Coreopsis tinctoria Nutt.,an edible flowering plant,belongs to the Chrysanthemum family and is mainly grown at high altitudes in Northwestern China.It is rich in polyphenolic compounds,particularly marein and flavomar...Coreopsis tinctoria Nutt.,an edible flowering plant,belongs to the Chrysanthemum family and is mainly grown at high altitudes in Northwestern China.It is rich in polyphenolic compounds,particularly marein and flavomarein,and possesses multiple health-promoting properties,such as antioxidant,hypoglycemic and vasorelaxant effects.Previous bioactivity investigations majorly focused on C.tinctoria and its crude extract.The aim of the present study was to prepare marein-dominant C.tinctoria flavonoids(CF),further investigate the CF protective effects of liver fibrosis induced by carbon tetrachloride and elucidate the associated molecular mechanisms.Results have demonstrated that CF effectively attenuated hepatofibrogenesis by increasing the activity of glutathione(GSH)and superoxide dismutase(SOD);suppressing the hepatic stellate cell(HSC)activation,inhibiting transforming growth factorβ(TGF-β)activation and the production ofα-smooth muscle actin(α-SMA),alleviating the phosphorylation of extracellular signal-regulated kinases1/2(ERK1/2)and small mothers against decapentaplegic1/2(Smad1/2),thus maintaining the collagen metabolic homeostasis in the liver.Our study revealed that CF possesses an efficacious protective effect against chronic hepatic fibrosis due to their strong inhibitory effects of oxidative stress and chronic inflammation.展开更多

Systematic bias is a type of model error that can affect the accuracy of data assimilation and forecasting that must be addressed.An online bias correction scheme called the sequential bias correction scheme(SBCS),was...Systematic bias is a type of model error that can affect the accuracy of data assimilation and forecasting that must be addressed.An online bias correction scheme called the sequential bias correction scheme(SBCS),was developed using the6 h average bias to correct the systematic bias during model integration.The primary purpose of this study is to investigate the impact of the SBCS in the high-resolution China Meteorological Administration Meso-scale(CMA-MESO)numerical weather prediction(NWP)model to reduce the systematic bias and to improve the data assimilation and forecast results through this method.The SBCS is improved upon and applied to the CMA-MESO 3-km model in this study.Four-week sequential data assimilation and forecast experiments,driven by rapid update and cycling(RUC),were conducted for the period from 2–29 May 2022.In terms of the characteristics of systematic bias,both the background and analysis show diurnal bias,and these large biases are affected by complex underlying surfaces(e.g.,oceans,coasts,and mountains).After the application of the SBCS,the results of the data assimilation show that the SBCS can reduce the systematic bias of the background and yield a neutral to slightly positive result for the analysis fields.In addition,the SBCS can reduce forecast errors and improve forecast results,especially for surface variables.The above results indicate that this scheme has good prospects for high-resolution regional NWP models.展开更多

This study investigated the electrophysiological characteristics of amphibian olfactory bulb in the period of hibernation and after hibernation and its effects on the start of hibernation and spontaneous awakening. The research showed four forms of spontaneous firings: single spontaneous firing, burst spontaneous firing, irregular spontaneous firing and consecutive single spontaneous firing. The single spontaneous firing includes slow depolarized spontaneous firing and fast depolarized spontaneous firing, and the slow depolarized spontaneous firing occurs only during the hibernation period. In hibernation, the low amplitude and low frequency firing with a longer duration may be relevant to maintaining the tonicity of the central nervous system in toads that are in hibernation, and this kind of firing may also provide an excited basis for their arousal from hibernation. After hibernation, the amplitude and frequency of firing increase, but the firing duration gets shorter. This form of short-term firing, which may be a phenomenon of sensory neurons fast adapting, is one of the neuronal mechanisms for the arousal of hibernating animals.展开更多
Model predictive control is widely used in the design of autonomous driving algorithms.However,its parameters are sensitive to dynamically varying driving conditions,making it difficult to be implemented into practice...Model predictive control is widely used in the design of autonomous driving algorithms.However,its parameters are sensitive to dynamically varying driving conditions,making it difficult to be implemented into practice.As a result,this study presents a self-learning algorithm based on reinforcement learning to tune a model predictive controller.Specifically,the proposed algorithm is used to extract features of dynamic traffic scenes and adjust the weight coefficients of the model predictive controller.In this method,a risk threshold model is proposed to classify the risk level of the scenes based on the scene features,and aid in the design of the reinforcement learning reward function and ultimately improve the adaptability of the model predictive controller to real-world scenarios.The proposed algorithm is compared to a pure model predictive controller in car-following case.According to the results,the proposed method enables autonomous vehicles to adjust the priority of performance indices reasonably in different scenarios according to risk variations,showing a good scenario adaptability with safety guaranteed.展开更多

