Ultra-high dose rate flash radiotherapy(FLASH-RT)has attracted wide attention in the field of radiotherapy in recent years.For FLASH-RT,radiation is delivered at a very high dose rate[usually thousands of times compared with conventional radiotherapy(CONV-RT)]in an extremely short time.This novel irradiation technique shows a protective effect on normal tissues,also known as the flash effect.At the same time,FLASH-RT is comparable to CONV-RT in terms of tumorkilling efficacy.As basic research dedicates to uncover the mechanisms by which FLASH-RT reduces radiation-induced normal tissue damage,clinical trials of FLASH-RT have been gradually conducted worldwide.This article systematically reviews the evidence of the feasibility and safety of FLASH-RT in clinical practice and offers insights into the future translation of this technology in clinic. 展开更多

Cumulative Evidence on Associations between Genetic Variants and Autoimmune Hepatitis

Genetic factors play a critical role in autoimmune hepatitis (AIH), and numerous studies have been conducted to identify variants associated with the risk of AIH. However, our knowledge of these genetic risk factors is still limited. In this study, we aim to provide a comprehensive synopsis of the genetic architecture of this disease. A systematic search was conducted to identify published studies on the associations between genetic variants and the risk of AIH. Meta-analyses were conducted to calculate the pooled odds ratio (OR) and 95% confidence interval (CI). Then, the cumulative evidence was evaluated for significant associations according to the Venice criteria and false-positive report probability. Finally, functional annotations and pathway analyses were conducted to identify potential pathogenic loci and related pathways. In total, 62 studies involving 11,068 cases and 45,482 controls were included to assess the association between 75 genetic variants and the risk of AIH. Among them, 24 variants were associated with the risk of AIH, and there is strong cumulative evidence supporting these associations. Importantly, HLA DRB1*0301 (OR: 3.023, 95% CI: 2.443 - 1.678, P = 2.81 × 10?24) and DRB3*0101 (OR: 3.667, 95% CI: 2.649 - 5.075, P = 4.69 × 10?15) are newly identified genome-wide significant risk loci. In addition, the rs3184504 variant (OR: 1.305, 95% CI: 1.122 - 1.516, P = 0.001) in the SH2B3 gene is a potential functional mutation. GO pathway analysis suggests that these genes are enriched in antigen processing and presentation, response to interferon-gamma, and immune response-regulating signaling pathways. This study comprehensively summarizes the genetic architecture of AIH and provides cumulative evidence. We have identified two new loci that exceed genome-wide significance. Since self-assembled peptide hydrogels can solve the problems such as low solubility, poor selectivity and serious adverse effects of traditional chemotherapy drugs, they have been widely used as carrier materials for drug delivery. In this study, we developed a novel and injectable drug delivery platform for the antitumor drug doxorubicin(DOX) using a p H-responsive ionic-complementary octapeptide FOE.This octapeptide could self-assemble into stable hydrogel under neutral conditions, while disassemble under the tumor microenvironment. Especially, at p H 5.8, its micromorphology displayed a transition from nanofibers to nanospheres with the change of secondary structure, which enhanced cellular uptake of DOX. In addition, FOE hydrogel serves as a smart drug reservoir by localized injection to achieve sustained drug release and improve antitumor efficacy. This octapeptide opens up new avenues for promoting the clinical translation of anticancer drugs on account of excellent injectable properties and economic benefits of simple and short sequence. 展开更多

Objective:Follistatin(FST)inhibits the action of activin by interfering with the binding of activin to its receptor.Although the prognostic value of FST in various cancers has been investigated previously,studies rarely focused on hypopharyngeal carcinoma(HPC).In our study,the effect of FST expression on HPC tissues and cell lines was investigated.Methods:A total of 60 patients with HPC were recruited for this study.Levels of FST mRNA and protein were measured by quantitative polymerase chain reaction(PCR)and immunohistochemistry in HPC tissue samples and by qPCR in the HPC FaDu cells,as well as immortal nasopharyngeal epithelial cell line NP-69 cells.After silencing the FST expression in FaDu cells using lentivirus-mediated siRNA that was specific for FST mRNA,cell proliferation was determined by a Celigo assay.Tumor growth was monitored in nude mice and viability was determined by a methylthiazoletetrazolium assay.The ratio of cell cycle arrest and apoptosis was evaluated by flow cytometry.The colony formation ability was performed using Giemsa staining.In addition,wound healing and Transwell migration and invasion assays were performed for the analysis of cell motility.Results:FST expression was significantly higher in human HPC tissue and FaDu cells than in normal tissue and NP-69 cells.A higher expression of FST in HPC samples was positively correlated with advanced tumors.Moreover,FST knockdown by shRNA significantly decreased cell growth,colony formation,migration and invasion.Furthermore,FST silencing increased the cell apoptosis percentage,and arrested cell cycle in the S phase in FaDu cells.In addition,FST silencing suppressed tumor growth in vivo.Conclusions:Our results indicated that the FST gene was associated with HPC progression and may serve as a potential therapeutic target for the treatment of HPC. 展开更多

