As a core course for the Engineering Cost major at Chongqing Institute of Engineering,Engineering Mechanics shoulders the dual mission of cultivating students’mechanical thinking,engineering practical abilities,and professional qualities.Centered on the deep integration of the outcome-based education(OBE)concept and curriculum-based ideological and political education,this paper systematically elaborates on the reform paths and implementation strategies from four dimensions-reconstruction of teaching content,innovation of assessment mechanisms,integration of ideological and political elements,and construction of teaching resources-by drawing on the teaching reform practices of Engineering Mechanics courses at multiple universities.Through modular teaching content design,a competency-oriented diversified assessment system,and a method of integrating ideological and political education with local characteristics and engineering case studies,the teaching objectives of“knowledge transmission,ability cultivation,and value shaping”are achieved in a three-in-one manner.Practical results indicate that after the reform,students’engineering practical abilities have significantly improved,with a course objective achievement rate exceeding 0.73,a 23%increase in student satisfaction with the course,and a 35%year-on-year increase in the number of awards won in professional competitions,providing a replicable paradigm for the teaching reform of engineering courses. 展开更多

Rechargeable zinc-ion batteries have emerged as one of the most promising candidates for large-scale energy storage applications due to their high safety and low cost.However,the use of Zn metal in batteries suffers from many severe issues,including dendrite growth and parasitic reactions,which often lead to short cycle lives.Herein,we propose the construction of functional organic interfacial layers(OIL)on the Zn metal anodes to address these challenges.Through a well-designed organic-assist pre-construction process,a densely packed artificial layer featuring the immobilized zwitterionic molecular brush can be constructed,which can not only efficiently facilitate the smooth Zn plating and stripping,but also introduce a stable environment for battery reactions.Through density functional theory calculations and experimental characterizations,we verify that the immobilized organic propane sulfonate on Zn anodes can significantly lower the energy barrier and increase the kinetics of Zn^(2+)transport.Thus,the Zn metal anode with the functional OIL can significantly improve the cycle life of the symmetric cell to over 3500 h stable operation.When paired with the H_(2)V_(3)O_(8)cathode,the aqueous Zn-ion full cells can be continuously cycled over 7000 cycles,marking an important milestone for Zn anode development for potential industrial applications. 展开更多

Treatment of malignant biliary obstruction by combined percutaneous transhepatic biliary drainage with local tumor treatment 被引量:31

AIM: To evaluate the utility of local tumor therapy combined with percutaneous transhepatic bilian/drainage (PTBD) for malignant obstructive bilian/disease. METHODS: A total of 233 patients with malignant billan/obstruction were treated in our hospital with PTBD by placement of metallic stents and/or plastic tubes. After PTBD, 49 patients accepted brachytherapy or extraradiation therapy or arterial infusion chemotherapy. The patients were followed up with clinical and radiographic evaluation. The survival and stent patency rate were calculated by Kaplan-Meier survival analysis. RESULTS: Twenty-two patients underwent chemotherapy (11 cases of hepatic carcinoma, 7 cases of pancreatic carcinoma, 4 cases of metastatic lymphadenopathy), and 14 patients received radiotherapy (10 cases of cholangiocardnoma, 4 cases of pancreatic carcinoma), and 13 patients accepted brachytherapy (7 cases of cholangiocarcinoma, 3 cases of pancreatic carcinoma, 4 cases of metastatic lymphadenopathy). The survival rate of the local tumor treatment group at 1, 3, 6, and 12 months was 97.96%, 95.92%, 89.80%, and 32.59% respectively, longer than that of the non treatment group. The patency rate at 1, 3, 6, and 12 months was 97.96%, 93.86%, 80.93%, and 56.52% respectively. The difference of patency rate was not significant between treatment group and non treatment group. CONCLUSION: Our results suggest that local tumor therapy could prolong the survival time of patients with malignant biliary obstruction, and may improve stent patency. 展开更多

