Objective: We investigated the correlation between the number of circulating tumor cells(CTCs) and wholebody metabolic tumor volume(WBMTV) measured by 18 F-fluorodeoxyglucose(FDG) positron emission tomography/computed...Objective: We investigated the correlation between the number of circulating tumor cells(CTCs) and wholebody metabolic tumor volume(WBMTV) measured by 18 F-fluorodeoxyglucose(FDG) positron emission tomography/computed tomography(PET/CT).The aim was to evaluate the value of the incorporation of CTC number and WBMTV in the prognostic prediction of stage III small-cell lung cancer(SCLC).Methods: One hundred and twenty-nine patients were enrolled in this study.All patients were treated with four cycles of a platinum-based regimen and concurrent chest irradiation,followed by prophylactic cranial irradiation.Blood samples for CTC analysis were obtained from 112 patients before the initiation of chemotherapy(as a baseline),after cycle 1 and after cycle 4.CTCs were measured using the CELLSEARCH? system.The patients underwent pretreatment FDG PET/CT WBMTV,which included all malignant lesions.The Spearman rank test was used to determine the correlation among CTC counts,WBMTV and disease stage.Overall survival(OS) and progression-free survival(PFS) curves were produced using the Kaplan-Meier method,and survival differences between groups were assessed by the log-rank test.Results: The number of CTCs at baseline did not correlate with WBMTV before the initiation of therapy(P=0.241).The number of CTCs at baseline and the WBMTV before the initiation of therapy were independent relevant factors for PFS and OS.The subgroup analysis(Group A: CTC count >19.5 and a WBMTV >266.5cm~3;Group B: CTC count >19.5 and a WBMTV ≤266.5cm~3; Group C: CTC count ≤19.5 and a WBMTV >266.5cm~3;Group D: CTC count ≤19.5 and a WBMTV ≤266.5cm~3) showed that the differences were statistically significant in the median PFS(Group A vs.D,P<0.001; Group B vs.D,P=0.018; Group C vs.D,P=0.029) and in the median OS(Group A vs.D,P<0.001; Group B vs.D,P=0.012).Conclusions: CTC number and WBMTV are related to progression and death in patients with SCLC.The incorporation of CTC number and WBMTV scans can provide a detailed prognostic prediction for SCLC.展开更多

Colorectal MALT ly...Mucosa-associated lymphoid tissue (MALT) lymphoma usually originates from the stomach and presents with low ^(18)F-fluorodeoxyglucose (FDG) avidity with average maximum standard uptake value of 3.6. Colorectal MALT lymphoma is a rare entity that contributes to 1.6% of all MALT lymphomas and < 0.2% of large intestinal malignancies. The case reported herein firstly revealed stage Ⅱ MALT lymphoma with unexpected higher ^(18)F-FDG avidity of 18.9 arising at the colorectal anastomosis in a patient with a surgical history for sigmoid adenocarcinoma, which was strongly suspected as local recurrence before histopathological and immunohistochemical examinations. After accurate diagnosis, the patient received four cycles of standard R-CVP regimen (rituximab, cyclophosphamide, vincristine and prednisone), combined target therapy and chemotherapy, instead of radiotherapy recommended by National Comprehensive Cancer Network guidelines. Objective Nonalcoholic fatty liver disease(NAFLD)is a common cause of clinical liver dysfunction and an important prepathological change of liver cirrhosis.Central obesity,type 2 diabetes mellitus,dyslipidemia,and met...Objective Nonalcoholic fatty liver disease(NAFLD)is a common cause of clinical liver dysfunction and an important prepathological change of liver cirrhosis.Central obesity,type 2 diabetes mellitus,dyslipidemia,and metabolic syndrome are the major risk factors for NAFLD.Sitagliptin(Sig)is a novel hypoglycemic agent that improves blood glucose levels by increasing the level of active incretin.Sig has been shown to prevent the development of fatty livers in mice on a fructose-rich diet.The purpose of this study was to observe the efficacy of Sig on NAFLD in type 2 diabetic mice.Methods The diet-induced obesity mouse model was established,and the diabetic mice were screened by an intraperitoneal glucose tolerance trial.The mice were randomly divided into four groups for 8 weeks of intervention:high-fat diet(HFD)group,Sig group,metformin(Met)group,and Sig+Met group.After the intervention,the liver function indexes as well as the blood glucose and blood lipid levels of the mice were measured.In addition,the wet weight of the liver was measured;the pathological sections of the liver tissues were stained to observe the hepatocyte fatty degeneration,inflammation,necrosis,and fibrosis;and the hepatic histological injury was recorded