BACKGROUND Primary liver cancer,predominantly hepatocellular carcinoma(HCC),is a major cause of cancer-related mortality.Transarterial chemoembolization(TACE)is a key palliative option for unresectable HCC.However,pro...BACKGROUND Primary liver cancer,predominantly hepatocellular carcinoma(HCC),is a major cause of cancer-related mortality.Transarterial chemoembolization(TACE)is a key palliative option for unresectable HCC.However,prognostic outcomes after TACE vary significantly.This study evaluated the prognostic value of the fibrinogen and neutrophil-to-lymphocyte ratio(F-NLR)score,serum alpha-fetoprotein(AFP),and prealbumin(PA)in patients undergoing TACE.AIM To investigate the prognostic significance of F-NLR score,AFP,and PA in patients undergoing TACE.METHODS Variables such as F-NLR score,AFP,PA,and other clinical indicators were assessed.Follow-ups determined prognosis as good or poor.Statistical asse-ssments,including receiver operating characteristic analyses,were performed to evaluate the prognostic significance and develop an integrated predictive model.RESULTS A retrospective analysis of 162 patients with primary liver cancer undergoing TACE was conducted.Low F-NLR scores and AFP levels and high PA were significantly associated with a good prognosis.The combined model,which integrated F-NLR,AFP,and PA,demonstrated a favorable prognostic predictive capability,with an area under the curve of 0.933.CONCLUSION Preoperative F-NLR,AFP,and PA are valuable prognostic predictors in patients with HCC undergoing TACE.展开更多
BACKGROUND Esophageal cancer patients had the highest intensive care unit(ICU)admitted rate in cancer patients.But their prognosis and evaluation methods were rarely studied.AIM To depict the short-term mortality outc...BACKGROUND Esophageal cancer patients had the highest intensive care unit(ICU)admitted rate in cancer patients.But their prognosis and evaluation methods were rarely studied.AIM To depict the short-term mortality outcome and identify the potential prognostic factors of esophageal cancer patients admitted into ICU.METHODS A multicenter cross-sectional study was performed from May 10,2021 to July 10,2021 at ICU departments of 37 cancer specialized hospitals in China.Patients aged≥14 years with ICU duration≥24 hours were included.Clinical records of patients with primary esophageal cancer diagnosis were reviewed.Patients were separated into groups according to the 90 days survival.Characteristics between groups were compared.Single and multi-variate regression tests were applied to analyze the correlated factors of ICU outcomes.Predictive values of disease severity scores were assessed using receiver operating characteristic curve analysis.RESULTS Total 180 esophageal cancer patients were included.The 90 days mortality was 22.2%.Patients with mortality outcome showed differences from those survived mostly in disease severity and unplanned transfer from clinical ward.The current evaluation tools,including Sequential Organ Failure Assessment and Acute Physiology and Chronic Health Evaluation II scores had low accuracy in prediction of short-term death.ICU admitted esophageal cancer patients have poor prognosis,especially those with acute illness.CONCLUSION The prognostic tools for these patients need to be further optimized.展开更多
Developing an accurate and visual sensing strategy for trace levels of fluoroquinolone residues that pose threat to food safety and human health is highly desired but remains challenging.Herein,a target selfcalibratio...Developing an accurate and visual sensing strategy for trace levels of fluoroquinolone residues that pose threat to food safety and human health is highly desired but remains challenging.Herein,a target selfcalibration ratiometric fluorescent sensing platform has been designed for sensitive visual detection of levofloxacin(LEV)based on fluorescent europium metal-organic framework(Eu-MOF)probe.Specifically,the Eu-MOF was facilely synthesized via directly mixing Eu^(3+)with 1,10-phenanthroline-2,9-dicarboxylic acid(PDA)ligand at room temperature,which exhibited well-stable red fluorescence at 612 nm.Upon the addition of target LEV,the significant fluorescence quenching from Eu^(3+)was observed owing to the inner filter effect between the Eu-MOF and LEV.While the intrinsic fluorescence for LEV at 462nm was gradually enhanced,thereby realizing the self-calibration ratiometric fluorescence responses to LEV.Through this strategy,LEV can be detected down to 27 nmol/L.Furthermore,a test paper-based Eu-MOF integrated with the smartphone assisted RGB color analysis was exploited for the quantitative monitoring of LEV through the multi-color changes from red to blue,thus achieved portable,convenient and visual detection of LEV in honey and milk samples.Therefore,the developed strategy could provide a useful tool for supporting the practical on-site test in food samples.展开更多
