目的探讨miR-218-5p通过靶向LIN28B调控慢性阻塞性肺疾病(COPD)时气道上皮细胞凋亡和炎症反应的作用及其机制。方法将人肺支气管上皮细胞BEAS-2B分为正常对照(NC)组、香烟烟雾提取物(CSE)组、miR-NC+CSE组、miR-218-5p+CSE组、si-NC+CS... 目的探讨miR-218-5p通过靶向LIN28B调控慢性阻塞性肺疾病(COPD)时气道上皮细胞凋亡和炎症反应的作用及其机制。方法将人肺支气管上皮细胞BEAS-2B分为正常对照(NC)组、香烟烟雾提取物(CSE)组、miR-NC+CSE组、miR-218-5p+CSE组、si-NC+CSE组、si-LIN28B+CSE组、miR-218-5p+pcDNA-NC+CSE组和miR-218-5p+pcDNA-LIN28B+CSE组。RT-qPCR检测miR-218-5p的表达水平;流式细胞术检测细胞凋亡;酶联免疫吸附实验(ELISA)检测白细胞介素6(IL-6)和转化生长因子-β1(TGF-β1)的表达水平;蛋白质印记(Western blot)检测LIN28B、裂解的天冬氨酸半胱氨酸蛋白酶(Cleaved-caspase-3)的表达水平;双荧光素酶报告基因实验验证miR-218-5p对LIN28B的靶向作用。结果与NC组比较,CSE组BEAS-2B细胞miR-218-5p表达降低,细胞凋亡率、LIN28B、Cleaved-caspase-3、IL-6和TGF-β1表达显著增加(均P<0.05)。与miR-NC+CSE组比较,miR-218-5p+CSE组BEAS-2B细胞凋亡率、Cleaved-caspase-3、IL-6和TGF-β1表达显著降低(均P<0.05)。与si-NC+CSE组比较,si-LIN28B+CSE组BEAS-2B细胞凋亡率、Cleaved-caspase-3、IL-6和TGF-β1表达显著降低(均P<0.05)。与miR-218-5p+pcDNA-NC+CSE组比较,miR-218-5p+pcDNA-LIN28B+CSE组BEAS-2B细胞细胞凋亡率、Cleaved-caspase-3、IL-6和TGF-β1表达显著增加(均P<0.05)。结论miR-218-5p通过靶向LIN28B可抑制CSE诱导的气道上皮细胞凋亡和炎症反应。因此,miR-218-5p可能是慢性阻塞性肺疾病的潜在治疗靶点。 展开更多

