With the TiB2−SiC powders after spray granulation and vacuum calcination as raw materials,the TiB2−SiC coating was prepared by supersonic atmospheric plasma spraying(SAPS).The effects of spraying power and spraying distance on the properties of the TiB2−SiC coating were investigated and the fabrication processing of SAPS was optimized.The results show that the sprayed powders after calcination have a uniform particle size distribution,good sphericity and enhanced fluidity.The coating prepared by the calcined powders has a dense structure and high deposition efficiency.When the calcined TiB2−SiC powders are used and the spraying power is 95 kW and the spraying distance is 150 mm during supersonic plasma spraying,the obtained TiB2−SiC coating behaves the best comprehensive performance with the porosity,microhardness,bonding strength and resistivity equal to 5.6%,3.57 GPa,18.3 MPa and 10.8 mΩ·cm,respectively. 展开更多

Downregulation of NFAT5 by RNA interference reduces monoclonal antibody productivity of hybridoma cells 被引量:4

However, the underlying mechanism is not well understood. In the present study, we hypothesize that the nuclear factor of activated T cells 5 (NFAT5)/tonicity enhancer binding protein (TonEBP) functions to increase the antibody productivity of hybridoma cells. NFAT5 is an osmosensitive mammalian transcription factor. However, its ubiquitous expression in various organs that are not bathed in hypertonic milieu suggests that NFAT5 may also regulate cell growth and function under isotonic conditions. In this study, we examined the expression of NFAT5 in hybridoma cells by Western blot analysis, and found that it increased significantly in hypertonic medium. To further define the function of NFAT5 in hybridoma cells, RNA interference technique was used to downregulate the expression of NFAT5 in SGB-8 cells (a hybridoma cell line). In isotonic medium, antibody productivity ofhybridoma cells was reduced by downregulation of NFAT5 while cell proliferation was not influenced. The results presented here demonstrate that NFAT5 not only plays an important role in osmotic stress response pathway in hybridoma cells but also is essential for optimal antibody productivity. 展开更多

半刚性钢框架-波纹钢板剪力墙结构抗震性能研究

为研究半刚性框架内填波纹钢板剪力墙对结构抗震性能影响,利用ABAQUS建立数值模型进行探索分析。通过设计3种不同构造的波纹钢板剪力墙,并与半刚性框架相结合,研究此结构体系抗震性能。首先建立单层单跨有限元模型,与现有文献中相关研... 为研究半刚性框架内填波纹钢板剪力墙对结构抗震性能影响,利用ABAQUS建立数值模型进行探索分析。通过设计3种不同构造的波纹钢板剪力墙,并与半刚性框架相结合,研究此结构体系抗震性能。首先建立单层单跨有限元模型,与现有文献中相关研究模型进行对比验证,确保模型精确性。然后在此基础上建立3种不同构造的波纹钢板剪力墙体,分别与此半刚性框架相结合形成半刚性框架-波纹钢板剪力墙结构体系,研究体系在侧向力作用下抗震性能,最后探究半刚性框架内嵌不同墙体的结构体系剪力分配规律。有限元研究结果表明:半刚性框架(KKJ)具有较好的延性,但其抗侧刚度较小,承载能力不高,能量耗散较稳定但耗能不强;半刚性框架内填墙体(KKJ-Q1)能大幅增加体系刚度、承载力及耗能能力,使其延性降低;墙体填充泡沫混凝土(KKJ-Q2)可抑制波纹钢板屈曲,提高墙体整体性能,其刚度、承载力、耗能能力进一步增加;墙体两端设置加强端(KKJ-Q3)能避免墙体过早发生破坏而失去承载力,刚度、承载力都有一定程度的提升,随着加强端高度增加,导致墙体塑性变形从两端向中心转移,墙体破坏模式从弯曲变为弯剪,刚度、承载力及耗能能力进一步提升;随着外荷载增加,墙体剪力呈现先上升后下降趋势,墙体剪力占比Q3>Q2>Q1,墙体刚度与剪力占比呈现正比关系。 展开更多

Magnetic tunnel junctions with ferroelectric barriers, often referred to as multiferroic tunnel junc- tions, have been proposed recently to display new functionalities and new device concepts. One of the notable predictions is that the combination of two charge polarizing states and the parallel and antiparallel magnetic states could make it a four resistance state device. We have recently studied the ferroelectric tunneling using a scanning probe technique and multiferroic tunnel junctions using ferromagnetic Lao.7Cao.3MnO3 and Lao.TSro.3MnO3 as the electrodes and ferroelectric (Ba, Sr)TiO3 as the barrier in trilayer planner junctions. We show that very thin (Ba, Sr)TiO3 films can sustain ferroelectricity up till room temperature. The multiferroic tunnel junctions show four resistance states as predicted and can operate at room temperatures. 展开更多