BACKGROUND Spontaneous esophageal rupture or Boerhaave’s syndrome is a rare and acute disease with a high incidence of misdiagnosis and mortality.Here,we aimed to explore the clinical characteristics,diagnosis,treatment,and prognosis of spontaneous esophageal rupture,and to analyze the causes of misdiagnosis during the treatment of spontaneous esophageal rupture.CASE SUMMARY The clinical features of the patient with spontaneous esophageal rupture misdiagnosed earlier as pleural effusion were retrospectively analyzed and the reasons for misdiagnosis are discussed based on a current review of the literature.The patient was admitted to a local hospital due to shortness of breath accompanied by vomiting and abdominal distension for five hours.Based on the computed tomography(CT)scan analysis,clinically,right pleural effusion was diagnosed.However,the patient was unwilling to undergo right closed thoracic drainage.The patient also had intermittent fevers against infection,and during the course of treatment,he complained of chest pain,following which,he was transferred to our hospital.Grapefruit-like residue drainage fluid was observed.Re-examination of the chest CT scans suggested the presence of spontaneous perforation in the upper left esophagus.Therefore,the patient underwent an urgent esophageal hiatus repair.Unfortunately,the patient died of infection and respiratory failure due to progressive dyspnea after surgery.CONCLUSION Spontaneous esophageal rupture is a rare disease associated with high fatality.The patients do not present typical clinical symptoms and the disease progresses rapidly.This case report highlights the importance of a dynamic review of chest CT scan,not only for the initial identification of segmental injury but also for prioritizing subsequent treatment strategies.Moreover,we have presented some clues for clinicians to recognize and diagnose spontaneous esophageal rupture at rare sites(upper-esophageal segment)through this case report of spontaneous esophageal rupture that caused the patient’s death.We have also summarized the reasons for the misdiagnosis and lessons learned. 展开更多

Onyx embolization of a spinal epidural hemorrhage caused by thoracic spinal epidural arteriovenous fistula:A case report and literature review

Spinal epidural hemorrhages(SEDH)caused by spinal epidural arteriovenous fistulas(SEAVFs)are rare;thus,their specific pathogenesis has not been explained.Furthermore,the standard treatment for SEAVFs has not yet been defined.Here we report the case of a 36-year-old Chinese man who experienced acute onset chest pain and tightness.His symptoms rapidly aggravated until the lower limbs were unable to support him.Spinal magnetic resonance angiography(MRA)revealed a localized SEAVF and a secondary spinal cord lesion at the T4 level.Digital subtraction angiography(DSA)confirmed the presence of the SEDH/SEAVF at the T3–4 level with the left radicular artery feeding the fistula.Based on DSA and MRA findings,SEDH,local spinal cord infarction,and spinal venous reflux disorder were conditionally diagnosed.Using the arterial route,Onyx-34 was injected into the fistula to embolize the feeding arteries and the venous system.Angiography was performed after the microcatheter was withdrawn,and no residual fistula or anterior spinal artery was observed.The six-week follow-up MRI showed acceptable healing of the SEAVF,and the patient improved neurologically.This case suggests that endovascular treatment with Onyx-34 embolization should be considered a promising treatment strategy for this type of complicated SEAVF. 展开更多

