Prevalence of fumonisin producing Fusarium verticillioides associated with cereals grown in Karnataka(India)

total of 135 cereal samples were collected from different districts of Karnataka state,India in which 69 samples were infected with Fusarium species.Amongthese 51 samples were having Fusarium verticillioides infection... total of 135 cereal samples were collected from different districts of Karnataka state,India in which 69 samples were infected with Fusarium species.Amongthese 51 samples were having Fusarium verticillioides infection and among them 42 samples were positive for fumonisin production.Per cent incidence and frequency were high in maize samples with 33.12%and 47.54%,respectively followed by paddy and sorghum,while pearl millet was free from F.verticillioides infection.Relative density of F.verticillioides association was 59.50%among the screened samples.A total of 326 Fusarium species were isolated by screening 135 cereal samples and among these 194 isolates of F.verticillioides scored positive for VERTF-1 and VERTR species-specific pair of primers.Further amplification with VERTF-1 and VERTF-2 pair of primers recorded 176 isolates of fumonisin producingF.verticillioides.The study revealed high incidence,frequency and relative density of fumonisin producingF.verticillioides and production of fumonisins in cereals.Itwas amplified using one forward and two reverse primers to discriminate fumonisin producing from fumonisin non-producing F.verticillioides which stresses the need for the development of managemental stratergies before they enter into the food chain. 展开更多

Mycotoxigenic fungi and their mycotoxins epitomize broad food spoilage and food safety problems worldwide.Fusarium contamination of foods and feeds has raised a concern associated with severe health issues causing economic burden.Fusarium equiseti species stand out among the group with lethal toxicity which can contaminate the cereal crops via trichothecenes production.A well-known food fermenting Lactic acid bacterium(LAB)has gained attention over the years for its anti-fungal and anti-mycotoxigenic capabilities.In the current study,20 LABs isolated from traditional herbal Amla juice were screened to determine the inhibition of the Fusarium equiseti.Among them,Limosilactobacillus fermentum MYSAGAM1 showed potent anti-fungal activity by exhibiting the morphological modifications of damaged mycelia and deformed conidia in Fusarium equiseti.The cell-free supernatant(CFS)from L.fermentum reduced mycelial biomass to 0.135 g compared to 2.464 g in control.The conidial germination was inhibited at 23.22%by using 20%CFS.The fungal growth inhibition was also observed in maize kernels over the control after the specified 7 days of incubation with CFS of MYSAGAM1.The inhibitory efficacy was displayed due to the presence of organic acids,which were analyzed through CFS characterization via LC-MS.Additionally,the strain MYSAGAM1 showed promising probiotic properties by tolerating acidic pH-2,exhibiting a survival rate of 73.50%at 2 h,34.18%at 4 h,bile tolerance of 77.52%at 2 h,and 80.76%at 4 h as well as the isolate also tolerated different concentration of NaCl and phenol.Further,the isolate was capable of colonizing intestinal epithelium cells with a strong adhesion of 44.77%for 5 h on the mucosal surface.Overall,the results conclude that Limosilactobacillus fermentum MYSAGAM1 is a potent probiotic organism with anti-fungal properties. 展开更多

This study investigated the antifungal activities of probiotic bacteria isolated from traditional herbal juice, which was a blend of Tulsi, Mint, Bael and Indian borage leaves. About 20 isolates were obtained from the herbal juice sample;these isolates were initially screened against fungal pathogens such as Aspergillus parasiticus , Aspergillus flavus , Aspergillus ochraceus , Aspergillus carbonarius and Aspergillus fumigatus . Of the 20 isolates obtained, MYSAGT3 and MYSAGT4 exhibited potential antifungal activity against the tested fungal pathogens. Moreover, MYSAGT3 and MYSAGT4 was found to survive acidic pH and bile conditions, but were susceptible to tested antibiotics. Both the isolates exhibited 40–60% of autoaggregation ability, >90% of cell surface hydrophobicity and tolerance to 0.4–0.6% phenol. The 16S rRNA sequencing results identified the isolates MYSAGT3 and MYSAGT4 as Lactiplantibacillus plantarum. The MYSAGT3 exhibited potent antagonistic and probiotic activity compared to MYSAGT4;hence MYSAGT3 was further used for the control of Aspergillus ochraceus. Initial antifungal activity of MYSAGT3 inhibited about 76.67% growth of Aspergillus ochraceus comparatively higher than other Aspergillus sp. tested. Co-inoculation and microdilution assay revealed the high inhibitory activity of Lactiplantibacillus plantarum MYSAGT3. Further, cell free supernatant (CFS) of Lactiplantibacillus plantarum MYSAGT3 reduced the growth of fungal hyphae and conidial germination of Aspergillus ochraceus in conidial germination assay and bio preservative activity on peanuts. The CFS characterization and Liquid chromatography-mass spectroscopy (LCMS) results showed that, the antifungal activity was due to different organic acids present in the CFS of MYSAGT3. MYSAGT3 and its CFS also reduced the growth of Aspergillus ochraceus in Poultry feeds and about 21.80% of ochratoxin A (OTA) degradation was attained after 24 h of incubation with MYSAGT3. The isolated Lactic acid bacteria are able to inhibit Aspergillus ochraceus and degrade OTA in-vitro . Hence, the results suggest Lactiplantibacillus plantarum MYSAGT3 as an efficient probiotic organism and possess antifungal properties. 展开更多
