Fibrotic scar formation after cerebral ischemic stroke:Targeting the Sonic hedgehog signaling pathway for scar reduction

Recent studies have shown that fibrotic scar formation following cerebral ischemic injury has varying effects depending on the microenvironment.However,little is known about how fibrosis is induced and regulated after cerebral ischemic injury.Sonic hedgehog signaling participates in fibrosis in the heart,liver,lung,and kidney.Whether Shh signaling modulates fibrotic scar formation after cerebral ischemic stroke and the underlying mechanisms are unclear.In this study,we found that Sonic Hedgehog expression was upregulated in patients with acute ischemic stroke and in a middle cerebral artery occlusion/reperfusion injury rat model.Both Sonic hedgehog and Mitofusin 2 showed increased expression in the middle cerebral artery occlusion rat model and in vitro fibrosis cell model induced by transforming growth factor-beta 1.Activation of the Sonic hedgehog signaling pathway enhanced the expression of phosphorylated Smad 3 and Mitofusin 2 proteins,promoted the formation of fibrotic scars,protected synapses or promoted synaptogenesis,alleviated neurological deficits following middle cerebral artery occlusion/reperfusion injury,reduced cell apoptosis,facilitated the transformation of meninges fibroblasts into myofibroblasts,and enhanced the proliferation and migration of meninges fibroblasts.The Smad3 phosphorylation inhibitor SIS3 reversed the effects induced by Sonic hedgehog signaling pathway activation.Bioinformatics analysis revealed significant correlations between Sonic hedgehog and Smad3,between Sonic hedgehog and Mitofusin 2,and between Smad3 and Mitofusin 2.These findings suggest that Sonic hedgehog signaling may influence Mitofusin 2 expression by regulating Smad3 phosphorylation,thereby modulating the formation of early fibrotic scars following cerebral ischemic stroke and affecting prognosis.The Sonic Hedgehog signaling pathway may serve as a new therapeutic target for stroke treatment. 展开更多

Small extracellular vesicles derived from hair follicle neural crest stem cells enhance perineurial cell proliferation and migration via the TGF-β/SMAD/HAS2 pathway

Peripheral nerve defect repair is a complex process that involves multiple cell types;perineurial cells play a pivotal role.Hair follicle neural crest stem cells promote perineurial cell proliferation and migration via paracrine signaling;however,their clinical applications are limited by potential risks such as tumorigenesis and xenogeneic immune rejection,which are similar to the risks associated with other stem cell transplantations.The present study therefore focuses on small extracellular vesicles derived from hair follicle neural crest stem cells,which preserve the bioactive properties of the parent cells while avoiding the transplantation-associated risks.In vitro,small extracellular vesicles derived from hair follicle neural crest stem cells significantly enhanced the proliferation,migration,tube formation,and barrier function of perineurial cells,and subsequently upregulated the expression of tight junction proteins.Furthermore,in a rat model of sciatic nerve defects bridged with silicon tubes,treatment with small extracellular vesicles derived from hair follicle neural crest stem cells resulted in higher tight junction protein expression in perineurial cells,thus facilitating neural tissue regeneration.At 10 weeks post-surgery,rats treated with small extracellular vesicles derived from hair follicle neural crest stem cells exhibited improved nerve function recovery and reduced muscle atrophy.Transcriptomic and micro RNA analyses revealed that small extracellular vesicles derived from hair follicle neural crest stem cells deliver mi R-21-5p,which inhibits mothers against decapentaplegic homolog 7 expression,thereby activating the transforming growth factor-β/mothers against decapentaplegic homolog signaling pathway and upregulating hyaluronan synthase 2 expression,and further enhancing tight junction protein expression.Together,our findings indicate that small extracellular vesicles derived from hair follicle neural crest stem cells promote the proliferation,migration,and tight junction protein formation of perineurial cells.These results provide new insights into peripheral nerve regeneration from the perspective of perineurial cells,and present a novel approach for the clinical treatment of peripheral nerve defects. 展开更多

