Establishment of a nested-ASP-PCR method to determine the clarithromycin resistance of Helicobacter pylori 被引量:4

AIM: To investigate clarithromycin resistance positions 2142, 2143 and 2144 of the 23 Sr RNA gene in Helicobacter pylori(H. pylori) by nested-allele specific primer-polymerase chain reaction(nested-ASP-PCR).METHODS: The gastric tissue and saliva samples from 99 patients with positive results of the rapid urease test(RUT) were collected. The nested-ASP-PCR method was carried out with the external primers and inner allele-specific primers corresponding to the reference strain and clinical strains. Thirty gastric tissue and saliva samples were tested to determine the sensitivity of nested-ASP-PCR and ASP-PCR methods. Then, clarithromycin resistance was detected for 99 clinical samples by using different methods, including nestedASP-PCR, bacterial culture and disk diffusion. RESULTS: The nested-ASP-PCR method was successfully established to test the resistance mutation points 2142, 2143 and 2144 of the 23 SrR NA gene of H. pylori. Among 30 samples of gastric tissue and saliva, the H. pylori detection rate of nested-ASP-PCR was 90% and 83.33%, while the detection rate of ASP-PCR was just 63% and 56.67%. Especially in the saliva samples, nested-ASP-PCR showed much higher sensitivity in H. pylori detection and resistance mutation rates thanASP-PCR. In the 99 RUT-positive gastric tissue and saliva samples, the H. pylori-positive detection rate by nested-ASP-PCR was 87(87.88%) and 67(67.68%), in which there were 30 wild-type and 57 mutated strains in gastric tissue and 22 wild-type and 45 mutated strains in saliva. Genotype analysis showed that three-points mixed mutations were quite common, but different resistant strains were present in gastric mucosa and saliva. Compared to the high sensitivity shown by nested-ASP-PCR, the positive detection of bacterial culture with gastric tissue samples was 50 cases, in which only 26 drug-resistant strains were found through analyzing minimum inhibitory zone of clarithromycin. CONCLUSION: The nested-ASP-PCR assay showed higher detection sensitivity than ASP-PCR and drug sensitivity testing, which could be performed to evaluate clarithromycin resistance of H. pylori. 展开更多

In clinical cancer research,it is quite promising to develop multimodal synergistic therapeutic strategies.Photodynamic and photothermal synergistic therapy is a very desirable multimodal therapy strategy.Herein,we report a facile and simple method to construct a nanotherapeutic agent for photodynamic and photothermal therapy.This nanotherapeutic agent(ZnO@Ce6-PDA)is composed of a ZnO nanoparticle core,an interlayer of photosensitizer chlorin e6(Ce6)and an outer layer of polydopamine(PDA).Due to the existence of Ce6,the ZnO@Ce6-PDA can efficiently generate singlet oxygen(1O2)under 660 nm laser irradiation.Moreover,the ZnO@Ce6-PDA can serve as a photothermal agent,because of the excellent photothermal conversion efficiency of the PDA coating layer in the presence of 780 nm laser.Experiment results demonstrated that the designed nanotherapeutic agent had outstanding phototoxicity upon the combination of laser irradiation at 660 and 780 nm.Thus,our work proves that the ZnO@Ce6-PDA is a promising photodynamic/photothermal dual-modal nanotherapeutic agent for enhanced cancer therapy. 展开更多

Towards a physics-constrained and interpretable datadriven parameterization scheme for mesoscale eddies in ocean modeling