Accurate liver injury assessment is essential for clinical evidence-based treatment. Liver biopsy is considered the gold standard method to stage liver disease, but it is not widely used in resource-limited settings. Therefore, non-invasive liquid biopsy tests are needed. AIM To assess liver injury in hepatitis B patients using quantified cell free DNA combined with other serum biomarker as a liquid biopsy-based method. METHODS A cohort of 663 subjects including 313 hepatitis B patients and 350 healthy controls were enrolled. Ultrasound-guided liver biopsies followed by histopathological assessments were performed for the 263 chronic hepatitis B patients to determine the degree of liver injury. Cell-free DNA was quantified using a novel duplex real-time polymerase chain reaction assay. RESULTS Compared with healthy controls, patients with hepatitis B virus (HBV) infection had significantly higher plasma DNA, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), bilirubin, and HBV DNA levels (P<0.01). Serum ALT, AST, bilirubin, and plasma DNA levels of patients with markedsevere inflammation were significantly higher than those with mild-moderate inflammation (P<0.01). There was a statistically significant correlation between hepatocyte inflammation severity and serum bilirubin (R^2=0.673, P<0.01) or plasma DNA (R^2=0.597, P<0.01) levels. The areas under the curves of serum ALT, bilirubin, plasma DNA, and their combination to distinguish between patients with mild–moderate and marked-severe inflammation were 0.8059, 0.7910, 0.7921, and 0.9564, respectively. CONCLUSION The combination of plasma DNA, serum ALT, and bilirubin could be a candidate liquid biopsy for non-invasive assessment of liver injury in hepatitis B patients. 展开更多

OBJECTIVE: To analyze the association between tongue manifestations and the levels of glucose (GLU), total cholesterol(TCH), and high-density lipoprotein cholesterol(HDL-C) in subjects with acute cerebral infarction.METHODS: Hospitalized patients with first unilateral cerebral infarction in the Neurological Department of Xuanwu Hospital were included and the correlation between tongue fur color, fur nature,and the levels of GLU,TCH, HDL-C were analyzed.RESULTS: HDL level in the thin fur group was higher than that in the thick fur group(P=0.02). The difference in the levels of GLU,TCH, and HDL-C among the groups was significant(P<0.05), classified in terms of slippery, moist, and dry fur. Further comparison between the groups by Student-Newman-Keuls test showed that GLU level in the dry fur group was the highest. Moreover, the TCH level in the slippery fur group was higher than the other two groups.CONCLUSION: A correlation between tongue manifestations and GLU, TCH, HDL-C was identified in the patients with acute cerebral infarction. 展开更多

碳酸盐岩储层经历了复杂的沉积、成岩和构造作用,内部发育着不同尺度的孔、洞、缝结构,往往表现出强烈的非均质性,常规力学实验难以全面评估其内部结构对岩石力学参数的影响。为此,以四川盆地震旦系灯影组灯四段藻白云岩为例,利用微米C... 碳酸盐岩储层经历了复杂的沉积、成岩和构造作用,内部发育着不同尺度的孔、洞、缝结构,往往表现出强烈的非均质性,常规力学实验难以全面评估其内部结构对岩石力学参数的影响。为此,以四川盆地震旦系灯影组灯四段藻白云岩为例,利用微米CT扫描和纳米压痕实验,系统表征了孔洞、藻凝块及白云石的微观结构特征和力学属性,然后结合均匀化理论提出了一种藻白云岩的岩石力学性质跨尺度评价方法,并揭示了白云石与藻凝块在力学性能上的差异及其对宏观力学参数的影响规律。研究结果表明:①白云石矿物展现出更高的微观强度、更低的塑性变形以及更强的抗裂缝扩展能力,而藻凝块的微观力学参数具有更强的分散性;②白云石与藻凝块在微观层面的弹性模量、硬度及断裂韧性之间呈现良好的线性相关性,通过均匀化方法预测得到的藻白云岩宏观弹性模量和泊松比分别是单轴实验测得值的1.90和1.39倍;③藻白云岩弹性模量随孔洞发育程度增加逐渐降低,其预测范围逐渐收窄,而泊松比与之相反,孔洞纵横比和微裂纹进一步控制宏观力学性质,当孔洞纵横比为0.087~0.202或裂缝线密度为0.30~0.63条/m时,均匀化预测结果与实验值一致。结论认为,该方法不仅避免了传统岩心采集中成本高昂和实验重复性差的问题,还有效降低了传统均匀化方法导致的高估现象,为复杂碳酸盐岩储层的精细力学参数表征提供了新思路,为复杂储层的力学建模与参数预测奠定了理论基础。 展开更多