本研究旨在设计针对牛链球菌和牛球虫的联合多表位疫苗.通过生物信息学筛选两种病原体的T/B细胞优势表位基因序列,串联拼接后引入霍乱毒素B亚单位(CTB)作为分子内佐剂,设计得到多表位重组抗原CARR(CTB-AMA1-RodA-RodA).经密码子优化后,... 本研究旨在设计针对牛链球菌和牛球虫的联合多表位疫苗.通过生物信息学筛选两种病原体的T/B细胞优势表位基因序列,串联拼接后引入霍乱毒素B亚单位(CTB)作为分子内佐剂,设计得到多表位重组抗原CARR(CTB-AMA1-RodA-RodA).经密码子优化后,将CARR基因克隆至pET28a(+)质粒,转化至大肠杆菌BL21(DE3),成功构建了稳定的表达工程菌.SDS-PAGE显示工程菌可高效表达约90 kDa目标蛋白,Western blot验证表达蛋白能与抗CTB及抗牛链球菌抗体特异性结合,且20代内工程菌质粒保有率超80%.纯化后的CARR蛋白免疫蛋鸡,7 wk后ELISA检测显示特异性IgY抗体效价达1∶12800.体外中和实验证实,该抗体显著抑制牛链球菌生长,说明其具备功能性中和活性.结果表明,CARR重组蛋白可有效快速激发宿主体液免疫应答,是牛链球菌和牛球虫联合疫苗的有效候选抗原靶点. 展开更多

低温火箭推进剂贮箱增压输送过程中,高速增压气体可能冲击液面,导致推进剂过量损失与增压失效问题。论文构建了液氢贮箱高液体充注率下的气体增压三维数值模型,研究了增压气流冲击下液面破碎和液体飞溅现象及其对增压效果的影响。采用... 低温火箭推进剂贮箱增压输送过程中,高速增压气体可能冲击液面,导致推进剂过量损失与增压失效问题。论文构建了液氢贮箱高液体充注率下的气体增压三维数值模型,研究了增压气流冲击下液面破碎和液体飞溅现象及其对增压效果的影响。采用三维壳传导模型代替固体壁面全结构模型,描述流固间耦合换热,对比分析了增压气体通过水平出口、半球形出口、垂直出口等不同消能器结构后气流冲击强度与增压性能。结果表明,垂直出口消能器注入高速气流直接冲击液面,发生液体飞溅及液相-气壁间歇接触现象,导致液体推进剂损失加剧。液相飞溅引起气液换热增强及气枕温度下降,造成贮箱压力下降与增压失效。增压效果的能量构成中,输入能量仅有18%~41%用于气枕增压,其余能量产生液相温升;对于冷气体增压方案,输入贮箱的能量由两部分构成,包括增压气体携带能量以及气枕-壁面换热;壁面向气枕的热量传递有利于提升增压性能,在输入能量中占比达9%~28%。 展开更多

The Si Tian project,designed to utilize 60 telescopes distributed across multiple sites in China,is a next-generation timedomain survey initiative.As a pathfinder for the Si Tian project,the Mini-Si Tian(MST)has been proposed and implemented to test the Si Tian’s brain and data pipeline,and to evaluate the feasibility of its technology and science cases.Mounted at the Xinglong Observatory,the MST project comprises three 30 cm telescopes and has been operated since 2022 November.Each telescope of the MST possesses a large field of view,covering 2°.29×1°.53 FOV,and is equipped withg',r'andi'filters,respectively.Acting as the pioneer of the forthcoming Si Tian project,the MST is dedicated to the discovery of variable stars,transients,and outburst events,and has already obtained some interesting scientific results.In this paper,we will summarize the first-two-year operation of the MST project. 展开更多

基于2000年和2010年湖北省102个县级行政区的规模以上工业总产值和县际迁入人口数据,采用空间自相关分析方法,探讨工业经济和迁入人口的空间格局及变化,以及两者之间的空间关联性;采用面板数据拟合联立方程模型,实证检验人口迁入与工业... 基于2000年和2010年湖北省102个县级行政区的规模以上工业总产值和县际迁入人口数据,采用空间自相关分析方法,探讨工业经济和迁入人口的空间格局及变化,以及两者之间的空间关联性;采用面板数据拟合联立方程模型,实证检验人口迁入与工业集聚之间的互动机制。结果表明:湖北省迁入人口和规模以上工业的县际分布之间存在显著的空间正向关联性,武汉市及其邻近县区一直是迁入人口和工业经济的高高集聚区,鄂西地域则是低低集聚区。人口迁入与工业集聚之间存在相互决定的互动关系,地区工业集聚度提高1%,其全部迁入人口和省内迁入人口分别增加0.544 9%和0.602 3%;全部迁入人口或省内迁入人口增加1%,其工业集聚度则分别提高0.294 0%和0.267 8%。社会经济发展水平相对落后的中西部省区应通过推进人口和非农产业协同集聚,培育地区经济增长极,促进地区经济增长和区域协调发展。 展开更多