Mesoscale eddies play a pivotal role in deciphering the intricacies of ocean dynamics and the transport of heat,salt,and nutrients.Accurate representation of these eddies in ocean models is essential for improving model predictions.In this study,we propose a convolutional neural network(CNN)that combines data-driven techniques with physical principles to develop a robust and interpretable parameterization scheme for mesoscale eddies in ocean modeling.We use a highresolution reanalysis dataset to extract subgrid eddy momentum and then applying machine learning algorithms to identify patterns and correlations.To ensure physical consistency,we have introduced conservation of momentum constraints in our CNN parameterization scheme through soft and hard constraints.The interpretability analysis illustrate that the pre-trained CNN parameterization shows promising results in accurately solving the resolved mean velocity and effectively capturing the representation of unresolved subgrid turbulence processes.Furthermore,to validate the CNN parameterization scheme offline,we conduct simulations using the Massachusetts Institute of Technology general circulation model(MITgcm)ocean model.A series of experiments is conducted to compare the performance of the model with the CNN parameterization scheme and high-resolution simulations.The offline validation demonstrates the effectiveness of the CNN parameterization scheme in improving the representation of mesoscale eddies in the MITgcm ocean model.Incorporating the CNN parameterization scheme leads to better agreement with high-resolution simulations and a more accurate representation of the kinetic energy spectra. 展开更多

Ischemia-reperfusion injury(IRI)remains an unavoidable challenge in liver surgery,with macrophages playing a critical role in its pathogenesis.However,the mechanisms by which macrophages regulate the pathogenesis of IRI are not well understood.Through a target-guided screening approach,we identified a small 3 k Da peptide(Sj DX5-271)from various schistosome egg-derived peptides that induced M2 macrophage polarization.SjDX5-271 treatment protected mice against liver IRI by promoting M2 macrophage polarization,and this protective effect was abrogated when the macrophages were depleted.Transcriptomic sequencing showed that the TLR signaling pathway was significantly inhibited in macrophages from the SjDX5-271 treatment group.We further identified that SjDX5-271 promoted M2 macrophage polarization by inhibiting the TLR4/My D88/NF-κB signaling pathway and alleviated hepatic inflammation in liver IRI.Collectively,SjDX5-271 exhibited some promising therapeutic effects in IRI and represented a novel therapeutic approach,potentially applicable to other immune-related diseases.The current study demonstrates the potential of new biologics from the parasite,enhances our understanding of host-parasite interplay,and provides a blueprint for future therapies for immunerelated diseases. 展开更多

AIM: To explore the effects of siRNA silencing of PIK3CA on proliferation, migration and invasion of gastric cancer cells and to investigate the underlying mechanisms. METHODS: The mutation of PIK3CA in exons 9 and 20 of gastric cancer cell lines HGC-27, SGC-7901, BGC-823, MGC-803 and MKN-45 was screened by polymerase chain reaction (PCR) followed by sequencing. BGC-823 cells harboring no mutations in either of the exons, and HGC-27 cells containing PIK3CA mutations were employed in the current study. siRNA targeting PIK3CA was chemically synthesized and was transfect- ed into these two cell lines in vitro . mRNA and protein expression of PIK3CA were detected by real-time PCR and Western blotting, respectively. We also measured phosphorylation of a serine/threonine protein kinase (Akt) using Western blotting. The proliferation, migration and invasion of these cells were examined separately by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltet-razolium bromide (MTT), wound healing and Transwell chambers assay. RESULTS: The siRNA directed against PIK3CA effectively led to inhibition of both endogenous mRNA and protein expression of PIK3CA, and thus significantly down-regulated phosphorylation of Akt (P < 0.05). Furthermore, simultaneous silencing of PIK3CA resulted in an obvious reduction in tumor cell proliferation activity, migration and invasion potential (P < 0.01). Intriguing, mutant HGC-27 cells exhibited stronger invasion ability than that shown by wild-type BGC-823 cells. Knockdown of PIK3CA in mutant HGC-27 cells contributed to a reduction in cell invasion to a greater extent than in non-mutant BGC-823 cells. CONCLUSION: siRNA mediated targeting of PIK3CA may specifically knockdown the expression of PIK3CA in gastric cancer cells, providing a potential implication for therapy of gastric cancer. 展开更多