AIM:To investigate age-related differences in the irislens angle(ILA)among patients with age-related cortical cataracts and elucidate the impact of age on lens stability.METHODS:A prospective observational study was conducted on patients with age-related cortical cataracts scheduled for phacoemulsification surgery.Preoperative ultrasound biomicroscopy(UBM)images were collected and analyzed.Initially,patients were stratified into two age groups:<60y and≥60y,with no significant intergroup differences in sex or eye laterality.For further analysis,participants were subdivided into three age strata:<60y,60-75y,and>75y.The ILA was measured in four quadrants(superior,inferior,nasal,and temporal).Intergroup differences in ILA were compared,and correlations between age and ILA parameters were analyzed using statistical methods.RESULTS:The sample data were categorized into three groups according to age,<60y(113 patients;55.8%female),60–75y(245 patients;61.0%female),and>75y(70 patients;50.2%female).The superior quadrant ILA increased progressively with age stratification(P=0.02),and the maximum ILA difference(ΔILA)was significantly higher in patients over 75y(P<0.01).Simple linear regression analysis demonstrated a positive correlation between age and ILA in the superior(Y=7.487+0.096X,R=0.191,P<0.001)and temporal(Y=10.254+0.052X,R=0.104,P=0.032)quadrants.Additionally,the mean ILA across all quadrants(ILAmean)andΔILA were positively correlated with age(ILAmean:Y=9.721+0.055X,R=0.138,P=0.004;ΔILA:Y=3.267+0.044X,R=0.006,P<0.05).CONCLUSION:In patients with age-related cortical cataracts,ILA increases with age,particularly in the superior and temporal quadrants,suggesting that advanced age is associated with greater lens deviation and decreased lens stability.UBM imaging can effectively evaluate the status of the zonule and lens stability,providing crucial evidence for personalized surgical planning based on patients’age. 展开更多

From perinodular to nodular tissues:aberrant accumulation of ornithine accelerates pulmonary fibrosis in silicosis

Silicosis is one of the most serious and prevalent occupational diseases globally,characterized by typical silicotic nodules and fibrosis.Recent studies suggest that the perinodular zone of the lung shares certain characteristics with the nodules themselves.In this study,a silicotic rat model was established via a single intratracheal in-stillation of a 50 mg/mL silica suspension.Pulmonary anatomical and pathological examinations revealed that silica deposition induced severe alterations in both the nodular and perinodular tissues.Subsequently,pseudo-targeted metabolomics analysis revealed that abnormally elevated ornithine levels were closely associated with the progression of silicosis,from normal to perinodular and finally to nodular tissues.Immunofluorescent stain-ing demonstrated that,in addition to M2 macrophages,silica exposure increased the protein levels of ARG1 in epithelial cells,a finding further confirmed by in vitro experiments using A549 and BEAS-2B cells.Moreover,accumulated ornithine induced epithelial-mesenchymal transition in vitro,increased extracellular matrix expres-sion in NIH 3T3 fibroblasts,and enhanced TGF-β1 levels in RAW264.7 cells.Co-exposure to ornithine and silica significantly induced the aberrant expression of fibrosis-associated proteins compared to silica exposure alone,characterized by increased levels of FN and𝛼-SMA,as well as decreased E-cad expression.These findings sug-gest that silica exposure up-regulates ARG1 in various cells,leading to ornithine accumulation,which in turn accelerates the progression of fibrosis. 展开更多

A greenhouse pot experiment was conducted to evaluate the potential of selected woody plants for revegetation in copper (Cu) and lead/zinc (Pb/Zn) mine tailing areas. Five woody species (Amorpha fruticosa Linn, Vitex trifolia Linn. var. simplicifolia Chum, Glochidion puberum (Linn.) Hutch, Broussonetia papyrifera, and Styrax tonkinensis) and one herbaceous species ( Sesbania cannabina Pets) were planted in Cu and Pb/Zn tailings to assess their growth, root morphology, nutrition uptake, metal accumulation, and translocation in plants. Amorpha fruticosa maintained normal growth, while the other species demonstrated stress related growth and root development. Sesbania cannabina showed the highest biomass among the plants, although it decreased by 30% in Cu tailings and 40% in Pb/Zn railings. Calculated tolerance index (TI) values suggested that A. fruticosa, an N-fixing shrub, was the most tolerant species to both tailings (TI values 0.92-1.01), while S. cannabina had a moderate TI of 0.65-0.81 and B. papyrifera was the most sensitive species, especially to Pb/Zn tailings (TI values 0.15-0.19). Despite the high concentrations of heavy metals in the mine tailings and plants roots, only a small transfer of these elements to the aboveground parts of the woody plants was evident from the low translocation factor (TF) values. Among the woody plants, V. trifolia var. simplicifolia had the highest TF values for Zn (1.32), Cu (0.78), and Pb/Zn (0.78). The results suggested that A. fruticosa and S. cannabina, which have the highest tolerance and biomass production, respectively, demonstrated the potential for tailings revegetation in southern China. 展开更多