as the NAFLD activity score(NAS).Results Compared with the normal control group,the body weight,liver weight,blood glucose level,insulin resistance(IR),blood lipid level,and transaminase level of the mice in the HFD group were significantly increased,showing typical metabolic syndrome.After treatment with Sig and/or Met,the mice gained less weight,had lower levels of blood glucose,triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),and transaminase,and had improved IR compared with the HFD group.The liver pathological NASs in the Sig group(P=0.01),Met group(P=0.028),and Sig+Met group(P<0.001)were lower than those in the HFD group(P<0.05),suggesting that the use of the two drugs alone or in combination can improve the state of liver inflammation.In terms of fibrosis,there was no fibrosis in the control group but there was significant fibrosis in the HFD group(P<0.001).There was no significant difference between the drug intervention groups and the HFD group,indicating that the drug therapy(Sig and/or Met)did not significantly improve the pre-existing fibrosis.Conclusion Our experiment proved that Sig can improve NAFLD,including improvement of the serum transaminase level,hepatic pathological inflammation level,and hepatocyte adiposis,suggesting that Sig may play a role by improving glucose and lipid metabolism,reducing the body weight and liver weight,improving insulin sensitivity,and inhibiting fatty liver inflammation.Sig may be a new direction for the treatment of patients with a nonalcoholic fatty liver and diabetes,delaying the progression of NAFLD.展开更多
The induced polarization relaxation time spectrum(RTS) reflects the distribution of rock pore size,which is a key factor in estimating the oil or water storage capacity of strata.However,as the data acquisition and ...The induced polarization relaxation time spectrum(RTS) reflects the distribution of rock pore size,which is a key factor in estimating the oil or water storage capacity of strata.However,as the data acquisition and transmission abilities of well logging instruments are much limited due to the underground environment,it is necessary to explore suitable sampling methods which can be used to obtain an accurate RST with less sampling data.This paper presents a uniform amplitude sampling method(UASM),and compares it with the conventional uniform time sampling method(UTSM) and logarithm time sampling method(LTSM) in terms of the adaptability to different strata,RTS inversion accuracy,and stratum vertical resolution.Numerical simulation results show that the UASM can obtain high inversion accuracy of RTS with different kinds of pore size distribution formation,with high dynamic ranges of pore size,and with a small number of sampling points.The UASM,being able to adapt to the attenuation speed of polarization curve automatically,thus has the highest vertical resolution.The inversion results of rock samples also show that the UASM is superior to the UTSM and LTSM.展开更多
F-2 toxin is an estrogenic mycotoxin that causes reproductive disorders in animals.Betulinic acid(BA)is a natural pentacyclic lupane-structure triterpenoid that has diverse pharmacological activities.In this study,the...F-2 toxin is an estrogenic mycotoxin that causes reproductive disorders in animals.Betulinic acid(BA)is a natural pentacyclic lupane-structure triterpenoid that has diverse pharmacological activities.In this study,the antioxidative and anti-inflammatory effects of BA and its underlying mechanism are explored in F-2 toxin-triggered mouse ovarian damage.We found that BA alleviated the F-2 toxin-induced ovarian impairment by stimulating follicle growth,reducing inflammatory cell infiltration,repairing damaged mitochondria and endoplasmic reticulum.Simultaneously,BA not only reversed F-2 toxin-induced reduction of follicle stimulating hormone(FSH)and luteinizing hormone(LH)levels in the serum,but also restrained the protein expression of the estrogen receptors a(ERa)and ERβ.Moreover,BA restored the balance of F-2 toxin-induced ovarian redox system disorders.Subsequently,we found that 0.25 mg/kg BA played an anti-inflammatory role in the F-2 toxin-induced ovarian impairment by decreasing interleukin-1β(IL-1β).IL-6,and tumor necrosis factor-α(TNF-α)mRNA expression,as well as inhibiting p38 protein expression.These data demonstrated that BA exerts its protective effect on F-2 toxin-induced ovarian oxidative impairment and inflammation by inhibiting p38 expression,which implies a natural product-based medicine to ameliorate F-2 toxin-caused female reproductive toxicity and provides a detoxifying method for food contaminated by mycotoxin.展开更多