目的:探究微小核糖核酸-17-5p(miR-17-5p)调控LIM激酶1(LIMK1)/丝切蛋白1(cofilin1)通路对子宫内膜异位症发生发展的影响。方法:选取行子宫切除术的30例子宫内膜异位症患者作为子宫内膜异位症在位内膜组,另选取30例非雌激素依赖性疾病... 目的:探究微小核糖核酸-17-5p(miR-17-5p)调控LIM激酶1(LIMK1)/丝切蛋白1(cofilin1)通路对子宫内膜异位症发生发展的影响。方法:选取行子宫切除术的30例子宫内膜异位症患者作为子宫内膜异位症在位内膜组,另选取30例非雌激素依赖性疾病患者作为正常子宫内膜组。对两组患者的在位内膜细胞中LIMK1、cofilin1表达量采用实时荧光定量PCR法检测,对所获取的在位内膜细胞进行转染,Transwell法检测细胞的侵袭能力;CCK-8法检测细胞的增殖能力;ELISA法检测子宫内膜细胞中细胞因子ICAM-1、VEGF、MMP-9表达。结果:在位内膜组患者的miR-17-5p表达水平低于正常子宫内膜组(P<0.05);在位内膜组的LIMK1蛋白及其mRNA、cofilin1蛋白及其mRNA相对表达水平均高于正常子宫内膜组(P<0.05);在位内膜组未转染组(EC组)及转染阴性对照(EC-C组)情况下细胞侵袭能力显著高于正常子宫内膜组(NC组),但转染情况下(EC-miR-17-5p-RNAi组)细胞侵袭能力显著低于正常子宫内膜组(NC-miR-17-5p-cDNA组),两组患者细胞转染阴性对照(EC-C组、NC-C组)与未转染状态(EC组、NC组)差异无统计学意义(P>0.05);转染情况在位内膜组患者(EC-miR-17-5p-RNAi组)细胞侵袭力低于未转染状态(EC组),但正常子宫内膜组(NC-miR-17-5p-cDNA组)高于未转染组(NC组,P>0.05)。EC组细胞增殖能力在24 h、48 h、72 h、96 h均高于NC组细胞(P<0.05);EC-miR-17-5p-RNAi组各时间段细胞增殖能力与EC组相比较均升高(P<0.05);EC组与EC-C组在各时间段的细胞增殖能力比较差异无统计学意义(P>0.05);NC-miR-17-5p-cDNA组在各时间段的增殖能力均低于NC组(P<0.05);NC-C组各时间段的细胞增殖能力与NC组比较差异无统计学意义(P>0.05)。基础状态下,EC组ICAM-1、MMP-9和VEGF水平高于NC组,EC-miR-17-5p-RNAi组与EC组比较明显升高(P<0.05);EC组与EC-C组各指标比较差异无统计学意义(P>0.05);NC组ICAM-1、MMP-9和VEGF水平明显高于NC-miR-17-5p-cDNA组(P<0.05),但与NC-C组比较差异无统计学意义(P>0.05)。结论:miR-17-5p在内异症细胞中表达水平下调,通过负向调控LIMK1/cofilin1通路抑制细胞增殖、侵袭,在内异症发生及进展过程中发挥基因调节功能。 展开更多

目的探讨老年胃癌患者血清长基因间非蛋白编码RNA(LINC)00511和LINC01133的表达及与临床特征的关系,分析二者单项及与糖类抗原(CA)72-4联合检测对老年人胃癌的临床诊断价值。方法选取老年胃癌患者100例作为实验组,再选取同期老年健康体... 目的探讨老年胃癌患者血清长基因间非蛋白编码RNA(LINC)00511和LINC01133的表达及与临床特征的关系,分析二者单项及与糖类抗原(CA)72-4联合检测对老年人胃癌的临床诊断价值。方法选取老年胃癌患者100例作为实验组,再选取同期老年健康体检者76例作为对照组,通过定量逆转录聚合酶链反应检测血清LINC00511和LINC01133的表达,分析二者血清表达水平的相关性及与肿瘤临床特征的关系;采用受试者工作特征(ROC)曲线评估其对胃癌的临床诊断效能;通过计算ROC曲线下面积(AUC)、敏感度、特异度、阳性预测值及阴性预测值,评估单项及联合检测的临床应用价值。结果实验组LINC00511和LINC01133表达量显著高于对照组(均P<0.01),实验组二者表达水平呈显著正相关(r=0.304,P<0.01)。LINC01133血清水平与性别、TNM临床分期和淋巴结转移均无统计学差异(P>0.05),但与肿瘤合并远处转移呈正相关(P<0.01)。LINC00511、LINC01133及CA72-4诊断胃癌的AUC分别为0.659、0.807和0.764,其中LINC01133诊断的敏感度(81.0%)最高,CA72-4诊断的特异度(84.2%)最高。LINC00511与LINC01133具有显著诊断互补性(P<0.05)。而LINC00511和CA72-4、LINC01133和CA72-4之间均无显著互补性(P>0.05)。LINC00511和LINC01133联合可将诊断的敏感度提高至92.0%,二者与CA72-4联合可将敏感度和阴性预测值分别提高至94.0%和85.7%。结论LINC00511和LINC01133在老年胃癌患者血清中高表达,与临床进展显著相关,可能是老年胃癌辅助诊断的潜在血清学标志物。 展开更多