Route A is the forward rolling; while during routes B and C the sheets are rotated 180o in rolling direction and normal direction, respectively; route D is the unidirectional rolling. The strain states of rolled sheets were analyzed by the finite element method, while the microstructure and texture were observed using optical microscopy, X-ray diffraction and electron back-scattered diffraction techniques, and the mechanical properties were measured by tensile test. The results show that route D produced the largest effective strain. Compared with other samples, sample D exhibited a homogeneous microstructure with fine grains as well as a weak and tilted texture, in corresponding, it performed excellent tensile properties, which suggested that route D was an effective way to enhance the strength and plasticity of AZ31 sheet. 展开更多

Inter-satellite link(ISL)scheduling is required by the BeiDou Navigation Satellite System(BDS)to guarantee the system ranging and communication performance.In the BDS,a great number of ISL scheduling instances must be addressed every day,which will certainly spend a lot of time via normal metaheuristics and hardly meet the quick-response requirements that often occur in real-world applications.To address the dual requirements of normal and quick-response ISL schedulings,a data-driven heuristic assisted memetic algorithm(DHMA)is proposed in this paper,which includes a high-performance memetic algorithm(MA)and a data-driven heuristic.In normal situations,the high-performance MA that hybridizes parallelism,competition,and evolution strategies is performed for high-quality ISL scheduling solutions over time.When in quick-response situations,the data-driven heuristic is performed to quickly schedule high-probability ISLs according to a prediction model,which is trained from the high-quality MA solutions.The main idea of the DHMA is to address normal and quick-response schedulings separately,while high-quality normal scheduling data are trained for quick-response use.In addition,this paper also presents an easy-to-understand ISL scheduling model and its NP-completeness.A seven-day experimental study with 10080 one-minute ISL scheduling instances shows the efficient performance of the DHMA in addressing the ISL scheduling in normal(in 84 hours)and quick-response(in 0.62 hour)situations,which can well meet the dual scheduling requirements in real-world BDS applications. 展开更多

A hybrid estimation of distribution algorithm (EDA) with iterated greedy (IG) search (EDA-IG) is proposed for solving the unrelated parallel machine scheduling problem with sequence-dependent setup times (UPMSP-SDST). For makespan criterion, some properties about neighborhood search operators to avoid invalid search are derived. A probability model based on neighbor relations of jobs is built in the EDA-based exploration phase to generate new solutions by sampling the promising search region. Two types of deconstruction and reconstruction as well as an IG search are designed in the IG-based exploitation phase. Computational complexity of the algorithm is analyzed, and the effect of parameters is investigated by using the Taguchi method of design-of-experiment. Numerical tests on 1640 benchmark instances are carried out. The results and comparisons demonstrate the effectiveness of the EDA-IG. Especially, the bestknown solutions of 531 instances are updated. BACKGROUND Developmental dysplasia of the hip is a developmental abnormality of the hip joint that results from hypoplasia during birth and continues to deteriorate after birth.AIM To observe the effects of magnesium sulfate wet compress,iodophor wet compress,and ice compress on reducing postoperative perineal swelling in children with developmental hip dislocation to provide effective nursing interventions in the clinic.METHODS A total of 120 children with hip dislocation after surgery in a third-class A hospital from January 2018 to January 2020 were randomly divided into four groups,the magnesium sulfate wet compress group,iodophor wet compress group,ice compress group and the control group.Data such as height,weight,age,duration of surgery,intraoperative blood loss,postoperative body temperature,swelling duration,pain score,and incidence of blisters were collected and analyzed.RESULTS There were no significant differences in height,weight,age,duration of surgery,intraoperative blood loss,and postoperative body temperature among the four groups of children.Statistical differences were observed between the intervention groups and the control group(P<0.05).CONCLUSION All three methods significantly reduced postoperative perineal swelling in children with developmental hip dislocation,reduced the duration of postoperative perineal swelling,reduced pain,and improved the quality of care. 展开更多