Viologens known as a kind of promising negolyte materials for aqueous organic redox flow batteries,face a critical stability challenge due to the S_N2 nucleophilic attack by hydroxide ions(OH-)during the battery cycli...Viologens known as a kind of promising negolyte materials for aqueous organic redox flow batteries,face a critical stability challenge due to the S_N2 nucleophilic attack by hydroxide ions(OH-)during the battery cycling.In this work,a N-cyclic quaternary ammonium-grafted viologen molecule,viz.1,1'-bis(4,4'-dime thylpiperidiniumyl)-4,4'-bipyridinium tetrachloride((DBPPy)Cl_(4)),is developed by the molecular engineering strategy.The obtained(DBPPy)Cl_(4) molecule shows a decent solubility of 1.84 M and a redox potential of-0.52 V vs.Ag/AgCl,Experimental and theoretical results reveal that the grafted N-cyclic quaternary ammonium groups act as the steric hindrance to prevent nucleophilic attack by OH~-,increasing the alkali resistance of the electroactive molecule.The symmetrical battery with 0.50 M(DBPPy)Cl4shows negligible decay during the 13-day cycling test.As demonstration,the flow battery utilizing 1.0 M(DBPPy)Cl_(4) as the negolyte and 1-(1-oxyl-2,2',6,6'-tetramethylpiperidin-4-yl)-1'-(3-(trimethylammonio)propyl)-4,4'-bipyridinium trichloride as the posolyte exhibits a high capacity retention rate of 99.99%per cycle at 60 mA cm^(-2).展开更多
Train timetables and operations are defined by the train running time in sections,dwell time at stations,and headways between trains.Accurate estimation of these factors is essential to decision-making for train delay...Train timetables and operations are defined by the train running time in sections,dwell time at stations,and headways between trains.Accurate estimation of these factors is essential to decision-making for train delay reduction,train dispatching,and station capacity estimation.In the present study,we aim to propose a train dwell time model based on an averaging mechanism and dynamic updating to address the challenges in the train dwell time prediction problem(e.g.,dynamics over time,heavy-tailed distribution of data,and spatiotemporal relationships of factors)for real-time train dispatching.The averaging mechanism in the present study is based on multiple state-of-the-art base predictors,enabling the proposed model to integrate the advantages of the base predictors in addressing the challenges in terms of data attributes and data distributions.Then,considering the influence of passenger flow on train dwell time,we use a dynamic updating method based on exponential smoothing to improve the performance of the proposed method by considering the real-time passenger amount fluctuations(e.g.,passenger soars in peak hours or passenger plunges during regular periods).We conduct experiments with the train operation data and passenger flow data from the Chinese high-speed railway line.The results show that due to the advantages over the base predictors,the averaging mechanism can more accurately predict the dwell time at stations than its counterparts for different prediction horizons regarding predictive errors and variances.Further,the experimental results show that dynamic smoothing can significantly improve the accuracy of the proposed model during passenger amount changes,i.e.,15.4%and 15.5%corresponding to the mean absolute error and root mean square error,respectively.Based on the proposed predictor,a feature importance analysis shows that the planned dwell time and arrival delay are the two most important factors to dwell time.However,planned time has positive influences,whereas arrival delay has negative influences.展开更多