通过优化产琥珀酸放线杆菌GXAS137发酵粗甘油产丁二酸的培养基,提高丁二酸产量,降低生产成本。先通过单因素试验对粗甘油发酵产丁二酸的电子受体、初始粗甘油质量浓度及氮源进行了优化,再利用响应面试验确定重要参数的最佳水平。结果表... 通过优化产琥珀酸放线杆菌GXAS137发酵粗甘油产丁二酸的培养基,提高丁二酸产量,降低生产成本。先通过单因素试验对粗甘油发酵产丁二酸的电子受体、初始粗甘油质量浓度及氮源进行了优化,再利用响应面试验确定重要参数的最佳水平。结果表明:二甲基亚砜(dimethyl sulfoxide,DMSO)为最适电子受体,玉米浆可替换酵母粉作为氮源,各因素的最佳条件为:初始粗甘油质量浓度55.43 g/L、DMSO质量浓度10.35 g/L、玉米浆质量浓度17.69 g/L。优化后丁二酸产量达到37.02 g/L,丁二酸得率为66.79%,生产强度为0.51 g/(L·h)。与初始条件下丁二酸产量(16.88 g/L)相比,优化后提高了1.19倍。本研究为微生物发酵粗甘油原料生产丁二酸提供了理论支持。 展开更多

丙酮酸是一种重要的有机酸,在聚合物、化妆品、食品添加剂、医药等领域具有广泛应用。酿酒酵母被认为是丙酮酸生产的潜在最适微生物,但其糖酵解过程产生的丙酮酸会在胞质内的丙酮酸脱羧酶催化作用下降解为CO2和乙醛,造成碳代谢流的损失... 丙酮酸是一种重要的有机酸,在聚合物、化妆品、食品添加剂、医药等领域具有广泛应用。酿酒酵母被认为是丙酮酸生产的潜在最适微生物,但其糖酵解过程产生的丙酮酸会在胞质内的丙酮酸脱羧酶催化作用下降解为CO2和乙醛,造成碳代谢流的损失。为将更多的碳代谢流引向丙酮酸合成,该研究通过敲除丙酮酸脱羧酶的3个结构基因(pdc1、pdc5和pdc6),显著地促进了丙酮酸积累,但也造成突变菌株XY-156生长缓慢。与进化之前相比,采用适应性进化策略驯化得到的菌株XY-156A,在细胞生长、葡萄糖消耗以及丙酮酸积累方面均获得显著提高;进一步利用2 L发酵罐进行批次补料发酵试验,发酵76 h丙酮酸产量可达105 g/L,生产强度为1.38g/(L·h),得率为0.5 g/g葡萄糖。结果表明,采用失活丙酮酸脱羧酶与适应性定向进化相结合的策略改造酿酒酵母,能够实现其高效积累丙酮酸,可为生物基丙酮酸工业化生产奠定坚实的基础。 展开更多

城中村居住对流动人口社会融合的影响机制研究——基于社会资本中介模型的广州实证

流动人口的社会融合已成为我国新型城镇化进程中的核心议题。城中村是流动人口典型聚居区,然而居住在城中村如何影响社会融合的内在机制尚未明晰。现有研究主要关注城中村的直接隔离效应,却未能充分探讨其中社会资本的作用机理,且现有... 流动人口的社会融合已成为我国新型城镇化进程中的核心议题。城中村是流动人口典型聚居区,然而居住在城中村如何影响社会融合的内在机制尚未明晰。现有研究主要关注城中村的直接隔离效应,却未能充分探讨其中社会资本的作用机理,且现有研究多聚焦于在地融合过程,忽视跨地域的互动视角。文章基于广州市问卷调查数据,引入社会资本视角,构建“城中村居住—社会资本—社会融合”的理论框架,并采用有调节的中介模型,系统分析居住在城中村对社会融合的影响路径,以及社会资本的中介与返乡频率的调节机制。研究发现:(1)居住在城中村显著降低流动人口社会融合水平,印证了其空间隔离效应;(2)社会资本在城中村与流动人口的社会融合之间发挥重要的中介作用,关系网络、信任与互惠规范成为3条核心中介路径传导间接效应;(3)返乡频率通过提供替代性支持,在一定程度上缓冲居住在城中村对社会资本和社会融合的负面影响,揭示了流动人口跨地域资源调配在社会融合过程中的调节效应。研究结果不仅深化了对城中村居住与社会融合关系的理解,还拓展了社会资本理论在流动人口研究中的应用,强调城中村是社会资本重构的重要空间,同时需要关注流动人口“原乡-异乡”联结的韧性,以更有效地推进“人的城镇化”。 展开更多