The improvement of mechanical properties of ZK60 processed by asymmetric reduction rolling (ARR) was investigated in this paper. The grain refinement and basal texture intensity decrease were attributed to the introduction of shear stress produced by ARR process. Compared to conventional symmetrical rolled (SR) ZK60 alloys, ARRed ZK60 exhibited finer, more homogeneous grains and higher mechanical properties. The intensity of basal texture of ARRed ZK60 after annealing was lower than that of SRed ZK60 after annealing. ZK60 sheet with good combination of strength and ductility could be obtained by ARR process. The yield strength (YS) and ultimate tensile strength (UTS) of the ARRed ZK60 sheet were increased 150% and 91.3%, compared to those of SRed ZK60 sheet, from 80 to 200 MPa and from 140 to 264 MPa, respectively. Simultaneously, the elongation to failure increased by 68.75% in the ARR sheet (27%) when compared to that of the SR sheet (16%). 展开更多

The investigation was accompanied with the calculation of Rayleigh numbers.It is found that Ti is the main segregating element and the content of Ti is the highest in the final liquid at the cooling rates of 3-6℃/min.The eta phase(η) precipitate presented in the residual liquid at the cooling rates higher than 6℃/min is responsible for the fluctuations in the curves of Ti content.The dendrite arm spacing is found to markedly decrease with the increase of cooling rate.The maximum relative Rayleigh number occurs at 10-20℃below the liquidus temperature at a cooling rate of 1℃/min,where the mushy zone is most unstable and fluid flow is most prone to occur. 展开更多

The influence of asymmetric reduction rolling (ARR) on the microstructure, texture and mechanical properties of AZ91 was investigated. The microstructural characteristics of the AZ91 sheet processed by symmetric roiling (SR) were the twins, intersection of twins and dynamic recrystalization (DRX) grains around the coarse grains and within the twins. However, the amount of twins and DRX grains in ARRed AZ91 was much smaller than that in SRed AZ91. The SRed AZ91 after annealing exhibited fine DRX grains and some coarse grains with a size of ~ 100 pro. The grains in ARRed AZ91 after annealing were much finer and more homogeneous than those in SRed AZ91 after annealing. The intensity of basal texture of ARRed AZ91 after annealing was lower than that of SRed AZ91 rolling after annealing. The average Schmid factor of ARRed AZ91 is 0.34, which is higher than that of SRed AZ91. The yield strength and ultimate tensile strength of the ARRed AZ91 sheet were inCreased to 16.1% and 31.8% compared to SRed AZ91 sheet, from 155 to 180 MPa, and from 220 to 290 MPa, respectively. The improvement of mechanical properties in ARRed AZ91 after annealing was attributed to much finer, more homogeneous DRX grains and weaker basal texture. 展开更多

Low temperature is a major stress that severely affects plant development,growth,distribution,and productivity.Here,we examined the function of a 2-oxoglutarate-dependent dioxygenase-encoding gene,SlF3HL,in chilling stress responses in tomato(Solanum lycopersicum cv.Alisa Craig[AC]).Knockdown(KD)of SlF3HL(through RNA interference)in tomato led to increased sensitivity to chilling stress as indicated by elevated levels of electrolyte leakage,malondialdehyde(MDA)and reactive oxygen species(ROS).In addition,the KD plants had decreased levels of proline and decreased activities of peroxisome and superoxide dismutase.The expression of four cold-responsive genes was substantially reduced in the KD plants.Furthermore,seedling growth was significantly greater in AC or SlF3HLoverexpression plants than in the KD plants under either normal growth conditions with methyl jasmonate(MeJA)or chilling stress conditions.SlF3HL appears to positively regulate JA accumulation and the expression of JA biosynthetic and signaling genes under chilling stress.Together,these results suggest that SlF3HL is a positive regulator of chilling stress tolerance and functions in the chilling stress tolerance pathways,possibly by regulating JA biosynthesis,JA signaling,and ROS levels. 展开更多

The paper discusses the mechanical and thermal performance manifested in natural nanorods attapulgite(ATP)reinforced Acrylonitrile butadiene styrene(ABS)nanocomposites in the process of fused deposition modeling(FDM).Molten extrusion technique was taken to manufacture the filaments of ABS/organic-attapulgite(OAT)nanocomposites with different mass fraction and the printing operation was made by one commercial FDM three-dimensional(3D)printer.Results indicate that the mechanical performance of these FDM 3D printed specimens are improved obviously via the introduction of OAT,and tensile strength of the ABS/OAT nanocomposites parts with only 2 wt%OAT addition is enhanced by 48.1%.At the same time,the addition OAT can reduce the linear expansion coefficient and creep flexibility,and improve the thermal stability and dimensional accuracy of these FDM 3D printed parts. 展开更多