During the life cycle of a plant,seed germination is crucial.Upon ingestion of water,the dry seeds resumed energy metabolism and cellular repair.To dissect the complex mechanisms at the very beginning of seed germinat...During the life cycle of a plant,seed germination is crucial.Upon ingestion of water,the dry seeds resumed energy metabolism and cellular repair.To dissect the complex mechanisms at the very beginning of seed germination,two approaches including transcriptome and small RNA sequencing were conducted during the water imbibition process of mung bean seeds compared with dry seed.The transcriptome sequencing analysis identified 10,108 differentially expressed genes(DEGs)between dry and imbibed mung bean seeds.KEGG enrichment analyses demonstrated numerous DEGs involved in hormone signaling pathways,carbohydrate,and energy metabolism.Out of the total DEGs,129 genes were investigated to involve in abscisic acid,gibberellin,and ethylene pathways,with 56 genes involved in glycolysis and TCA cycle.Through the analysis of small RNA sequencing among different samples,there were 284 miRNAs found in the imbibed and dried seeds of mung bean containing 213 known and 71 novel miRNAs.From comparison between the two types of seeds,51 miRNAs were differentially expressed.The integrated analyses of transcriptome and miRNAome data showed that 23 DEGs are the putative target genes of 8 miRNAs including miR156,miR171b-3p,miR166e-3p,miR169-1,etc.QRT-PCR was used to validate several DEGs and miRNAs.The data suggested that the seed imbibition process is regulated by a variety of DEGs and microRNAs.Meanwhile,we hope to gain a deeper insight into how mung bean seeds germinate in order to optimize seed growth.展开更多
Multiple myeloma(MM)is one of the most common hematological malignancies and to date,it remains an incurable disease.In this study,we evaluated the inhibitory effect of 5-acetyloxy-6,7,8,4′-tetramethoxyflavone(5-ATMF...Multiple myeloma(MM)is one of the most common hematological malignancies and to date,it remains an incurable disease.In this study,we evaluated the inhibitory effect of 5-acetyloxy-6,7,8,4′-tetramethoxyflavone(5-ATMF),a compound from aged citrus peel extracts,on the MM U266 cell line.We found that the compound inhibited cell growth and induced cell apoptosis in multiple apoptotic assays.The apoptotic proteins caspase-3,caspase-9,and PARP(poly ADP-ribose polymerase)were cleaved when cells were treated with 5-ATMF.Along with the apoptosis process,the anti-apoptotic protein,Bcl-2(B-cell lymphoma-2),was significantly downregulated and the pro-apoptotic protein,Bax(Bcl-2 associated X protein),upregulated along with the release of cytochrome C(Cyt c)and the reduction of mitochondrial membrane potential(MMP).Notably,we found that the phosphorylation of Bad(Bcl-2/Bcl-XL-associated death promoter)was decreased but Bad remained unchanged in this process.On adding 5-ATMF to U266 cells,we observed that 5-ATMF repressed the phosphorylation of Akt(PKB Protein Kinase B PKB),thereby increasing the release of Cyt c and inhibiting the phosphorylation of Bad.These effects were enhanced by Ly294002,an inhibitor of Akt.These results suggested that 5-ATMF exerts a pro-apoptotic effect on U266 cells,possibly associated with the mitochondrial apoptotic pathway induced by the PI3K/Akt/Bad pathway.展开更多