Lithium–sulfur(Li–S)batteries are supposed to be one of the most potential next-generation batteries owing to their high theoretical capacity and low cost.Nevertheless,the shuttle effect of firm multi-step two-electron reaction between sulfur and lithium in liquid electrolyte makes the capacity much smaller than the theoretical value.Many methods were proposed for inhibiting the shuttle effect of polysulfide,improving corresponding redox kinetics and enhancing the integral performance of Li–S batteries.Here,we will comprehensively and systematically summarize the strategies for inhibiting the shuttle effect from all components of Li–S batteries.First,the electrochemical principles/mechanism and origin of the shuttle effect are described in detail.Moreover,the efficient strategies,including boosting the sulfur conversion rate of sulfur,confining sulfur or lithium polysulfides(LPS)within cathode host,confining LPS in the shield layer,and preventing LPS from contacting the anode,will be discussed to suppress the shuttle effect.Then,recent advances in inhibition of shuttle effect in cathode,electrolyte,separator,and anode with the aforementioned strategies have been summarized to direct the further design of efficient materials for Li–S batteries.Finally,we present prospects for inhibition of the LPS shuttle and potential development directions in Li–S batteries. 展开更多

The loss-of-function mutations in the ALPL result in hypophosphatasia(HPP), an inborn metabolic disorder that causes skeletal mineralization defects. In adults, the main clinical features are early loss of primary or secondary teeth, osteoporosis, bone pain,chondrocalcinosis, and fractures. However, guidelines for the treatment of adults with HPP are not available. Here, we show that ALPL deficiency caused a reduction in intracellular Ca2+ influx, resulting in an osteoporotic phenotype due to downregulated osteogenic differentiation and upregulated adipogenic differentiation in both human and mouse bone marrow mesenchymal stem cells(BMSCs). Increasing the intracellular level of calcium in BMSCs by ionomycin treatment rescued the osteoporotic phenotype in alpl+/- mice and BMSC-specific(Prrx1-alpl-/-) conditional alpl knockout mice. Mechanistically, ALPL was found to be required for the maintenance of intracellular Ca2+ influx, which it achieves by regulating L-type Ca2+ channel trafficking via binding to the α2δsubunits to regulate the internalization of the L-type Ca2+ channel. Decreased Ca2+ flux inactivates the Akt/GSK3β/β-catenin signaling pathway, which regulates lineage differentiation of BMSCs. This study identifies a previously unknown role of the ectoenzyme ALPL in the maintenance of calcium channel trafficking to regulate stem cell lineage differentiation and bone homeostasis. Accelerating Ca2+ flux through L-type Ca2+ channels by ionomycin treatment may be a promising therapeutic approach for adult patients with HPP. 展开更多

The safety issues and lower energy density of the lithium metal batteries are the two main challenges that hinder their applications in the fields of electric vehicles and portable devices.In this work,the semi-interpenetrated polyvinylidene fluoride-hexafluoropropylene(PVdF-HFP)-based gel polymer electrolyte was synthesized through UV-curing method by employing the ethoxylated trimethylolpropane triacrylate(ETPTA)monomer.The semi-interpenetrating networks formed by polymerization of ETPTA and the high liquid absorption rate of the PVdF-HFP impart the as-prepared electrolyte with a high room temperature ionic conductivity of 3.17×10-3 s cm^(-1)and a high mechanical strength of 3.46 MPa.LiFePO4 was selected as cathode materials,and the active material loading of the cathode is about 4.2 mg cm-2.The electrolyte shows superior long-term cycling properties(127 mAh g^(-1)after 200 cycles at 0.5 C),excellent rate performance(113 mAh g^(-1)at1 C,80 mAh g^(-1)at 2 C,and the discharge capacity of 135 mAh g^(-1)can be restored when the rate goes back to 0.1 C)as well as good ability to inhibit the growth of lithium dendrite(about 150 h).The facile synthesis strategy and great electrochemical performance of the electrolyte make it a potential candidate for lithium metal batteries. 展开更多