[Objectives] This study was conducted to investigate the effects of different proportions of spent Pleurotus ostreatus substrate on the germination and seedling growth of mung beans. [Methods] The cellulose-degrading bacteria HB8 and HF1 were mixed with a commercially available microbial composting agent, respectively, for the composting of spent P. ostreatus substrate. Mung beans were cultivated with different proportions of spent mushroom substrate compost and soil. The seed germination rate, plant height, fresh weight and chlorophyll content of mung bean were used as indicators to investigate the effects of the organic fertilizer from spent P. ostreatus substrate on the growth of mung bean seedlings. [Results] The addition of cellulose-degrading bacteria can significantly improve the composting effect of the spent mushroom substrate. After 8 d of cultivation of mung beans with different ratios of the mushroom substrate organic fertilizer, 50% of the organic fertilizer can make the plant height, fresh weight and leaf chlorophyll content of mung bean seedlings reach the highest value and was suitable for mung bean breeding and cultivation. [Conclusions] This study provides scientific basis and technical indicators for the rapid and harmless treatment of spent mushroom substrate and its application in crop cultivation and nursery. 展开更多

In the refinery scheduling, operational transitions in mode switching are of great significance to formulate dynamic nature of production and obtain efficient schedules. The discrete-time formulation meets two main challenges in modeling: discrete approximation of time and large size of mixed-integer linear problem(MILP).In this article, a continuous-time refinery scheduling model, which involves transitions of mode switching, is presented due to these challenges. To reduce the difficulty in solving large scale MILPs resulting from the sequencing constraints, the global event-based formulation is chosen. Both transition constraints and production transitions are introduced and the numbers of key variables and constraints in both of the discrete-time and continuous-time formulations are analyzed and compared. Three cases with different lengths of time horizons and different numbers of orders are studied to show the efficiency of the proposed model. 展开更多

Fast optical wavefront engineering for controlling light propagation in dynamic turbid media

While propagating inside the strongly scattering biological tssue,photons lose their incident directions beyond one transport mean free path(TMFP,~1 millimeter(mm)),which makes it challenging to achieve optical focusing or clear imaging deep inside tissue.By manipulating many degrees of the incident optical wavefront,the latest optical wavefront engineering(WFE)technology compensates the wavelfront distortions caused by the scattering media and thus is toward breaking this physical limit,bringing bright perspective to many applications deep inside tissue,eg,high resolution functional/molecular imaging,optical excitation(optogenetics)and optical tweezers.However,inside the dynamic turbid media such as the biological tissue,the wavefront distortion is a fast and continuously changing process whose decorrelation rate is on timescales from milliseconds(ms)to microseconds(μs),or even faster.This requires that the WFE technology should be capable of beating this rapid process.In this review,we discuss the major challenges faced by the WFE technology due to the fast decorrelation of dynamic turbid media such as living tissue when achieving light focusing/imaging and summarize the research progress achieved to date to overcome these challenges. 展开更多

Stroke is the second leading cause of disability and mortality worldwide,imposing a substantial socioeconomic burden on individuals and healthcare systems.Annually,approximately 14 million people experience stroke,with ischemic stroke comprising nearly 85%of cases,of which 10%to 20%involve large vessel occlusions.Currently,recombinant tissue plasminogen activator(tPA)remains the only approved pharmacological intervention.However,its utility is limited due to a narrow therapeutic window and low recanalization rates,making it applicable to only a minority of patients.Therefore,there is an urgent need for novel therapeutic strategies,including pharmacological advancements and combinatory treatments.Small-molecule natural medicines,particularly those derived from traditional Chinese herbs,have demonstrated significant therapeutic potential in ischemic stroke management.These compounds exert multiple neuroprotective effects,such as antioxidation,anti-inflammatory action,and inhibition of apoptosis,all of which are critical in mitigating stroke-induced cerebral damage.This review comprehensively examines the pathophysiology of acute ischemic stroke(AIS)and highlights the recent progress in the development of small-molecule natural medicines as promising therapeutic agents for cerebral ischemic stroke. 展开更多