A numerical model and transmission characteristic analysis of DPSK (differential phase shift keying) pressure signals in mud channels is introduced. With the control logic analysis of the rotary valve mud telemetry, a logical control signal is built from a Gate function sequence according to the binary symbols of transmitted data and a phase-shift function is obtained by integrating the logical control signal. A mathematical model of the DPSK pressure signal is built based on principles of communications by modulating carrier phase with the phase-shift function and a numerical simulation of the pressure wave is implemented with the mathematical model by MATLAB programming. Considering drillpipe pressure and drilling fluid temperature profile along drillpipes, the drillpipe of a vertical well is divided into a number of sections. With water-based drilling fluids, the impacts of travel distance, carrier frequency, drillpipe size, and drilling fluids on the signal transmission were studied by signal transmission characteristic analysis for all the sections. Numerical calculation results indicate that the influences of the viscosity of drilling fluids and volume fraction of gas in drilling fluids on the DPSK signal transmission are more notable than the others and the signal will distort in waveform with differential attenuations of the signal frequent component. 展开更多

Diabetic nephropathy(DN)is one of the most common complications of diabetes mellitus,which is characterized in renal tubulointerstitial fibrosis(TIF).The current study was designed to investigate the protective effect of Jujuboside A(Ju A)on TIF in type 2 diabetes(T2DM)mice,and explore its underlying anti-fibrosis mechanism.A mouse T2DM model was established using high fat diet(HFD)feeding combined with intraperitoneal injection of streptozotocin(STZ).Then,diabetic mice were treated with Ju A(10,20 and 40 mg·kg^(−1)·d^(−1),i.g.)for 12 weeks.Results showed that administration of Ju A not only down-regulated fasting blood glucose(FBG)levels,but also improved hyperlipidemia and renal function in diabetic mice.Moreover,the reduced ECM accumulation was observed in the renal cortex of Ju A treated diabetic mice,while the TIF progression was also attenuated by Ju A through blocking the epithelial-to-mesenchymal transition(EMT)of renal tubular epithelial cells(RTECs).Further mechanism studies showed that Ju A treatment effectively down-regulated the protein expression and subsequent nuclear translocation of Yin Yang 1(YY1)in the renal cortex of diabetic mice,and reduced the levels of transforming growth factor-β1(TGF-β1)in the serum and renal cortex of Ju A treated mice.According to in vitro studies,the up-regulated YY1/TGF-β1 signaling pathway was restored by Ju A in high glucose(HG)cultured HK-2 cells.Taken together,these findings demonstrated that Ju A can ameliorate the TIF of DN through down-regulating the YY1/TGF-β1 signaling pathway. 展开更多

To assess the effects of straw return coupled with deep nitrogen(N)fertilization on grain yield and N use efficiency(NUE)in mechanical pot-seedling transplanting(MPST)rice,the seedlings of two rice cultivars,i.e.,Yuxiangyouzhan and Wufengyou 615 transplanted by MPST were applied with N fertilizer at 150 kg/hm2 and straw return at 6 t/hm2 in early seasons of 2019 and 2020.The experiment comprised of following treatments:CK(no fertilizer and no straw return),MDS(deep N fertilization and straw return),MBS(broadcasting fertilizer and straw return),MD(deep N fertilization without straw return),MB(broadcasting fertilizer without straw return).Results depicted that the MDS treatment significantly increased the rice yield by 41.69%-72.22%due to total above-ground biomass,leaf area index and photosynthesis increased by 54.70%-55.80%,38.52%-52.17%and 17.89%-28.40%,respectively,compared to the MB treatment.In addition,the MDS treatment enhanced the total N accumulation by 37.74%-43.69%,N recovery efficiency by 141.45%-164.65%,N agronomic efficiency by 121.76%-134.19%,nitrate reductase by 46.46%-60.86%and glutamine synthetase by 23.56%-31.02%,compared to the MB treatment.The average grain yield and NUE in both years for Yuxiangyouzhan were higher in the MDS treatment than in the MD treatment.Hence,deep N fertilization combined with straw return can be an innovative technique with improved grain yield and NUE in MPST in South China. 展开更多