XRD and FT-IR are employed to characterize its structure and show that ammonium is successfully incorporated into HT. Meanwhile, the HT still retains the typical structure of hydrotalcite-like compounds. NOx storage activity tests indicate Mg/Al/Cu-NH3·H2O HT shows the significant NOx removal activity at 156℃ with a little Oxygen existence. TGA and DTG reveal the reaction of the released ammonium with NOx and storage NOx as nitrates will be responsible for the high NOx storage activity. Additionally, the temperature programmed desorption test of NOx adsorbed sample under 175℃ proves NOx is stored successfully into Mg/Al/Cu-NH3·H2O HT further.展开更多

Pulmonary fibrosis(PF)is sexually dimorphic,with a relatively high prevalence and severity in males;however,the mechanism remains unclear.Our study revealed pronounced sexual dimorphism of immune cell genes in the lun...Pulmonary fibrosis(PF)is sexually dimorphic,with a relatively high prevalence and severity in males;however,the mechanism remains unclear.Our study revealed pronounced sexual dimorphism of immune cell genes in the lung,among which grancalcin(GCA)showed profound sex differences.GCA was produced by lung-infiltrating bone marrow macrophages triggered by heightened inflammation in the lung.However,a unique HTR2C+alveolar macrophage population enriched in female lungs metabolically reprogramed bone marrow-derived macrophages and constrained local GCA amplification.As a novel chemokine,GCA bound to protein tyrosine phosphatase receptor type T(PTPRT)in Th17 cells and facilitated pathogenic lung infiltration by activating the ROCK1-MLC pathway,thus aggravating lung fibrosis.Notably,both GCA and Th17 cells abundantly accumulated in lung biopsies from male PF patients but not in those from female patients.GCA-neutralizing antibodies in combination with pirfenidone,a prescribed medication for treating fibrosis,provided superior effectiveness and survival rates against PF compared with treatment with pirfenidone alone.Overall,our findings reveal that sex-biased lung fibrosis is shaped by lung immuneregulatory units,which could be targeted to limit lung fibrosis.展开更多

In hibernation, the low amplitude and low frequency firing with a longer duration may be relevant to maintaining the tonicity of the central nervous system in toads that are in hibernation, and this kind of firing may also provide an excited basis for their arousal from hibernation. After hibernation, the amplitude and frequency of firing increase, but the firing duration gets shorter. This form of short-term firing, which may be a phenomenon of sensory neurons fast adapting, is one of the neuronal mechanisms for the arousal of hibernating animals.

文摘Model predictive control is widely used in the design of autonomous driving algorithms.However,its parameters are sensitive to dynamically varying driving conditions,making it difficult to be implemented into practice.As a result,this study presents a self-learning algorithm based on reinforcement learning to tune a model predictive controller.Specifically,the proposed algorithm is used to extract features of dynamic traffic scenes and adjust the weight coefficients of the model predictive controller.In this method,a risk threshold model is proposed to classify the risk level of the scenes based on the scene features,and aid in the design of the reinforcement learning reward function and ultimately improve the adaptability of the model predictive controller to real-world scenarios.The proposed algorithm is compared to a pure model predictive controller in car-following case.According to the results,the proposed method enables autonomous vehicles to adjust the priority of performance indices reasonably in different scenarios according to risk variations,showing a good scenario adaptability with safety guaranteed.

文摘Traumatic brain injury(TBI)is closely related to neuro-inflammatory response and causes a complex pathological process and serious consequence.In this study,we explored whether the formatted citrus peel extract gold lotion(GL)could have a therapeutic effect on rats suffered TBI.TBI rat model was prepared by the electric Cortical Contusion Impactor(eCCI)device.Prevention against TBI by GL was assessed by the behavioral,organizational of rats and the molecular markers in the brain injury area.The results showed that GL could reduce nerve damage and neurological dysfunction.The mechanisms probably due to that GL could not only reduce the associated damage markers including GFAP,OX-42,TNF-α,COX-2,NF-κB and TLR4 but also promote the expression of VEGF which is responsible for protecting neuronal cells in the injury area.These findings revealed that GL could be a promising preparation for TBI prevention.