BACKGROUND Postoperative enterostomy is increasing in patients with colorectal cancer,but there is a lack of a model that can predict the probability of early complications.AIM To explore the factors influencing early...BACKGROUND Postoperative enterostomy is increasing in patients with colorectal cancer,but there is a lack of a model that can predict the probability of early complications.AIM To explore the factors influencing early postoperative stoma complications in colorectal cancer patients and to construct a nomogram prediction model for predicting the probability of these complications.METHODS A retrospective study of 462 patients who underwent postoperative ostomy for colorectal cancer in the Gastrointestinal Department of the Anhui Provincial Cancer Hospital.The patients’basic information,surgical details,pathological results,and preoperative inflammatory and nutritional indicators were reviewed.We used univariate and multivariate logistic regression to analyze the risk factors for early postoperative stoma complications in colorectal cancer patients and constructed a nomogram prediction model to predict the probability of these complications.RESULTS Binary logistic regression analysis revealed that diabetes[odds ratio(OR)=3.088,95%confidence interval(CI):1.419-6.719],preoperative radiotherapy and chemotherapy(OR=6.822,95%CI:2.171-21.433),stoma type(OR=2.118,95%CI:1.151-3.898),Nutritional risk screening 2002 score(OR=2.034,95%CI:1.082-3.822)and prognostic nutritional index(OR=0.486,95%CI:0.254-0.927)were risk factors for early stoma complications after colorectal cancer surgery(P<0.05).On the basis of these results,a prediction model was constructed and the area under the re-ceiver operating characteristic curve was 0.740(95%CI:0.669-0.811).After internal validation,the area under the receiver operating characteristic curve of the validation group was 0.725(95%CI:0.631-0.820).The calibration curves for the modeling group and validation group are displayed.The predicted results have a good degree of overlap with the actual results.CONCLUSION A previous history of diabetes,preoperative radiotherapy and chemotherapy,stoma type,Nutritional risk screening 2002 score and prognostic nutritional index are risk factors for early stoma complications after colorectal cancer surgery.The nomogram prediction model constructed on the basis of the results of logistic regression analysis in this study can effectively predict the probability of early stomal complications after colorectal cancer surgery.展开更多
Objective To report the development,validation,and findings of the Multi-dimensional Attention Rating Scale(MARS),a self-report tool crafted to evaluate six-dimension attention levels.Methods The MARS was developed ba...Objective To report the development,validation,and findings of the Multi-dimensional Attention Rating Scale(MARS),a self-report tool crafted to evaluate six-dimension attention levels.Methods The MARS was developed based on Classical Test Theory(CTT).Totally 202 highly educated healthy adult participants were recruited for reliability and validity tests.Reliability was measured using Cronbach's alpha and test-retest reliability.Structural validity was explored using principal component analysis.Criterion validity was analyzed by correlating MARS scores with the Toronto Hospital Alertness Test(THAT),the Attentional Control Scale(ACS),and the Attention Network Test(ANT).Results The MARS comprises 12 items spanning six distinct dimensions of attention:focused attention,sustained attention,shifting attention,selective attention,divided attention,and response inhibition.As assessed by six experts,the content validation index(CVI)was 0.95,the Cronbach's alpha for the MARS was 0.78,and the test-retest reliability was 0.81.Four factors were identified(cumulative variance contribution rate 68.79%).The total score of MARS was correlated positively with THAT(r=0.60,P<0.01)and ACS(r=0.78,P<0.01)and negatively with ANT's reaction time for alerting(r=−0.31,P=0.049).Conclusion The MARS can reliably and validly assess six-dimension attention levels in real-world settings and is expected to be a new tool for assessing multi-dimensional attention impairments in different mental disorders.展开更多

Therapeutic benefits of using RDS for both disease control and prevention, in the effort to contain the coronavirus disease 2019(COVID-19), have been shown. However,the biochemically active constituents of RDS and their mechanisms of action are still unclear. The goal of the present study is to clarify the material foundation and action mechanism of RDS.Methods: To conduct an analysis of RDS, an integrative analytical platform was constructed, including target prediction, protein–protein interaction(PPI) network, and cluster analysis;further, the hub genes involved in the disease-related pathways were identified, and the their corresponding compounds were used for in vitro validation of molecular docking predictions. The presence of these validated compounds was also measured in samples of the RDS formula to quantify the abundance of the biochemically active constituents. In our network pharmacological study, a total of 26 bioinformatic programs and databases were used, and six networks, covering the entire Zang-fu viscera, were constructed to comprehensively analyze the intricate connections