BACKGROUND: The availability of novel direct-acting antivirals (DAAs) represents a new era of curative hepatitis C virus (HCV) treatment, with over 95% of patients infected with HCV genotype 1 achieving sustained virological response (SVR). Nevertheless, the majority of patients globally are unable to access these treatments because of cost and infrastructure constraints and, thus, remain untreated and uncured. DATA SOURCE: Relevant articles of peginterferon (PegIFN)-based treatments in HCV and sofosbuvir-based treatments, simeprevir, daclatasvir/asunaprevir, ritonavir-boosted paritaprevir/ombitasvir/dasabuvir, and grazoprevir/elbasvir, were searched in PubMed database, including general population and special population. RESULTS: PegIFN in combination with ribavirin remains an important and relevant option for some patients, achieving SVR rates of up to 79% in genotype 1 and 89% in genotype 2 or 3 infections, which increases for patients with favorable IL28B genotypes. Triple therapy of DAA plus PegIFN/ribavirin is effective in treating difficult-to-cure patients infected with HCV genotype 3 or with resistance-associated variants. Owing to its long history in HCV management, the efficacy, tolerability and long-term outcomes associated with PegIFN alfa-2a are well established and have been validated in large-scale studies and in clinical practice for many populations. Furthermore, emerging data show that IFN-induced SVR is associated with lower incidences of hepatocellular carcinoma compared with DAAs. On the contrary, novel DAAs have yet to be studied in special populations, and long-term outcomes, particularly tumor development and recurrence in patients with cirrhosis and/or hepatocellular carcinoma, and reactivation of HBV in dually infected patients, are still unclear. CONCLUSION: In this interferon-free era, PegIFN-based regimens remain a safe and effective option for selected HCV patients. 展开更多

BACKGROUND Prompt and effective cardiopulmonary resuscitation(CPR)can promote the recovery of spontaneous circulation to some extent and can save patients’lives.The minimum target of cardiac resuscitation is the restoration of spontaneous circulation(ROSC).However,owing to prolonged sudden cardiac arrest,there is relatively high mortality within 24 h after cardiac resuscitation.Moreover,severe cerebral anoxia can deteriorate the prognosis of patients.Therefore,it is important to adopt an effective clinical evaluation of acute myocardial infarct(AMI)patients’prognosis after cardiac resuscitation for the purpose of prevention and management.AIM To investigate early CPR effects on human myeloperoxidase(MPO),soluble ST2(sST2),and hypersensitive C-reactive protein(hs-CRP)levels in AMI patients.METHODS In total,54 patients with cardiac arrest caused by AMI in our hospital were selected as the observation group,and 50 other patients with AMI were selected as the control group.The differences in serum levels of MPO,sST2,and hs-CRP between the observation group and the control group were tested,and the differences in the serum levels of MPO,sST2,and hs-CRP in ROSC and non-ROSC patients,and in patients who died and in those who survived,were analyzed.RESULTS Serum levels of MPO,sST2,hs-CRP,lactic acid,creatine kinase isoenzyme(CKMB),and cardiac troponin I(cTnI)were significantly higher in the observation group than in the control group(P<0.05).Serum levels of MPO,sST2,hs-CRP,lactic acid,CK-MB,and cTnI in the observation group were lower after CPR than before CPR(P<0.05).In the observation group,MPO,sST2,hs-CRP,lactic acid,CK-MB,and cTnI serum levels were lower in ROSC patients than in non-ROSC patients(P<0.05).MPO,sST2,hs-CRP,and lactic acid serum levels of patients who died in the observation group were higher than those of patients who survived(P<0.05).The areas under receiver operating characteristic curve predicted by MPO,sST2,hs-CRP,lactic acid,CK-MB,and cTnI were 0.616,0.681,0.705,0.704,0.702,and 0.656,respectively(P<0.05).The areas under receiver operating characteristic curve for MPO,SST2,hs-CRP,and lactic acid to predict death were 0.724,0.800,0.689,and 0.691,respectively(P<0.05).Logistic regression analysis showed that MPO,sST2,and hs-CRP were the influencing factors of ROSC[odds ratios=1.667,1.589,and 1.409,P<0.05],while MPO,sST2,hs-CRP,and lactic acid were the influencing factors of death(odds ratios=1.624,1.525,1.451,and 1.365,P<0.05).CONCLUSION Serum levels of MPO,sST2,hs-CRP,and lactic acid have a certain value in predicting recovery and prognosis of patients with ROSC. 展开更多