This paper presents a comprehensive analysis of the photometric system of the University of Chinese Academy of Sciences 70 cm Telescope located at the Yan-qi Lake campus of the University of Chinese Academy of Sciences.We evaluated the linearity,bias stability,and dark current of the camera.Utilizing the Johnson-Cousins Blue-Visible-Red-Infrared filter system and an Andor DZ936 charge-coupled device camera,we conducted extensive observations of Landolt standard stars to determine the color terms,atmospheric extinction coefficients,photometric zero-points,and the sky background brightness.The results indicate that this telescope demonstrates excellent performance in photometric calibration and good system performance overall,meeting the requirements for limited scientific research and teaching purposes. 展开更多

为研究川续断提取物对氟苯尼考在鸡体内药物动力学影响,将30只肉鸡(2.0 kg±0.1 kg)随机分成2组,每组15只。试验组连续7 d分别灌服川续断提取物(0.3 g/kg,1次/d),对照组给予相同体积的生理盐水,第8天2个组各10只鸡灌服氟苯尼考(30 m... 为研究川续断提取物对氟苯尼考在鸡体内药物动力学影响,将30只肉鸡(2.0 kg±0.1 kg)随机分成2组,每组15只。试验组连续7 d分别灌服川续断提取物(0.3 g/kg,1次/d),对照组给予相同体积的生理盐水,第8天2个组各10只鸡灌服氟苯尼考(30 mg/kg)后按时间点连续采血,采用高效液相色谱法检测氟苯尼考血药浓度,数据采用DAS2.0进行分析。各组剩下的5只鸡,处死鸡剖检取肝脏和空肠,荧光定量PCR检测组织中CYP3A37、MDR1和CXR mRNA表达。试验组和对照组的氟苯尼考主要药物动力学参数为:C_(max)分别为20.829 mg/L±5.267 mg/L、18.429 mg/L±8.365 mg/L,T_(max)分别为0.688 h±0.139 h、1.250 h±0.141 h,AUC_(0-∞)分别为57.907 mg/L·h±8.12 mg/L·h、47.729 mg/L·h±11.601 mg/L·h,T_(1/2)分别为3.649 h±0.193 h、2.520 h±0.255 h,MRT分别为3.885 h±0.487 h、3.151 h±1.604 h,Vd分别为1.338 L/kg±0.492 L/kg、2.139 L/kg±1.182 L/kg,CL分别为0.426 L/h·kg±0.071 L/h·kg、0.657L/h·kg±0.148 L/h·kg。川续断提取物的连续使用,引起了氟苯尼考药物动力学特征明显改变,AUC_(0-∞)和T_(1/2)增加(P<0.05),T_(max)和CL降低(P<0.05)。荧光定量PCR检测结果显示,川续断提取物给药组抑制了CXR和CYP3A37在肉鸡肝脏和空肠中mRNA的表达量(P<0.05)。结果表明,川续断提取物连续给药7 d后,增加了氟苯尼考在鸡体内的吸收速度,延缓了代谢,提示两者在临床上合用有潜在的增效作用。 展开更多

We assess the detectability of tidal disruption events(TDEs)using mock observations from the Mini-SiTian Array.We select 100 host galaxy samples from a simulated galaxy catalog based on specific criteria such as redshift,black hole mass,and event rate.Taking into account the site conditions and survey strategy,we simulate observations over a 440 deg^(2)field.The results indicate that 0.53±0.73 TDEs can be detected per year when observing in both g and r bands with 300 s exposures every 3 days.Applying this method to the SiTian project,we expect to discover approximately 204 TDEs annually,heralding a new era in TDE science. 展开更多