among the compounds-targets-disease pathways-meridians of RDS.Results: For all 1071 known chemical constituents of the nine ingredients in RDS, identified from established TCM databases, 157 passed drug-likeness screening and led to 339 predicted targets in the constituent–target network. Forty-two hub genes with core regulatory effects were extracted from the PPI network, and 134 compounds and 29 crucial disease pathways were implicated in the target–constitu ent–disease network. Twelve disease pathways attributed to the Lung–Large Intestine meridians, with six and five attributed to the Kidney–Urinary Bladder and Stomach–Spleen meridians, respectively. Onehundred and eighteen candidate constituents showed a high binding affinity with SARS-coronavirus-23-chymotrypsin-like protease(3 CLpro), as indicated by molecular docking using computational pattern recognition. The in vitro activity of 22 chemical constituents of RDS was validated using the 3 CLproinhibition assay. Finally, using liquid chromatography mass spectrometry in data-independent analysis mode,the presence of seven out of these 22 constituents was confirmed and validated in an aqueous decoction of RDS, using reference standards in both non-targeted and targeted approaches.Conclusion: RDS acts primarily in the Lung–Large Intestine, Kidney–Urinary Bladder and Stomach–Spleen meridians, with other Zang-fu viscera strategically covered by all nine ingredients. In the context of TCM meridian theory, the multiple components and targets of RDS contribute to RDS’s dual effects of healthstrengthening and pathogen-eliminating. This results in general therapeutic effects for early COVID-19 control and prevention.展开更多

Potential relevance between the placental inflammation and CMV-related autism has been reported by clinical observation. Meanwhile, abnormal expression of Toll-like receptor 2 (TLR2) and TLR4 in placenta of patients with chorioamnionitis was observed in multiple studies. IL-6 and IL- 10 are two important maternal inflammatory mediators involved in neurodevelopmental disorders. To investigate whether murine CMV (MCMV) infection causes alterations in placental IL-6/10 and TLR2/4 levels, we analyzed the dynamic changes in gene expression of TLR2/4 and IL-6/10 in placentas following acute MCMV infection. Mouse model of acute MCMV infection during pregnancy was created, and pre-pregnant MCMV infected, lipopolysaccharide (LPS)-treated and uninfected mice were used as controls. At E13.5, E 14.5 and E 18.5, placentas and fetal brains were harvested and mRNA expression levels of placental TLR2/4 and IL-6/10 were analyzed. The results showed that after acute MCMV infection, the expression levels of placental TLR2/4 and IL-6 were elevated at E13.5, accompanied by obvious placental inflammation and reduction of placenta and fetal brain weights. However, LPS 50 ktg/kg could decrease the IL-6 expression at E13.5 and E14.5. This suggests that acute MCMV infection during pregnancy could up-regulate the gene expression of TLR2/4 in placental trophoblasts and activate them to produce more pro- inflammatory cytokine IL-6. High dose of LPS stimulation (50 gg/kg) during pregnancy can lead to down-regulation of IL-6 levels in the late stage. Imbalance of IL-6 expression in placenta might be associated with the neurodevelopmental disorders in progeny.展开更多
Absent in melanoma 2(AIM2)inflammasome is a crucial link bridging the innate host defense and the subsequent adaptive immunity when activated by exogenous double stranded DNA(dsDNA).Through establishing models of diss...Absent in melanoma 2(AIM2)inflammasome is a crucial link bridging the innate host defense and the subsequent adaptive immunity when activated by exogenous double stranded DNA(dsDNA).Through establishing models of disseminated murine cytomegalovirus(MCMV)infection in BALB/c and C57BL/6 mice,we evaluated dynamic expression of AIM2 inflammasome components and its relationship with pathological damage and viral replication,trying tofigure out whether AIM2 inflammasome is related to the chronic mechanism of MCMV.BALB/c and C57BL/6 mice were sacrificed on day 0,1,3,7,14 and 28 post infection.Expression levels of AIM2,pro-caspase-1,caspase-1 p20,pro-IL1β and mature IL1β in primary peritoneal macrophages(PMs)and spleens were detected by Western blotting.Contents of IL18 in the serum were detected by ELISA.Pathological examinations of livers were performed,and mRNA levels of MCMV glycoprotein B(gB)in salivary glands also assessed.Results showed that expression levels of AIM2 in PMs and spleens of C57BL/6 mice increased on day 3,even continued to day 28;caspase-1 p20 and mature IL1β increased on day 7,14 and 28;the persistently high expression of IL1β in the serum started on day 1,showing a double peak curve.As for BALB/c mice,expression of AIM2 in PMs increased on day 1 and day 7,while contents of AIM2 in spleens increased on day 1 and day 3;caspase-1 p20 and mature ILip merely increased 7 days fter infection.Thereafter,expression levels of AIM2,caspase-1 p20,mature IL1β and IL18 were limited;the duration of AIM2 inflammasome activation in BALB/c mice was much shorter than that in C57BL/6 mice.The severer pathological damage and more viral replications in BALB/c mice further proved the deficient antiviral immunity to MCMV.In conclusion,the activation of AIM2 inflammasome in BALB/c mice was short-lived,which is quite possibly related to the chronicity of MCMV infection.展开更多

