Rice transformation with cell wall degrading enzyme genes from Trichoderma atroviride and its effect on plant growth and resistance to fungal pathogens

Three genes encoding for fungal cell wall degrading enzymes (CWDE), ech42, nag70 and gluc78 from the biocontrol fungus Trichoderma atroviride were inserted into the binary vector pCAMBIA1305.2 singly and in all possible combinations. The coding sequences were placed downstream of the rice actin promoter and all vectors were used to transform rice plants. A total of more than 1,800 independently regenerated plantlets in seven different populations (for each of the three genes and each of the four gene combinations) were obtained. Expression in plant was obtained for all the fungal genes used singly or in combinations. The ech42 gene encoding for an endochitinase increased resistance to sheath blight caused by Rhizoctonia solani, while the exochitinase-encoding gene, nag70, had a lesser effect. The expression level of endochitinase but not of the exochitinase was correlated with disease resistance. Nevertheless, exochitinase enhanced the positive effect of endochitinase on disease resistance when two genes were co-expressed in transgenic rice. Improved resistance to Magnaporthe grisea was found in all types of regenerated plants, including those with the gluc78 gene alone, while a few lines expressing either ech42 or nag70 appeared to be immune to this pathogen. Transgenic plants expressing the gluc78 gene alone were stunted and only few of them survived, even though they showed resistance to M. grisea. However, combination with either one of the two other genes (ech42, nag70) as included in the same T-DNA region, reduced the negative effect of gluc78 on plant growth. This is the first report of single or multiple of expression of transgens encoding CWDEs that results in resistance to blast and sheath blight in rice. 展开更多

Weak-Localization Effect on the Density of States in Disordered d-wave Superconductors

The weak-localization effect on the quasiparticle density of states (DOS) is studied with the diagrammatic technique in the binary-alloy model of disordered two-dimensional d-wave superconductors both in the Born and the unitary limits. We derive in details the expressions of the Goldstone modes (cooperon and diffuson) for quasiparticle diffuson. For generic Fermi surfaces, the DOS is shown to be subject to a quantum interference correction of logarithmic suppression. In the combined limit of unitarity and nested Fermi surface (the UN limit), it is found that the self-energy diagrams with two π-mode diffusons make additional contributions to the weak-localization effect, which has not been considered in the previous diagrammatic analysis. Due to the contributions of these new diagrams, the DOS in the UN limit is shown to have also a negative logarithmic correction, which is qualitatively different from the previous prediction. 展开更多

Objective: to analyze the influence of optimized recruitment mode of component blood donation on blood donor retention and blood safety. Methods: 100 randomly selected blood donors recruited from machine collected components were studied. The medical observation began in March 2021 and ended in March 2022. They were divided into the control group (50 cases, conventional blood donation recruitment mode) and the observation group (50 cases, optimized blood donation recruitment mode) by double blind method, and the effects of the two groups were compared. Results: the qualified rate of primary screening test and the reaction rate of blood test in the observation group were better than those in the control group (P < 0.05). Conclusion: the optimized recruitment model of component blood donation can increase the number of blood donors and ensure the safety of blood supply, which is worthy of promotion. 展开更多

Observation of the Causes and Effects of Adverse Reactions of Unpaid Blood Donors

Objective: to analyze the causes of adverse reactions after unpaid blood donation. Methods: the data of 24114 unpaid blood donors were analyzed retrospectively, and the number of adverse reactions was counted. Single factor and multifactor logistic regression analysis was used to identify the causes of adverse reactions. Results: among 24114 blood donors who donated blood free of charge from June 2020 to June 2021 in this station, 178 had adverse reactions according to statistics, with the incidence rate of 0.74%. Class A, B, C and D accounted for 45.51%, 30.90%, 12.36% and 11.24% respectively. The final analysis results showed that the weight of the blood donors, the time and season of blood donation, as well as the food status, sleep quality, anxiety and social support level before blood donation were the main causes of adverse reactions to blood donation (P0.05). Conclusion: low personal weight, inappropriate time or season of blood donation, blood donation on an empty stomach and poor physical and mental status of unpaid blood donors are important reasons for adverse reactions. 展开更多