Wheat is an important cereal crop used to produce diverse and popular food worldwide because of its high grain yield(GY)and grain protein content(GPC).However,GY and GPC are usually negatively correlated.We previously...Wheat is an important cereal crop used to produce diverse and popular food worldwide because of its high grain yield(GY)and grain protein content(GPC).However,GY and GPC are usually negatively correlated.We previously reported that favorable alleles of the wheat domestication gene Q can synchronously increase GY and GPC,but the underlying mechanisms remain largely unknown.In this study,we investigate the regulatory network involving Q associated with GY and GPC in young grains through DNA affinity purification sequencing and transcriptome sequencing analyses,electrophoretic mobility shift and dualluciferase assays,and transgenic approaches.Three Q-binding motifs,namely TTAAGG,AAACA[A/T]A,and GTAC[T/G]A,are identified.Notably,genes related to photosynthesis or carbon and nitrogen metabolism are enriched and regulated by Q.Moreover,Q is revealed to bind directly to its own gene and the glutamine synthetase gene GSr-4D to increase expression,thereby influencing nitrogen assimilation during the grain filling stage and increasing GPC.Considered together,our findings provide molecular evidence of the positive regulatory effects of Q on wheat GY and GPC.展开更多
Cytoskeletal proteins are involved in neuronal survival.Brain-derived neurotrophic factor can increase expression of cytoskeletal proteins during regeneration after axonal injury.However,the effect of neural stem cell...Cytoskeletal proteins are involved in neuronal survival.Brain-derived neurotrophic factor can increase expression of cytoskeletal proteins during regeneration after axonal injury.However,the effect of neural stem cells genetically modified by brain-derived neurotrophic factor transplantation on neuronal survival in the injury site still remains unclear.To examine this,we established a rat model of traumatic brain injury by controlled cortical impact.At 72 hours after injury,2 × 10~7 cells/m L neural stem cells overexpressing brain-derived neurotrophic factor or naive neural stem cells(3 m L) were injected into the injured cortex.At 1–3 weeks after transplantation,expression of neurofilament 200,microtubule-associated protein 2,actin,calmodulin,and beta-catenin were remarkably increased in the injury sites.These findings confirm that brain-derived neurotrophic factor-transfected neural stem cells contribute to neuronal survival,growth,and differentiation in the injury sites.The underlying mechanisms may be associated with increased expression of cytoskeletal proteins and the Wnt/β-catenin signaling pathway.展开更多
AIM:To evaluate the value of positron emission tomography(PET)/computerized tomography(CT)in surveillance of colorectal cancer(CRC)patients with different carcinoembryonic antigen(CEA)concentrations.METHODS:One hundre...AIM:To evaluate the value of positron emission tomography(PET)/computerized tomography(CT)in surveillance of colorectal cancer(CRC)patients with different carcinoembryonic antigen(CEA)concentrations.METHODS:One hundred and six postoperative CRC patients who had suspected recurrence or metastasis and received fluorodeoxyglucose(FDG)PET/CT within one week were included in this study.The final diagnosis was confirmed by histological examination or clinicalfollow-up over at least six months.RESULTS:The sensitivity,specificity,and accuracy of FDG PET/CT were 95.2%,82.6%,and 92.5%,and94.8%,81.4%and 92.8%,respectively,in the caseand lesion-based analyses.The sensitivity and accuracy of FDG PET/CT significantly differed from CT in both analyses(χ2=8.186,P=0.004;χ2=6.201,P=0.013;χ2=13.445,P=0.000;χ2=11.194,P=0.001).In the lesion-based analysis,the sensitivity,specificity,and accuracy of FDG PET/CT in the abnormal CEA group were97.8%,82.6%,and 95.6%,compared with 81.3%,80%,and 80.6%for patients with normal CEA levels.In case-based analysis,the sensitivity,specificity,and accuracy of FDG PET/CT were 97.2%,77.8%,and 95%in abnormal CEA group.Only in lesion-based analysis,the sensitivity and accuracy of FDG PET/CT in the abnormal CEA group were significantly superior to those in the normal CEA group(χ2=6.432,P=0.011;χ2=7.837,P=0.005).FDG PET/CT changed the management in 45.8%of patients with positive scans.CONCLUSION:FDG PET/CT showed superior diagnostic value and is an advisable option in surveillance of postoperative CRC patients with a vague diagnosis.展开更多