The calcium-sensing receptor (CaSR) is expressed in testicular tissues and plays a pivotal role in calcium homeostasis by activating cellular signaling pathways, but its role in testicular damage induced by diabetes remains unclear. A diabetic model was established by a single intraperitoneal injection of streptozotocin (STZ, 40 mg kg-1) in Wistar rats. Animals then received GdCl3 (an agonist of CaSR, 8.67 mg kg-1), NPS-2390 (an antagonist of CaSR, 0.20 g kg-~), or a combination of both 2 months after STZ injection. Diabetic rats had significantly lower testes weights and serum levels of testosterone compared to healthy rats, indicating testicular damage and dysfunction in STZ-induced diabetic rats. Compared with healthy controls, the testicular tissues of diabetic rats overexpressed the CaSR protein and had higher levels of malondialdehyde (MDA), lower superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity, and higher numbers of apoptotic germ cells. The testicular tissues from diabetic rats also expressed lower levels of Bcl-2 and higher levels of Bax and cleaved caspase-3 in addition to higher phosphorylation rates of c-Jun NH2-terminal protein kinase (JNK), p38, and extracellular signaling-regulated kinase (ERK) 1/2. The above parameters could be further increased or aggravated by the administration of GdCI3, but could be attenuated by injection of NPS-2390. In conclusion, the present results indicate that CaSR activation participates in diabetes-induced testicular damage, implying CaSR may be a potential target for protective strategies against diabetes-induced testicular damage and could help to prevent infertility in diabetic men.展开更多

The differences in the levels of hk10 and DJ-1 in preoperative serum as well as the expression of ovarian cancer-related proliferation genes and invasion genes in intraoperative lesion tissue were compared between the two groups. Correlation analysis was used to assess the inner link of serum hk10 and DJ-1 levels with tumor malignancy in patients with ovarian cancer.Results: Serum hk10 and DJ-1 levels of ovarian cancer group were higher than those of ovarian cyst group;proliferation genes Clusterin, FUNDC1, AKT, TNFAIP8 and HBXIP mRNA expression in lesion tissues were higher than those of ovarian cyst group;invasion genes Beclin1, BRMS1 and SIRT2 mRNA expression in lesion tissues were lower than those of ovarian cyst group whereas EFEMP1 mRNA expression was higher than that of ovarian cyst group. The Pearson test showed that serum hk10 and DJ-1 levels in ovarian cancer were directly correlated with the expression of proliferation genes and invasion genes. Conclusion: hk10 and DJ-1 are highly expressed in serum of patients with ovarian cancer, and their levels are directly correlated with the proliferation and invasion activity of cancer cells.展开更多

The ELF3 activity is subjected to attenuation in response to warm temperature;however,how the protein level of ELF3 is regulated at warm temperature remains less understood. Here, we report that the E3 ligase XB3 ORTHOLOG 5 IN ARABIDOPSIS THALIANA, XBAT35, mediates ELF3 degradation. XBAT35 interacts with ELF3 and ubiquitinates ELF3. Loss-of-function mutation of XBAT35 increases the protein level of ELF3 and confers a short-hypocotyl phenotype under warm temperature conditions. Thus, our findings establish that XBAT35 mediates ELF3 degradation to lift the inhibition of ELF3 on PIF4 for promoting thermoresponsive hypocotyl growth in plants.展开更多

文摘BACKGROUND Primary liver cancer,predominantly hepatocellular carcinoma(HCC),is a major cause of cancer-related mortality.Transarterial chemoembolization(TACE)is a key palliative option for unresectable HCC.However,prognostic outcomes after TACE vary significantly.This study evaluated the prognostic value of the fibrinogen and neutrophil-to-lymphocyte ratio(F-NLR)score,serum alpha-fetoprotein(AFP),and prealbumin(PA)in patients undergoing TACE.AIM To investigate the prognostic significance of F-NLR score,AFP,and PA in patients undergoing TACE.METHODS Variables such as F-NLR score,AFP,PA,and other clinical indicators were assessed.Follow-ups determined prognosis as good or poor.Statistical asse-ssments,including receiver operating characteristic analyses,were performed to evaluate the prognostic significance and develop an integrated predictive model.RESULTS A retrospective analysis of 162 patients with primary liver cancer undergoing TACE was conducted.Low F-NLR scores and AFP levels and high PA were significantly associated with a good prognosis.The combined model,which integrated F-NLR,AFP,and PA,demonstrated a favorable prognostic predictive capability,with an area under the curve of 0.933.CONCLUSION Preoperative F-NLR,AFP,and PA are valuable prognostic predictors in patients with HCC undergoing TACE.
