The duration of incubation varied considerably among the six temperature treatments, whereas hatching success did not. The mean incubation length decreased as temperature increased in a nonlinear way, and increased as the thermal variance increased. Incubation temperature affected the body size (linear length and mass) and shape of hatchlings, with eggs incubated at 26, 28, and 30 ℃ producing larger and heavier hatchlings than did those incubated at 24 ℃, 32 ℃, or fluctuating temperatures. Our results showed that exposure of P. chinensis eggs to extreme temperatures for brief periods of time did not increase embryonic mortality and, in the fluctuating-temperature treatment, the thermal variance affected hatchling morphology more evidently than the thermal mean. Our results highlight the importance of the thermal variance in affecting embryonic development and hatchling morphology, and add further evidence that temperatures within the range of 26-30 ℃ are optimal for P. chinensis embryos.展开更多
The species diversity and altitudinal distribution of amphibians along an eleva tional gradient of 200-1600 m in the Xianxialing and Wuyishan Mountain Ranges in Southeastern China were investiga ted through time-const...The species diversity and altitudinal distribution of amphibians along an eleva tional gradient of 200-1600 m in the Xianxialing and Wuyishan Mountain Ranges in Southeastern China were investiga ted through time-constra ined visual surveys along 32 transect lines in 9 survey areas,in which the ha bitat types were also recorded.A total of 27 amphibian species belonging to 19 genera,7 families,and 2 orders were found.The species diversity of the amphibians plateaued at low elevation,and the altitudinal boundary of their distribution was at 800 m.Their species compositions were dissimilar in the two mountain ranges probably beca use the annual average temperature and annual rainfall were different in both areas.The eleva tional Rapoport's rule demonstrated that the species range size of the amphibians expanded as the elevation increased in both mountain ranges.The results of the cross species method supported the rule only when the influence of the low-frequency occurrence proba bility of an investigated species was excluded,whereas those of the Steven's method strongly corroborated the rule rega rdless of the incidental occurrence or absence of the species.展开更多

Age-related variation in electrophoretic profiles was found in both species and proteolytic and fibrinogenolytic activity was higher in neonate than adult venoms. Neonate D. acutus venom had higher 5' nucleotidase, PLA2, hyaluronidase and gelatinolytie activity, but lower esterolytic activity, than adult venom. Neonate and adult D. acutus venoms showed identical phosphomonoesterase, LAO and fibrinolytic activities. Neonate N. atra venom had higher phosphomonoesterase and LAO activity, but lower 5' nucleotidase, PLA2, hyaluronidase and Ache activities than adult venom. Neonate and adult N. atra venoms showed similar gelatinolytic activity. Further, age-dependent immunoreactivity was found in both species, and cross-reactions between homologous venoms and antiserums were closely related to venom composition. The Chinese cobra(Naja atra) is a large sized, venom...Detailed information on venom yield is helpful in preparing antivenoms and treating snakebites, but such information is lacking for many species of venomous snakes. The Chinese cobra(Naja atra) is a large sized, venomous snake commonly found in southeastern China, where it causes a heavy burden of snakebites. To examine the effects of various factors(morphology, sex, age, season, and geographical origin) on the venom yield in this snake, we collected venom samples of 446 individuals(426 adults and 20 neonates) from 10 populations of N. atra over an eightyear period. We used two variables, lyophilized venom mass(venom yield) and solid content of venom(% solids), to quantify the venom yield. We used linear regression analysis to check if venom yield was related to morphological factors, one-way ANOVA and one-way ANCOVA to detect the sexual, ontogenetic, and geographic variation in venom yield, and repeated-measures ANOVA to examine seasonal shifts in venom yield. Our results indicate that venom yield of N. atra is positively related to the morphological traits examined, with male snakes expelling more venom than females. Venom yield in N. atra was age-related, with elder snakes always expelling more venom than younger ones. Geographic variation in venom yield was also observed, while seasonal variation was not. The solid content of venom was lower in males than in females, but this was not related to morphology, season, age, or geography. Our findings suggest that venom yield in N. atra is influenced by multiple factors, as well as by the interactions among these factors.展开更多