Therefore, it may become an effective means for monitoring the anesthetic degree of anesthetic drug in the operation. OBJECTIVE: To observe the dose-effect relationship of propofol influencing β2-band power of quantitative pharmaco-electroencephalography in rabbits. DESIGN: A randomized block design. SETTING: Department of Anesthesiology, Xuzhou Medical College. MATERIALS: Thirty-six healthy adult rabbits of either gender, weighing (2.4 ± 0.5 ) kg, of clean grade, were provided by the Laboratory Animal Center of Xuzhou Medical College. The involved rabbits were randomly divided into 3 groups with 12 in each by table of random digit: high-dose propofol group, moderate-dose propofol group, and low-dose propofol group. The protocol was carried out in accordance with animal ethics guidelines for the use and care of animals. METHODS: This study was carried out in the Department of Anesthesiology, Xuzhou Medical College between August 1999 and April 2000. Rabbits in the high-dose propofol group, moderate-dose propofol group, and low-dose propofol group were injected with 10, 5 and 2.5 mg/kg propofol (ZENECA Company, British, Batch No. 032000), respectively. Before and after intravenous administration of propofol, percentage of β2-band power of quantitative pharmaco-electroencephalography was measured, and the latent and persistent periods when rabbit righting reflex disappeared were observed by quantitative pharmaco-electroencephalography and power spectrum analysis. MAIN OUTCOME MEASURES: (1) Percentage of β2-band power of quantitative pharmaco-electroencephalography. (2)Latent period and persistent period of abolition of righting reflex of rabbits. RESULTS: Thirty-six rabbits were involved in the final analysis. (1)Effect of propofol on righting reflex of rabbits: Righting reflex disappeared within 1 minute after the rabbits being intravenously injected with propofol. The higher dose, the shorter latent period (r = - 0.94, P 〈 0.01), and the longer persistent period (r =0.79, P 〈 0.01). Both latent period and persistent period had good correlation with propofol dose. (2) Effect of propofol on the percentage of β2-band power of quantitative pharmaco-electroencephalography: In the low-dose propofol group, no significant changes in the percentage of β2-band power of quantitative pharmaco-electroencephalography existed between before and after administration within 30 minutes (P 〉 0.05). In the moderate-dose propofol group, the percentage of β2-band power of quantitative pharmaco-electroencephalography in each brain region except for left and right frontal areas was significantly decreased within 30 s to 5 minutes (P 〈 0.05), and recovered to the level before administration 10 minutes later. In the high-dose propofol group, the percentage of β2-band power of quantitative pharmaco-electroencephalography in each brain region was significantly decreased from 20 s to 5 minutes after administration (P 〈 0.05 - 0.01), and that was gradually recovered to the level before administration 5 to 10 minutes after administration. This tendency was basically the same as the changes of latent period and persistent period. CONCLUSION: Propofol decreases the percentage of β2-band power of quantitative pharmacoelectroencephalography in dose-dependent manner, It indicates that β2-band power might become one of indexes for reflecting the anesthetic degree of propofol,展开更多

文摘F-2 toxin is an estrogenic mycotoxin that causes reproductive disorders in animals.Betulinic acid(BA)is a natural pentacyclic lupane-structure triterpenoid that has diverse pharmacological activities.In this study,the antioxidative and anti-inflammatory effects of BA and its underlying mechanism are explored in F-2 toxin-triggered mouse ovarian damage.We found that BA alleviated the F-2 toxin-induced ovarian impairment by stimulating follicle growth,reducing inflammatory cell infiltration,repairing damaged mitochondria and endoplasmic reticulum.Simultaneously,BA not only reversed F-2 toxin-induced reduction of follicle stimulating hormone(FSH)and luteinizing hormone(LH)levels in the serum,but also restrained the protein expression of the estrogen receptors a(ERa)and ERβ.Moreover,BA restored the balance of F-2 toxin-induced ovarian redox system disorders.Subsequently,we found that 0.25 mg/kg BA played an anti-inflammatory role in the F-2 toxin-induced ovarian impairment by decreasing interleukin-1β(IL-1β).IL-6,and tumor necrosis factor-α(TNF-α)mRNA expression,as well as inhibiting p38 protein expression.These data demonstrated that BA exerts its protective effect on F-2 toxin-induced ovarian oxidative impairment and inflammation by inhibiting p38 expression,which implies a natural product-based medicine to ameliorate F-2 toxin-caused female reproductive toxicity and provides a detoxifying method for food contaminated by mycotoxin.