The role of autologous hematopoietic stem cell transplantation(auto-HSCT)following high-dose chemotherapy has been validated and accepted as a standard treatment for patients with relapsed diffuse large B-cell lymphoma(DLBCL).However,its clinical efficacy as frontline therapy remains to be elucidated.This study aimed to examine the feasibility of frontline auto-HSCT for newly diagnosed intermediate/high-risk DLBCL patients.We retrospectively reviewed the data of 223 patients treated with frontline auto-HSCT or chemotherapy alone(year 2008-2014)from four hospitals.The median follow-up time was 29.4 months.Between the two treatment arms among the intermediate/high-risk DLBCL patients,the 3-year overall survival(OS)and progression-free survival(PFS)rates of patients given frontline auto-HSCT were 87.6%and 81.9%,respectively,and the chemotherapy-alone group showed 3-year OS and PFS rates of 64.9%and 59.59%,respectively.Compared with the chemotherapy-alone group,the frontline auto-HSCT could eliminate the adverse impact of non-germinal center B-cell(GCB)type.In addition,in the frontline auto-HSCT group,patients who achieved complete response(CR)at auto-HSCT had a longer survival time than those who did not achieve CR.Our results suggested that frontline auto-HSCT could improve the prognosis of intennediate/high-risk DLBCL patients. 展开更多

Acute myeloid leukemia(AML) is a heterogeneous disease characterized by the accu.mulation of immature myeloid progenitor cells in the bone marrow,compromising of normal hematopoi.esis and ultimately resulting in bone marrow failure.Chemotherapy is the mainstay treatment for all AML patients,however,drug resistance and clinical relapse limits its efficacy.The 5-year survival rate of AML patients is only 26.6%.Survival rates are even lower among patients ages 65 to 74 years(5.3%) and 75 years or older(1.6%).Therefore,exploring novel therapeutic agents is urgent for improving the outcome of patients with AML.Saponins are amphipathic glycosides found in traditional Chinese medicines.In the present study,we isolated a panel of saponins from Paris forrestii(Takht.) H.Li,a unique plant found in Tibet and Yunnan provinces,China.By examining their activities in suppressing acute myeloid leukemia cell proliferation,total saponins from Paris forrestii(TSPf) displayed more potent activity than individual ones.TSPf induced more than 40% AML cell apoptosis within 24 h and decreased the viability of all leukemia cell lines.TSPf-induced apoptosis was confirmed by both Annexin V staining and caspase-3 activation.TSPf downregulated pro-survival proteins Mcl-1,Bcl-xL and Bcl-2,but upreg.ulated the expression of tumor suppressor proteins p53,p27,Bax and Beclin 1.The AKT/mTOR signaling pathway is frequently over activated in various AML cells,and TSPf was found to suppress the activa.tion of both AKT and mTOR,but had no effects on their total protein expression.This was further con.firmed by the inactivation of 4 EBP-1 and p70 S6 K,two typical downstream signal molecules in the AKT/mTOR pathway.More specifically,TSPf-inactivated AKT/mTOR signaling was found to be associated with downregulated RNF6,a recently identified oncogene in AML.RNF6 activated AKT/mTOR,and consistently,knockdown of RNF6 led to inactivation of the AKT/mTOR pathway.Furthermore,TSPf suppressed the growth of AML xenografts in nude mice models.Oral administration of 100 mg · kg^(-1) body weight almost fully suppressed tumor growth within 14 d,without gross toxicity.This study thus demonstrated that TSPf displays potent anti-AML activity by suppressing the RNF6/AKT/mTOR pathway.Given its low toxicity,TSPf could be developed for the treatment of AML. 展开更多