The hysteretic behavior and nonlinearity of the equivalent material coefficient of macro fiber composites(MFC) under staircase input conditions are investigated using the Preisach model.Based on a database of first order reversal curves, formulas are derived to predict the hysteresis of strain output and nonlinearity of the equivalent piezoelectric coefficient of MFCs. Formulae are verified by comparing the predicted strains with the measured strains of three MFC specimens,which are driven by a random sequence of staircase voltage inputs. The coefficients obtained by the formulae and experimentation coincide. Further results indicate that the equivalent piezoelectric strain coefficient depends greatly on the value of drive voltage across the entire input range, and the coefficient is asymmetric across the negative and positive input ranges. Deflection testing of an MFC composite cantilever demonstrates the importance of taking the nonlinearity of the equivalent piezoelectric coefficient into consideration in the application of actuation. 展开更多

目的探讨急性脑梗死(AS)患者血清Ghrelin水平与炎性因子及颈动脉粥样硬化斑块的关系。方法筛选郑州大学附属郑州中心医院2014年1月—2016年1月收治的急性脑梗死患者122例,根据血浆Ghrelin水平[(117.0±37.2)pmol/l],大于(117.0+37.2... 目的探讨急性脑梗死(AS)患者血清Ghrelin水平与炎性因子及颈动脉粥样硬化斑块的关系。方法筛选郑州大学附属郑州中心医院2014年1月—2016年1月收治的急性脑梗死患者122例,根据血浆Ghrelin水平[(117.0±37.2)pmol/l],大于(117.0+37.2)pmol/L为治疗组,共61例;低于(117.0-37.2)pmol/L分为对照组,用ELISA法测血浆肿瘤坏死因子α(TNFα)、白介素-8(IL-8)、单核细胞趋化蛋白-1(MCP-1)水平,用彩色多普勒超声检测颈总动脉、颈内动脉颅外段及分叉处的内-中膜厚度(intima-media thickness,IMT),测量动脉粥样硬化(Athrosclerotic plaque,ASP)斑块面积占血管管腔总面积的百分比;并分析Ghrelin水平与炎性因子及颈动脉斑块之间关系。结果血浆炎性因子随着血清Ghrelin水平增加而降低,差异有统计学意义(P<0.05);对照组IMT明显增厚,颈动脉粥样硬化斑块检出及不稳定性斑块发生率高于治疗组,差异有统计学意义(P<0.05)。结论急性脑梗死患者的血清Ghrelin水平与炎性因子及颈动脉斑块显著相关,Ghrelin可能是急性脑梗死患者的一个重要保护因子。 展开更多

The constructed potential-pH diagrams of Li-Ni(Co,Mn)-H_(2)O system indicate that the LiNiO_(2),LiCoO_(2)and LiMnO_(2)are thermodynamically stable in aqueous solution within the temperature range of 25-200℃and the activity range of 0.01-1.00.A predominant co-region of LiNiO_(2),LiCoO_(2)and LiMnO_(2)oxides(Li-Ni-Co-Mncomposite oxide)is found in the Li-Ni-Co-Mn-H_(2)O potential-pH diagrams,in which the co-precipitation region expands towards lower pH with rising temperature,indicating the enhanced possibility of synthesizing Li-Ni-Co-Mn composite oxide in aqueous solution.The experimental results prove that it is feasible to prepare the LiNi_(0.5)Co_(0.2)Mn_(0.3)O_(2)cathode materials(NCM523)by an aqueous routine.The as-prepared lithiated precursor and NCM523 both inherit the spherical morphology of the hydroxide precursor and the obtained NCM523 has a hexagonalα-NaFeO_(2)structure with good crystallinity.It is reasonable to conclude that the aqueous routine for preparing NCM cathode materials is a promising method with the guidance of the reliable potential-pH diagrams to some extent. 展开更多