文摘Wheat is an important cereal crop used to produce diverse and popular food worldwide because of its high grain yield(GY)and grain protein content(GPC).However,GY and GPC are usually negatively correlated.We previously reported that favorable alleles of the wheat domestication gene Q can synchronously increase GY and GPC,but the underlying mechanisms remain largely unknown.In this study,we investigate the regulatory network involving Q associated with GY and GPC in young grains through DNA affinity purification sequencing and transcriptome sequencing analyses,electrophoretic mobility shift and dualluciferase assays,and transgenic approaches.Three Q-binding motifs,namely TTAAGG,AAACA[A/T]A,and GTAC[T/G]A,are identified.Notably,genes related to photosynthesis or carbon and nitrogen metabolism are enriched and regulated by Q.Moreover,Q is revealed to bind directly to its own gene and the glutamine synthetase gene GSr-4D to increase expression,thereby influencing nitrogen assimilation during the grain filling stage and increasing GPC.Considered together,our findings provide molecular evidence of the positive regulatory effects of Q on wheat GY and GPC.

文摘Cytoskeletal proteins are involved in neuronal survival.Brain-derived neurotrophic factor can increase expression of cytoskeletal proteins during regeneration after axonal injury.However,the effect of neural stem cells genetically modified by brain-derived neurotrophic factor transplantation on neuronal survival in the injury site still remains unclear.To examine this,we established a rat model of traumatic brain injury by controlled cortical impact.At 72 hours after injury,2 × 10~7 cells/m L neural stem cells overexpressing brain-derived neurotrophic factor or naive neural stem cells(3 m L) were injected into the injured cortex.At 1–3 weeks after transplantation,expression of neurofilament 200,microtubule-associated protein 2,actin,calmodulin,and beta-catenin were remarkably increased in the injury sites.These findings confirm that brain-derived neurotrophic factor-transfected neural stem cells contribute to neuronal survival,growth,and differentiation in the injury sites.The underlying mechanisms may be associated with increased expression of cytoskeletal proteins and the Wnt/β-catenin signaling pathway.
文摘AIM:To evaluate the value of positron emission tomography(PET)/computerized tomography(CT)in surveillance of colorectal cancer(CRC)patients with different carcinoembryonic antigen(CEA)concentrations.METHODS:One hundred and six postoperative CRC patients who had suspected recurrence or metastasis and received fluorodeoxyglucose(FDG)PET/CT within one week were included in this study.The final diagnosis was confirmed by histological examination or clinicalfollow-up over at least six months.RESULTS:The sensitivity,specificity,and accuracy of FDG PET/CT were 95.2%,82.6%,and 92.5%,and94.8%,81.4%and 92.8%,respectively,in the caseand lesion-based analyses.The sensitivity and accuracy of FDG PET/CT significantly differed from CT in both analyses(χ2=8.186,P=0.004;χ2=6.201,P=0.013;χ2=13.445,P=0.000;χ2=11.194,P=0.001).In the lesion-based analysis,the sensitivity,specificity,and accuracy of FDG PET/CT in the abnormal CEA group were97.8%,82.6%,and 95.6%,compared with 81.3%,80%,and 80.6%for patients with normal CEA levels.In case-based analysis,the sensitivity,specificity,and accuracy of FDG PET/CT were 97.2%,77.8%,and 95%in abnormal CEA group.Only in lesion-based analysis,the sensitivity and accuracy of FDG PET/CT in the abnormal CEA group were significantly superior to those in the normal CEA group(χ2=6.432,P=0.011;χ2=7.837,P=0.005).FDG PET/CT changed the management in 45.8%of patients with positive scans.CONCLUSION:FDG PET/CT showed superior diagnostic value and is an advisable option in surveillance of postoperative CRC patients with a vague diagnosis.

