Objective:To predict couplet medicinals Cuscuta-Medlar(CM)in treat male infertility and analyze their potential mechanisms by the network pharmacology method.Method:TCMSP,BATMAN and literature were used to obtain comp...Objective:To predict couplet medicinals Cuscuta-Medlar(CM)in treat male infertility and analyze their potential mechanisms by the network pharmacology method.Method:TCMSP,BATMAN and literature were used to obtain components of CM,and male infertility targets were predicted by OMIM and GeneCards databases.Cytoscape software was used to construct the"component-target-disease"network.The STRING web site was used for the analysis of Protein-to-protein interaction(PPI)data between compounds and disease.DAVID 6.8 was carried on GO analysis and KEGG pathway analysis.Result:A total of 52 active ingredients and 242 potential targets of CM were obtained,with 7661 male infertility related genes retrieved from disease database.Then 113 common targets were obtained.Biological processes were related to enzyme binding,signaling receptor binding and molecular function regulator.The biological pathways mainly included TNF signaling pathway,apoptosis-related signaling pathway,PI3K/AKT signaling pathway,FoxO signaling pathway,Thyroid hormone signaling pathway and toll-like receptor signaling pathway.Conclusion:CM may treat male infertility by inhibiting inflammation,inhibiting apoptosis and promoting spermatogenesis.展开更多
Objective:To evaluate the effect of ischemic stroke(is)combined with hyperlipidemia(HLP)on erectile function in rats and its mechanism.Methods:Thirty male SD rats were randomly divided into normal group and HLP group....Objective:To evaluate the effect of ischemic stroke(is)combined with hyperlipidemia(HLP)on erectile function in rats and its mechanism.Methods:Thirty male SD rats were randomly divided into normal group and HLP group.The model of is combined with HLP was established by feeding high-fat diet.The stroke model group and sham operation group were established by operation method.The contents of testosterone(T),D-Dimer(D-D)and cyclic guanosine(cGMP)were detected by ELISA.Western blot and real-time PCR were used to detect the expression of VEGF protein and mRNA,and hematoxylin and eosin(He)staining of penile tissue were observed to evaluate its erectile function.Results:Compared with group C,the levels of cGMP and T in group M decreased,while VEGF and D-D increased.He staining showed that the tissue structure of corpus cavernosum in model group was obviously damaged.Conclusion:The model of ischemic stroke combined with hyperlipidemia has certain effect on erectile function of rats,and the mechanism may be related to the expression of cGMP,VEGF related protein and mRNA.展开更多
Objective:To study the therapeutic effect of Huoxue Tongluo Decoction(HXTLD)on erectile dysfunction caused by ischemic stroke and identify the mechanisms involved.Methods:Network pharmacology was used to predict the k...Objective:To study the therapeutic effect of Huoxue Tongluo Decoction(HXTLD)on erectile dysfunction caused by ischemic stroke and identify the mechanisms involved.Methods:Network pharmacology was used to predict the key active ingredients and targets of HXTLD.Surgical methods were used to create a rat model of ischemic stroke.The rats were then given a suspension of HXTLD by ig administration.Erectile function was evaluated by Apomorphine(APO)induction.Real-time fluorescence quantitative reverse transcription-polymerase chain reaction(Real-time PCR)and Western blotting were used to detect the expression of related m RNAs and proteins in rat penile corpus cavernous tissue and brain tissue.Hematoxylin&Eosin(HE)staining was used to investigate structural changes in the penile cavernous tissue.Results:Network pharmacology showed that tumor necrosis factor(TNF),nitric oxide synthase 3(eNOS),and vascular endothelial growth factor(VEGF)were the key targets of HXTLD in the treatment of erectile dysfunction caused by ischemic stroke.Experimental studies showed that HXTLD improved erectile dysfunction caused by ischemic stroke.HE results showed that HXTLD improved the structure of the corpus cavernosa.HXTLD also inhibited the expression of TNF and VEGF proteins in penile tissue(P<0.05)and enhanced the expression of e NOS protein in penile tissue(P<0.05).Conclusion:HXTLD improved the erectile function of rats with erectile dysfunction caused by ischemic stroke by regulating the m RNA and protein levels of TNF,e NOS and VEGF.展开更多