文摘Viologens known as a kind of promising negolyte materials for aqueous organic redox flow batteries,face a critical stability challenge due to the S_N2 nucleophilic attack by hydroxide ions(OH-)during the battery cycling.In this work,a N-cyclic quaternary ammonium-grafted viologen molecule,viz.1,1'-bis(4,4'-dime thylpiperidiniumyl)-4,4'-bipyridinium tetrachloride((DBPPy)Cl_(4)),is developed by the molecular engineering strategy.The obtained(DBPPy)Cl_(4) molecule shows a decent solubility of 1.84 M and a redox potential of-0.52 V vs.Ag/AgCl,Experimental and theoretical results reveal that the grafted N-cyclic quaternary ammonium groups act as the steric hindrance to prevent nucleophilic attack by OH~-,increasing the alkali resistance of the electroactive molecule.The symmetrical battery with 0.50 M(DBPPy)Cl4shows negligible decay during the 13-day cycling test.As demonstration,the flow battery utilizing 1.0 M(DBPPy)Cl_(4) as the negolyte and 1-(1-oxyl-2,2',6,6'-tetramethylpiperidin-4-yl)-1'-(3-(trimethylammonio)propyl)-4,4'-bipyridinium trichloride as the posolyte exhibits a high capacity retention rate of 99.99%per cycle at 60 mA cm^(-2).

文摘Train timetables and operations are defined by the train running time in sections,dwell time at stations,and headways between trains.Accurate estimation of these factors is essential to decision-making for train delay reduction,train dispatching,and station capacity estimation.In the present study,we aim to propose a train dwell time model based on an averaging mechanism and dynamic updating to address the challenges in the train dwell time prediction problem(e.g.,dynamics over time,heavy-tailed distribution of data,and spatiotemporal relationships of factors)for real-time train dispatching.The averaging mechanism in the present study is based on multiple state-of-the-art base predictors,enabling the proposed model to integrate the advantages of the base predictors in addressing the challenges in terms of data attributes and data distributions.Then,considering the influence of passenger flow on train dwell time,we use a dynamic updating method based on exponential smoothing to improve the performance of the proposed method by considering the real-time passenger amount fluctuations(e.g.,passenger soars in peak hours or passenger plunges during regular periods).We conduct experiments with the train operation data and passenger flow data from the Chinese high-speed railway line.The results show that due to the advantages over the base predictors,the averaging mechanism can more accurately predict the dwell time at stations than its counterparts for different prediction horizons regarding predictive errors and variances.Further,the experimental results show that dynamic smoothing can significantly improve the accuracy of the proposed model during passenger amount changes,i.e.,15.4%and 15.5%corresponding to the mean absolute error and root mean square error,respectively.Based on the proposed predictor,a feature importance analysis shows that the planned dwell time and arrival delay are the two most important factors to dwell time.However,planned time has positive influences,whereas arrival delay has negative influences.

文摘During the life cycle of a plant,seed germination is crucial.Upon ingestion of water,the dry seeds resumed energy metabolism and cellular repair.To dissect the complex mechanisms at the very beginning of seed germination,two approaches including transcriptome and small RNA sequencing were conducted during the water imbibition process of mung bean seeds compared with dry seed.The transcriptome sequencing analysis identified 10,108 differentially expressed genes(DEGs)between dry and imbibed mung bean seeds.KEGG enrichment analyses demonstrated numerous DEGs involved in hormone signaling pathways,carbohydrate,and energy metabolism.Out of the total DEGs,129 genes were investigated to involve in abscisic acid,gibberellin,and ethylene pathways,with 56 genes involved in glycolysis and TCA cycle.Through the analysis of small RNA sequencing among different samples,there were 284 miRNAs found in the imbibed and dried seeds of mung bean containing 213 known and 71 novel miRNAs.From comparison between the two types of seeds,51 miRNAs were differentially expressed.The integrated analyses of transcriptome and miRNAome data showed that 23 DEGs are the putative target genes of 8 miRNAs including miR156,miR171b-3p,miR166e-3p,miR169-1,etc.QRT-PCR was used to validate several DEGs and miRNAs.The data suggested that the seed imbibition process is regulated by a variety of DEGs and microRNAs.Meanwhile,we hope to gain a deeper insight into how mung bean seeds germinate in order to optimize seed growth.