文摘BACKGROUND Conservative treatments have been reported to diminish or resolve clinical symptoms of lumbar intervertebral disc herniation(LIDH)within a few weeks.CASE SUMMARY Computed tomography and magnetic resonance imaging(MRI)of the lumbar region of a 25-yearold male diagnosed with LIDH showed prolapse of the L5/S2 disc.The disc extended 1.0 cm beyond the vertebral edge and hung along the posterior vertebral edge.The patient elected a conservative treatment regimen that included traditional Chinese medicine(TCM),acupuncture,and massage.During a follow-up period of more than 12 mo,good improvement in pain was reported without complications.MRI of the lumbar region after 12 mo showed obvious reabsorption of the herniation.CONCLUSION A conservative treatment regimen of TCM,acupuncture,and massage promoted reabsorption of a prolapsed disc.

文摘BACKGROUND: Quantitative pharmaco-electroencephalography can be used for studying the dose-effect and time-effect relationships of drugs affecting central nervous system. Therefore, it may become an effective means for monitoring the anesthetic degree of anesthetic drug in the operation. OBJECTIVE: To observe the dose-effect relationship of propofol influencing β2-band power of quantitative pharmaco-electroencephalography in rabbits. DESIGN: A randomized block design. SETTING: Department of Anesthesiology, Xuzhou Medical College. MATERIALS: Thirty-six healthy adult rabbits of either gender, weighing (2.4 ± 0.5 ) kg, of clean grade, were provided by the Laboratory Animal Center of Xuzhou Medical College. The involved rabbits were randomly divided into 3 groups with 12 in each by table of random digit: high-dose propofol group, moderate-dose propofol group, and low-dose propofol group. The protocol was carried out in accordance with animal ethics guidelines for the use and care of animals. METHODS: This study was carried out in the Department of Anesthesiology, Xuzhou Medical College between August 1999 and April 2000. Rabbits in the high-dose propofol group, moderate-dose propofol group, and low-dose propofol group were injected with 10, 5 and 2.5 mg/kg propofol (ZENECA Company, British, Batch No. 032000), respectively. Before and after intravenous administration of propofol, percentage of β2-band power of quantitative pharmaco-electroencephalography was measured, and the latent and persistent periods when rabbit righting reflex disappeared were observed by quantitative pharmaco-electroencephalography and power spectrum analysis. MAIN OUTCOME MEASURES: (1) Percentage of β2-band power of quantitative pharmaco-electroencephalography. (2)Latent period and persistent period of abolition of righting reflex of rabbits. RESULTS: Thirty-six rabbits were involved in the final analysis. (1)Effect of propofol on righting reflex of rabbits: Righting reflex disappeared within 1 minute after the rabbits being intravenously injected with propofol. The higher dose, the shorter latent period (r = - 0.94, P 〈 0.01), and the longer persistent period (r =0.79, P 〈 0.01). Both latent period and persistent period had good correlation with propofol dose. (2) Effect of propofol on the percentage of β2-band power of quantitative pharmaco-electroencephalography: In the low-dose propofol group, no significant changes in the percentage of β2-band power of quantitative pharmaco-electroencephalography existed between before and after administration within 30 minutes (P 〉 0.05). In the moderate-dose propofol group, the percentage of β2-band power of quantitative pharmaco-electroencephalography in each brain region except for left and right frontal areas was significantly decreased within 30 s to 5 minutes (P 〈 0.05), and recovered to the level before administration 10 minutes later. In the high-dose propofol group, the percentage of β2-band power of quantitative pharmaco-electroencephalography in each brain region was significantly decreased from 20 s to 5 minutes after administration (P 〈 0.05 - 0.01), and that was gradually recovered to the level before administration 5 to 10 minutes after administration. This tendency was basically the same as the changes of latent period and persistent period. CONCLUSION: Propofol decreases the percentage of β2-band power of quantitative pharmacoelectroencephalography in dose-dependent manner, It indicates that β2-band power might become one of indexes for reflecting the anesthetic degree of propofol,