Rectal cancer(RC)is the third most commonly diagnosed cancer and has a high risk of mortality,although overall survival rates have improved.Preoperative assessments and predictions,including risk stratification,responses to therapy,long-term clinical outcomes,and gene mutation status,are crucial to guide the optimization of personalized treatment strategies.Radiomics is a novel approach that enables the evaluation of the heterogeneity and biological behavior of tumors by quantitative extraction of features from medical imaging.As these extracted features cannot be captured by visual inspection,the field holds significant promise.Recent studies have proved the rapid development of radiomics and validated its diagnostic and predictive efficacy.Nonetheless,existing radiomics research on RC is highly heterogeneous due to challenges in workflow standardization and limitations of objective cohort conditions.Here,we present a summary of existing research based on computed tomography and magnetic resonance imaging.We highlight the most salient issues in the field of radiomics and analyze the most urgent problems that require resolution.Our review provides a cutting-edge view of the use of radiomics to detect and evaluate RC,and will benefit researchers dedicated to using this state-of-the-art technology in the era of precision medicine. 展开更多

This study investigates the long-term changes of monthly sea surface wind speeds over the China seas from 1988 to 2015. The 10-meter wind speeds products from four major global reanalysis datasets with high resolution are used: Cross-Calibrated Multi-Platform data set(CCMP), NCEP climate forecast system reanalysis data set(CFSR),ERA-interim reanalysis data set(ERA-int) and Japanese 55-year reanalysis data set(JRA55). The monthly sea surface wind speeds of four major reanalysis data sets have been investigated through comparisons with the longterm and homogeneous observation wind speeds data recorded at ten stations. The results reveal that(1) the wind speeds bias of CCMP, CFSR, ERA-int and JRA55 are 0.91 m/s, 1.22 m/s, 0.62 m/s and 0.22 m/s, respectively.The wind speeds RMSE of CCMP, CFSR, ERA-int and JRA55 are 1.38 m/s, 1.59 m/s, 1.01 m/s and 0.96 m/s,respectively;(2) JRA55 and ERA-int provides a realistic representation of monthly wind speeds, while CCMP and CFSR tend to overestimate observed wind speeds. And all the four data sets tend to underestimate observed wind speeds in Bohai Sea and Yellow Sea;(3) Comparing the annual wind speeds trends between observation and the four data sets at ten stations for 1988-1997, 1988–2007 and 1988–2015, the result show that ERA-int is superior to represent homogeneity monthly wind speeds over the China seaes. 展开更多

African swine fever(ASF)is an acute,highly contagious and deadly viral disease in swine that jeopardizes the worldwide pig industry.Unfortunately,there are no authoritative vaccine and antiviral drug available for ASF control.African swine fever virus(ASFV)is the etiological agent of ASF.Among the ASFV proteins,p72 is the most abundant component in the virions and thus a potential target for anti-ASFV drug design.Here,we con-structed a luciferase reporter system driven by the promoter of p72,which is transcribed by the co-transfected ASFV RNA polymerase complex.Using this system,we screened over 3200 natural product compounds and obtained three potent candidates against ASFV.We further evaluated the anti-ASFV effects and proved that among the three candidates,ailanthone(AIL)inhibits the replication of ASFV at the nanomolar concentration(IC_(50)=15 nmol/L).Our in vitro experiments indicated that the antiviral effect of AIL is associated with its inhibition of the HSP90-p23 cochaperone.Finally,we showed the antiviral activity of AIL on Zika virus and hepatitis B virus(HBV),which supports that AIL is a potential broad-spectrum antiviral agent. 展开更多