文摘BACKGROUND Esophageal cancer patients had the highest intensive care unit(ICU)admitted rate in cancer patients.But their prognosis and evaluation methods were rarely studied.AIM To depict the short-term mortality outcome and identify the potential prognostic factors of esophageal cancer patients admitted into ICU.METHODS A multicenter cross-sectional study was performed from May 10,2021 to July 10,2021 at ICU departments of 37 cancer specialized hospitals in China.Patients aged≥14 years with ICU duration≥24 hours were included.Clinical records of patients with primary esophageal cancer diagnosis were reviewed.Patients were separated into groups according to the 90 days survival.Characteristics between groups were compared.Single and multi-variate regression tests were applied to analyze the correlated factors of ICU outcomes.Predictive values of disease severity scores were assessed using receiver operating characteristic curve analysis.RESULTS Total 180 esophageal cancer patients were included.The 90 days mortality was 22.2%.Patients with mortality outcome showed differences from those survived mostly in disease severity and unplanned transfer from clinical ward.The current evaluation tools,including Sequential Organ Failure Assessment and Acute Physiology and Chronic Health Evaluation II scores had low accuracy in prediction of short-term death.ICU admitted esophageal cancer patients have poor prognosis,especially those with acute illness.CONCLUSION The prognostic tools for these patients need to be further optimized.

文摘Developing an accurate and visual sensing strategy for trace levels of fluoroquinolone residues that pose threat to food safety and human health is highly desired but remains challenging.Herein,a target selfcalibration ratiometric fluorescent sensing platform has been designed for sensitive visual detection of levofloxacin(LEV)based on fluorescent europium metal-organic framework(Eu-MOF)probe.Specifically,the Eu-MOF was facilely synthesized via directly mixing Eu^(3+)with 1,10-phenanthroline-2,9-dicarboxylic acid(PDA)ligand at room temperature,which exhibited well-stable red fluorescence at 612 nm.Upon the addition of target LEV,the significant fluorescence quenching from Eu^(3+)was observed owing to the inner filter effect between the Eu-MOF and LEV.While the intrinsic fluorescence for LEV at 462nm was gradually enhanced,thereby realizing the self-calibration ratiometric fluorescence responses to LEV.Through this strategy,LEV can be detected down to 27 nmol/L.Furthermore,a test paper-based Eu-MOF integrated with the smartphone assisted RGB color analysis was exploited for the quantitative monitoring of LEV through the multi-color changes from red to blue,thus achieved portable,convenient and visual detection of LEV in honey and milk samples.Therefore,the developed strategy could provide a useful tool for supporting the practical on-site test in food samples.
文摘BACKGROUND Postoperative enterostomy is increasing in patients with colorectal cancer,but there is a lack of a model that can predict the probability of early complications.AIM To explore the factors influencing early postoperative stoma complications in colorectal cancer patients and to construct a nomogram prediction model for predicting the probability of these complications.METHODS A retrospective study of 462 patients who underwent postoperative ostomy for colorectal cancer in the Gastrointestinal Department of the Anhui Provincial Cancer Hospital.The patients’basic information,surgical details,pathological results,and preoperative inflammatory and nutritional indicators were reviewed.We used univariate and multivariate logistic regression to analyze the risk factors for early postoperative stoma complications in colorectal cancer patients and constructed a nomogram prediction model to predict the probability of these complications.RESULTS Binary logistic regression analysis revealed that diabetes[odds ratio(OR)=3.088,95%confidence interval(CI):1.419-6.719],preoperative radiotherapy and chemotherapy(OR=6.822,95%CI:2.171-21.433),stoma type(OR=2.118,95%CI:1.151-3.898),Nutritional risk screening 2002 score(OR=2.034,95%CI:1.082-3.822)and prognostic nutritional index(OR=0.486,95%CI:0.254-0.927)were risk factors for early stoma complications after colorectal cancer surgery(P<0.05).On the basis of these results,a prediction model was constructed and the area under the re-ceiver operating characteristic curve was 0.740(95%CI:0.669-0.811).After internal validation,the area under the receiver operating characteristic curve of the validation group was 0.725(95%CI:0.631-0.820).The calibration curves for the modeling group and validation group are displayed.The predicted results have a good degree of overlap with the actual results.CONCLUSION A previous history of diabetes,preoperative radiotherapy and chemotherapy,stoma type,Nutritional risk screening 2002 score and prognostic nutritional index are risk factors for early stoma complications after colorectal cancer surgery.The nomogram prediction model constructed on the basis of the results of logistic regression analysis in this study can effectively predict the probability of early stomal complications after colorectal cancer surgery.