Using GIS-based and phylogenetic comparative methods, we investigated global patterns of variation in climatic niche breadth in lacertid lizards to test the following three hypotheses about climatic niche widths. First, does a species' temperature or precipitation niche breadth relate to its temperature or precipitation niche position(the mean value of annual mean temperature or annual precipitation across sampled localities in the range of each species)? Second, are there trade-offs between a species' temperature niche breadth and precipitation niche breadth? Third, does a species' temperature or precipitation niche breadth relate to altitude or latitude? We expect that:(1) species distributed in cold regions are specialized for low-temperature environments(i.e. narrow niche breadth center around low temperatures);(2) a negative relationship between species niche breadth on temperature and precipitation axes according to the tradeoff hypothesis(i.e. species that tolerate a broad range of precipitation regimes cannot also tolerate a broad range of temperatures);(3) precipitation niche breadth decreases with altitude or latitude, whereas temperature climatic niche breadth increases with altitude or latitude. In this study, we measured samples of unha...Changes in structure and composition of the eggshell resulting from embryonic mobilization of minerals from the eggshell are found in all oviparous reptiles studied thus far. In this study, we measured samples of unhatched and hatched eggshells of the Chinese cobra Naja atra to determine the percentage of ash and the phase composition of calcium carbonate. The mean percentage of ash was significantly higher in unhatched eggshells (24.6%) than in hatched eggshells (22.3%). The dominant phase in unhatched eggshells was the calcite form of calcium carbonate. In addition to the peaks of calcite, a few small peaks were found to be caused by the aragonite and vaterite phases of calcium carbonate, implying that there are small amounts of aragonite and vaterite in the eggshell. The concentration of the various phases calculated from the intensity of the X-ray diffraction spectra allowed the estimation that percentages of calcite, aragonite and vaterite were about 92%, 4% and 4%, respectively. Hatched eggshells produced similar spectral characteristics as unhatched eggshells, with one exception. The dominant phase composition in the hatched eggshell was also calcite, but the amount of the aragonite phase had a marked increase. Our study adds evidence that embryonic mobilization of minerals from the eggshell may result in changes in structure of the eggshell.展开更多

The mitogenome has ...We describe the complete mitochondrial genome of a small-scaled forest agamid (Pseudocalotes microlepis), which is 17 873 bp in size, containing 13 PCGs, 2 rRNAs, 22 tRNAs and non-coding regions. The mitogenome has a typical gene order among squamates. 13 PCGs include 2 start codons (ATG and ATA), 3 stop codons (TAG, TAA and AGG), and an incomplete stop codon (T-). Codon usage analyses showed that CUA-Leul and CGG-Arg are the most frequently and rarely used codon, respectively. All 22 tRNAs were predicted to form canonical cloverleaf secondary structures, except for two tRNAs (tRNAcys and tRNASer (AGY)) lacking the dihydorouridine (DHU) arm. The large non- coding region (control region) is 2 687 bp long (28.3% T, 18.2% C, 42.3% A, and 11.2% G), with four different types of repeating sequences. The phylogenetic tree resulting from BEAST analyses based on concatenated 2 rRNAs and 13 PCGs in sequence revealed that the newly sequenced P. microlepis, where the genus Acanthosaura were aggregated. Together with Calotes versicolor, they constitute the subfamily Draconinae.展开更多

MHC genes show the most prominent genetic diversity in vertebrates,reflecting the adaptation of populations to their evolving environment, population survival and reproduction. In the present study, we used nextgeneration sequencing(NGS) to study the loci polymorphism of exon 3 of the MHC class Ⅰ genes in an ovoviviparous skink, the many-lined sun skink,Eutropis multifasciata and five other species of Scincidae, to quantify genetic variation. In addition,we genotyped the same MHC class Ⅰ genes of E.multifasciata using clone sequencing, to directly compare the effectiveness of both analytical techniques for MHC genotyping. NGS detected 20MHC class Ⅰ alleles in E. multifasciata, and 2 to 15 alleles in the other five Scincidae species. However,clone sequencing detected only 15 of those MHC class Ⅰ alleles in E. multifasciata. In addition, transspecies polymorphism of MHC class Ⅰ genes was studied by constructing a phylogenetic tree using the gene sequences obtained by NGS. Phylogenetic analysis revealed that MHC class I alleles were shared among different species of Scincidae with trans-species polymorphism, and did not exhibit specific genealogical inheritance. These results have important implications for understanding polymorphism interspecies diversity in the MHC genes of Scincidae, and the evolution of the MHC more broadly.展开更多