This study was conducted to investigate the association between ideal CVH and subclinical atherosclerosis in a population cohort of Chinese adults aged ≥ 40 years. Methods This study was designed as a cross-sectional analysis of 8,395 participants who had complete data at baseline and a prospective analysis of 4,879 participants who had complete data at 4.3 years of follow-up. Ideal CVH metrics were defined according to the American Heart Association. Subclinical atherosclerosis was evaluated by plaques in carotid arteries, carotid intima-media thickness(CIMT), brachial-ankle pulse wave velocity(baPWV), and urinary albumin-to-creatinine ratio(UACR). Results Both the prevalence and incidence of atherosclerosis measures were found to be decreased with increasing numbers of ideal CVH metrics at baseline(all P values for trend < 0.01). The levels of CIMT and UACR at follow-up showed an inverse and significant association with the numbers of ideal CVH metrics at baseline(both P values for trend < 0.05) but a borderline significant association with baPWV(P for trend = 0.0505). Taking participants with 0-1 ideal metric as reference, we found that participants with 5-6 ideal metrics had significantly lower risks of developing carotid plaques(odds ratio, OR = 0.46; 95% confidence interval, CI 0.27-0.79), increased CIMT(OR = 0.60; 95% CI 0.42-0.84), and increased baPWV(OR = 0.57; 95% CI 0.34-0.97) after full adjustments. A significant interactive effect of age and CVH was detected on CIMT and baPWV progression(both P values for interaction < 0.05). Conclusion The numbers of ideal CVH metrics showed a significant and inverse association with the risk of developing subclinical atherosclerosis in middle-aged and elderly Chinese adults, whereas its dose-response effect was attenuated in individuals aged ≥ 60 years and partially weakened in male participants. 展开更多

Newly-acquired seismic data reveal widespread carbonate deposits covering a large part of the northwestern South China Sea margin.Three carbonate platforms are identified to have developed on the topographic highs inherited from tectonic deformation and volcanic accretion.Across the carbonate platforms,the Miocene strata are characterized by high-amplitude seismic reflections and distinct platform architecture that overlaps older strata.The Guangle and Xisha carbonate platforms grew on faulted blocks due to South China Sea continental rifting,while the Zhongjian carbonate platform developed on a structural high induced by early Miocene volcanism.During the late Miocene,partial drowning resulted in the inhibition of platform growth,eventual platform drowning and termination of most carbonate deposition.The drowning of the Guangle and Zhongjian carbonate platforms is shown by the supply of siliciclastic sediments during the late Miocene and seems to be closely linked to late Neogene volcanic activity,whilst the drowning of the Xisha carbonate platform is primarily related to relative eustatic changes.Our results imply that tectonic activity,volcanism and eustasy are the dominant controls on the evolution of carbonate platforms on the northwestern margin of the South China Sea. 展开更多

AIM:To evaluate scleral buckling(SB)surgery using a noncontact wide-field viewing system and 23-gauge intraocular illumination for the treatment of rhegmatogenous retinal detachment in silicone oil(SO)-filled eyes.METHODS:Totally 9 patients(9 eyes)with retinal detachment in SO-filled eyes were retrospectively analyzed.All patients underwent non-contact wide-field viewing system-assisted buckling surgery with 23-gauge intraocular illumination.SO was removed at an appropriate time based on recovery.The patients were followed up for at least 3mo after SO removal.Retinal reattachment,complications,visual acuity and intraocular pressure(IOP)before and after surgery were observed.RESULTS:Patients were followed up for a mean of 8.22mo(3-22mo)after SO removal.All patients had retinal reattachment.At the final follow-up,visual acuity showed improvement for 8 patients,and no change for 1 patient.The IOP was high in 3 patients before surgery,but it stabilized after treatment;it was not affected in the other patients.None of the patients had infections,hemorrhage,anterior ischemia,or any other complication.CONCLUSION:This new non-contact wide-field viewing system-assisted SB surgery with 23-gauge intraocular illumination is effective and safe for retinal detachment in SO-filled eyes. 展开更多