文摘Objective To report the development,validation,and findings of the Multi-dimensional Attention Rating Scale(MARS),a self-report tool crafted to evaluate six-dimension attention levels.Methods The MARS was developed based on Classical Test Theory(CTT).Totally 202 highly educated healthy adult participants were recruited for reliability and validity tests.Reliability was measured using Cronbach's alpha and test-retest reliability.Structural validity was explored using principal component analysis.Criterion validity was analyzed by correlating MARS scores with the Toronto Hospital Alertness Test(THAT),the Attentional Control Scale(ACS),and the Attention Network Test(ANT).Results The MARS comprises 12 items spanning six distinct dimensions of attention:focused attention,sustained attention,shifting attention,selective attention,divided attention,and response inhibition.As assessed by six experts,the content validation index(CVI)was 0.95,the Cronbach's alpha for the MARS was 0.78,and the test-retest reliability was 0.81.Four factors were identified(cumulative variance contribution rate 68.79%).The total score of MARS was correlated positively with THAT(r=0.60,P<0.01)and ACS(r=0.78,P<0.01)and negatively with ANT's reaction time for alerting(r=−0.31,P=0.049).Conclusion The MARS can reliably and validly assess six-dimension attention levels in real-world settings and is expected to be a new tool for assessing multi-dimensional attention impairments in different mental disorders.

文摘Serine protease 50(PRSS50/TSP50)is highly expressed in spermatocytes.Our study investigated its role in testicular development and spermatogenesis.Initially,PRSS50 knockdown was observed to impair DNA synthesis in spermatocytes.To further explore this,we generated PRSS50 knockout(Prss50^(−/−))mice(Mus musculus),which exhibited abnormal spermatid nuclear compression and reduced male fertility.Furthermore,dysplastic seminiferous tubules and decreased sex hormones were observed in 4-week-old Prss50^(−/−)mice,accompanied by meiotic progression defects and increased apoptosis of spermatogenic cells.Mechanistic analysis indicated that PRSS50 deletion resulted in increased phosphorylation of extracellular signal-regulated protein kinases 1 and 2(ERK1/2)and elevated levels of MAP kinase phosphatase 3(MKP3),a specific ERK antagonist,potentially accounting for testicular dysplasia in adolescent Prss50−/−mice.Taken together,these findings suggest that PRSS50 plays an important role in testicular development and spermatogenesis,with the MKP3/ERK signaling pathway playing a significant role in this process.

文摘Objective: Lung-toxin Dispelling Formula No. 1, referred to as Respiratory Detox Shot(RDS), was developed based on a classical prescription of traditional Chinese medicine(TCM) and the theoretical understanding of herbal properties within TCM. Therapeutic benefits of using RDS for both disease control and prevention, in the effort to contain the coronavirus disease 2019(COVID-19), have been shown. However,the biochemically active constituents of RDS and their mechanisms of action are still unclear. The goal of the present study is to clarify the material foundation and action mechanism of RDS.Methods: To conduct an analysis of RDS, an integrative analytical platform was constructed, including target prediction, protein–protein interaction(PPI) network, and cluster analysis;further, the hub genes involved in the disease-related pathways were identified, and the their corresponding compounds were used for in vitro validation of molecular docking predictions. The presence of these validated compounds was also measured in samples of the RDS formula to quantify the abundance of the biochemically active constituents. In our network pharmacological study, a total of 26 bioinformatic programs and databases were used, and six networks, covering the entire Zang-fu viscera, were constructed to comprehensively analyze the intricate connections among the compounds-targets-disease pathways-meridians of RDS.Results: For all 1071 known chemical constituents of the nine ingredients in RDS, identified from established TCM databases, 157 passed drug-likeness screening and led to 339 predicted targets in the constituent–target network. Forty-two hub genes with core regulatory effects were extracted from the PPI network, and 134 compounds and 29 crucial disease pathways were implicated in the target–constitu ent–disease network. Twelve disease pathways attributed to the Lung–Large Intestine meridians, with six and five attributed to the Kidney–Urinary Bladder and Stomach–Spleen meridians, respectively. Onehundred and eighteen candidate constituents showed a high binding affinity with SARS-coronavirus-23-chymotrypsin-like protease(3 CLpro), as indicated by molecular docking using computational pattern recognition. The in vitro activity of 22 chemical constituents of RDS was validated using the 3 CLproinhibition assay. Finally, using liquid chromatography mass spectrometry in data-independent analysis mode,the presence of seven out of these 22 constituents was confirmed and validated in an aqueous decoction of RDS, using reference standards in both non-targeted and targeted approaches.Conclusion: RDS acts primarily in the Lung–Large Intestine, Kidney–Urinary Bladder and Stomach–Spleen meridians, with other Zang-fu viscera strategically covered by all nine ingredients. In the context of TCM meridian theory, the multiple components and targets of RDS contribute to RDS’s dual effects of healthstrengthening and pathogen-eliminating. This results in general therapeutic effects for early COVID-19 control and prevention.