Forty-tw...Body size and female reproduction in the water adult females larger than 500 mm SVL and 32 adult males Ledong, Hainan under permit to our laboratory in Hainan monitor lizard (Varanus salvator) were studied. Forty-two larger than 400 mm SVL were donated by local people in in 2013 and 2014. The largest male and female measured 745 and 755 mm SVL, respectively. The mean SVL was greater in adult females than in adult males. Males had larger heads (head width) than females of the same SVL. The smallest reproductive female in our sample was 565 mm SVL. Females produced a single clutch of 17.1 (10-23) pliable-shelled eggs per breeding season stretching from mid-June and mid-September. Clutch size and clutch mass were all positively related to female SVL. However, there was no significant linear relationship between egg mass and female SVL. Larger females generally produced more eggs, and thus heavier clutches than did smaller ones. There was no significant linear relationship between relative clutch mass and female SVL. Phylogenetic generalized least squares (PGLS) analysis, accounting for phylogenetic relationships, showed that clutch size was positively correlated with mean maternal SVL in varanid lizards. PGLS analysis showed that phylogenetic relationships did not affect clutch (or/and egg) mass and the SVL although there were significant linear relationship between clutch (or/and egg) mass and mean maternal SVL. Therefore, we could draw some general conclusions about the body size and reproductive tactics in varanid lizards that larger females generally produced more eggs, larger eggs and thus heavier clutches than did smaller ones.展开更多
We used mitochondrial cytochrome b and ND4 genes and 9 microsatellite loci to determine genetic diversity,population structure,evolutionary history,and migration patterns within the Reeves’butterfly lizard Leiolepis ...We used mitochondrial cytochrome b and ND4 genes and 9 microsatellite loci to determine genetic diversity,population structure,evolutionary history,and migration patterns within the Reeves’butterfly lizard Leiolepis reevesii(Agamidae).Considering molecular-based phylogeographical lineages,we then performed niche equivalency and similarity tests between divergent lineages.Phylogenetic analyses based on mitochondrial DNA(mtDNA)data revealed 2 lineages(A and B)diverging≈0.84 million years ago and,respectively,restricted to the northern and southern portions of the Wuzhishan and Yinggeling mountain ranges.Lineage B contains individuals from southern Hainan;Lineage A includes individuals from all other localities and can be further divided into 3 clusters according to microsatellite data.The null hypothesis that the 2 lineages shared identical niches was rejected in all niche equivalency tests,indicating niche shifts during genetic divergence.Similarity tests provided evidence of niche conservatism,suggesting that the 2 lineages share more characteristics of their niche spaces than randomly expected.The niche similarity and equivalency tests indicated a complex niche pattern in which both lineages share a main portion of their ecological spaces.The climatic niche of Lineage B represented a marginal and specialized fraction of the entire ecological space of the climatic niche of Lineage A,with warmer conditions.Isolation caused by orogenesis and subsequent niche divergence,together with local adaptation,may have led to genetic differentiation and further lineage sorting in L.reevesii.展开更多

文摘We describe the complete mitochondrial genome of a small-scaled forest agamid (Pseudocalotes microlepis), which is 17 873 bp in size, containing 13 PCGs, 2 rRNAs, 22 tRNAs and non-coding regions. The mitogenome has a typical gene order among squamates. 13 PCGs include 2 start codons (ATG and ATA), 3 stop codons (TAG, TAA and AGG), and an incomplete stop codon (T-). Codon usage analyses showed that CUA-Leul and CGG-Arg are the most frequently and rarely used codon, respectively. All 22 tRNAs were predicted to form canonical cloverleaf secondary structures, except for two tRNAs (tRNAcys and tRNASer (AGY)) lacking the dihydorouridine (DHU) arm. The large non- coding region (control region) is 2 687 bp long (28.3% T, 18.2% C, 42.3% A, and 11.2% G), with four different types of repeating sequences. The phylogenetic tree resulting from BEAST analyses based on concatenated 2 rRNAs and 13 PCGs in sequence revealed that the newly sequenced P. microlepis, where the genus Acanthosaura were aggregated. Together with Calotes versicolor, they constitute the subfamily Draconinae.