Ischemic stroke often elicits profound white matter lesions, which results in poor neurological outcomes and impairing the recovery in post-stroke. However, few studies have focused on white matter lesions caused by cerebral ischemia. The present study was aimed to investigate the effects of cornel iridoid glycoside(CIG), a main active component extracted from Cornus officinalis, on the white matter injury induced by ischemic stroke. CIG(60 and 120 mg·kg-1) were administered intragastrically 6 h after middle cerebral artery occlusion reperfusion(MCAO) surgery once a day for 7 d. A series of behavioral tests were carried out to evaluate the neurological function of MCAO rats. White matter structure was detected by luxol fast blue staining and transmission electron microscopy. Immunohistochemical staining was used to assess myelin loss, oligodendrocytes maturation and glial activation. Results showed that CIG remarkably decreased neurological deficit score, accelerated the recovery of somatosensory and motor functions, and ameliorated the memory deficit in MCAO rats. CIG alleviated white matter lesions and demyelination, increased myelin basic protein expression and the number of mature oligodendrocytes in the corpus callosum of MCAO rats. Besides, CIG inhibited the activation of microglia and astrocytes. Further data obtained by western blot analysis indicated that CIG increased the expression of brain-derived neurotrophic factor(BDNF)/p-Trk B, neuregulin-1/Erb B, and PI3 K p110α/p-Akt/p-m TOR in the corpus callosum of MCAO rats. Our findings demonstrated that CIG protected against white matter lesions induced by cerebral ischemia and the mechanisms were partially contributed to increasing BDNF and activating neuregulin-1/Erb B signaling and its downstream PI3 K/Akt/m TOR pathway in white matter. 展开更多

Phosphorus(P)is an essential element for plant growth but is often limiting in ecosystems;therefore,improving the P fertilizer use efficiency is important.Biochar and arbuscular mycorrhizal fungi(AMF)may enhance P cycling in paddy soils that contain high content of total P but low content of available P(AP).In this study,the effects of biochar addition and Rhizophagus irregularis inoculation on the organic and inorganic P contents and phosphatase activities in paddy soils,rice seedling growth,and AMF colonization were investigated.Compared with no biochar addition,biochar addition enhanced the percentage of spore germination at day 7,hyphal length,most probable number,and mycorrhizal colonization rate of R.irregularis by 32%,662%,70%,and 28%on average,respectively.Biochar and R.irregularis altered soil P cycling and availability.Biochar and R.irregularis,either individually or in combination,increased soil AP content by 2%–48%.Rice seedlings treated with biochar and R.irregularis produced greater biomass,improved root morphology,and increased nutrient uptake compared with those of the control without biochar and R.irregularis.The results suggest that combined application of biochar and R.irregularis is beneficial to rice cultivation in paddy soils with high content of total P but low content of AP. 展开更多

An attempt was made to prepare carbon coated tourmaline by mixing tourmaline powders and polyvinyl alcohol (PVA), followed by heat treatment in argon atmosphere. All samples were characterized by powder X-ray diffraction, high-resolution transmission electron microscopy, UV diffuse reflectance spectroscopy. Results showed that the residual carbon content was influenced by heat treatment temperature and the amount of PVA. The degradation of methylene blue by Carbon-coated tourmaline was also studied. The experiments pointed out that the carbon coated effects are best when the heating temperature was 900℃ and the weight content of PVA was 70%. And the tourmaline prepared under 900℃ in the oxidation atmosphere has the best degrade efficiency. The results also proved that the infrared radiation of tourmaline is not effect in the degrade progress. 展开更多

Accurate mapping and timely monitoring of urban redevelopment are pivotal for urban studies and decisionmakers to foster sustainable urban development.Traditional mapping methods heavily depend on field surveys and subjective questionnaires,yielding less objective,reliable,and timely data.Recent advancements in Geographic Information Systems(GIS)and remote-sensing technologies have improved the identification and mapping of urban redevelopment through quantitative analysis using satellite-based observations.Nonetheless,challenges persist,particularly concerning accuracy and significant temporal delays.This study introduces a novel approach to modeling urban redevelopment,leveraging machine learning algorithms and remote-sensing data.This methodology can facilitate the accurate and timely identification of urban redevelopment activities.The study’s machine learning model can analyze time-series remote-sensing data to identify spatio-temporal and spectral patterns related to urban redevelopment.The model is thoroughly evaluated,and the results indicate that it can accurately capture the time-series patterns of urban redevelopment.This research’s findings are useful for evaluating urban demographic and economic changes,informing policymaking and urban planning,and contributing to sustainable urban development.The model can also serve as a foundation for future research on early-stage urban redevelopment detection and evaluation of the causes and impacts of urban redevelopment. 展开更多
