positive clinical cases dropped from 15378 to 960 between 2002 and 2017 in the country[1].There are seven anopheline species as primary or secondary vectors of malaria in Iran including,Anophele(An.)culicifacies Giles(s.l.),An.dthali Patton,An.fluviatilis James(s.l.),An.maculipennis Meigen(s.l.),An.sacharovi Favre,An.stephensi Liston and An.superpictus Grassi(s.l.).These suitable vectors exist in several provinces[3]. 展开更多