文摘The analysis of available Nd isotope data from the Tanzania Craton places important constraints on the crest-mantle separation ages,and events marking juvenile crustal addition and crustal recycling.Nd model ages date the oldest crust extraction to 3.16 Ga in the Tanzania Craton,although a rock record of such antiquity is yet to be found there.The most significant period of juvenile crustal addition as well as crustal recycling is 2.7-2.6 Ga.The Nd isotopes of marie samples show that chemical heterogeneity existed in the mantle beneath the Tanzania Craton,with some samples originating from significantly depleted mantle, and most samples originating from the mixture of primitive mantle and depleted mantle.The Nd isotope section reveals significant differences in Nd isotopes between the north eraton and central craton;compared to the north craton,the central craton yields a Nd model age that is approximately 100 Ma older, and its εNd(t)values are more negative,indicating that the two parts of the craton have different mantle source regions.Different types of granitoids are distributed in the Tanzania Craton,such as high-K and low-Al granite,ealc-alkaline granite,peraluminous granite and transitional types of tonalite-trondhjemite- granodiorites (TTGs).Most of the granitoids formed later than the marie rocks in syn-collision and postcollision events.
文摘Increasing evidence has revealed that maternal cytomegalovirus (CMV) infection may be associated with neurodevelopmental disorders in offspring. Potential relevance between the placental inflammation and CMV-related autism has been reported by clinical observation. Meanwhile, abnormal expression of Toll-like receptor 2 (TLR2) and TLR4 in placenta of patients with chorioamnionitis was observed in multiple studies. IL-6 and IL- 10 are two important maternal inflammatory mediators involved in neurodevelopmental disorders. To investigate whether murine CMV (MCMV) infection causes alterations in placental IL-6/10 and TLR2/4 levels, we analyzed the dynamic changes in gene expression of TLR2/4 and IL-6/10 in placentas following acute MCMV infection. Mouse model of acute MCMV infection during pregnancy was created, and pre-pregnant MCMV infected, lipopolysaccharide (LPS)-treated and uninfected mice were used as controls. At E13.5, E 14.5 and E 18.5, placentas and fetal brains were harvested and mRNA expression levels of placental TLR2/4 and IL-6/10 were analyzed. The results showed that after acute MCMV infection, the expression levels of placental TLR2/4 and IL-6 were elevated at E13.5, accompanied by obvious placental inflammation and reduction of placenta and fetal brain weights. However, LPS 50 ktg/kg could decrease the IL-6 expression at E13.5 and E14.5. This suggests that acute MCMV infection during pregnancy could up-regulate the gene expression of TLR2/4 in placental trophoblasts and activate them to produce more pro- inflammatory cytokine IL-6. High dose of LPS stimulation (50 gg/kg) during pregnancy can lead to down-regulation of IL-6 levels in the late stage. Imbalance of IL-6 expression in placenta might be associated with the neurodevelopmental disorders in progeny.

文摘Absent in melanoma 2(AIM2)inflammasome is a crucial link bridging the innate host defense and the subsequent adaptive immunity when activated by exogenous double stranded DNA(dsDNA).Through establishing models of disseminated murine cytomegalovirus(MCMV)infection in BALB/c and C57BL/6 mice,we evaluated dynamic expression of AIM2 inflammasome components and its relationship with pathological damage and viral replication,trying tofigure out whether AIM2 inflammasome is related to the chronic mechanism of MCMV.BALB/c and C57BL/6 mice were sacrificed on day 0,1,3,7,14 and 28 post infection.Expression levels of AIM2,pro-caspase-1,caspase-1 p20,pro-IL1β and mature IL1β in primary peritoneal macrophages(PMs)and spleens were detected by Western blotting.Contents of IL18 in the serum were detected by ELISA.Pathological examinations of livers were performed,and mRNA levels of MCMV glycoprotein B(gB)in salivary glands also assessed.Results showed that expression levels of AIM2 in PMs and spleens of C57BL/6 mice increased on day 3,even continued to day 28;caspase-1 p20 and mature IL1β increased on day 7,14 and 28;the persistently high expression of IL1β in the serum started on day 1,showing a double peak curve.As for BALB/c mice,expression of AIM2 in PMs increased on day 1 and day 7,while contents of AIM2 in spleens increased on day 1 and day 3;caspase-1 p20 and mature ILip merely increased 7 days fter infection.Thereafter,expression levels of AIM2,caspase-1 p20,mature IL1β and IL18 were limited;the duration of AIM2 inflammasome activation in BALB/c mice was much shorter than that in C57BL/6 mice.The severer pathological damage and more viral replications in BALB/c mice further proved the deficient antiviral immunity to MCMV.In conclusion,the activation of AIM2 inflammasome in BALB/c mice was short-lived,which is quite possibly related to the chronicity of MCMV infection.

