目的:探讨鼠李糖(rhamnose,Rha)修饰对曲妥珠单抗(trastuzumab,Tra)Fc效应功能的影响。方法:将二硫键还原的Tra分子与含交联剂的Rha衍生物分子偶联,构建Tra-Rha偶联物。使用聚丙烯酰胺凝胶电泳、免疫印迹和高效液相分子排阻色谱对偶联... 目的:探讨鼠李糖(rhamnose,Rha)修饰对曲妥珠单抗(trastuzumab,Tra)Fc效应功能的影响。方法:将二硫键还原的Tra分子与含交联剂的Rha衍生物分子偶联,构建Tra-Rha偶联物。使用聚丙烯酰胺凝胶电泳、免疫印迹和高效液相分子排阻色谱对偶联物进行表征,并通过流式细胞术评估偶联物的HER2抗原亲和水平,免疫荧光和流式细胞术评估偶联物的抗Rha抗体募集能力。最后,通过在体外检测偶联物介导的补体依赖的细胞毒作用(complement-dependent cytotoxicity,CDC)和抗体依赖细胞介导的吞噬作用(antibody-dependent cell-mediated phagocytosis,ADCP),评估其Fc效应功能的增强水平。结果:成功构建了一款Tra-Rha偶联物。亲和实验和抗体募集实验表明Tra-Rha偶联物保留了良好的HER2抗原结合能力,并能将高水平抗Rha抗体选择性募集至靶细胞表面。CDC实验显示,Tra-Rha偶联物可介导显著的CDC效应对HER2阳性的SKBR3和SKOV3细胞进行杀伤,最高杀伤率分别为75%和70%,而相同条件下未修饰Rha的Tra则不能介导CDC。ADCP实验则显示,在SKBR3和SKOV3细胞中,Tra-Rha介导的ADCP水平较Tra分别提高了2.3倍和1.2倍。结论:Rha修饰可作为一种经济高效的策略,用于同时增强Tra的多种Fc效应功能,进而改善Tra的治疗指数。 展开更多

【目的】探究加压滴灌系统灌水时间及轮灌组优化模型。【方法】通过水力解析的方法,分析轮灌制度参数与管网主要水力性能参数的关系,提出了灌水量偏差最小为目标的数学模型,考虑了灌水时间及轮灌周期约束、滴头压力约束以及轮灌组流量... 【目的】探究加压滴灌系统灌水时间及轮灌组优化模型。【方法】通过水力解析的方法,分析轮灌制度参数与管网主要水力性能参数的关系,提出了灌水量偏差最小为目标的数学模型,考虑了灌水时间及轮灌周期约束、滴头压力约束以及轮灌组流量等约束,采用遗传算法求解模型。【结果】在同一工程案例中,与W1方案相比,W2、W3方案滴灌系统完成1次灌水过程灌水量偏差率分别降低27%、37%,缩短滴灌系统1次灌水延续时间约15、20 h,节约用电能耗15%、20%,提高水资源利用效率约14%、19%,减少作物生育期所水费80、111元/hm^(2),电费72~86、100~120元/hm^(2)。【结论】基于传统滴灌工程设计采用加压滴灌系统灌水时间及轮灌组优化模型,可降低滴灌系统灌水量偏差率,缩短滴灌系统1次灌水延续时间,减少作物生育期水费和电费。 展开更多

Tripterygium hypoglaucum(Levl.)Hutch,a traditional Chinese medicinal herb with a long history of use,is widely distributed in China.One of its main active components,celastrol,has great potential to be developed into anti-cancer and anti-obesity drugs.Although it exhibits strong pharmacological activities,there is a lack of sustainable sources of celastrol and its derivatives,making it crucial to develop novel sources of these drugs through synthetic biology.The key step in the biosynthesis of celastrol is considered to be the cyclization of 2,3-oxidosqualene into friedelin under the catalysis of 2,3-oxidosqualene cyclases.Friedelin was speculated to be oxidized into celastrol by cytochrome P450 oxidases(CYP450s).Here,we reported a cytochrome P450 ThCYP712K1 from Tripterygium hypoglaucum(Levl.)Hutch that catalyzed the oxidation of friedelin into polpuonic acid when heterologously expressed in yeast.Through substrate supplementation and in vitro enzyme analysis,ThCYP712K1 was further proven to catalyze the oxidation of friedelin at the C-29 position to produce polpunonic acid,which is considered a vital step in the biosynthesis of celastrol,and will lay a foundation for further analysis of its biosynthetic pathway. 展开更多

It is desired that the model can be well applicable to conventional diamond turningand the model may be employed as a tool in the design of diamond tools. This approach is quitedifferent from traditional investigations primarily based on empirical studies. As the depth of cutbecomes the same order as the rounded cutting edge radius, sliding along the clearance face due toelastic recovery of workpiece material and plowing due to the rounded cutting edge may becomeimportant in micro-machining, the forces acting on the cutting edge rounding and the clearance facecan not be neglected. Effect of Stratified Nursing Model on Nutritional Status and Quality of Life of Patients with Uremia Undergoing Hemodialysis

Objective: to carry out stratified nursing care for uremic patients undergoing hemodialysis, and analyze the effect of nursing care on nutritional status and quality of life of patients. Methods: a total of 80 patients admitted in this study were uremic patients treated with hemodialysis in Tongji hospital from May 2020 to May 2021. All patients were equally divided into two groups according to the ball-touching grouping method: observation group (40 cases) and control group (40 cases). Patients in the control group received routine care during the treatment, while patients in the observation group received stratified care during the treatment. After completing the nursing, the data of the two groups of patients were collected, including the patients' nutritional status, emotional status, quality of life and patients' compliance with the treatment. Results: the nutritional status of the patients in the observation group was significantly higher than that of the patients in the control group (P < 0.05). The scores of emotional state indexes in the observation group were significantly lower than those in the control group (P < 0.05). The quality of life index of the observation group was significantly higher than that of the control group (P < 0.05). The total treatment compliance of the observation group was 97.5%, which was higher than 82.5% of the control group (P < 0.05). Conclusion: layered nursing model can help uremic patients to improve their quality of life, and is worthy of promotion. 展开更多

Objective: to explore the clinical application value of hemiplegic rehabilitation decoction in the rehabilitation of acute cerebral infarction. Methods: 86 patients with acute cerebral infarction were randomly divided into the same number of control group and study group (n 43). They were treated with conventional western medicine and additional hemiplegic rehabilitation decoction, respectively. The therapeutic conditions of the two groups were observed and compared. Results: compared with the control group, the effective rate of the study group was higher (P < 0.05). Before and after treatment, the increase of BI score, NIHSS score, TCM syndrome score and the level of inflammatory factors in the study group decreased significantly (P < 0.05). Conclusion: the application of hemiplegic rehabilitation decoction during the rehabilitation treatment of patients with acute cerebral infarction can achieve ideal effect, improve the neurological function defects of patients and enhance their daily living ability, which is worth promoting and applying. 展开更多