AIM:To identify the association of the vitreoretinal surgeons'experience with the time interval between parsplana vitrectomy(PPV)and cataract extraction(CE).METHODS:Eyes with prior PPV and following CE were included in this retrospective cohort study.The years of practice and the annual case volume were used to describe the surgeons'experience.Multivariable linear regression analysis was used to investigate the relationship between surgeons'experience and the time interval adjusted for the patients age,gender,intraocular tamponade,and case complexity.RESULTS:Of 132430 eyes,1445 eyes were included in this study.In multivariable linear regression analysis,cases performed by surgeons with>20 practice years had longer time intervals compared with surgeons with<10 practice years after adjusted for other variables(β=0.329,95%CI:0.113 to 0.549,P=0.003).No difference in time interval was detected for comparing the lowest with the highest volume groups(β=0.089,95%CI:-0.164 to 0.343,P=0.343).The surgeons'practice years were not directly with the volume.For complicated surgery,the higherpractice-year surgeons had longer time interval than lowerpractice-year surgeons.CONCLUSION:The time intervals from PPV to CE is longer in higher-practice-year surgeons.The surgeons'practice years may have a greater effect on the time interval than annual case volume in high-complexity cases.Matching the complexity of vitreoretinal diseases with the surgeons'practice year should be considered. 展开更多

BACKGROUND The obesity rate of adolescents is gradually increasing,which seriously affects their mental health,and sleep plays an important role in adolescent obesity.AIM To investigate the relationship between sleep rhythm and obesity among adolescents and further explores the interactive effect of sleep rhythm and gender on adolescent obesity,providing a theoretical basis for developing interventions for adolescent obesity.METHODS Research data source Tianjin Mental Health Promotion Program for Students.From April to June 2022,this study selected 14201 students from 13 middle schools in a certain district of Tianjin as the research subject using the convenient cluster sampling method.Among these students,13374 accepted and completed the survey,with an effective rate of 94.2%.The demographic data and basic information of adolescents,such as height and weight,were collected through a general situation questionnaire.The sleep rhythm of adolescents was evaluated using the reduced version of the morningness-eveningness questionnaire.RESULTS A total of 13374 participants(6629 females,accounting for 49.56%;the average age is 15.21±1.433 years)were analyzed.Among them,the survey showed that 2942 adolescent were obesity,accounting for 22%and 2104 adolescent were overweight,accounting for 15.7%.Among them,1692 male adolescents are obese,with an obesity rate of 25.1%,higher than 18.9%of female adolescents.There is a statistically significant difference between the three groups(χ2=231.522,P<0.000).The obesity group has the smallest age(14.94±1.442 years),and there is a statistical difference in age among the three groups(F=69.996,P<0.000).Obesity rates are higher among individuals who are not-only-child,have residential experience within six months,have family economic poverty,and have evening-type sleep(P<0.05).Logistic regression analysis shows a correlation between sleep rhythm and adolescent obesity.Evening-type sleep rhythm can increase the risk of obesity in male adolescents[1.250(1.067-1.468)],but the effect on female obesity is not remarkable.Further logistic regression analysis in the overall population demonstrates that the interaction between evening-type sleep rhythm and the male gender poses a risk of adolescent obesity[1.122(1.043-1.208)].CONCLUSION Among adolescents,the incidence of obesity in males is higher than in females.Evening-type sleep rhythm plays an important role in male obesity but has no significant effect on female obesity.Progressive analysis suggests an interactive effect of sleep rhythm and gender on adolescent obesity,and the combination of evening-type sleep and the male gender promotes the development of adolescent obesity.In formulating precautions against adolescent obesity,obesity in male adolescents with evening-type sleep should be a critical concern. 展开更多

However, there is increasing evidence to indicate that arousal may be more relevant at some stages of affective processing. The present study employed a combined priming and cue-target paradigm and examined event-related potentials (ERPs) in order to explore the effects of emotional conflict of arousal on attention allocation. The background context of arousal was manipulated (using photos of facial expressions) while subjects performed a cognitive task in which a central arrow cue indicated the location of a peripheric target square, and a response was made according to the direction of the square. There was no main effect between incongruent emotion and congruent emotion in the response time, which means the resolution of the emotional conflict facilitate the participant behavioral response. There was a main effect of different emotional states on ERP component. In the present study, incongruent arousal also triggered greater positive potential compared with congruent arousal. 展开更多