文摘The genes of the major histocompatibility complex(MHC) encode cell surface proteins that are essential for adaptive immunity. MHC genes show the most prominent genetic diversity in vertebrates,reflecting the adaptation of populations to their evolving environment, population survival and reproduction. In the present study, we used nextgeneration sequencing(NGS) to study the loci polymorphism of exon 3 of the MHC class Ⅰ genes in an ovoviviparous skink, the many-lined sun skink,Eutropis multifasciata and five other species of Scincidae, to quantify genetic variation. In addition,we genotyped the same MHC class Ⅰ genes of E.multifasciata using clone sequencing, to directly compare the effectiveness of both analytical techniques for MHC genotyping. NGS detected 20MHC class Ⅰ alleles in E. multifasciata, and 2 to 15 alleles in the other five Scincidae species. However,clone sequencing detected only 15 of those MHC class Ⅰ alleles in E. multifasciata. In addition, transspecies polymorphism of MHC class Ⅰ genes was studied by constructing a phylogenetic tree using the gene sequences obtained by NGS. Phylogenetic analysis revealed that MHC class I alleles were shared among different species of Scincidae with trans-species polymorphism, and did not exhibit specific genealogical inheritance. These results have important implications for understanding polymorphism interspecies diversity in the MHC genes of Scincidae, and the evolution of the MHC more broadly.

文摘Body size and female reproduction in the water adult females larger than 500 mm SVL and 32 adult males Ledong, Hainan under permit to our laboratory in Hainan monitor lizard (Varanus salvator) were studied. Forty-two larger than 400 mm SVL were donated by local people in in 2013 and 2014. The largest male and female measured 745 and 755 mm SVL, respectively. The mean SVL was greater in adult females than in adult males. Males had larger heads (head width) than females of the same SVL. The smallest reproductive female in our sample was 565 mm SVL. Females produced a single clutch of 17.1 (10-23) pliable-shelled eggs per breeding season stretching from mid-June and mid-September. Clutch size and clutch mass were all positively related to female SVL. However, there was no significant linear relationship between egg mass and female SVL. Larger females generally produced more eggs, and thus heavier clutches than did smaller ones. There was no significant linear relationship between relative clutch mass and female SVL. Phylogenetic generalized least squares (PGLS) analysis, accounting for phylogenetic relationships, showed that clutch size was positively correlated with mean maternal SVL in varanid lizards. PGLS analysis showed that phylogenetic relationships did not affect clutch (or/and egg) mass and the SVL although there were significant linear relationship between clutch (or/and egg) mass and mean maternal SVL. Therefore, we could draw some general conclusions about the body size and reproductive tactics in varanid lizards that larger females generally produced more eggs, larger eggs and thus heavier clutches than did smaller ones.

文摘We used mitochondrial cytochrome b and ND4 genes and 9 microsatellite loci to determine genetic diversity,population structure,evolutionary history,and migration patterns within the Reeves’butterfly lizard Leiolepis reevesii(Agamidae).Considering molecular-based phylogeographical lineages,we then performed niche equivalency and similarity tests between divergent lineages.Phylogenetic analyses based on mitochondrial DNA(mtDNA)data revealed 2 lineages(A and B)diverging≈0.84 million years ago and,respectively,restricted to the northern and southern portions of the Wuzhishan and Yinggeling mountain ranges.Lineage B contains individuals from southern Hainan;Lineage A includes individuals from all other localities and can be further divided into 3 clusters according to microsatellite data.The null hypothesis that the 2 lineages shared identical niches was rejected in all niche equivalency tests,indicating niche shifts during genetic divergence.Similarity tests provided evidence of niche conservatism,suggesting that the 2 lineages share more characteristics of their niche spaces than randomly expected.The niche similarity and equivalency tests indicated a complex niche pattern in which both lineages share a main portion of their ecological spaces.The climatic niche of Lineage B represented a marginal and specialized fraction of the entire ecological space of the climatic niche of Lineage A,with warmer conditions.Isolation caused by orogenesis and subsequent niche divergence,together with local adaptation,may have led to genetic differentiation and further lineage sorting in L.reevesii.