文摘Multiple myeloma(MM)is one of the most common hematological malignancies and to date,it remains an incurable disease.In this study,we evaluated the inhibitory effect of 5-acetyloxy-6,7,8,4′-tetramethoxyflavone(5-ATMF),a compound from aged citrus peel extracts,on the MM U266 cell line.We found that the compound inhibited cell growth and induced cell apoptosis in multiple apoptotic assays.The apoptotic proteins caspase-3,caspase-9,and PARP(poly ADP-ribose polymerase)were cleaved when cells were treated with 5-ATMF.Along with the apoptosis process,the anti-apoptotic protein,Bcl-2(B-cell lymphoma-2),was significantly downregulated and the pro-apoptotic protein,Bax(Bcl-2 associated X protein),upregulated along with the release of cytochrome C(Cyt c)and the reduction of mitochondrial membrane potential(MMP).Notably,we found that the phosphorylation of Bad(Bcl-2/Bcl-XL-associated death promoter)was decreased but Bad remained unchanged in this process.On adding 5-ATMF to U266 cells,we observed that 5-ATMF repressed the phosphorylation of Akt(PKB Protein Kinase B PKB),thereby increasing the release of Cyt c and inhibiting the phosphorylation of Bad.These effects were enhanced by Ly294002,an inhibitor of Akt.These results suggested that 5-ATMF exerts a pro-apoptotic effect on U266 cells,possibly associated with the mitochondrial apoptotic pathway induced by the PI3K/Akt/Bad pathway.
文摘Ammonium containing catalyst Mg/Al/Cu-NH3·H2O hydrotalcite (HT) is prepared via co-precipitation, which is effective for NOx storage under 160℃. XRD and FT-IR are employed to characterize its structure and show that ammonium is successfully incorporated into HT. Meanwhile, the HT still retains the typical structure of hydrotalcite-like compounds. NOx storage activity tests indicate Mg/Al/Cu-NH3·H2O HT shows the significant NOx removal activity at 156℃ with a little Oxygen existence. TGA and DTG reveal the reaction of the released ammonium with NOx and storage NOx as nitrates will be responsible for the high NOx storage activity. Additionally, the temperature programmed desorption test of NOx adsorbed sample under 175℃ proves NOx is stored successfully into Mg/Al/Cu-NH3·H2O HT further.

文摘Hide-line-suture-prosthesis repair was introduced by Yibing Li from Shenzhen TCM Anorectal Hospital.In his early years,it was used in rectovaginal fistula,in which it achieved good curative effect.As the pathology of rectovaginal fistula and anal fistula is similar,Yibing Li innovated and applied it to anal fistula cases.This is a case report of a patient with low complex anal fistula who was treated in Shenzhen TCM Anorectal Hospital after two surgeries.The purpose of this report is to share the diagnosis and treatment experience in this case,so as to provide reference for clinicians.

文摘Pulmonary fibrosis(PF)is sexually dimorphic,with a relatively high prevalence and severity in males;however,the mechanism remains unclear.Our study revealed pronounced sexual dimorphism of immune cell genes in the lung,among which grancalcin(GCA)showed profound sex differences.GCA was produced by lung-infiltrating bone marrow macrophages triggered by heightened inflammation in the lung.However,a unique HTR2C+alveolar macrophage population enriched in female lungs metabolically reprogramed bone marrow-derived macrophages and constrained local GCA amplification.As a novel chemokine,GCA bound to protein tyrosine phosphatase receptor type T(PTPRT)in Th17 cells and facilitated pathogenic lung infiltration by activating the ROCK1-MLC pathway,thus aggravating lung fibrosis.Notably,both GCA and Th17 cells abundantly accumulated in lung biopsies from male PF patients but not in those from female patients.GCA-neutralizing antibodies in combination with pirfenidone,a prescribed medication for treating fibrosis,provided superior effectiveness and survival rates against PF compared with treatment with pirfenidone alone.Overall,our findings reveal that sex-biased lung fibrosis is shaped by lung immuneregulatory units,which could be targeted to limit lung fibrosis.