Simultaneous and quantitative detection of multiple exosomal micro RNAs(miRNAs)was successfully performed by a surface-enhanced Raman scattering(SERS)assay consisting of Raman probes and capture probes.In this design,the asymmetric core-shell structured Au@Au@Ag nanoparticles were first synthesized by layer-by-layer self-assembly method and modified with different Raman molecules and recognition sequences(poly A-DNA)to prepare the surface-enhanced Raman probes.Then,the streptavidinmodified magnetic beads were used to immobilize the biotinylated DNA capture sequences(biotin-DNA)to obtain capture probes.In the presence of target exosomal miRNAs,the Raman probes and capture probes could bind to the target exosomal miRNAs in the partial hybridization manner.Thus,the developed SERS sensor could indicate the target miRNAs levels in the buffer solution.Using breast cancerrelated miRNAs as model targets,the limits of detection of this sensor were determined to be 1.076 fmol/L for synthetic miR-21,0.068 fmol/L for synthetic miR-126,and 4.57 fmol/L for synthetic miR-1246,respectively.Such SERS sensors were further employed to detect the miR-21 in 20%human serum and the extraction solution of exosomes,respectively.Therefore,simultaneous and multiplex detection of cancerrelated exosomal miRNAs by this assay could provide new opportunities for further biomedical applications. 展开更多

By using swap test,a quantum private comparison(QPC)protocol of arbitrary single qubit states with a semi-honest third party is proposed.The semi-honest third party(TP)is required to help two participants perform the comparison.She can record intermediate results and do some calculations in the whole process of the protocol execution,but she cannot conspire with any of participants.In the process of comparison,the TP cannot get two participants’private information except the comparison results.According to the security analysis,the proposed protocol can resist both outsider attacks and participants’attacks.Compared with the existing QPC protocols,the proposed one does not require any entanglement swapping technology,but it can compare two participants’qubits by performing swap test,which is easier to implement with current technology.Meanwhile,the proposed protocol can compare secret integers.It encodes secret integers into the amplitude of quantum state rather than transfer them as binary representations,and the encoded quantum state is compared by performing the swap test.Additionally,the proposed QPC protocol is extended to the QPC of arbitrary single qubit states by using multi-qubit swap test. 展开更多

Zinc-finger proteins play different roles in cancer;however,the function of zinc-finger protein ZNF575 in cancer remains unclear.In the present study,we aimed to determine the function and expression of ZNF575 in colorectal cancer.Proliferation assay,colony formation assay,and tumor model in mice were used to investigate the function of ZNF575 after ectopic expression of ZNF575 in colorectal cancer(CRC)cells.RNA sequencing,ChIP,and luciferase assays were used to investigate the mechanism behind ZNF575 regulation of CRC cell growth.The expression of ZNF575 was determined by IHC staining in 150 pairs of malignant CRC tissues,followed by prognosis analysis.We indicated that ectopic expression of ZNF575 inhibited CRC cell proliferation,colony formation and promoted cell apoptosis in vitro.Tumor growth in CRC was also impaired by ZNF575 in mice.RNA sequencing,follow-up western blotting,and qPCR results demonstrated the increase of p53,BAK,and PUMA in ZNF575-expressing CRC cells.Further results indicated that ZNF575 directly targeted the p53 promoter and promoted the transcription of p53.Downregulation of ZNF575 was confirmed in malignant tissues,and ZNF575 expression was positively correlated with the prognosis of CRC patients.The present study demonstrated the function,underlying mechanism,expression,and the prognosis-predicting role of ZNF575 in CRC,which indicated that ZNF575 would be a potential prognostic predictor and therapeutic target for CRC and other cancers. 展开更多

However, until recently more and more studies show the convincing evidence that it most probably originates from tubal cells. It has a close relationship with the development of serous tumors, especially low-grade serous carcinoma. The lesion does cause symptoms and signs, such as chronic abdominal pain or tumor-like mass, though it is often found accidently for other gynecologic problems. Occasionally atypical endosalpingiosis needs to be differentiated from a malignancy when it appears in an unusual site with worrying morphologic presentations or under some special circumstances. Owing to the facts that it may evolve quietly and continuously towards serous tumors, and that the lesions at different sites may evolve independently, the outcomes of its evolution can eventually kill the patient. Therefore, a proper recognition of the lesion will translate it into an adequate care for those patients. This review summarizes the most recent findings and makes thoughtful comments. 展开更多