文摘Male infertility caused by testicular damage is one of the complications of diabetes mellitus. The calcium-sensing receptor (CaSR) is expressed in testicular tissues and plays a pivotal role in calcium homeostasis by activating cellular signaling pathways, but its role in testicular damage induced by diabetes remains unclear. A diabetic model was established by a single intraperitoneal injection of streptozotocin (STZ, 40 mg kg-1) in Wistar rats. Animals then received GdCl3 (an agonist of CaSR, 8.67 mg kg-1), NPS-2390 (an antagonist of CaSR, 0.20 g kg-~), or a combination of both 2 months after STZ injection. Diabetic rats had significantly lower testes weights and serum levels of testosterone compared to healthy rats, indicating testicular damage and dysfunction in STZ-induced diabetic rats. Compared with healthy controls, the testicular tissues of diabetic rats overexpressed the CaSR protein and had higher levels of malondialdehyde (MDA), lower superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity, and higher numbers of apoptotic germ cells. The testicular tissues from diabetic rats also expressed lower levels of Bcl-2 and higher levels of Bax and cleaved caspase-3 in addition to higher phosphorylation rates of c-Jun NH2-terminal protein kinase (JNK), p38, and extracellular signaling-regulated kinase (ERK) 1/2. The above parameters could be further increased or aggravated by the administration of GdCI3, but could be attenuated by injection of NPS-2390. In conclusion, the present results indicate that CaSR activation participates in diabetes-induced testicular damage, implying CaSR may be a potential target for protective strategies against diabetes-induced testicular damage and could help to prevent infertility in diabetic men.
文摘Objective:To discuss the value of serum hk10 and DJ-1 levels in patients with ovarian cancer for evaluating the malignant biology of tumor.Methods:A total of80 patients with ovarian cancer who underwent surgical treatment in our hospital between August 2015 and February 2018 were selected as ovarian cancer group, and 67 patients with ovarian cyst were selected as ovarian cyst group. The differences in the levels of hk10 and DJ-1 in preoperative serum as well as the expression of ovarian cancer-related proliferation genes and invasion genes in intraoperative lesion tissue were compared between the two groups. Correlation analysis was used to assess the inner link of serum hk10 and DJ-1 levels with tumor malignancy in patients with ovarian cancer.Results: Serum hk10 and DJ-1 levels of ovarian cancer group were higher than those of ovarian cyst group;proliferation genes Clusterin, FUNDC1, AKT, TNFAIP8 and HBXIP mRNA expression in lesion tissues were higher than those of ovarian cyst group;invasion genes Beclin1, BRMS1 and SIRT2 mRNA expression in lesion tissues were lower than those of ovarian cyst group whereas EFEMP1 mRNA expression was higher than that of ovarian cyst group. The Pearson test showed that serum hk10 and DJ-1 levels in ovarian cancer were directly correlated with the expression of proliferation genes and invasion genes. Conclusion: hk10 and DJ-1 are highly expressed in serum of patients with ovarian cancer, and their levels are directly correlated with the proliferation and invasion activity of cancer cells.

文摘The research for three-dimension(3D)printing carbon and carbide energy storage devices has attracted widespread exploration interests.Being designable in structure and materials,graphene oxide(GO)and MXene accompanied with a direct ink writing exhibit a promising prospect for constructing high areal and volume energy density devices.This review not only summarizes the recent advances in 3D printing energy storage devices including printing methods,ink rheological properties,and different energy storage systems,but also discusses the printing methods related to energy storage.In addition,the binder or additive free of two-dimensional carbide materials is quite important for the present electrochemical energy storage devices,which also are presented.

文摘BACKGROUND Colorectal cancer(CRC)is one of the most common cancers and CRC patients are among the most common intensive care unit(ICU)admitted cancer patients.However,their prognosis and evaluation methods are rarely studied.AIM To determine the short-term mortality outcome and identify the potential prognostic factors of CRC cancer patients admitted to the ICU.METHODS A multicenter cross-sectional study was performed from May 10,2021 to July 10,2021 at the ICU departments of 37 cancer specialized hospitals in China,and included patients aged≥14 years with ICU duration≥24 hours.Clinical records of patients with a primary CRC diagnosis were reviewed.Patients were separated into groups according to 90-day survival.Characteristics between groups were compared.Univariate and multivariate regression tests were used to analyze the correlated factors of ICU outcomes.Predictive values of disease severity scores were assessed using receiver operating characteristic curve analysis.RESULTS In total,189 CRC patients were included in the study.The 90-day mortality was 12.2%.Patients who died showed differences compared to patients who survived mostly in terms of disease severity and ICU complications.It appears that patients admitted to the ICU from a clinical ward due to emergencies may have a higher risk of mortality while surgical management was associated with better survival.In multivariate analysis,only chemotherapy,elective surgery and conventional oxygen therapy were identified as independently correlated with 90-day mortality.Sequential organ failure assessment and acute physiology and chronic health evaluation II scores had moderate accuracy in predicting short-term mortality.CONCLUSION ICU admitted CRC patients appear to have low short-term mortality which requires further confirmation in prospective studies.The prognostic tools for these patients need further optimization.
